Ionic Dependence and Modulatory Factors of the Background Current Activated by Isoprenaline in Rabbit Ventricular Cells

  • In order to elucidate the properties of the background current whole cell patch clamp studies were performed in rabbit ventricular cells. Ramp pulses of ${\pm}80\;mV$ from holding potential of 40 mV(or 20 mV) at the speed of 0.8 V/sec were given every 30 sec(or 10 sec) and current-voltage diagrams(I-V curve) were obtained. For the activation of the background current isoprenaline, adenosine 3',5'-cyclic monophosphate(dBcAMP), guanosine 3',5'-cyclic monophosphate(cGMP), and $N^6$-2'-o-dibutyryladenosine 3',5'-cyclic monophosphate(dBcAMP) were applied after all known current systems were blocked with 2mM Ba, 1 mM Cd ,5 mM Ni, 10 ${\mu}M$ diltiazem, 10 ${\mu}m$ ouabain, and 20 mM tetraethylammonium(TEA). The conductance of background current in control was $0.65{\pm}0.69$ nS at 0 mV, its I-V curves was almost linear and reversed near 50 mV. When there was no taurine in pipette solution, isoprenaline hardly activated the background current but when taurine existed in pipette solution, isoprenaline activated the larger background current. Cyclic AMP or cyclic GMP alone had little effect on the activation of the background current, while cGMP potentiated cGMP effect. When the background current was activated with cGMP and cAMP, isoprenaline could not further increased the background current. The background current activated by isoprenaline depended on extracellular $Cl^-$ concentration and its reversal potential was shifted according to chloride equilibrium potential. The change of extracellular $Na+$ concentration had little effect on reversal potential of the background current activated by isoprenaline.

${\alpha}$-Galactosidase from Bacillus megaterium VHM1 and Its Application in Removal of Flatulence-Causing Factors from Soymilk

  • A bacterial strain capable of producing extracellular ${\alpha}$-galactosidase was isolated from a sample of sugarcane industrial waste. Microbiological, physiological, and biochemical studies revealed that the isolate belonged to Bacillus sp. Furthermore, based on a 16S rDNA sequence analysis, the new isolate was identified as Bacillus megaterium VHM1. The production of ${\alpha}$-galactosidase was optimized based on various physical culture conditions. Guar gum and yeast extract acted as the best carbon and nitrogen sources, respectively. The optimum pH was 7.5 and the enzyme remained stable over a pH range of 5-9. The enzyme was optimally active at $55^{\circ}C$ and thermostable with a half-life of 120 min, yet lost 90% of its residual activity within 120 min at $60^{\circ}C$. One mM concentrations of $Ag^2$, $Cu^2$, and $Hg^{2+}$ strongly inhibited the ${\alpha}$-galactosidase, whereas the metal ions $Fe^2$, $Mn^{2+}$, and $Mg^{2+}$ had no effect on the ${\alpha}$-galactosidase activity, and $Zn^{2+}$, $Ni^{2+}$, and $Ca^{2+}$ reduced the enzyme activity slightly. When treated with the B. megaterium VHM1 enzyme, the flatulence-causing sugars in soymilk were completely hydrolyzed within 1.5 h.

  • Background: Many critically ill patients require transfer to a higher-level hospital for complex medical care. Despite the publication of the American Academy of Pediatrics guidelines for pediatric interhospital transportation services and the establishment of many pediatric transport programs, adverse events during pediatric transport still occur. Purpose: To determine the incidence of adverse events occurring during pediatric transport and explore their complications and risk factors. Methods: This prospective observational study explored the adverse events that occurred during the interhospital transport of all pediatric patients referred to the pediatric intensive care unit of Ramathibodi Hospital between March 2016 and June 2017. Results: There were 122 pediatric transports to the unit. Adverse events occurred in 25 cases (22%). Physiologic deterioration occurred in 15 patients (60%). Most issues (11 events) involved circulatory problems causing patient hypotension and poor tissue perfusion requiring fluid resuscitation or inotropic administration on arrival at the unit. Respiratory complications were the second most common cause (4 events). Equipment-related adverse events occurred in 5 patients (20%). The common causes were accidental extubation and endotracheal tube displacement. Five patients had both physiologic deterioration and equipment-related adverse events. Regarding transport personnel, the group without complications more often had a physician escort than the group with complications (92% vs. 76%; relative risk, 2.4; P=0.028). Conclusion: The incidence of adverse events occurring during the transport of critically ill pediatric patients was 22%. Most events involved physiological deterioration. Escort personnel maybe the key to preventing and appropriately monitoring complications occurring during transport.

