Expression Analysis of ESTs Derived from the Four-Year Root of Chunpoong (Panax ginseng C.A. Meyer)

  • Expressed sequence tags (EST) are help to quickly identify functions of expressed genes and to understand the complexity of gene expression. To assist genetic study of the root development in Panax ginseng, which is one of the most important medicinal plant, expressed sequence tags (EST) analysis was carried out. We constructed a CDNA library using the 4-year Chunpoon root. Partial sequences were obtained from 3,841 clone. The ESTs could be clustered into 2,056 (64%) non-redundant groups. Similarity search of the non-redundant ESTs against public non-redundant databases of both protein and DNA indicated that 1,498 groups show similarity to genes of known function. These ESTs clones were divided into eighteen categories depending upon gene function. The most abundant transcripts were major latex protein (41), ribonuclease 2 (36), metallothionein 2(35). Our extensive EST analysis of genes expressed in 4-year Chunpoong root not only contributes to the understanding of the dynamics of genome expression patterns in root organ development but also adds data to the repertoire of all genomic genes.

  • In this paper, we presented a strengthening plan to prevent personal information leakage incidents by securing legal compliance for personal information impact assessment and suggesting measures to strengthen privacy during personal information impact assessment. Recently, as various services based on big data have been created, efforts are being made to protect personal information, focusing on the EU's GDPR and Korea's Personal Information Protection Act. In this society, companies entrust processing of personal information to provide customized services based on the latest technology, but at this time, the problem of personal information leakage through consignees is seriously occurring. Therefore, the use of personal information by trustees.

  • This paper presents a general purpose data acquisition system based on the microprocessor system with M 6800. instrument and lograrithmic amplifiers and A/D converters are used to implement a signal conditioner for a various kinds of signals. The proposed system has a function such that the processor may select the one of the input signals as will and also it is proved that the system may control the control signal and digital converted signal through I/O port of PIA. Practical measurement by the proposed system shows in good results.

Regulation of Insulin-Sensitive Cyclic Nucleotide Phosphodiesterase in Adipocytes of Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 당뇨병을 유발시킨 흰쥐의 지방세포에서 일어나는 Insulin-Sensitive Phosphodiesterase의 조절에 관한 연구)

  • Possible changes in the role of insulin-sensitive cyclic nucleotide phosphodiesterase(PDE) in mediating the antilipolytic action of insulin were investigated in adipocytes from streptozotocin-induced diabetic rats. Isolated adipocytes prepared from epididymal adipose tissue were incubated, with or without insulin, at $37^{\circ}C$ for 15 min following pretreatment with various drugs or toxins, and three (plasma membranes, microsomal membranes, and cytosol) fractions prepared by differential centrifugation were then assayed for cAMP phosphodiesterase activity. The PDE activities only in the crude microsomal (P2) fractions were activated by insulin both in diabetic and control rats. The basal PDE activities in P2 fractions of adipocytes from diabetic rats were higher than those from control rats, although the maximal effects observed at 2 nM of insulin, $100\;{\mu}M$ of isoproterenol or the combination of both were not significantly different from each other. The insulin-stimulated PDE activities in P2 fractions of adipocytes from diabetic rats were not changed by PIA, a $A_{1}$ adenosine receptor agonist, whereas they were decreased to the basal PDE activities in those from control rats. In addition, the adipocytes from diabetic rats showed an increased sensitivity to pertussis toxin compared to those from controls. There were no differences between diabetic and control rats in the sensitivity of adipocytes to cholera toxin. These data indicate that the impaired signalling through inhibitory receptors such as adenosine receptors in adipocytes from streptozotocin-induced diabetes relates to the loss or the decreased function of $G_i$ proteins, and leads to the increased activity of the insulin-dependent PDE at the basal states.

  • To investigate the origin of backyeoncho (Opuntia ficus-indica var. saboten), we isolated 685 bp clone using ITS primer pairs. The rDNA consists of the genes coding for the partial 54 bp 185, 162 bp 5.8S, and partial 56 bp 26S. The coding regions are interrupted by two internal transcribed spacers, 193 bp ITS1 and 220 bp ITS2. The ITS2 of backnyeoncho in length was shorter than that previously registered in Cucurbitoideae plants. The GC contents was 66.8% in ITS1, and 67.7% in ITS2. The rDNA of backnyeoncho matched to the previously reported genes and showed a high similarity with the 95% identity with Pereskiopsis porteri (L708037). In the phylogenetic analysis, the backnyeoncho rDNA was clustered with Pereskiopsis porteri (L708037).

