Anti-obesity effects of Glycyrrhiza uralensis ethanol extract on the inhibition of 3T3-L1 adipocyte differentiation in high-fat diet-induced C57BL/6J mice (감초 주정추출물의 3T3-L1 지방세포 분화 억제 및 고지방 식이로 유도된 C57BL/6J 마우스에 대한 항비만 효과)

  • The anti-adipogenic activity of Glycyrrhiza uralensis was investigated by examining the effects of its ethanol extract (GUE) on a mouse model with a high-fat diet (HFD) and 3T3-L1 preadipocytes during adipocyte differentiation. GUE administration for eight weeks significantly reduced weight gain in mice fed an HFD. GUE effectively inhibited 3T3-L1 preadipocyte differentiation and lipid droplet accumulation. This inhibitory effect is associated with the downregulation of key adipogenic regulators, including PPARγ and C/EBPα, and the modulation of adipose metabolism regulators, such as Fasn and Fabp4. LC-Q-TOF-MS analysis identified twelve phenolic and flavonoid compounds, including liquiritigenin and licorice saponin, in the GUE. These findings demonstrate that the anti-obesity effect of the GUE is attributed to the biological activity of its phenolic and flavonoid compounds. Therefore, the GUE has potential anti-obesity activity. Moreover, further studies on the isolation of bioactive components from the GUE and the investigation of the underlying molecular mechanisms of the GUE are required to establish its efficacy in metabolic disorders, including obesity.

Antioxidant activity and comparative analysis of major functional compounds in liqueur using coffee and coffee-ground (커피와 커피박 침출주의 항산화 활성 및 주요 생리활성 물질의 비교 분석)

  • Sensory evaluation, in vitro antioxidant activities and main compounds of coffee water-extract, coffee liqueur (CL) and coffee-ground liqueur (CGL) were investigated to consider their industrialization. Sensory evaluation showed that all groups of CGL without 25% CGL (3 month) were relatively higher than CL groups. Total phenolic compounds and in vitro antioxidant activities such as 1,1-diphenyl-2picryl-hydrazyl (DPPH) radical scavenging activity and ferric reducing/antioxidant power (FRAP) were also performed. The group of 35% CGL had higher total phenolic compounds than others, and the result of DPPH radical scavenging activity was similar to that of total phenolic compounds. In addition, 35% CGL is comparable to the FRAP of coffee water extract (CE). Qualitative and quantitative analysis using high-performance liquid chromatography (HPLC) were performed, and chlorogenic acid as a ployphenolic compound and caffeine as a nonpolyphenolic compound were detected in all samples. Moreover, the HPLC analysis showed that CGLs contain a larger amounts of chlorogenic acid (difference of 0.3~10.5%) and also greater amounts of caffeine (difference of 10.0~18.2%) more then CE. Consequently, these results suggest that coffee-ground as coffee by-products could be used as commercially available food substances because of its physiological molecules remained.

Variation in bioactive principles and bioactive compounds of Rosa rugosa fruit during ripening (해당화 열매 성숙단계에 따른 생리활성 및 기능성 물질 변화 분석)

  • Fruit ripening is a genetically programmed process involving a number of biochemical and physiological processes assisted by variations in gene expression and enzyme activities. This process generally affects the phytochemical profile and the bioactive principles in fruits and vegetables. To appraise the variation in bioactive principles of fruits from Rosa rugosa during its ripening process, we analyzed the changes in antioxidant and anti-elastase activities and polyphenolic compounds during the four ripening stages of fruits. Overall, an extract of unripe fruits contained the highest levels of total phenolic and flavonoid contents, radical scavenging activity, reducing power, oxygen radical antioxidant capacity, and elastase inhibitory activity, compared with the extracts of fruits at other stages of ripening. Additionally, we found that the reduction of flavonoid content occurs because of decreased transcriptional levels of genes involved in flavonoid biosynthesis pathway during the ripening process. Based on HPLC analysis, we found that the extract of unripe fruits contained the highest amount of myricetin, caffeic acid, chlorogenic acid, syringic acid, and p-coumaric acid and suggested that the antioxidant and anti-elastase activities of the extract obtained from stage 1, should be mediated by the presence of these compounds. Additionally, we analyzed the interaction sites and patterns between these compounds and elastase using the structure-based molecular docking approach, and suggested that chlorogenic acid strongly interacted with elastase. Together, these findings suggest that the maturity of fruits has profound effects on the pharmaceutical value of R. rugosa.

