한방산업의 지역개발유형연구 (Study in the Local Developing Form of Oriental Medicine Industry)

  • According to each developing type, we study a concept, contents and a consideration about in oriental medicine industry. The divisions of each developing speciality are followings: Urban industry type - The development of urban industry type has difficulties; The lack of technical development and social adaptation, etc. If it overcomes these difficulties, it will be activated in the oriental medicine industry. Rural industry type - The oriental medicine industry of rural type needs the model of rural oriental medicine industry which leads an activity and an independence of the rural region. Within the industrialization of the oriental medicine industry, if rural resource makes a specialized brand, it can have great effects in the future industry. Resort industry type - When The resort type is related the characters of natural circumstances, cultural industry and health management, it has many effects. If we gain the clear direction which is activated the regional specialities, the oriental medicine industry of resort type can estabilishs the high value industry. Event industry type - The event type is a festival, an exhibition, etc. Within this event, the promoter must provide visitors the specialized sights, the food things, the enjoyable things. And using traditional image, we have the regional resource is recognized good things and the nomination of open place can inform.

  • 본 실험은 한국산 잠견에 기생하는 세균을 분리하여 생리학적 형태학적 및 배양적인 특성을 파악하여 그 미생물을 동정하고 저장중인 잠견에 미생물 침식을 방지하는 과학적인 근거를 모색했다. 그 실험결과는 다음과 같다. 1. 잠견으로부터 분리된 12 strain의 Bacteria의 형태학적인 실험결과 colony의 형태는 table II와 같다. 종합적인 형태적 특성은 table III과 같으며 gram stain과 spore stain은 Fig, I, II와 같다. 2. 분리된 12 strain의 배양적인 특성은 table IV, V, Ⅵ와 같다. 3. 분리된 12 strain의 생리적인 특성은 table Ⅶ와 같다. 4. 이상, 형태학적 배양적 생리적인 특성에 의거하여 분리균주의 유연관계를 추정하면 다음과 같다. Prevalence of Disc Degeneration in Asymptomatic Korean Subjects. Part 2 : Cervical Spine

  • Objective : Similar to back pain, neck pain has recently shown to have increasing prevalence. Magnetic resonance imaging (MRI) is useful in identifying the causes of neck pain. However, MRI shows not only pathological lesions but also physiological changes at the same time, and there are few Korean data. The authors have attempted to analyze the prevalence of disc degeneration in highly selective asymptomatic Korean subjects using MRI. Methods : We performed 3 T MRI sagittal scans from C2 to T1 on 102 asymptomatic subjects (50 men and 52 women) who visited our hospital between the ages of 14 and 82 years (mean age 46.3 years). All images were read independently by three observers (two neurosurgeons and one neuroradiologist) who were not given any information about the subjects. We classified grading for cervical disc herniation (HN), annular fissure (AF), and nucleus degeneration (ND), using disc degeneration classification. Results : The prevalence of HN, AF, and ND were 81.0%, 85.9%, and 95.4%, respectively. High prevalence of HN, AF, and ND was shown compared to previous literature. Conclusion : In asymptomatic Korean subjects, the abnormal findings of 3 T MRI showed a high prevalence in HN, AF, and ND. Several factors might play important roles in these results, such as population-specific characters, MRI field strength, and disc degeneration grading system.

Influence of Different Treatment Regimes of Phytoecdysteroid on Silkworm, Bombyx mori L.

  • The response of silkworm, Bombyx mori L. to phytoecdysteroid (PE) when administered at different ages of $5^{th}$ instar was studied in the popular bivoltine ($CSR2{\times}CSR4$) and multi${\times}$bivoltine ($PM{\times}CSR2$) silkworm hybrids, reared on the Victory-1 variety of mulberry leaves. PE was administered to $5^{th}$ instar silkworm per os at a rate of $250{\mu}g$ per 100 larvae to different batches of silkworm at 0, 24, 48, 72, 96, 120, 144 hrs and at the onset of cocoon spinning when a few larvae were ripe. The larval and mounting duration, cocoon yield and cocoon characters were influenced by PE. The intensity of influence was dependent on the time of application. The larvae treated at the beginning of the instar, improved the economic traits significantly with a marginal increase in larval duration. In the larvae treated at the middle of the instar, larval duration was shortened remarkably but the economic traits were adversely affected. This particular treatment can become a good management strategy in the case of mulberry leaf shortage or disease incidence. In the larvae treated at the onset of cocoon spinning, the mounting duration was substantially reduced without much effect on the cocoon traits which would be a big benefit in commercial sericulture. The physiological significance of varied response of silkworm to PE administration is discussed.

