Photostability evaluation of Jawarishe Jalinoos

  • Jawarishe Jalinoos (JJ) is an orally used formulation available in semisolid dosage form, prepared with powdered plant materials mixed in honey or sugar syrup. It has many admirable pharmacological effects and used in Unani medicine to treat various acute and chronic disorders since ancient times. The ICH Harmonised Tripartite Guideline stated that photostability testing should be an essential part of stability testing to confirm that light exposure does not result in an unacceptable change in drugs substance and finished products. To date, the effect of light on JJ is not studied, in this study photostability evaluation of JJ was carried out. The test sample was manufactured with genuine ingredients in the in-door pharmacy of the National Institute of Unani Medicine. JJ was packed in two transparent polyethylene terephthalate airtight containers. The first sample was analysed at zero-day and the second sample was placed in a stability chamber subjected to light challenge with an overall illumination of 1.2 million lux hours combined with near ultraviolet energy of 200-watt hours per square meter by using option 2, along with 30±2℃ temperature and relative humidity 70±5%. Analysis of both finished products showed no considerable changes in organoleptic characters. Less than 5% variation was observed in physicochemical parameters. HPTLC fingerprinting showed justifiable variation. Microbial load and specific counts were within the limit prescribed by WHO. As no unacceptable changes were noted in JJ subjecting to light challenge, it is concluded that JJ is a photostable Unani compound formulation.

Fine Structure of Blue-green Algae, Microcystis aeruginosa Kutzing (남조(藍藻) Microcystis aeruginosa Kutzing의 미세구조(微細構造)에 관(關)한 연구(硏究))

  • In order to understand the morphological differences between two different organic loadings by its upstream, and to compare with other algal groups with references, the fine structure of blue-green algae, Microcystis aeruginosa Kitzing, taken from two branches, Tongbok and Bosung stream of Lake Chuam, Korea pennisula was examined. It showed extinct differences in most physicochemical factors between both branches, except water temperature and pH values. The concentrations of total phosphorus in Tongbok branch were twice as those of Bosung. M. aeruginosa cells were enumerated totally $1.2X10^4cells/ml$ and these individuals in branch of Tongbok were close to two times as much as Bosung. In light and electron microscopy, natural M. aeruginosa colonies formed irregular shape and non-directional array in amorphous matrix. They were consisted of many kinds of cells, youngs or olds in cell division, solitary, and various size of cells. Each cell ranged from 2.61 to $5.40{\mu}m$ in diameter, and averaged as $3.54{\pm}0.19{\mu}m$. In cytoplasm, they contained a number of inclusions in various size, shape and appearances. Among them, polyhedral bodies or carboxysomes, a structured granules, photosynthetic lamellae or thylakoids, and gas vacuoles were prominent and easy to recognize. Although it was failed to find the definable morphological variations in the ultrastructure of M. aeruginosa in terms of algal habitual environments, some useful characters were founded, outer layer of cell wall, polyhedral bodies and gas vacuoles, in blue-green algal classification and taxonomy.

Influence of Silicate Application on Rice Grain Quality (규산 시용이 미입의 품질에 미치는 영향)

  • To evaluate the characters related to rice grain quality as affected by silicate application, the ripened color, appearence and physicochemical properties of rice grain which were produced at the silt-loam paddy field of National Yeongnam Agricultural Exeperiment Station with a little poor drainage for trials on long-term effects of continuous application of same fertilizer were analyzed comparing with the NPK fertilizer an the NPK+compost in 1995. The whiteness value and color space value in ripened color of rice hull by silicate application were higher than those in the plots of NPK and NPK+compost, because of low percentage of rusty grain and sooty grain, The percentage of perfect grain in brown rice and in milled rice by the silicate application increased by 6.1~7.5% and by 1.8~3.5% respectively, as compared with the NPK application and white core and white belly of milled rice were decreased, The amylose content, protein content and starch-iodine blue value of milled rice by silicate application were lower than those in the plots of NPK and NPK+compost, while the peak point and breakdown viscosity of gelatinized rice flour on amylograph, and tastes value evaluated as a indicater of pannel test by nireco tester were higher. By silicate application these factors could be affected toward better in eating quality.

