PURPOSE: The purpose of this study was to determine the effect of different frequencies (4Hz and 100Hz) of transcutaneous electrical nerve simulation (TENS) on pain relief using c-fos expression in the spinal cord of rat osteoarthritis to investigate the appropriate frequency for pain relief. METHODS: Total of 30 Sprague-Dawley rats was used and randomly divided 2 groups according TENS frequency and applicate the TENS during 3 period (3 days, 7 days, 10 days). The induction of osteoarthritis by 3mg monosodium iodoacetat was injected into the right knee joint of rats. Three days later, commercially available TENS unit was used for stimulation was set to 20minutes on 3, 7, 10 days after surgery. Western blot analysis system was used to detect immunoreactive proteins. The thickness of the bands were photographically measured by Scion Image. RESULTS: When investigating the c-fos expression of TENS on spinal cord in OA knee over 10 days, between-groups differences in c-fos expression reached a significant level by day 10. For within-groups comparisons, the c-fos expression decreased significantly across days in low- and high-frequency TENS groups. CONCLUSION: Whether at low- and high-frequency, the TENS as a therapy obtained beneficial effects of pain relief and TNES at high-frequency is more beneficial effects on the pain relief when TENS applied at injury site.
Expression of c-fos, an immediate early gene, has accepted to be a marker of functional activity in neurons. This study was aimed to investigate the effects of cold therapy on the expression of spinal cord c-fos in rats of carrageenan-induced muscle pain. Muscle pain was induced in male Sprague-Dawley rats by intra-muscular injection of gastrocnemius with $2\%$ carrageenan. The paw withdrawal latency (PWL) and tail flick test (TFT) responses to heat stimuli were used to detect secondary hyperalgesia produced by the muscle pain and measured to assess the effects of cold. The expression of c-fos was determined in the lumbar regions of the spinal cord by reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry assays. The secondary hyperalgesia to heat simuli (PWL and TFT) were significantly reduced in cold therapy compared with that in the controls. In RT-PCR assays the expression of c-fos mRNA was down-regulated in the lumbar spinal cord in cold group. In addition, Fos immunoreactivity in the dorsal horn of the lumbar spinal cord was decreased in cold group. These results suggested that application of cold attributed to increase PWL and TFT responses and to decrease expression of the c-fos produced by muscle pain.
This study was performed to investigate the effect of low power laser irradiation on Substance P(SP) expression in the burned skin of the rats. Burns of about 3cm in diameter were created with $75^{\cric}C$ water on the back of the rats, and the lesion of experimental group were irradiated on days 1, 2, 3 and 4 postwounding. Control leasions were not irradiated. After burns, low power laser irradiation was applied by using 1000Hz, 830nm GaAlAs(Gallium-aluminum-arsenide) semiconductor diode laser. The expression of evaluated Substance P(SP) immunohistochemistry on rabbit anti-SP The results of this study wereas follows 1. The Substance P was expressed in the lamina I and II of dorsal horn of spinal cord. In expression of SP, the lesion of control group made SP to more induce significantly than experimental leasions. 2. SP immunoreactivity in burned leasion of spinal cord were decreased markedly 4 days after burns, and decreased gradually from 1 day to 2 days in burns which is laser irradiation These data suggest that low power laser have a pain release effect in the burned skin of the rats.
PURPOSE: This study examined the effects of treadmill exercise of diverse intensities on the expression of IL-$1{\beta}$ (interleukine-$1{\beta}$) in the spinal cord in osteoarthritis rats. METHODS: The authors applied treadmill exercise of diverse intensity for 4 weeks to Sprague-Dawley rats to which intra-articular injection of monosodium iodoacetate(MIA, $3mg/50{\mu}l$, diluted in saline) was applied in the right knee joint to induce osteoarthritis. The four-week exercise was not applied to the control group(CG, n=15), while exercise of applicable intensity was applied to the low-intensity exercise group(LEG, n=15), moderate-intensity exercise group (MEG, n=15), and high-intensity exercise group(HEG, n=15) for four weeks. Observations were made of expression of IL-$1{\beta}$ in the spinal cord in osteoarthritis rats using western blot analysis. RESULTS: There were significant differences(p<.05) in the comparison of expression of IL-$1{\beta}$ in the spinal cord between the four groups involved. And the LEG and MEG had reduced expression of IL-$1{\beta}$ significantly than the CG(p<.05); in particular, the MEG showed the lowest expression. On the other hand, the HEG had more elevated expression of IL-$1{\beta}$ significantly than the CG(p<.05). CONCLUSION: As a result, factors that induce neuropathic pain such as IL-$1{\beta}$ are reduced; thus, the recovery of damaged neurons is improved and neuropathic pain is reduced. Further, when prescribing exercise to treat osteoarthritis patients, exercise of moderate intensity suitable for patients' physical conditions, rather than high intensity, maximizes the effects of this therapy.
