A Gram (-), rod-shaped bacterium in size of 1.6-2.8 $\times$ 0.4 μm was isolated from a eutrophic reservoir, which exhibited growth-inhibiting effect against a bluegreen alga (Anabaena cylindrica). This isolate showed positive reactions for catalase and oxidase, and optimal conditions of 35-40°C and pH 9.0. This isolate was designated AC-1 in this manuscript. In a mixed-culture of A. cylindrica and AC-1, their growth patterns were inversely correlated and the bluegreen algal vegetative cells completely disappeared within 24-36 hours. AC-1 showed similar lytic activity in natural water as in an artificial medium. The lytic activity of AC-1 was dependent on the photosynthetic activity of A. cylindrica. When observed under phase contrast microscope, the isolate lysed vegetative cells of A. cylindrica in scattered state in a liquid medium, whereas heterocysts have not been lysed.

Carotenoids produced by non-photosynthetic bacteria protect organisms against lethal photodynamic reactions and scavenge oxygenic radicals. However, the carotenoid produced by Gordonia alkanivorans SKF120101 is coupled to reducing power generation. SKF120101 selectively produces carotenoid under light conditions. The growth yield of SKF120101 cultivated under light conditions was higher than that under dark condition. In the cyclic voltammetry, both upper and lower voltammograms for neutral red (NR) immobilized in intact cells of SKF120101 were not shifted in the condition without external redox sources but were commonly shifted downward by glucose addition and light. Electric current generation in a biofuel cell system (BFCS) catalyzed by harvested cells of SKF120101 was higher under light than dark condition. The ratio of electricity generation to glucose consumption by SKF120101 cultivated in BFCS was higher under light than dark condition. The carotenoid produced by SKF120101 catalyzes production of reducing power from light energy, first evaluated by the electrochemical technique used in this research.
Two ferredoxin (Fd) fractions, namely, Fd-A and Fd-B were isolated from Heliobacillus mobilis cells, and purified by ammonium sulfate fractionation, DEAE, gel-permeation and Phenyl-Superose column chromatographies under anaerobic conditions. Their absorption spectra were typical of 2[4Fe-4S] cluster type Fds with peaks at about 385 and 280 nm and a shoulder at about 305 nm. Their N-terminal amino acid sequences were determined, which showed that both of them contain a [4Fe-4S] cluster binding motif. Fd-B was sensitive to oxygen, and itsA$_{385}$ value decreased by about 50% in 2 h at 4$^{\circ}C$ under aerobic conditions. In contrast, $A_{385}$ of Fd-A was essentially unchanged up to 24 h under the same conditions.
The effectiveness of resting cells of a photosynthetic bacterium, Rhodospirillum rubrum N-1, was investigated on the production of extracellular ${\delta}-aminolevulinic$ acid(ALA). The ALA generating system required 3hr-incubation in the presence of 10mg of resting cells per ml to obtain the maximal yield of extracellular ALA. and also, under this condition the effect of ALA inducers, i.e., 30mM levulinic acid (LA) and L-glutamic acid($C_5$ pathway precursor) was relatively higher than that of produced extracellular ALA($83{\mu}M$). The volume of system and proper cell density appeared to be important factors for the effective production of extracellular ALA.

Photosynthetic bacterium strain N-1 was isolated from the eutrophic wet soil and identified as Rhodospirillum rubrum. The optimum conditions for the cultivation of R. rubrum N-1 were estimated as 0.2% (w/v) of sodium acetate, 0.2% (w/v) of sodium propionate, 0.2% (w/v) of sodium butyrate in the Lasscelles basal medium at 30$circ$C, pH 7.0 under 4000 lux of the illumination. The removal efficiencies of total organic acid and chemical oxygen demand (COD$_{Cr}$) in swine wastewater were 80% and 87%, respectively, when 10% (v/v) of R. rubrum N-1 was inoculated.