Characterization of exopolysaccharide-producing lactic acid bacteria from Taiwanese ropy fermented milk and their application in low-fat fermented milk

  • Objective: The aim of this study was to characterize the exopolysaccharides (EPS)-producing lactic acid bacteria from Taiwanese ropy fermented milk (TRFM) for developing a clean label low-fat fermented milk. Methods: Potential isolates from TRFM were selected based on the Gram staining test and observation of turbid suspension in the culture broth. Random amplified polymorphic DNA-polymerase chain reaction, 16S rRNA gene sequencing, and API CHL 50 test were used for strain identification. After evaluation of EPS concentration, target strains were introduced to low-fat milk fermentation for 24 h. Fermentation characters were checked: pH value, acidity, viable count, syneresis, and viscosity. Sensory evaluation of fermented products was carried out by 30 volunteers, while the storage test was performed for 21 days at 4℃. Results: Two EPS-producing strains (APL15 and APL16) were isolated from TRFM and identified as Lactococcus (Lc.) lactis subsp. cremoris. Their EPS concentrations in glucose and lactose media were higher than other published strains of Lc. lactis subsp. cremoris. Low-fat fermented milk separately prepared with APL15 and APL16 reached pH 4.3 and acidity 0.8% with a viable count of 9 log colony-forming units/mL. The physical properties of both products were superior to the control yogurt, showing significant improvements in syneresis and viscosity (p<0.05). Our low-fat products had appropriate sensory scores in appearance and texture according to sensory evaluation. Although decreasing viable cells of strains during the 21-day storage test, low-fat fermented milk made by APL15 exhibited stable physicochemical properties, including pH value, acidity, syneresis and sufficient viable cells throughout the storage period. Conclusion: This study demonstrated that Lc. lactis subsp. cremoris APL15 isolated from TRFM had good fermentation abilities to produce low-fat fermented milk. These data indicate that EPS-producing lactic acid bacteria have great potential to act as natural food stabilizers for low-fat fermented milk.

Relation of Correlation about Rice quality related Characters in Condition storage of Unhulled rice (정조 저장조건에서 식미관련특성에 대한 상관성 정도)

  • This study was carried on cool and RT(room temperature) storage of unhulled rice. In RT storage of an analysis of coefficient relation, high significant positive coefficients were observed in toyo index and breakdown, setback and protein content. high significant negative coefficients were showed setback and breakdown, breakdown and protein content. In cool storage of an analysis of coefficient relation, high significant positive coefficients were observed in toyo index and amylose content and gelatinization start temperature and protein content and high significant negative coefficients were showed toyo index and whiteness, toyo index and gelatinization start temperature, gelatinization start temperature and amylose content. In RT storage of a path coefficient analysis, a highest positive direct influence was observed in amylose content and a highest negative direct influence was protein content. Positive indirect influence was high revealed breakdown and protein content and negative indirect influence was gelatinization start temperature and Mg/K ratio. In cool storage of a path coefficient analysis, a highest positive direct influence was whiteness and a highest positive indirect influence was gelatinization start temperature. Positive indirect influence was high revealed gelatinization start temperature and amylose content, negative indirect influence was whiteness and gelatinization start temperature. In RT storage of Multiple regression equation of Toyo index based on physicochemical properties of unhulled rice, a highest coefficient of determination was revealed among five facters of whiteness, protein content, Mg/K ratio, amylose content and gelatinization start temperature. In cool storage of Multiple regression equation of toyo index based on physicochemical properties of unhulled rice, highest coefficient of determination was revealed among five facters of moisture content, amylose content, gelatinization start temperature, breakdown and setback.

Effects of Storage Temperature on Physicochemical and Sensory Characteristics of Soybean Sprouts (콩나물의 저장 중 온도변이에 따른 품질변화)

