Hormonal Study to Induce Direct Organ Differentiation of Kalanchoe pinnata by Tissue Culture (조직배양으로 Kalanchoe pinnata의 직접기관분화를 유도하기 위한 호르몬 연구)

  • Plant cells have a totipotencial capacity, the ability of each cell to produce a new complete individual through development. By applying this, several technologies are being developed for widespread application of somatic embryogenesis by processing hormones in vitro as a method of propagation of plants. In order to use this technology, in Kalanchoe pinnata, a plant capable of asexual reproduction with more regular cell division, kinetin belonging to cytokinin and picloram among hormones belonging to auxin were added in combination and treated for 8 weeks, and then the typical performance was evaluated. As a result of our experiment, the rooting effect in leaf slices showed a 70% incidence rate at a picloram concentration of 0.1 mg/L. It has been proven that a concentration difference of 1:5-1:10 in the ratio of kinetin and picloram is effective. It is the experimental result that the effect of auxin is essential for the development of Kalanchoe roots. As for the effect of shooting, the incidence rate was 60% at the picloram concentration of 0.5 mg/L. The kinetin concentration from 0.5 and 1.0 mg/L and has a significant effect on development. It has been proven that the ratio of kinetin to picloram is effective with a concentration difference of 1:1-1:2. These results show that the combination of cytokinin and auxin is crucially important for shooting. It is thought that it can be the basis of a technology for inducing mass proliferation in vitro by inducing direct organogenesis with a combination of hormones.

Establishment of Efficient Callus Induction System with Picloram Hormone in Lily Plants (Picloram을 이용한 효율적인 백합 캘러스 유도 체계 확립)

  • Lily is one of the most important 5 cut flowers in international flower market and lilies are distributed in Asia, Eurasia and North America. To develop a new lily cultivar, in addition to hybridization, mutation and selection methods, biotechnological techniques including tissue culture are also required. Establishment of tissue culture system is one of the requirement for the breeding program in Lily. Among many fields of plant tissue culture, establishment of regeneration system via embryogenic calluses are studied in many crops. In this study, research was carried out to decide the proper concentration of picloram which is used for the induction of embryogenic calluses. As a result, 3 different types of callused were observed after 3-4 weeks. They were CEC (compact embryogenic callus), FEC (friable embryogenic callus) and white callus type. 1.0 mg /l of picloram showed the best result for the production of embryogenic callus, however, due to its higher rate of browning in this concentration, 0.75 mg/l of picloram was selected as a proper concentration of picloram for the induction of CEC and FEC in Lily. These results can be contributed to the establishment of both regeneration system and mass propagation in lily in the future.

Effect of Plant Growth Regulators on Calls Initiation and Organogenesis from Tissue Culture of Arabidopsis thaliana Stem (애기장대 줄기 조직배양에 있어서 식물생장조절제가 캘러스 형성과 기관분화에 미치는 영향)

  • This experiment was carried out to investigate the effects of plant growth regulators on the organogenesis from the tissue culture of Arabidopsis thaliana stem, and the origin of the callus development. When the stem segments were cultured on medium with 2mg/L IAA or NAA, adventitious roots and trichomes were differentiated after 11 days of culture. Callus vigorously formed on medium with 2/L2,4 after 7 days of culture, but adventitious roots and trichomes were not differentiated from callus after 10 days of culture. This results suggesting that picloram is very effective auxin for the callus formation and organogenesis. Callus weakly formed on 0.05mg/L kinetin, and formed on combination of auxins(2mg/L) with 0.05mg/L kinetin. But the effect of combination of auxins and kinetin the callus formation was less than 2,4-D or picloram alone. A histological examination of callus formed on picloram showed that phloram showed that phloem parenchyma cells were divided and enlarged after 2 days of culture. Cortex parenchyma cells were divided and meristematic nodules were developed from these cells after 4 days of culture. Finally, callus formed on outside of cortex and epidermis by division of meristematic nodules after 7 days of culture.

Factors Affecting plant Regeneration in Unpollinated Ovary Culture of Rice (벼의 미수분 자방배양에 영향을 미치는 요인)

  • The optimal conditions for callus formation and plant regeneration were determined by the manipulation of culture method in unpollinated ovary culture of rice. The effect of cold pretreatment on callus formation and plant regeneration varied with duration of pretreatment. The maximum frequency (38.7%) of plant regeneration was obtained from the unpollinated ovary pretreated for 10 days at $12^{\circ}C$. The ability of callus formation and plant regeneration was higher on the medium with picloram (1 mg/L) than that of 2, 4-D (1 mg/L). However, the high concentration of picloram increased markedly the frequency of albino plant from unpollinated ovary-derived callus. Floral parts inoculated as a unit play important roles in callus formation and plant regeneration. Best result was obtained when ovary with partial cut glume, pedicel and secondary branch as a unit was cultured.