  • A large scale of riverside rearrangement has been recently done in the major rivers in Korea. So inundation possibility in agricultural area closed by these rivers has been higher than the possibility a few years ago. However, land use in this area has not been adjusted to a change of this situation near the rivers. Therefore, when typhoon or heavy rain is happened on this area, it can cause a large damage in agricultural area. This study analyzed inundation potentiality in agricultural area at Ibang-myeon, Changnyeong-gun, Kyeongnam-province, Korea by using the logistic regression model and the piecewise regression model. The first thing we did was to transfer the inundation area per elevation to the accumulated inundation area per elevation. This accumulated inundation area per elevation as an distribution function could be described by the logistic regression model(LRM), and piecewise regression model(PRM) could make it much more accurate to analyze the inundation area per elevation. As a result, the regression models derived from LRM and PRM showed $R^2$ over 0.950. The models derived from LRM and PRM in Ibang-myeon noted that frequently inundated area(FIA) was shown up to 12.12m in elevation, and potentially inundated area(PIA) was shown up to 14.60m in elevation. In FIA, regular agricultural activity would be impossible. And It would be not easy to continue the regular agricultural activity in PIA. So, this land should be rearranged to be used for a buffer zone for ecosystem protection, landscape conservation and things like that in riverside.

  • Hela cells, a transformed human epithelial cell line, attach to various substrata but subsequent spreading is specific to collagen or gelatin. The spreading is initiated by the activation of phospholipase $A_2$ (PLA$_2$) which produces arachidonic acid (AA) as a consequence of cell surface collagen receptor clustering. This study examines the mechanism of PLA$_2$activation and which phospholipids are hydrolyzed by PIA$_2$ to release AA in response to Hela cell adhesion to a gelatin substratum. The levels of phosphatidyicholine decreases, among various phospholipids, during attachment and spreading of Hela cells. Lysophosphatidyicholine Is the only lysophospholipids formed during ileLa cell adhesion indicating that clustered collagen receptors activate PLA$_2$to hydrolyze posphatidylcholine to AA and lysophosphatidylcholine. Among various molecular entitles which are known to regulate PLA$_2$ activation, we have previously shown that PLA2 activation is not mediated by either changes in $Ca_2$+ levels, alkalinization of cytoplasmic p11, or activation of protein hinase C. It is also likely that PIA2 activation is not mediated by either pertussis or cholera toxinsensitive G proteins as those toxins do not affect both AA release and cell spreading.

Effects of Antiiflammatory Agents on Acetaldehyde Induced Cytotoxicity (Acetaldehyde 유도 세포독성에 대한 항염증제의 영향)

  • In order to get infonnations on the development of alcohol induced cardiovascular disorders, primary cultured vascular smooth muscle cells (PVSMC) were treated with acetaldehyde, one of the most reactive metabolites of ethanol. Acetaldehyde caused the striking release of lactate dehydrogenase (LDH) from PVSMC and it stimulated the prostaglandin synthesis in the same system. But it didn't induce cyclooxygenase activity. lipoxygenase inhibitors-propyl gallate and nordihydroguaiaretic acid could reverse the effect of acetaldehyde, but dexamethasone, a phospholipase $A_2\;(PIA_2)$ inhibitor and cyclooxygenase inhibitors except indomethacin could not protect the cells from acetaldehyde toxicity. These results indicate that enhanced prostaglandin synthesis by acetaldehyde is not a direct cause of cell death, but secondary effect due to the activation of PIAl and also, the roles of the lipoxygenase metabolites and/or $PIA_2$ activity itself might be more important in the cytotoxicity of acetaldehyde.

  • A full-length cDNA encoding ribulose-1,5-bisphosphate carboxylase small subunit (rbcS) has been isolated and its nucleotide sequence determined from root in ginseng plant (Panax ginseng). The rbcS cDNA of ginseng is 790 nucleotides long and has an open reading frame of 549 bp with deduced amino acid of 183 residues (pI 8.37), 20.5 kDa. The deduced amino acid sequence of rbcS matched to the previously reported rbcS protein genes and showed a high similarity with the $78\%$ identity with rbcS of Helianthus annuus (CAA68490). In the phylogenetic analysis based on the amino acid residues, the ginseng rbcS was clustered with H. annuus (CAA68490), C. morifolium (AA025119) and L. sativa (Q40250).