  • Soybean sprouts produced at optimal temperature are placed or displayed for several days in market shelf of relatively cool temperature (ca. $13^{\circ}C$). During this period a number of changes occur including changes in color, smell, taste, nutritional quality, etc. In order to investigate the changes of these factors, soybean sprouts packed in plastic film bag (OPP+PE) were stored at the two different temperature ($3^{\circ}C$ and $13^{\circ}C$). Morphological characters, physicochemical changes and enzymes activity related to visible quality (color) of soybean sprouts were examined. The numbers of fine roots were greater and hypocotyls were longer in soybean sprouts stored at $13^{\circ}C$, although there was no significant difference in diameter, fresh weight and dry weight of hypocotyls between the two storage temperatures. Browning of hypocotyl, as an indicator of a typical deterioration in sprout quality, was highly dependent on the activity of polyphenol oxidase (PPO). Considering the low level of soluble protein in hypocotyls, the relatively higher activity of PPO suggested a critical role of PPO in stored soybean sprouts. PPO activity of sprouts stored at $13^{\circ}C$ was 2-fold higher than that of sprouts stored at $3^{\circ}C$ after 4 days. In sprouts stored at $13^{\circ}C$, the PPO activity was increased from day 0 until 6 days and since then, it was not detected. Crude protein content was increased to 30.9~35.4% based on dry weight with extended storage period. The change in crude protein was greater in sprouts stored at high temperature ($13^{\circ}C$). Total free amino acid content was increased in both temperatures. However, the changing rate of free amino acid was greater in sprouts stored at $13^{\circ}C$.

Diversification of Rice Quality for Processing. Physicochemical Characteristics and Inheritance of Floury Endosperm Mutants (특수 가공용 미질개발 : 분상질배유 돌연변이 계통의 이화학적특성과 유전)

  • This study was carried out to assess the agronomic characters and physicochemical properties of floury and chalky-endosperm mutant lines induced by chemical mutagen treatment to rice varieties, Hwacheongbyeo and IR24. Linkage analysis of a floury-endosperm gene was carried out using linkage testers. The grain size of brown rice of the mutants was smaller than that of the original varieties. The l, 000-grain and 1$\ell$ weight were lighter in the mutants compared with those in the original varieties. The compound starch granules in the endosperm cell of the mutants showed a loosely-packed crystalline structure. Amylose contents in mutants ranged from 16.9 to 28.5%. Crude protein contents of the mutants were not significantly different from the original rice variety, Hwacheongbyeo, but white core mutant(line 47106) derived from IR24 showed higher protein(l1.32%) compared with IR24(8.30%). The mutants showed slightly harder gel characteristics, and much lower viscosity in Amylograph than original varieties. Steamed rice-cakes from mutant lines showed greater volume than those from original varieties. During the process of alcohol fermentation, Brix in the mutants(especially floury mutants) decreased faster and the alcohol production after 10-day fermentation was much greater in the mutants than in the original varieties. Three different gene loci for floury endosperm characteristics were identified from the allelism test among mutant lines, and the genes were tentatively symbolized as flo-a, flo-b and flo-c, respectively. A floury gene, flo-a, was linked with lg(liguleless) gene in the linkage group N, with R.V. 5.76$\pm$1.72%.

Variation of Grain Quality and Grain Filling Rapidity Milyang 23 / Gihobyeo Recombinant Inbred Lines (벼 밀양 23호$\times$기호벼의 재조합 자식계통에서 초기급속등숙과 미질 특성)

  • This study was carried out to get the basic informations regarding the varietal variations for the physicochemical properties such as protein content, amylose content, fatty acid content, grain quality values and color properties such as lightness value, chroma and hue for the 164 recombinant inbred lines(RILs) of Milyang 23 and Gihobyeo(M/G) at the experimental farm in the Sangji University. The principal component analysis and heritability study were conducted for this experiments. The rapidity of grain filling(RCF) for the 164 M/G RILs could be classified into four groups such as slow maturing group less than 41%, mid-slow maturing group 41∼60%, fast maturing group 61∼80% and very fast maturing group more than 81% based on the rapidity of grain filling rate. The slow maturing group of RGF showed a little bit higher protein content 9.1%, compared to the other RGF groups. However, the amylose content of all the RGF groups revealed the same content by the groups. The very fast maturing group of RGF showed longer grain length in brown rice compared to other RGF varietal groups, in case of grain width in brown rice showed shorter than any other groups. The alkali digestive value which was so much related to gelatinization temperature showed 3.40 degree at fast maturing group of RGF in M/G RILs. However, the very fast maturing group of RGF revealed 4.31 degree of alkali digestive value. The principal component analysis was performed by the chemical and color properties such as quality value, protein content, amylose content, alkali digestive value, fatty acid content, lightness value, chroma and hue for M/G RILs. The first principal component was able to explained upto 36% to total informations. It was corresponded to quality value, protein content, amylose content, fatty acid content, lightness value and a-value(green -1 red). The characters regarding grain quality showed high heritable properties more than 75% of heritability, but color characters appeared relatively lower heritability compared to grain quality.

