Effect of Ginseng Total Saponin on the Development of Psychic and Physical Dependence on Nalbuphine

  • This study was undertaken to estimate whether nalbuphine, a mixed agonist/antagonist opioid analgesic produced psychic dependence. Moreover, the physical dependence liability of nalbuphine was compared with that of morphine after 7 days administrations of the drugs in mice and rats, and the effects of ginseng total saponin (GTS) on the development of physical dependence on nalbuphine were also studied. Nalbuphine did not produce psychic dependence. However, various abstinence signs precipitated by naloxone were observed in nalbuphine-dependent mice and rats. As the nature of the dependence syndrome produced by nalbuphine 30 mg/kg under these conditions seems similar to that induced by morphine 10 mg/kg, it is clear that nalbuphine possesses the substantial abuse potential. Therefore, nalbuphine may be needed to initiate more stringent controls for the prevention of nalbuphine abuse. On the other hand, GTS inhibited the development of physical dependence on nalbuphine and reduced the contents of dopamine and its metabolite in the brains of mice. Accordingly, results of this study suggest that the inhibitory effects of GTS on the development of physical dependence on nalbuphine may involve dopaminergic mechanism. GTS may be useful for the therapy of physical dependence on nalbuphine.

Physical Dependence on DA-5018, a Non-narcotic Analgesic Agent (비 마약성 진통제 DA-5018의 신체의존성)

  • The physical dependence potency of DA-5018, a non-narcotic analgesic agent, was tested in mice dosed with 0.5 and 4 mg/kg/day for 2 months and daily increasing doses of 1, 2, 4, 6, 8 and 10 mg/kg over 10 days. Physical dependence was assessed taking natural withdrawal induced morphine-type abstinence (jumping, falling, biting or backward locomotion, rearing etc.) as well as barbiturates-type abstinence (body weight reduction, convulsion, ataxia etc.) into consideration. The results were compared with those after the same daily increasing doses of morphine. DA-5018 did not show evidence of physical dependence liability or abuse potential as measured by morphine-type or barbiturate-type abstinence signs following daily increasing or 2-month repeated administration. On the other hand, daily increasing doses of morphine produced physical dependence and the dependent state disappeared about 6 hours after the start of withdrawal signs. In the single dose suppression test, a single dose of morphine completely suppressed natural withdrawal signs that appeared in morphine-dependent animals. Therefore, these results indicate that DA-5018 does not have abuse potential and physical dependence liability.

The Effect of Physical Self-concept on Sportswear Dependence and Psychological Happiness (신체적 자기개념이 스포츠웨어 의존성 및 심리적 행복감에 미치는 영향)

  • This study investigates the effect of physical self-concept and sportswear dependence on psychological happiness for women over 20 years of age who participate in life sports. Survey questionnaires were developed and implemented to collect data. Three hundred and thirty nine responses were analyzed to test the structural equation model using SPSS 21.0 and Amos 21. The results were as follows. First, physical selfconcept had a significant positive effect on psychological happiness and sportswear dependence mediated the relationship between them. Second, sports competence had a significant effect on psychological happiness and sportswear dependence respectively. Third, appearance satisfaction did not significantly affect psychological happiness and had a significant positive effect on sportswear dependence. Fourth, physical strength satisfaction did not affect psychological happiness and sportswear dependence. Fifth, health condition had a significant positive effect on psychological happiness and negatively influenced sportswear dependency. In conclusion, physical self-concept and sportswear dependence plays an important role in psychological happiness that requires a differentiated marketing strategy in the sportswear market.

Effects of Ginseng Saponin on Morphine Physical Dependence (몰핀의 신체적 의존성에 미치는 인삼 Saponin의 효과)

  • The present experiments were performed to investigate the effects of the ginseng total saponin on the development of physical dependence on morphine via intracerebroventricular (i.c.v) route. Morphine (10 $\mu\textrm{g}$/${mu}ell$/hr) was continuously infused via osmotic minipumps into lateral cerebral ventricle of male Sprague Dawley rats for 7 days. Concurrent ginseng total saponin (100, 200 $\mu\textrm{g}$/10${mu}ell$/hr) was infused intraperitoneally (i.p) via osmotic pumps for 7 days. Treatment with ginseng total saponin (200$\mu\textrm{g}$/10${mu}ell$/hr) significantly diminished jumping, teeth chattering, hypothermia and weight loss precipitated by naloxone, compared with those animals received only morphine infusion. These results suggest that ginseng total saponin has central effect on the inhibition of physical dependence on morphine, as systemic ginseng total saponin inhibits the development of physical dependence in rats infused with morphine intracerebroventricularly.

Effects of Glycine on the Development of Analgesic Tolerance to and Physical Dependence on Morphine in Mice

  • This study was performed to investigate the effects of glycine on the development of tolerance to and physical dependence on morphine. Repeated administration of morphine (10 mg/kg) developed tolerance and physical dependence. Glycine (100, 200 and 400 mg/kg) was administered intraperitoneally (i.p.) to mice for 7 days prior to the morphine injection. Analgesic effects were estimated by the tail flick methods. The inhibitory degree of the development of morphine-induced analgsic tolerance by i.p. administration of glycine was evidenced by the increase in analgesic response to morphine. Glycine inhibited the development of tolerance to morphine. In addition, we separately measured jumping response as the naloxone-precipitated withdrawal sign in mice that had received the same morphine. Glycine reduced the number of jumping behaviors in morphine dependent mice. These results suggest that glycine might be useful the prevention or treatment of morphine tolerance and physical dependence.

  • This review focuses on finding neurochemical changes in physical dependence on butorphanol, a relatively potent mixed agonist-antagonist opioid analgesic agent that is five times more potent than morphine in antinociceptive effects. The chronic administration of butorphanol induces physical dependence. Withdrawal from such dependence can be reliably precipitated by administration of a narcotic antagonist, e.g., naloxone. Evidence for critical involvement of excitatory aminoacid (glutamate), opioid receptors, and phosphorylation of proteins in these phenomena is summarized.

  • The present study was undertaken to determine the inhibitory effects of orally administered ginseng saponins (GS), protopanaxadiol saponins (PD), protopanaxatriol saponins (PT) and ginseng ether fraction (GE) on the development of morphine induced tolerance and physical dependence in mice and also to determine the hepatic glutathione contents. GS, PD and PT inhibited significantly the development of morphine induced tolerance and physical dependence, but GE was effective only on the inhibition of the development of morphine induced physical dependence. GS, PD, PT and GE also inhibited the hepatic glutathione level decrease induced by morphine multiple injections.

Statistical Analysis on the Temperature Dependence and Long-Term Change of Relative Humidity Sensors (상대습도계의 온도 의존성과 경년변화의 통계적 분석)

  • We have investigated temperature dependence and long-term change of humidity measurement from 32 relative humidity sensors. The readings of the humidity sensors depended not only the reference humidity, but also temperature of the chamber. Approximately, the temperature dependence of the humidity sensor in average was 0.05 %R.H./$^{\circ}C$ in the temperature range from $5^{\circ}C$ to $55^{\circ}C$. For humidity sensors that have an internal temperature compensation circuit, the resulting temperature dependence was weaker by 20%. It should be also noted that for the humidity sensors used in this work underwent ${\pm}3$ %R.H. change per year for level of confidence of 95%. The users of relative humidity sensors may refer this value as a minimum change when they set the calibration interval of the humidity sensors.
