Liquid Crystal Alignment and Generation of Pretilt Angle by Using Photo-alignment Techniques on Different Polymer Molecules (광배향기술을 이용한 액정배향의 기구 및 폴리이미드의 분자구조가 프리틸트각에 미치는 영향)

  • In this paper, we investigated the liquid crystal(LC) alignment and generation of pretilt angle by using photo-alignment techniques on two kinds of polyimide(PI) surface, It was found that the uniform alignment for nematic(N) LC is obtained in a cell with slanted UV light irradiation on PI surface without side chain. We successfully observed that the pretilt angle of NLC is generated about $3^{\circ}$ with an incident angle of 70 degree on the PI surface without side chain. It is considered that the pretilt angle generation in NLC is attributed to interaction between the LC molecules and the polymer surfaces.

  • We investigated the electro-optical(EO) performances of the super twisted nematic liquid crystal display(STN-LCD) on the polyimide(PI) surface with polymer film using rubbing and photoalignment method. Monodomain alignment of the plastic STN-LCD can be observed. A faster response time for the photoaligned plastic STN-LCD on the polyimide(PI) surfaces can be achieved than rubbingaligned plastic STN-LCD.

Pathological findings of experimentally infected piglets with a porcine epidemic diarrhea virus isolated in Korea (국내에서 분리된 돼지 유행성 설사증 바이러스 접종자돈의 병리학적 소견)

  • Twelve colostrum-deprived piglets were inoculated oral1y with porcine epidemic diarrhea virus isolated from Korea. The piglets were euthanatized from at 12 hour postinoculation(PI) at 6-hour intervals. At 24 to 36 hours PI, all infected piglets showed severe yellowish to watery diarrhea, dehydration, depression, and anorexia. At necropsy, the intestinal wall looked thin, mesenteric vessels congested, and mesenteric lymph nodes edematous. The histological findings showed marked villous atrophy and fusion, severe degeneration of enterocytes and monomuclear cell infiltration in the lamina propria. On electron microscopy, villous epithelial cells of infected jejunum contained viral particles at 18 hour PI. The viral particles were pleomorphic spheres with a mean diameter of 95∼180nm including 18nm projections.

  • The mammalian glycosylphosphatidylinositol (GPI) anchor consists of three mannoses attached to acylated GlcN-(acyl)PI to form $Man_3$-GlcN-(acyl)PI. The first of the three mannose groups is attached to an intermediate to generate Man-GlcN-(acyl)PI by the first mannosyltransferase (GPI-MT-I). Mammalian and protozoan GPI-MT-I have different substrate specificities. PIG-M encodes the mammalial GPI-MT-I which has 423 amino acids and multiple transmembrane domains. In this work we cloned PIG-M homologues from humans, Plasmodium falciparum (PfPIG-M), and Saccharomyces cerevisiae (GPI14), to test whether they could complement GPI-MT-I-deficient mammalian cells, since this biosynthetic step is likely to be a good target for selective screening of inhibitors against many pathogenic organisms. PfPIG-M partially restored cell surface expression of the GPI-anchored protein CD59 in PIG-M deficient mammalian cells, and first mannose transfer activity in vitro; however, this was not the case for GPI14.

Pretilt Angle Generation of Photo-aligned TN-LCD Using Oblique Non-polarized UV Light Irradiation on Polymer Surface (경사진 자외선조사를 이용한 네마틱액정의 프리틸각의 발생 및 광배향 TN-LCD의 전기광학특성)

  • In this paper, we have investigated the generation of pretilt angle for nematic liquid crystal (NLC) in a cell with oblique non-polarized ultraviolet(UV) light irradiation polyimide (PI) surface. It was found that the monodomain alignment of the NLC is obtained with an angle of incidence of 70∼85。 on PI surface. It is considered that the monodomain alignment of NLC is attributed to anisotropic of NLC is about 3。 with and angle of incidence of 70∼75。. Finally, we observed that the voltage-transmittance, response time, and voltage-holding-ratio(VHR) characteristics of photo-aligned twisted nematic(TN)-LCD with and angle of incidence of 85。 on PI surface were almost same compared to rubbing -treated TN-LCD.

  • In this paper, we obtained the monodomain alignment of nematic liquid Crystal(NLC) in the cell fabricated by transcription alignment method on polyimide(PI) surface with side chain. It is considered that the LC alignment produced by the transcription alignment method is attributed to a memory effect of the NLC on PI surfaces. Also we observed that the generated pretilt angle of NLC is about $3.7^{\circ}$ with transcription alignment on PI surface. Next, we measured that the voltage-transmittance characteristics of transcription-aligned TN-LCD are almost same compared to rubbing-aligned TN-LCD. Also, we measured that the curve of transcription-aligned TN-LCD is less sharp than that of the rubbing-aligend TN-LCD in the decay time characteristics. It is considered that the response time characteristics of transcription-aligned TN-LCD are attributed to the weak anchoring strength between the LC molecules and the polymer surface.