Comparison of Ingredients and Antioxidant Activity of the Domestic Regional Wolfiporia extensa (국내 지역별 매립 복령의 성분 및 항산화 활성 비교)

  • This study was conducted for comparison of ingredients, phytochemical compounds and antioxidant activity of Wofiporia extensa cultured in Gangwon-do, Gyeongsang-do, and Jeolla-do. Three contents of Wofiporia extensa were analyzed as oxygen (46~48%), carbon (38~39%), hydrogen (6.05~6.1%) and nitrogen (0.17~0.21%). The mineral contents of 50% ethanol Wofiporia extensa extracts were measured as sulfur (S) 145~149 ppm, Magnesium (Mg) 69~72 ppm, phosphorus (P) 122~154 ppm and calcium (Ca) 210.61~509.98 ppm. Wofiporia extensa from Gyeongsang-do (509.98 ppm) contained a significantly higher quantity of Ca than that from Gangwon-do (210.62 ppm) and Jeolla-do (223.88 ppm). In the gas chromatograph-mass spectrometry (GC-MS) analysis, oleic acid was identified in three 50% ethanol Wofiporia extensa extracts. In the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) assay for antioxidant activity, the $IC_{50}$ values of Wofiporia extensa cultured in Gangwon-do, Gyeongsang-do and Jeolla-do were calculated as 2.966 mg/mL, 23.03 mg/mL, and 4.16 mg/mL and 3.521 mg/mL, 12.17 mg/mL, and 7.40 mg/mL. In the ferric reducing antioxidant power (FRAP) assay, the $IC_{50}$ values of Wofiporia extensa cultured in Gangwon-do, Gyeongsang-do, Jeolla-do were 6.585 mg/mL, 19.06 mg/mL, and 18.97 mg/mL, respectively. In summary, Wofiporia extensa cultured in Gangwon-do had stronger antioxidant activity and higher concentration of oleic acid than that of Geyongsang-do and Jeolla-do. However, Wofiporia extensa cultured in Geyongsang-do contained a much higher concentration of Ca than that of Gangwon-do and Jeolla-do.

Increased Anti-oxidative Activity and Whitening Effects of a Saposhnikovia Extract Following Bioconversion Fermentation using Lactobacillus plantarum BHN-LAB 33 (Lactobacillus plantarum BHN-LAB 33의 생물전환공정을 통한 방풍 발효 추출물의 항산화 활성 및 미백 활성 증대 효과)

  • Saposhnikovia has been used as a traditional medicinal herb in Asia because of the reported anti-inflammatory, anti-allergic rhinitis, pro-whitening, anti-atopy, anti-allergy, and anti-dermatopathy effects of the phytochemical compounds it contains. In this study, we investigated the antioxidant effects of a Saposhnikovia extract after fermentation by Lactobacillus plantarum BHN-LAB 33. Saposhnikovia powder was inoculated with L. plantarum BHN-LAB 33 and fermented at $37^{\circ}C$ for 72 hr. After fermentation, the total polyphenol content of the Saposhnikovia extract increased by about 14%, and the total flavonoid content increased by about 9%. The superoxide dismutase-like activities, DPPH radical scavenging, ABTS radical scavenging, reducing power activity, and tyrosinase inhibition activity also increased after fermentation by approximately 70%, 80%, 45%, 39%, and 44%, respectively. The results confirmed that fermentation of a Saposhnikovia extract by L. plantarum BHN-LAB 33 is an effective way to increase the antioxidant effects of the extract. The bioconversion process investigated in this study may have the potential to produce phytochemical-enriched natural antioxidant agents with high added value from Saposhnikovia matrices. These results can also be applied to the development of improved foods and cosmetic materials.

