Effects of Shading Treatments on Growth and Physiological Characteristics of Aruncus dioicus var. kamtschaticus (Maxim.) H. Hara Seedling (차광처리가 눈개승마 유묘의 생장 및 생리적 특성에 미치는 영향)

  • Background: This study was conducted to investigate the changes in the photosynthetic parameters, chlorophyll content, chlorophyll fluorescence, and growth characteristics of Aruncus dioicus var. kamtschaticus seedlings under different shading treatments. Methods and Results: The shading treatment was regulated with the shading level (non-shaded, 35%, 55%, and 75% shading). Photosynthetic activities, such as net photosynthetic rate, stomatal conductance, stomatal transpiration rate, and performance index on absorption basis ($PI_{ABS}$)were the highest under 35% shading ($4.36{\mu}mol\;CO_2{\cdot}m^{-2}{\cdot}s^{-1}$, $54.2mmol\;H_2O{\cdot}m^2{\cdot}s^{-1}$, $0.66mmol\;H_2O{\cdot}m^{-2}{\cdot}s^{-1}$, and 1.3, respectively), and the lowest under 75% shading. This implies that the decrease in net photosynthetic rate may be due to an inability to regulate water and $CO_2$ exchanged through the stomata. Thechlorophylla, b, and a + b contents were increased with elevating shading level and the chlorophyll a/b ratio showed non-significant differences. It was found that the dry weight (leaf, shoot, and whole) was the highest (1.14 g, 0.49 g, and 2.31 g, respectively) under 35% shading and the t/R ratio was the highest under 75% shading. Conclusions: It is concluded that 75% shading exhibited a strong reduction of photosynthetic activity, and 35% shading showed the best conditions for the early growth and cultivation of A. dioicus var. kamtschaticus.

Photosynthetic and respiratory responses of the surfgrass, Phyllospadix japonicus, to the rising water temperature (수온 상승에 따른 게바다말의 광합성 및 호흡률 변화)

  • Photosynthesis and respiration of seagrasses are mainly controlled by water temperature. In this study, the photosynthetic physiology and respiratory changes of the Asian surfgrass Phyllospadix japonicus, which is mainly distributed on the eastern and southern coasts of Korea, were investigated in response to changing water temperature (5, 10, 15, 20, 25, and 30℃) by conducting mesocosm experiments. Photosynthetic parameters (maximum photosynthetic rate, Pmax; compensation irradiance, Ic; and saturation irradiance, Ik) and respiration rate of surfgrass increased with rising water temperature, whereas photosynthetic efficiency (α) was fairly constant among the water temperature conditions. The Pmax and Ik dramatically decreased under the highest water temperature condition (30℃), whereas the Ic and respiration rate increased continuously with the increasing water temperature. Ratios of maximum photosynthetic rates to respiration rates (Pmax : R) were highest at 5℃ and declined markedly at higher temperatures with the lowest ratio at 30℃. The minimum requirement of Hsat (the daily period of irradiance-saturated photosynthesis) of P. japonicus was 2.5 hours at 5℃ and 10.6 hours at 30℃ for the positive carbon balance. Because longer Hsat was required for the positive carbon balance of P. japonicus under the increased water temperature, the rising water temperature should have negatively affected the growth, distribution, and survival of P. japonicus on the coast of Korea. Since the temperature in the temperate coastal waters is rising gradually due to global warming, the results of this study could provide insights into surfgrass responses to future severe sea warming and light attenuation.

  • 미래산업의 중요한 자원인 광합성세균(purple non-sulfur photosynthetic bacteria)은 해양에 대한 연구정도가 미비하기 때문에 이 연구를 수행하였다. 현재 총 47개의 mud 시료중에서 15개의 광합성세균으로 추정되는 strain을 분리하였다. 이후의 실험은 RAPD PCR을 이용하여 중복되는 strain의 유무를 확인하고, 165 rDNA의 염기분석을 통하여 분류학적인 위치를 연구할 계획이다.