Characteristics and Genesis of Terrace Soils in Yeongnam Area -V : Soil Genesis and Classification (영남지역(嶺南地域)에 분포(分布)된 단구지토양(段丘地土壤)의 특성(特性)과 생성연구(生成硏究) -제(第)5보(報) : 토양생성(土壤生成)과 분류(分類))

  • A series of studies on the properties of clayey terrace soils distributed at the inland (Yeongcheon) and coastal (Yeongjil) regions in Yeongnam district was carried out. On the base of the facts found and already reported about the macro morphological features as well as on pedological characters in micro scale, physicochemical properties, mineralogical characteristics etc., the present study dealt with soil genesis and tried to classify the soils for reasonable use and managements. 1. Although the both regions belonged to "Mesic" soil temperature regime and "red and yellow earths" areas of "Thornthwaite" pedo-climatic diagram, climatic indices as a soil forming factor indicate that the coastal Yeongil had milder than the inland Yeongcheon. 2. All the terrace soils had developed soil profiles with an "Argrllic B". Upyeong soils in Yeongil region had "Argillans" even in the "II B horizons" that possibly be "Paleo-argillic". 3. The bisequum profiles of Bancheon in Yeongcheon and Upyeong in Yeongil revealed that they were developed on Late Mesozoic shale and on semiconsolidated Tertiary deposits respectively, therefore the overlying clayey terrace deposits were assumed to be originated from the Early Quaternary deposits, Diluvium. 4. To supplement the Soil Taxonomy of USDA, the terrace soils with different degrees of gleyzation were classified as follows; Deogpyeong and Hwadong soils which have less than 50cm of paddified gley horizons (redness less than 0.5) in the upper part of the profiles by artificial surface irrigation, tentatively classified into "Anthrepiaquic Hapludalfs" and the Geugrag soils that have more than 50cm of paddified gley horizons within 1.2m of the profiles, into "Anthr-aquic Ochraqualfs" while the Upyeong soils that had greyish mottles in subsoils by natural ground water remain as an "Aquic Hapludalfs" the same as present. The Bancheon soils with free mottles are into "Typic Hapludalfs" as used at present.

Major Characteristics Related on Eating Quality and Classification of Inbred Lines of Waxy Corn (찰옥수수 자식계통 식미관련 특성 및 계통 분류)

  • Objectives of this study were to select inbred lines which have a good eating quality and desirable segregates during inbreeding of waxy corn. The 64 inbred lines showed a large variance in their kernel shape and weight. 100-kernel weight, pericarp thickness, kernel length, kernel width, and kernel thickness ranged $11.7\~37.3g,\;11\~77{\mu}m,\;5.8\~9.6mm,\;6.5\~10.0mm$, and $4.1\~6.8mm$, respectively. The physicochemical analysis of 64 waxy corn inbred lines showed crude protein, crude fat, free sugar, and amylopectin content ranging $8.7\~15.8\%,\;2.3\~5.8\%,\;1.1\~11.0\%,\;and\;78.5\~93.8\%$, respectively. The texture property analysis of 64 inbred lines by texture analyzer showed a big difference. Gumminess, hardness, and chewiness of 64 inbred lines ranged $91\~383,\;181\~394,\;and\;73\~370$, respectively. The principal component analysis for 14 characteristics related to kernel quality showed that $73.1\%$ of the total variation could be attributed to the first five principal components. Biological meaning of the principal component was explained clearly by the correlation coefficient between principal components and characters. The first principal component appeared to correspond to small kernel and bad eating quality, The second principal component appeared to correspond to large kernel and good eating quality. The 64 inbred lines were classified into 8 groups by the cluster analysis using the first and second principal component. Among the groups, group VII and VIII included inbred lines with good eating quality that had thin pericarp thickness, low protein content, large kernel, and soft tenderness.