Correlation between phytochemical contents in peel and flesh of cold-stored Fuji apple (저온 저장 후지 사과의 과피와 과육 중 파이토케미컬 함량의 상관관계)

  • This study determined the relationships between the total anthocyanin content in apple skin and the total polyphenols, flavonoids, proanthocyanidins, and soluble solids contents in the flesh of cold-stored Fuji apples. Total anthocyanin content in apple skin ranged from 0.130±0.005 mg CE/g fw to 0.262±0.028 mg CE/g fw, and the overall average was 0.200±0.008 mg CE/g fw. The total polyphenolic compounds in the flesh was ranged from a 4.283±0.141 mM GAE/g fw to 8.207±0.234 mM GAE/g fw, and the average content was 6.275±0.177 mM GAE/g fw. The total flavonoid content ranged from 4.510±0.080 mM QE/g fw to 2.467±0.458 mM QE/g fw, and the average total flavonoid content was about 3.586 mM QE/g fw. The total proanthocyanidin content was relatively high, ranging from 3.475±0.577 mM EE/g fw to 6.816±0.277 mM EE/g fw, and the soluble solid in the flesh was about 12 °Brix to 14 °Brix. The DPPH radical scavenging activity of extracts from apple flesh ranged from 66.36% to 94.99%, and the ascorbate equivalent concentration was 0.482 mM. The ABTS radical scavenging activity was 99.12% to 99.9%, indicating a higher inhibitory activity than the DPPH inhibitory activity, and the ascorbate equivalent concentration was 0.486 mM. The correlation between the total anthocyanin and total polyphenolic compounds was y = 15.192x + 3.2169 (R2=0.2748), but the concentration of total polyphenolic compounds increased when the total anthocyanin content was increased. The correlation equation of total anthocyanin with total flavonoids was y = 15.18x + 0.5555 (R2=0.6226), with total proanthocyanin was y = 14.918x + 2.3422 (R2=0.3372), and with soluble solid was y = 10.558x + 11.126 (R2=0.1925), indicating that the correlation of total anthocyanin with total flavonoid was higher than that with soluble solid.

Collagen Increasing and Hair Growth Effect of Supramolecular Cosmetic Materials Containing Phytochemicals (식물성 유용물질을 함유하는 초분자체 화장제재의 콜라겐 증가 및 육모효과)

  • Medicinal herbs have been shown to have protective functions for skin and hair. We investigated the effects of complex of soluble ${\beta}$-cyclodextrin and phytochemicals on the functions of skin and hair. In previous report, we evaluated the safety of supramolecules and found their anti-microbial effects and anti-fungal effect against Gram (+) and Malassezia furfur which is known to cause dandruff. Here we present that functional supramolecules-containing cream promotes the biological skin activity by inducing the collagen formation. And treatment of supramolecules-containing hair tonic increased the rate of hair growth of mouse. Taken together, supramolecular cosmetic compounds containing water insoluble phytochemicals and water soluble ${\beta}$-cyclodextrin exhibit the potential ability for hair growth promotion and delaying the aging of skin.

Anticancer Effects of Thymoquinone, Caffeic Acid Phenethyl Ester and Resveratrol on A549 Non-small Cell Lung Cancer Cells Exposed to Benzo(a)pyrene