To treat chromaticity contained in effluents of dyeing wastewater efficiently, potent dye-degrading microorganisms were isolated from influent water, aeration- tank sludge, recycle water and settling-tank sludge located in leather and dyeing treatment plant. Six potent strains were finally isolated and identified as Comamonas testosteroni, Methylobacteriaceae bacterium, Stenotrophomonas sp., Kluyveromyces fragilis, Ascomycetes sp. and Basidiomycetes sp. When Basidiomycetes sp. was inoculated into ME medium containing basal mixed-dyes, 93% of color was removed after 8 days incubation. In the same experiment, the 1:1 mixed culture of Basidiomycetes sp. and photosynthetic bacterium exhibited 88% of color removal; however, it showed better color removal for single-color dyes. The aeration-tank and settling-tank samples revealed higher color removal (95-96%) for black dyes. The settling-tank sample also revealed higher color removal on basal mixed-dyes, which resulted in 90% color removal after 6-h incubation. From the above results, it is expected to achieve a higher color removal using the mixed microorganisms that were isolated from aeration-tank and settling-tank samples.
To improve the hydrogen yield from biological fermentation of organic wastewater, a co-culture system of dark- and photo-fermentation bacteria was investigated. In a pure-culture system of the dark-fermentation bacterium Clostridium butyricum, a pH of 6.25 was found to be optimal, resulting in a hydrogen production rate of 18.7 ml-$H_2/l/h$. On the other hand, the photosynthetic bacterium Rhodobacter sphaeroides could produce the most hydrogen at 1.81mol-$H_2/mol$-glucose at pH 7.0. The maximum specific growth rate of R. sphaeroides was determined to be 2.93 $h^{-1}$ when acetic acid was used as the carbon source, a result that was significantly higher than that obtained using either glucose or a mixture of volatile fatty acids (VFAs). Acetic acid best supported R. sphaeroides cell growth but not hydrogen production. In the co-culture system with glucose, hydrogen could be steadily produced without any lag phase. There were distinguishable inflection points in a plot of accumulated hydrogen over time, resulting from the dynamic production or consumption of VFAs by the interaction between the dark- and photo-fermentation bacteria. Lastly, the hydrogen production rate of a repeated fed-batch run was 15.9 ml-$H_2/l/h$, which was achievable in a sustainable manner.

Effect of $(NH_4)_2SO_4$ precipitation and heat-treatment on hydrogenase which was extracted from the cytoplasmic fraction of the phototrophic purple sulfur bacterium Thiocapsa roseopersicina NCIB 8347 was studied. Crude enzyme extract was prepared by centrifugation($28,000{\times}g$, $400,000{\times}g$) after sonication of cells grown under photosynthetic condition for 96 hrs. Various conditions of $(NH_4)_2SO_4$ precipitation and heat-treatment were examined and the effect of protein concentration was analyzed by SDS-electrophoresis between the treatments. Optimum conditions for $(NH_4)_2SO_4$ precipitation and heat-treatment for evolution hydrogenase activity were 40-60% saturation and $60^{\circ}C$ for 20 min, respectively, which exhibited the specific hydrogenase activity of 0.78 U/mg-protein. Specific hydrogenase activity was decreased to 31.6% when the heat-treatment at $60^{\circ}C$ increased from 20 min to 5 hrs.
Photosynthetic bacteria, Rhodospirillum rubrum, have been reported to change their metabolic patterns depend-ing on the light condition. The genetic approach for such a metabolic change is one of main subject in pho-tosynthetic bacteria. It has been reported that the extrachromosomal plasmid might be related to this metabolic regulation. In this study, we have determined the partial sequences of R. rubrum plasmid pKYl with HindIII fragments and the predicted pKYl ORFs and physical map. We found the 8 putative proteins related to the genetic recombination of bacterium, which is reported to the alternative gene expression. Our results suggest that the genes located in pKYl are possibly involved in the metabolic switch according to the photocondition.