Endothelial Ca2+ signaling-dependent vasodilation through transient receptor potential channels

  • Ca2+ signaling of endothelial cells plays a critical role in controlling blood flow and pressure in small arteries and arterioles. As the impairment of endothelial function is closely associated with cardiovascular diseases (e.g., atherosclerosis, stroke, and hypertension), endothelial Ca2+ signaling mechanisms have received substantial attention. Increases in endothelial intracellular Ca2+ concentrations promote the synthesis and release of endothelial-derived hyperpolarizing factors (EDHFs, e.g., nitric oxide, prostacyclin, or K+ efflux) or directly result in endothelial-dependent hyperpolarization (EDH). These physiological alterations modulate vascular contractility and cause marked vasodilation in resistance arteries. Transient receptor potential (TRP) channels are nonselective cation channels that are present in the endothelium, vascular smooth muscle cells, or perivascular/sensory nerves. TRP channels are activated by diverse stimuli and are considered key biological apparatuses for the Ca2+ influx-dependent regulation of vasomotor reactivity in resistance arteries. Ca2+-permeable TRP channels, which are primarily found at spatially restricted microdomains in endothelial cells (e.g., myoendothelial projections), have a large unitary or binary conductance and contribute to EDHFs or EDH-induced vasodilation in concert with the activation of intermediate/small conductance Ca2+-sensitive K+ channels. It is likely that endothelial TRP channel dysfunction is related to the dysregulation of endothelial Ca2+ signaling and in turn gives rise to vascular-related diseases such as hypertension. Thus, investigations on the role of Ca2+ dynamics via TRP channels in endothelial cells are required to further comprehend how vascular tone or perfusion pressure are regulated in normal and pathophysiological conditions.

  • Behavioral characteristics of Attention Deficit Hyperactivity Disorder(ADHD) is one of the most common mental disorders among children.child psyachiatry. Inattention, Hyperactivity that is done by hyperkinesis or minimal brain dysfunction is major sypmton in ADHD, But etiology and pathological facor of ADHD is very much or unkown.. We brought to about a Study on the Oriental - medical pathologic Understanding about Inattention, Hyperactivity symptom in ADHD within Don yui bo gam Book are as follows. 1. Oriental medical pathologic concepts about Inattention, Hyperactivity are continuous with process of Yang Qi(陽氣), an unbalance of qi(氣) and shen ming(神明), excess of seven emotions(七情), pathology of Huo(火). 2. Immanent factors in inattention, Hyperactivity are improper diet, overtiredness and seven emotions, are continuous with pathological process of the heart, liver, gall bladder, spleen, stomach, kidneys. 3. In oriental medicine, considered as a child's qi of shao yang, dynamic physiological feature, excess and want of yin and yang, organs and bowels, immanently imbalance in growth rather than a child's mental disorder 4. Inattention, looseness in ADHD-PI type are continuous with forgetfulness, improper overtiredness, shortage of qi, the interior heat syndrome due to yin deficiency within Don yui bo gam Book 5. Hyperactivity, impulsive actions in ADHD-C type are continuous with sudden palpitation, severe palpitation, delirium, fidgeting due to deficiency, fidgetiness, hyperactivity of huo due to yin deficiency, fever, febrile disease with accumulation of blood.

Development on the Questionnaire of Cold-Heat Pattern Identification Based on Usual Symptoms for Health Promotion - Focused on Reliability Study (건강 증진을 위한 평소 증상 기반의 한열변증 설문지 개발 - 신뢰도를 중심으로)

  • This study was performed to develop questionnaire of cold-heat pattern identification(PI) based on usual symptoms for general people, and we analyzed reliability of the questionnaire. We reviewed cold-heat PI questionnaire of advanced research and selected twenty-three items through three time experts meeting. Three hundred and fifty nine healthy subjects were joined in this study. As a results, Cronbach's α of cold-heat PI questionnaire was 0.79 and 0.83. According to the factor analysis about fifteen-chosen cold-heat PI questionnaires, each cold-heat consisted of two factors and each Convergence was 56.46% and 65.93%. Intra-class Correlation Coefficient was 0.67-0.83. Based on the median of samples of primary source, we classified subjects into four category as Cold, Heat, No Cold-Heat, and Cold-heat complex. We examined agreement of diagnosis and coefficient of kappa, and agreement rate of diagnosis was 64.2%, and coefficient of kappa was 0.51. Based on research result, we expect that validity study about questionnaire of cold-heat PI based on usual symptoms will be continued, and hope to be used as subsidiary diagnosis in clinical practice.

Study on Deficiency-Excess Pattern Questionnaire Development Possibility (허실 변증 설문지 개발 가능성에 대한 고찰)