This study examined the effect of parents' neglect behavior and children's irrational beliefs on children's anger expression style. The participants were 379 5th and 6th grade elementary school children. The major findings were as follows. Children tended to perceive higher levels of their parents' cognitive and emotional neglect than physical and home environmental neglect. Boys perceived higher levels of their parents' physical, supervisory and home environmental neglect than girls, and girls showed higher level of excessively self-expecting belief than boys. There were statistically significant correlations between the parents' neglect behavior, their child's irrational beliefs and anger expression style. A regression analysis showed that parents' physical and cognitive neglect were predictors of children's anger control. Children's excessive apprehension and avoidance beliefs were the best predictors of children's anger-in, while children's deterministic belief was the best predictor of children's anger-out. Parents' physical neglect behavior appeared to be powerful predictor.
The purpose of this study was to investigate the effect of electrical stimulation(EST) on MAP2(Microtubule Associated Protein 2) expression in cerebral cortex following sciatic nerve crush injury in rats. Twelve Sprague-Dawley adult female rats, six for control and six for experimental, were anesthetized and their sciatic nerves were crushed. The electrical stimulation (EST) was applicated with 3 Hz for 10 minuties in a day for muscles innervated sciatic nerve. The MAP2 expression in cerebral cortex was identified from immunohistochemistry against MAP2. The result of this study were as follow: 1) In control group, MAP2 immunoreactive neurons were observed but there no significant increase for 3 days. 2) MAP2 immunoreactive neurons were increased markably in experimental group than control group. 3) MAP2 immunoreactive neurons were increased markably after applicating with EST in sciatic nerve crush injury induced group from 2nd day. This study showed that the application of EST for muscles after sciatic nerve crushed injury made MAP2 immunoreactive neurons in the cerebral cortex increased. Therefore, the electrical stimulation on the peripheral site, denervated muscle, may facilitate MAP2 expression in the cerebral cortex.
Rats that are fed a fructose-rich diet develop hypertension, insulin resistance, and hypertriglyceridemia. To elucidate whether altered expression levels of endothelin-1 and nitric oxide synthase are related to the development of insulin-resistant hypertension, we examined the present study. Male Sprague-Dawley rats were fed a fructose-rich diet for 5 weeks. Systolic blood pressure significantly increased in fructose-fed rats. While serum free fatty acid and plasma nitrite/nitrate levels did not significantly differ between the fructose-fed and control groups, plasma insulin and serum triglyceride concentrations significantly increased in the former. Endothelin-1 mRNA expression in the aorta increased in fructose-fed rats. Neither the protein expression of constitutive nitric oxide synthase nor that of inducible nitric oxide synthase were significantly affected by fructose feeding. However, nitrite/nitrate levels in the aorta were significantly increased. These results suggest that an increase in vascular endothelin-1 is an important contributing factor to the development of hypertension in fructose-fed rats. However, the vascular nitric oxide pathway may not be causally related to the development of fructose-induced hypertension.

Objectives: The study aims to look into how the application of EA(Electro-acupuncture) in the initial step of ischemic brain injury affects the changes in Bax and Caspase-3 expression among forebrain apoptosis. Methods: It caused GI by using common carotid artery occlusion and conducted the test by dividing into the no applied EA group after 6 hours and 12 hours, the LI 4 EA group that applied EA to LI4, and GV 20 EA group that was applied to GV 20. After that, the following results were obtained by comparing Bax and Caspase-3 expression, which were apoptosis-related factors, with its application time through immunohistochemical staining after extracting brain of each group. Results: According to each group's Bax expression by the application time, the order of expression increase after 6 hours is shown to be the no applied EA group, LI 4 group, and GV 20 group. The stimulation after 12 hours was most lowly expressed in the GV 20 group, and the no applied EA group and LI 4 group showed similar level. Conclusions: EA stimulation in the initial step after ischemia seems to affect positively Bax and Caspase-3 expression that are index of forebrain apoptosis.
Muscle contusion has a negative effect on muscle function. Although several studies showed that pain control and muscle recovery is facilitated by pulsed electromagnetic fields (PEMF), there has not been much research regarding the specific effects of PEMF on them. The aim of the present study is to investigate effects of PEMF on pain and muscle recovery following extensor digitorum longus (EDL) contusion injury through measuring the expression of the c-fos proto-oncogene and nerve growth factor (NGF). Significantly reduced c-fos expression in the spinal cord was shown in PEMF groups compared with control (CON) groups. There was no significant difference between PEMF1 and CON1, but significantly increased NGF expression was shown in PEMF3 and PEMF5 compared with in CON groups, where the numbers in the group names are the days from contusion. In conclusion, PEMF could be used to not only reduce pain in muscle injuries by down-regulating c-fos expression in the spinal cord, but it could also influence muscle healing through increasing NGF expression in the injured muscle.