  • Peptidoglycan (PG), the gram positive bacterial pathogen-associated molecular patterns (PAMP), is detected in a high proportion in macrophage-rich atheromatous regions, and expression of chemokine CXCL8, which triggers monocyte arrest on early atherosclerotic endothelium, is elevated in monocytes/ macrophages in human atherosclerotic lesion. The aim of this study was to investigate whether PG induced CXCL8 expression in the cell type and to determine cellular signaling pathways involved in that process. Exposure of THP-1 cell, human monocyte/macrophage cell line, to PG not only enhanced CXCL8 release but also profoundly induced il8 gene transcription. PG-induced release of CXCL8 and induction of il8 gene transcription were blocked by OxPAPC, an inhibitor of TLR-2/4 and TLR4, but not by polymyxin B, an inhibitor of LPS. PG-mediated CXCL8 release was significantly attenuated by inhibitors of PI3K-Akt-mTOR pathways. PKC inhibitors, MAPK inhibitors, and ROS quenchers also significantly attenuated expression of CXCL8. The present study proposes that PG contributes to inflammatory reaction and progression of atherosclerosis by inducing CXCL8 expression in monocytes/macrophages, and that TLR-2, PI3K-Akt-mTOR, PKC, ROS, and MAPK are actively involved in the process.

STAT6 Expression and IL-13 Production in Association with Goblet Cell Hyperplasia and Worm Expulsion of Gymnophalloides seoi from C57BL/6 Mice

  • In intestinal helminth infections, Th2 immune respones are generally associated with mucin secretion for worm expulsion from the host intestine. In particular, IL-4 and IL-13 are the important cytokines related with intestinal mucus production via STAT6 signalling in nematode infections. However, this perspective has never been studied in Gymnophalloides seoi infection. The present study aimed to observe the STAT6 signalling and cytokine responses in C57BL/6 mice, a mouse strain resistant to infection with this trematode. The results showed that worm expulsion occurred actively during days 1-2 post-infection (PI), when goblet cells began to proliferate in the small intestine. The STAT6 gene expression in the mouse spleen became remarkable from day 2 PI. Moreover, G. seoi infection induced a significant increase of IL-13 from day 4 PI in the spleen of infected mice. Our results suggested that goblet cell hyperplasia and worm expulsion in G. seoi-infected mice should be induced by STAT6 signalling, in which IL-13 may be involved as a dominant triggering cytokine.

  • Protein kinase C an enzyme of signal transduction has been known to regulate cell proliferation activation as well as apoptosis in some cancer cell lines. To explore the role of PKC in the course of cell mediated autoimmune disease such as experimental autoimmune encephalomyelitis (EAE) EAE was induced in Lewis rats(6-8 weeks old) with immunization of myelin basic protein supplemented with complete Freund's adjuvants and affected spinal cords were sampled at days 13 postimmunization(PI) as peak stage of EAE and at days 21 PI as recovery stage. The spinal cords with EAE were subjected to Northern blot analysis and insitu hybridization of PKC delta which is one of prominant isotypes of PKC in the haematopoietic cells. Northern blot analysis showed that levels of PKS delta mRNA in the spinal cords of rats withEAE was significantly increased at days 13 PI in which inflammatory cells including T cells and macrophages in the EAE lesions appeared. however the stage. By in situ hybridization signals of PKC delta in EAE lesions was intensely expressed on the delta is also expressed on some brain cells in normal rat central nervous system This finding suggests that PKC plays an important role on either activation of inflammatory cells including encephalitogenic T cells and macrophages or apoptotic elimination of some inflammatory cells depending on the stge of EAE.

A Study of Neuroproctective Effect of Bupleuri Radix on Hippocampal Neurons (시호(柴胡)의 뇌해마 신경세포 보호효능에 대한 연구)

  • Objective : This study was performed to investigate neuroprotective effects of Bupleuri Radix against oxidative and ischemic damages. Method : To observe the neuroprotective effects against ischemic damage, ischemic insult was induced by oxygen/glucose deprivation (OGD) on organotypic hippocampal slice cultures (OHSC) from 1 week-old Sprague-Dawley rats. Propidium iodide (PI) fluorescence-stained neuronal dead-cell areas, area percentages and TUNEL-positive apoptotic cells in CA1 and dentate gyrus, and LDH levels in culture media of the OHSC were measured following Bupleuri Radix extract treatment. Result : The following results were obtained: (1) The $5\;{\mu}g/ml$ of Bupleuri Radix treatment demonstrated a significant decrease in PI fluorescence-stained neuronal dead-cell areas and area percentage in CA1 region of the OHSC from 18 hrs to 48 hrs following the OGD. The $50\;{\mu}g/ml$ of Bupleuri Radix treatment was also significant from 6 hrs to 48 hrs following the OGD and was more effective. (2) The 5 and $50\;{\mu}g/ml$ of Bupleuri Radix treatment demonstrated a significant decrease in PI fluorescence-stained neuronal dead-cell areas and area percentage in DG region of the OHSC from 6 hrs to 48 hrs following the OGD. The $50\;{\mu}g/ml$ treatment was more effective than the $5\;{\mu}g/ml$ treatment. (3) Bupleuri Radix treatment demonstrated a significant decrease in TUNEL-positive apoptotic cells in CA1 region (with 5 and $50\;{\mu}g/ml$) and in DG region (with $50\;{\mu}g/ml$) of the OHSC damaged by the OGD. (4) Bupleuri Radix treatment demonstrated a significant decrease in LDH concentrations in culture media of the OHSC damaged by the OGD. Conclusion : These results suggest that Bupleuri Radix has neuroprotective and control effects on inflammatory and immune responses where there has been ischemic damage to the central nervous system.