  • Deficiency and excess pattern differentiation is unique characteristic of traditional oriental medicine on diagnosis and treatment. This differentiation is accomplished through pulse, tongue diagnosis and question examination, but most of these processes need to be objectified for efficacious treatment and traditional medicine development. In this study, we developed the deficiency and excess pattern questionnaire for objectification of question examination. The deficiency and excess pattern questionnaire was made out through The Traditional Oriental Medical Literature with Delphi Technique. Patients who visited oriental medical hospital filled out the questionnaire by themselves. Diagnosis of deficiency and excess pattern are conducted separately by oriental medical doctors with more than 5 years' clinical experience. Various physical condition factors were derived for the deficiency and excess pattern questionnaire. (Ordinary health degree, pain pattern, fatigue, weight change, sweating, uncomfortable awareness on chest and abdomen) Deficiency symptoms group acquired internal consistency, but excess symptoms group did not. (Cronbach's ${\alpha}$ > 0.6) There were significant associations between doctor's diagnosis and deficiency and excess symptoms in 'ordinary heath degree', 'voice weakening', and 'chest distress' (p-value < 0.1) There were significant differences between deficiency and excess syndrome patients groups in deficiency questionnaire score but there were no significant differences between deficiency and excess syndrome patients groups in excess questionnaire score. We acquired the internal consistency and significant result of deficiency pattern questionnaire, but we can find out some difficulties in development of the excess pattern questionnaire. These difficulties are associated with insufficiency description of traditional literature and small number of patients diagnosed as excess pattern.

Hair-growth Promoting Effect of Microneedle Roller Therapy (미세침요법의 모발성장효과)

  • Micro needle roller therapy has been used for cosmetic purposes, such as reducing skin winkles and improving elasticity of skin. It is claimed that micro needle roller therapy has potentials for connective tissue regeneration by facilitating collagen synthesis. Therefore, there seems to be a possibility that connective tissue regenerating potential of micro needle roller therapy could influence the hair growth cycle. This study, we investigated the hair growth-promoting effects of micro needle roller therapy. C57BL/6 mice were devided into three groups as follows: normal saline-treated, minoxidil-treated, and micro needle roller therapy-received group. Hair growth activity was evaluated by handscopic and microscopic observations. Sections of dorsal skin were stained with hematoxylin and eosin. Expression of BrdU, FGF, and VEGF was detected by immunohistochemical staining. Micro needle roller therapy enhanced the development of hair follicle during anagen. Immunohistochemical analysis revealed that micro neeld roller therapy incresed the expression of BrdU and FGF in the hair follicles of C57BL/6 mice. Furthermore, micro needle roller therapy upregulated mRNA expression of VEGFR-2, FGF-2, EGF - growth factors that play a central role in hair follicle development during anagen. These results suggest that Micro needle roller therapy can potentially be used for the treatment of alopecia.

Comparative Study on the Configuration Factors and Measurement Method of Yangsaeng Gigong and Psychological Skills Training (양생기공과 심리기술훈련의 구성요인과 측정평가방법의 비교 연구)

  • If we can evaluate the training results more appropriately and analyze the psychological changes in more objectively, the training can be more efficient and it can help to solve the problems during the training. This research analyzed the similarity and differences of Yangseng Qigong and psychological skill training's components and measurement methods. Also, the research further studied psychological skill training's measurement method. Results : The measurement type of Yangseng Qigong are Sagong, a record of Sagong, Damgong, IIgi, and Sogyeol. Similarly, the measurement type of psychological skills training are questionnaire, interview, personal profile, and observation of a single subject. Despite the similarities between the measurement type of Yangsaeng Qigong and psychological skills training, unfortunately there are differences in the contents. Thus, If we introduce strengths of each method to the other, we may have better measurement methods. The questionnaire method of psychological skills training is the most commonly used method, because it gives concrete, objective, reliable and highly reproducible results. If we refer to the questionnaire development process in psychological skills training, we can have objective and concrete measurement tool for Yangsaeng Qigong training. In the psychological skills training measurement tool development process, there are 5 stages; test conception, test preparation, test execution, question analysis, and test modification. Furthermore, the requirements for the good survey are reliability, validity, practicality, and cultural appropriateness. Through this study, we reached the conclusion that we need to combine two methods together to develop a better method because Yangsaeng Qigong places importance on comprehensive method and realization while psychological skills training places importance on analytical method and objective evaluation. And It seems that a practical study is needed to resolve the concerete problems of Yansaeng Qigong through these concrete cases.

Gallic Acid Inhibits STAT3 Phosphorylation and Alleviates DDS-induced Colitis via Regulating Cytokine Production

  • Signal transducer and activator of transcription 3 (STAT3) is associated with various human diseases, such as cancer, auto-immune disease, and intestinal inflammation. The limited and inadequate effect of standard approaches for treating inflammatory bowel disease (IBD) has prompted to develop alternative anti-colitis agents through inhibition of STAT3. Here, we show that gallic acid (GA), a 3,4,5-trihydroxybenzoic acid, markedly reduced phosphorylation of STAT3. Among the derivatives of benzoic acids, GA showed significant inhibition on STAT3 phosphorylation. In addition, GA ameliorated the dextran sodium sulfate (DSS)-induced acute colitis as determined by the measurement of symptomatic and histological indices. The suppression of DSS-induced acute colitis by GA treatment may be related to the regulation of cytokines and growth factors. Furthermore, GA inhibited phosphorylation of STAT3 in the colon tissue of DSS-treated mice. These findings may be useful in comprehending the molecular action of GA on STAT3 phosphorylation and provide novel insights into the potential application of GA in the treatment of STAT3-related inflammatory disease, such as IBD.