  • 항생제에 대한 내성균의 출현은 매년 급격하게 증가되고 있으며 이에 대한 약제 개발이 요구되고 있다. 특히 메치실린내성 황색 포도상구균(MRSA)은 여러 항생물질에 대하여 다제내성을 나타내므로 감염증치료에 어려운 문제점으로 남아 있다. 이러한 감염증치료에 사용할 목적으로 임상 가검물에서 분리한 MRSA를 시험균으로 사용하여 항균 활성이 강한 방선균인 YSK-681을 선별하였다. 균주 동정을 위하여 선별된 YSK-681을 형태적, 배양학적, 생리적, 화학적 분석을 하였으며 41개의 단위 항목을 조사하여 TAXON program으로 수리 동정한 결과 주군집 29개에 속하는 Streptomyces lydicus로 동정하였다.

  • 산업적인 응용을 위한, 생 전분 분해력이 높고 액체 배양에 적용 가능한 균을 토양으로부터 분리한 결과, 옥수수 생 전분 당화 효소 생산력이 높은 곰팡이 No.55 균주를 분리하고 이 균주의 형태적, 생리적, 배양특성을 조사, 균 동정을 행한 결과 No.55 분리균은 Aspergillus niger 또는 그 유연균으로 동정되었다.

  • 유기농 인증을 받아서 3년 이상이 경과한 농가의 토양을 수집하여 근권 세균을 450균주 분리하였고, 여러 가지 식물 병원균의 균사 생장을 억제하는 세균 KB-401 균주를 선발하였다. KB-401 균주는 특히 감귤 검은점무늬병균(Diaporthe citri)의 균사 생장과 포자 발아를 억제하며, 발아하는 포자에 처리하였을 때 발아관의 선단이 팽윤되는 현상을 관찰할 수 있었다. 본 균주는 생리적인 특성과 16S rDNA의 염기서열을 분석한 결과, Bacillus subtilis로 동정되었다.

Influence of preserved brewing yeast strains on fermentation behavior and flocculation capacity

  • Preservation methods on the physiological and brewing technical characters in bottom and top brewing yeast strains were investigated. The preserved yeasts were reactivated after 24 months storage and grown up to stationary phase. The samples of filter paper storage indicated a higher cell growth and viability during propagation than those of nitrogen and lyophilization storage independent on propagation temperature. In addition, the filter paper storage demonstrated a faster absorption of free amino nitrogen and a highest level of higher aliphatic alcohols production during propagation than other preservation methods, which can be attributed to intensive cell growth during propagation. Moreover, the filter paper storage showed a faster accumulation for glycogen and trehalose during propagation, whereas, in particular, lyophilization storage noted a longer adaptation time regarding synthesis of glycogen and trehalose with delayed cell growth. In beer analysis, the filter paper storage formed an increased higher aliphatic alcohols than control. In conclusion, the preservation of filter paper affected positively on yeast growth, viability and beer quality independent on propagation temperature. In addition, in this study, it was obtained that the HICF and Helm-test can be involved as rapid methods for determination of flocculation capacity.

Characterization of In vitro Propagated Plants Via Somatic Embryo Formation from Old Wild Panax ginseng

  • Wild Korean ginseng has been recognized as highly precious medicine since ancient times. Nowadays, the population of wild ginseng in the forest of Korean peninsula is very rare due to indiscreet harvest. In this work, we investigated the plant regeneration via somatic embryogenesis from embryogenic callus of old wild ginseng (more than 50 years-old) and compared the features of plants regenerated from 5-years old and 50 years-old ginseng. Induction of embryogenic callus from adventitious roots of 50 year-old wild ginseng required 83 weeks of culture, but only 10 weeks were sufficient for 5 year-old ginseng. Height and width of plants derived from the old wild ginseng was smaller and slender compared to the plantlets derived from 5 year-old ginseng. Total chlorophyll contents was 2-6 time lower in plantlets regenerated from 50 year-old wild ginseng than those from 5 year-old ginseng, but anthocyanin content was higher in 50 year-old ginseng. Our results revealed that plants regenerated from old wild ginseng have different morphological and physiological characters probably due to age-dependent phenomenon.

Moderate 호염성 세균의 Plasmid 유전자 분리 (Isolation of Plasmids from the Moderately Halophilic Bacteria)

  • 천일염으로 부터의 $10\%$ NaCl 농도에서 생육할 수 있는 moderate 호염성 세균들을 대상으로하여 간단한 boiling 법으로 plasmid 유전자를 함유하는 6종을 선별하여 Alkaline-Lysis 법과 고분자 plasmid 추출법인 D. Portnoy & F. White 방법에 따라 plasmid를 추출하여 agarose gel 전기영동으로 확인하였고 각 균을 동정하였다. 그중 Vibrio sp. 14는 최소 7.2kbp, Alcaligenes sp.63은 6.4 kbp, Pseudomonas sp.11은 6.35 kbp, Flavohacterium sp.38은 8.5 kbp, Bacillus sp.16은 8.75 kbp, Alcaligenes sp.52는 6.8kbp 크기의 plasmid를 가지고 있는 것으로 나타났다.
