The effluent sediment from the land-based seawater fish farms of Jeju consists of proteins, fats, ash and moisture. An evaluation of the effluent sediment as substrate for growth of phototrophic or lactic acid bacteria revealed that the sediment supported the growth of phototrophic bacteria but could support lactic acid bacteria only on supplementation with sugar. The possibility of using phototrophic bacteria for recycling the land-based seawater fish farm effluent is shown.
Single cell of an indigenous phototrophic bacterium, Rhodovulum sulfidophilum, was incorporated in commercial fish feed for Oreochromis niloticus. The bacterial cell was analyzed for nutritional value and tested for toxicity and acceptability as an aquaculture feed supplement. The results showed higher survival rate and significantly higher growth rate (p<0.001) in O. niloticus fed with the bacteria incorporated fish feed. It is suggested that R sulfidophilum can be utilized as an aquaculture feed supplement.
This study was carried out to find deodorization effect of swine-slurry by addition of phototrophic bacteria(PTB). The pilot-scale reactors operation conditions was designed by the inoculum amounts of PTB and light-conditions. Treatment conditions was divided into 3 types; 106 MPN/ml$.$Dark(T-1), 108 MPN/ml$.$Dark(T-2), 108 MPN/ml$.$Natural light(T-3). The changes of the concentration of volatile fatty acids(VFAs), hydrogen sulfide(H2S), ammonia (NH3) and odor intensity were analyzed during the treatment period(35 days). From results of this study, the maximum intensity of odor in the headspace of the reactor T-1 was 4.82 and T-2, T-3 was 2.63, respectively. In swine-slurry of reactors used, it almost took 10 days until to be stabilized with solid and liquid phase. Intensity of odor in headspace was mainly derived from the liquid phase. The PTB inoculum method to swine-slurry was very effective in reduction of VFAs, H2S and Sulfate-reducing bacteria(SRB) concentration. Expecially, It was interested in reverse growth behaviour of SRB and PTB in these conditions.
We used phototrophic bacteria to remove low concentrated organic materials (CODCr), nitrogen and phosphorus. We applied $COD_{Cr}$ 37.3 mg/L, $NH_3-N$ 4.0 mg/L, and $PO_4^{3-}-P$ 1.0 mg/L (C:N:P=100:10:1) in the batch test, and the removal efficiencies were shown as follow: $COD_{Cr}$ 87.4%, $NH_3-N$ 46.3%, $PO_4^{3-}-P$ 79.7%. The aerobic process with mixed phototrophic bacteria, ceramic media, and media KSP01 showed the removal efficiencies of $COD_{Cr}$, $NH_3-N$, and $PO_4^{3-}-P$, each as 72.7% and 79.2%, respectively in the lab-scale reactor. The maximum $PO_4^{3-}-P$ removal efficiency reached 92.6% by adjusting pH. There were three conditions used to remove $NH_3-N$. The highest removal efficiency was 98.5% with 10.2 L/min of aeration in 1-2 reactors, and the result of applying river-water showed the high removal efficiency of $NH_3-N$ (82.8%). Therefore, this purification system may be useful to control nitrogen and phosphorus at low concentration in field.
Twenty nine strains of photosynthetic purple nonsulfur bacteria were isolated from Kyonggi area in Korea. The isolated strains were identified as Rhodopseudomonas blastica, Rhodocyclus gelatinosus, Rhodocyclus tenuis, and Rhodopseudomonas rutila. The enhanced nutrients removal system for wastewater using phototrophic purple non-sulfur bacteria was developed. Experiments were performed into two Phases and the results were compared: the synthetic wastewater was tested for the removal efficiency of nutrients and organics during Phase 1 and the real wastewater during Phase2. Results showed that 97∼99% of organics were removed during Phase 1 and 96∼99% during Phase 2. Nutrients (nitrogen and phosphorus) were also removed efficiently: 85∼91% removal of T-N and 78∼92% removal of T-P were achieved for Phase 1, and 76∼89% removal of T-N and 73∼88% removal of T-P for Phase 2.