Photosynthesis of Guard Cell Chloroplast

  • Chlorophasts are a central structural feature of stomatal guard cells. Guard cell chloroplasts have both photosystems I and II (PS I and II), carry out O2 evoluation , cyclic and noncyclic photophosporylation, and possess the Calvin-Benson cycle enzymes involved in CO2 fixation. These imply that guard cell chloroplasts have a normal photosynthetic carbon reduction pathway just like their mesophyll counterparts, indicating similar fuctional organization of thylakoid membranes in both types of mesophyll and guard cell chloroplasts. It has been, however, found that guard cell chloroplasts have distinctive and comparative properties in their photosynthetic performance. In this article, I review the intrinsic features on the light reaction of and carbon reduction by guard cell chloroplasts.

  • The soluble cytochrome c-551 of photosynthetic bacterium, Rhodopseudomonas gelatinosa ATCC 17013 was purified through a sequene of four step chromatography including CM-cellulose ion-exchange chromatography, DEAE-Sephacel chromatography, Sephacryl s-200 gel permeation chromatography, and HPLC (SP-5PW). The molecular weight of the purified cytochrome c-551 was 14, 600 Da, and this protein shows the absorption peak at 551 nm, 522 nm, and 417 nm as the reduced form, and at 412 nm as the oxidized form. The cytochrome c-551 seems to be a substrate for the terminal oxidase in the electron transport chain.

  • The effects of environmental factors on the extracellular release of organic carbon by Scenedesmus qudricauda were studied. The PER (percentage extracellular release) was greater at high temperature and at high concentration of nitrogen and phosphate. The PER variation according to the change of M/P ratio showed high values at each extreme N/P ratio. This result suggested that the limitation of nitrogen or phosphorous resulted in the accumulation of carbohydrates as photosynthetic products, and the products in high concentration were excreted through algal cell membrane.

Photosynthetic Performances of Temperate Sargassum and Kelp Species Growing in the Same Habitat

  • Characteristics of photosynthesis-light and photosynthesis-temperature relationships were seasonally compared among 4 species, two temperate Sargassurn (S. ringgoldianum and S. spathulophyllum) and two temperate kelp (Ecklonia cava and Eisenia bicyclis), growing in the same habitat in Oura Bay, Shimoda, central Japan. The photosynthesis-irradiance (P-I) curve of each species at the in situ temperature and the photosynthesis-temperature (P-T) curve at near saturation irradiance (400μmol·m^(-2)·s^(-1)) were determined by using differential gas-volumeters. Maximum photosynthetic rates (Prnax) for P-I curves of the two Sargassurn species were higher from summer to autumn than from winter to spring, while those of the two kelp species showed little difference among seasons. Net photosynthetic rates (Pn) at 100-400μmol·m^(-2)·s^(-1) of the Sargassurn species were higher than those of the kelp species in autumn, spring and summer, while in winter the rates were about the same between the Sargassurn and kelp species. Among seasons, the light saturation index (1k) values, dark respiration rates and light compensation points of Sargassurn species differed more than those of kelp species. Optimum temperature for P-T curves of Sargassurn species was 29℃ and that of kelp species was 27℃ in summer, while in winter the former was 27℃ and the latter was 25℃. Pn at 400 μmol·m^(-2)·s^(-1) at 10-33℃ of Sargassurn species were considerably higher than those of kelp species from spring to summer, while from autumn to winter the rates at 5-27℃ were about the same between the two. At supra-optimum temperatures, Pn of kelp species decreased more sharply than those of Sargassurn species in each season. These facts indicate that the two temperate Sargassurn species have a higher potential photosynthetic performance under warmer conditions than the two temperate kelp species even though they grow in the same habitat.

  • This study was performed to investigate the growth and photosynthetic rate of ten-years old Pinus koraiensis and Pinus rigida in Banwol industrial region comparing with those of the control site in Gwangju, Gyeonggi Province. Surface soil of the damaged site in pH 4.2 was more acidic than control site in pH 4.8, assuming the accelerated soil deterioration by acid deposition. The shoot growth, needle survival rate and net photosynthetic rate of P. koraiensis and P. rigida at the damaged site were lower than at the control site. In addition, the quantum yield and chlorophyll a+b and b contents reduced in damaged treatment. These results suggest that an inhibition of photochemical reaction of P. koraiensis and P. rigida at the damaged site of Banwol industrial region could be attributed to combined influence of atmospheric pollution and soil acidification.