  • Background: Phytochemical compounds are emerging as a new generation of anticancer agents with limited toxicity in cancer patients. The purpose of this study was to investigate the potential effcts of thymoquinone, caffeic acid phenylester (CAPE) and resveratrol on inflammatory markers, oxidative stress parameters, mRNA expression levels of proteins and survival of lung cancer cells in Vitro. Materials and Methods: The A549 cell line was treated with benzo(a)pyrene, benzo(a)pyrene plus caffeic acid phenylester (CAPE), benzo(a)pyrene plus resveratrol (RES), and benzo(a)pyrene plus thymoquinone (TQ). Inflammatory markers, oxidative stress parameters, mRNA expression levels of apoptotic and anti-apoptotic proteins and cell viability were assessed and results were compared among study groups. Results: TQ treatment up-regulated Bax and down-regulated Bcl2 proteins and increased the Bax/Bcl2 ratio. CAPE and TQ also up-regulated Bax expression. RES and TQ down-regulated the expression of Bcl-2. All three agents decreased the expression of cyclin D and increased the expression of p21. However, the most significant up-regulation of p21 expression was observed in TQ treated cells. CAPE, RES and TQ up-regulated TRAIL receptor 1 and 2 expression. RES and TQ down-regulated the expression of NF-kappa B and IKK1. Viability of CAPE, RES and TQ treated cells was found to be significantly decreased when compared with the control group (p=0.004). Conclusions: Our results revealed up-regulation of the key upstream signaling factors, which ultimately cause increase in their regulatory p53 levels affecting the induction of G2/M cell cycle arrest and apoptosis. Overall these results provide mechanistic insights for understanding the molecular basis and utility of the anti-tumor activity of TQ, RES and CAPE.

Anti-bacterial Effects of Aqueous Extract Purified from the Immature Cone of Red Pine (Pinus densiflora) (미성숙 솔방울 열수추출물의 항균성)

  • Red pine (Pinus densiflora) is widely used traditional medicine, pharmacological and nutritional values from which the phytochemical compounds are derived. The present study was aimed to examine the antibacterial effects in the absence and presence of a immature red pine cone extract against 13 microorganisms. The components in the aqueous extract from immature red pine cone were identified by GC-MS. About 1.4% of total polyphenolic compound was measured in aqueous extract collected from immature red pine cone. Also, the high concentration of ${\beta}$-phellenandrene, ${\alpha}$-pinene, limonene, bornyl acetate and aldehyde was detected in total ion chromatograms. Of total 13 microorganisms, 4 microorganisms including Pseudomonas aeruginosa, Vibrio cholera, Listeria monocytogenes, Klebsiella pneumonia were effectively killed by aqueous extract of immature red pine cone. The highest anti-bacterial effect was detected in P. aeruginosa, followed by V. cholera, L. monocytogenes and K. pneumonia. In case of P. aeruginosa, the largest diameter of inhibition zone was maintained to 1/2 solution treated cells and slightly decreased at 1/4 and 1/8 solution treated cells. Also, in test used V. cholera and L. monocytogenes, the inhibition zone was strongly formed in only 1 and 1/2 solution treated cells, while K. pneumonia showed the very small diameter of inhibition zone in all concentrations. Therefore, these results suggested that the aqueous extracts of immature red pine cone should be considered as a new and potentially important anti-bacterial substrate to effectively prevent the microbial infection and penetration.

Evaluation of Biological Activity in Pepper (Capsicum annuum L.) Breeding Lines (육성계통에 따른 고추의 생리활성 평가)

  • The purpose of this study was to determine the capsaicinoid and antioxidant compounds and to evaluate the biological activity of 40 different pepper (Capsicum annuum L.) breeding lines. Capsaicinoid and ascorbic acid content were measured with HPLC. Total polyphenolic and flavonoid contents and antioxidant activity were measured with spectrophotometric methods. The antiproliferative qualities of the samples against human breast tumor cells (MCF7) were assessed with a MTT assay. The nitric oxide content in the culture media was measured to evaluate the anti-inflammatory qualities of the samples using Raw264.7 macrophages. The capsaicinoid, ascorbic acid, polyphenolic and flavonoid content of the 40 pepper breeding lines were 0.1~204.2 mg/100 g (dry weight basis, DWB), 279.1~1695.5 mg/g (DWB), 2.6~10.2 mg/g (DWB), and 1.4~5.7 mg/g (DWB), respectively. The highest antioxidant, antiproliferative, and anti-inflammatory qualities were found in breeding line numbers 2500, 3201, and 3232, respectively. This study provides basic information useful to plant breeders and biotechnologists who are planning to breed pepper genotypes with high phytochemical content.