In an effort to identify possible microbes for seeking bioagents for remediation of herbicide-contaminated soils, seven species of phototrophic nonsulfur bacteria (Rhodobacter capsulatus and sphaeroides, Rhodospirillum rubrum, Rhodopseudomonas acidophila, blastica and viridis, Rhodomicrobium vannielii) were grown in the presence of the herbicide, butachlor, and bacterial growth rates and nitrogen fixation were measured with different carbon sources. Under general conditions, all species showed 17-53% reductions in growth rate following butachlor treatment. Under nitrogen-fixing conditions, Rb. capsulatus and Rs. rubrum showed 1-4% increases in the growth rates and 2-10% increases in nitrogen-fixing abilities, while the other 5 species showed decreases of 17-47% and 17-85%, respectively. The finding that Rp. acidophila, Rp. blastica, Rp. viridis and Rm. vannielii showed stronger inhibitions of nitrogenase activity seems to indicate that species in genera Rhodobacter and Rhodospirillum are less influenced by butachlor than those in Rhodopseudomonas and Rhodomicrobium in terms of nitrogen-fixing ability. Overall, nitrogenase activity was closely correlated with both growth rate and glutamine synthetase activity (representing nitrogen metabolism). When the carbon sources were compared, pyruvate (three carbons) was best for all species in terms of growth rate and nitrogen fixation, with malate (four carbons) showing intermediate values and ribose(five carbons) showing the lowest; these trends did not change in response to butachlor treatment. We verified that each of the 7 species had a plasmid ($12.2{\sim}23.5\;Kb$). We found that all 7 species could use butachlor as a sole carbon source and 3 species were controlled by plasmid-born genes, but it is doubtful whether plasmid-born genes were responsible to nitrogen fixation.
Phototrophic bacteria T-20 investigated in the present report was isolated from Thailand oil, Cells of the bacterium were rod-shaped. measured $0.6{\sim}0.7{\mu}m$ wide, and $2.3{\mu}m$ long. A color of cell suspension was red, and the long wavelength absorption maxima of the bacteriochlorophyll measured in 870nm. Substrates utilized as carbon sources: citrate, aspirate, glutamate, and fructose: unique for this strain is the liquefaction of gelatine, Ubiquinone Q-8 was also found. Rhodocyclus gelatinosus, which was reported in Bergey's Manual of Systematics Bacteriology, was corresponded to T-20.
Thiocapsa roseopersicina NCIB 8347은 purple sulfur bacteria이며 광합성종속영양 조건에서는 nitrogenase 효소계가 유도되어 질소를 고정하며, 수소를 발생한다. 또한 광합성독립영양 조건에서는 hydrogenase 효소계가 유도되어 3~4개 종류의 특성이 다른 hydrogenase가 membrane에 결합되어 있거나, cytoplasma에 존재하며, 이 중의 일부는 산소농도와 온도의 상승에도 비교적 안정하다. 본 연구에서는 T. roseopersicina NCIB 8347이 광합성종속영양 조건에서 수소를 생산할 수 있는 제반 배양조건을 최적화하고, nitrogenase와 일부 hydrogenase역가를 측정하여 purple non-sulfur bacteria, Rhodobacter sphaeroides KD131의 nitrogenase와 비교하여 수소생산을 최적화하였다. 할로겐램프를 8-9 $Klux/m^2$로 조사할 때와 배양온도 $26{\sim}30^{\circ}C$, 배양시간 72시간에서 균체 성장과 수소생산이 가장 높았다. T. roseopersicina NCIB 8347는 광합성 독립영양, 종속영양 조건에서 모두 성장 할 수 있었다.
Chromatium sp., densely populated on the bacterial plate of Lake Kaiike throughout the seasons, possibly fix N2 and concurrently produce H2 N2 Fixation and H2 production by Chromatium sp. were performed under photoautotrophic growth condition, and of which rates were much higher and showed expontial growth phase. Bacterial plater samples from Lake Kaiike collected on July 27, 1994 were used to know the effect of the light or H2S on N2 fixation and H2 productin by the bacteria. At low light intensity (250 lux), low rates of N2 fixation and H2 production were detected after 18 hours. However, high rates of the production were observed under the condition of high light intensity (1000 lux). On the other hand, a very low rate of N2 fixation was observed without an addition of H2S, while the bacterial rapilly increased N2 fixation and H2 production after adding H2S and the highest rate was observed in case of adding 20mg H2S-S/$m\ell$ to the bacterial plates.

Cyanobacteria are the dominant phototrophic bacteria in water environments. Here, the diversity of cyanobacteria in seven Korean reservoir waters where different levels of algal blooms were observed during the summer of 2002, was examined by T-RFLP analysis. The number of T-RF bands in the HaIII T-RFLP profiles analyzed from those water samples ranged from 20 to 44. Of these, cyanobacteria accounted for 6.1 to 27.2% of the total bacteria. The water samples could be clustered into 2 groups according to the Dice coefficient of the T -RF profiles. The eutrophic Dunpo and oligotrophic Chungju reservoirs were selected, and several representative clones from both reservoir waters analyzed for the nucleotide sequences of their 16S rDNA. The major clones were found to belong to the Microcystis and Anabaena species in the waters from the Dunpo and Chungju reservoirs, respectively, which was in agreement with the T-RFLP result. That is, the Microcystis and Anabaena species were dominant in the eutrophic and polluted Dunpo and oligotrophic Chungju reservoir waters, respectively. These results indicated that there is a correlation between prevalence of cyanobacterial species and levels of pollution in reservoir waters.