Effect of picloram and 2,4-D on plant regeneration from mature and immature embryos of moroccan durum wheat varieties

  • An efficient genetic transformation protocol is a fundamental requirement for high regeneration capacity from cultivated durum wheat (Triticum durum) varieties. In this study, wereportedtheeffectsoftwoauxins,2,4-dichlorophenoxyaceticacid(2,4-D)and4-amino-3,5,6-trichloropicoli nicacid(picloram), at a concentration of 2 mg/Laloneandincombination on the embryogenic callus and plantlet regeneration of four durum wheat varieties (Amria, Chaoui, Marouane, and Tomouh) using mature embryos (MEs) and immature embryos (ImEs). Significanteffectsofvariety,culturemedium(theauxinused),andvariety-mediuminteraction were observed on the callus weight and plantlet regeneration of both MR and ImE explants. The medium used for callus induction significantly affected plantlet regeneration (p < 0.001). Comparedto2,4-D, picloram led to a higher plantlet regeneration rate in both ME and ImE explants (19.8% and 40.86%, respectively). Plantlet regeneration also varied significantly depending on the variety and medium used. PicloramledtohighplantletregenerationofbothME and ImE explants in all varieties except Tomouh, which showed high plantlet regeneration of ME explants in 2,4-D. A comparison of ME and ImE responses indicated that ImEs are the best explants for high plantlet regeneration in durum wheat. Ourfindingssuggestthatpicloramisthebestauxin and should be used instead of 2,4-D due to its positive effect on increasing plant regeneration of durum wheat ME and ImE explants.

Improvement of Regeneration Efficiency from Mature Embryo and Leaf Base Segment in Korean Oat Genotypes

  • Mature embryo and leaf base segment of Korean oat were used as materials in an experiment to check plant regeneration efficiency. MS media supplemented with 2,4-dichlorophenoxyacetic acid (2,4-D), kinetin, and picloram were used for callus induction from mature embryos and leaf base segments. Three mg/l of 2,4­D and 3 mg/l of picloram in callus induction medium showed high frequency for plant regeneration from mature embryos. Leaf base segments were transferred to callus induction medium and incubated at $25^{\circ}C$ in 16/8 hr light/dark cycle for 3 weeks. Callus induction from leaf base segments of Malgwiri showed high efficiency in medium containing 3 mg/l of 2,4-D and 1 mg/l of kinetin $(91.8\%)$. In case of Samhangwiri, the combinations of phytohormones did not show significant difference. Regeneration from leaf base segments showed high frequency in shoot medium containing 1 mg/l of antiauxin, tri-iodobenzoic acid (TIBA) and 1 mg/l of 6-benzyladenine (BA). Calli induced from leaf base segments of Samhangwiri and Malgwiri in media containing 3 mg/l of 2,4-D and 3 mg/l of picloram showed high regeneration frequency. It appears that the callus initiation medium may be an important factor for subsequent plant regeneration.

Callus Induction and Plant Regeneration from Leaf Tissue. Culture of $\emph{Aralia elata}$ S. (두릅의 엽조직배양에 의한 Callus유기 및 식물체 재분화)

  • This study was conducted to determine the optimum culture conditions for inducing callus and regenerating plantlets from cultured leaf tissues of Aralia elata. Young leaf tissues(1cm) of A. elata plant were cultured on MS medium supplemented with 2, 4-D and Thidiazuron. Embryogenic callus was induced along the leaf veins, more efficiently on the medium containing 1.0mg /1 Thidiazuron in 4 weeks after culture initiation. Calli were subcultured to proliferate on MS media containing 2, 4-D, Dicamba, Picloram, and Thidiazuron. Callus was better proliferated on the medium containing Dicamba than on the others.. However, callus subcultured on the medium containing Thidiazuron was more embryogenic and light green-colored, of which some showed embryoid-like structure on the surface. Hormone-free medium was more efficient to regenerate plantlets than media supplemented with Kinetin, BA, and Thidiazuron.

Effect of Some Factors Affecting Callus Formation of Red Clover (레드 클로버의 캘러스 유도에 미치는 몇가지 요인의 영향)

  • This experiment was conducted to determine factors affecting callus formation of some varieties of red clover. Growth regulators. basal medium, medium pH, agar concentration, and plant age for explant were investigated. The result obtained were as follows. As auxin source, 0.1 mg/ l picloram was found to be effective on callus formation. Callus formation was the highest on PC medium among several growth media. The callus produced on PC medium was more friable than those produced on the other media. The medium pH 5.8 gave the best response for callus formation. The effective seedling age for callus formatiom was around 5-7 days old. In agar concentration, 0.5-0.8%(WN) was suitable for callus formation.

  • This study was carried out to produce haploid plants to verify a systematic breeding program and genetic analysis. Effect of developmental stage of pollen grains and pre-treatment temperature on production of haploid plants was investigated. Microscopic investigation of the explants (Lilium Asiatic hybrid 'reamland' revealed that the length of flower bud at 23.0-24.9, 25.0-26.9, and 27.0-28.9 mm long coincided with tetrad, uninucleate, and binucleate, respectively. When the efficiency of the anther culture from microgametogenetic stages was tested, late uninucleate to early binucleate stage, having the length of 23.0 to 28.0 mm long flower bud, was the best. The frequencies of the callus induction and plant regeneration from the stage mentioned above were 17.8 and 6.7%, respectively. When calli were cultured on the MS medium containing picloram and zeatin at $25^{\circ}C$, shoots were obtained. Roots of regenerated plantlets were confirmed as haploid through an microscopic observation.
