위암 환자에서 위 점막 인지질 분획의 지방산 조성 (Gastric Mucosal Fatty Acid Composition of Phospholipids in Gastric Cancer)

  • Although the age-standardized incidence of gastric cancer has decreased in Korea, it remains the second most common type of cancer. The purpose of this study was to analyze the phospholipid fatty acid compositions of gastric mucosa in gastric cancer. Cancerous mucosa and noncancerous mucosa adjacent to cancerous tissues were obtained from 29 patients who had undergone gastrectomy for gastric adenocarcinoma. Phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylinositol (PI), and phosphatidylserine (PS) were separated from phospholipids by thin-layer chromatography, and fatty acids were analyzed by gas chromatography. In cancerous mucosa, saturated fatty acids of total phospholipids and stearic acid of PE and PC contents as well as total phospholipids were significantly more abundant than in noncancerous tissues. The ratios of ω6 fatty acid products to linoleic acid of PC, PE, PI, and PS contents as well as total phospholipids were significantly higher in cancerous mucosa than in noncancerous mucosa. Arachidonic acid levels of PE and PI were significantly higher, but the PC level was lower in cancerous mucosa. These results suggest that the characteristic differences in fatty acid compositions of phospholipids and their subfractions shown in gastric cancerous mucosa may be affected by changes in lipid metabolism in gastric carcinogenesis. Further studies on structural and functional changes in phospholipids related to gastric carcinogenesis will be needed.

인체적혈구막(人體赤血球膜) 안정화(安定化)에 미치는 인지질(燐脂質)의 영향(影響) (The Stabilizing Effects of Phospholipids on the Human Erythrocyte Membranes)

  • Phospholipids were examined for their capacity to protect human erythrocytes against hemolysis induced by hypotonic solution, p-hydroxymercuribenzoate or hematin. The following results were obtained. 1. Phosphatidyl choline, lysophosphatidyl choline and phophatidyl ethanoleamine as well as chlorpromazine prevented the osmotic hemolysis of human erythrocytes which occurred due to water influx into erythrocytes from medium, but showed no effect on hematin-induced hemolysis which occurred without the volume change of erythrocytes. 2. Human erythrocytes were found to be most sensitive to the antihemolytic action of phospholipids among mammalian erythrocytes from sheep, rabbit, rat and mouse. 3. Phospholipids at the concentrations showing their strong antihemolytic effect on human erythrocytes against osmotic hemolysis had no influence on methylene blue uptake and volume change of erythrocytes in hypotonic solution. 4. Phospholipids increased erythrocyte deformability 2 to 3 times over control group and there was aclose relationship between their antihemolytic action and increase of deformability as a function of their concentrations. 5. The phospholipids increased the resistance to osmotic hemolysis of human erythrocytes by increasing membrane elasticity through their incorporation into lipid bilayer without altering glucose metabolism and water influx to erythrocytes.

Evaluation of antioxidant property of heat shock protein 90 from duck muscle

  • Objective: The objectives of this study were to investigate the direct antioxidative effect of 90 Kda heat shock protein (Hsp90) obtained from duck muscle. Methods: The interaction of Hsp90 with phospholipids and oxidized phospholipids was studied with surface plasmon resonance (SPR), and their further oxidation in the presence of Hsp90 was evaluated with thiobarbituric acid reactive substances (TBARS) assay. The scavenging effect on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis (3-ethylbenzthiazoline-6-sulfonic acid (ABTS) was measured, and the electron paramagnetic resonance (EPR) spectroscopy in combination with 5-tert-Butoxycarbonyl-5-methyl-1-pyrroline-N-oxide and 2-phenyl-4,4,5,5,-tetramethylimidazoline-1-oxyl-3-oxide (PTIO) was utilized to determine the abilities of Hsp90 in scavenging hydroxyl and PTIO radicals. Results: SPR showed Hsp90 could bind with both phospholipids and oxidized phospholipids, and prevent their further oxidation by the TBARS assay. The DPPH and ABTS scavenging activity increased with Hsp90 concentration, and could reach 27% and 20% respectively at the protein concentration of 50 μM. The EPR spectra demonstrated Hsp90 could directly scavenge ·OH and PTIO· radicals. Conclusion: This suggests that Hsp90, a natural antioxidant in meat, may play an important role in cellular defense against oxidative stress, and may have potential use in meat products.

  • The amount of total lipids nad phospholipids in the rat liver and brain after sodium nitrite treatment was measured together with the composition ratio of phospholipids. The results obtained were as follows : 1) The amount of total lipids and phospholipids was decreased significantly and this decrease was more outstanding in the rat brain liver. 2) The amount of phosphatidylcholine, phosphatidylethanolamine, and sphingomyelin was decreased, whereas that of phosphatidylinositol and diphosphatidylglycerol nitrite treatment of 120mg/kg/day concentration brings inhibition of lipid metabolism in the rat liver and brain.

Substrate Specificity of Cabbage Phospholipase D with Phospholipids Having Different Head Groups

  • A substrate specificity of cabbage phospholipase D (PLD) was studied using the synthetic phospholipids having different head groups. The phospholipids were synthesized from phosphatidylcholine and appropriate bases by transphosphatidylation of PLD. The bases used were ethanolamine, serine, ethanol and γ-hydroxybutyric acid. The phosphatidic acid, the product of PLD, was separated in TLC and measured densitometrically. The kinetic parameters were estimated for each substrate and the effects of pH, SDS, Ca2+ and other metal ions were examined. Vmax values found were 3.75, 2.36, 5.59, 1.63, 2.30 nmol/min/μg protein for phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, phosphatidylethanol, and phosphatidylburytic acid, respectively. These results indicate a broad specificity of cabbage PLD toward phospholipids with different head groups. Particularly phosphatidylserine was most easily hydrolyzed by PLD and its activity did not depend on Ca2+.

  • The simple binary mixtures of pure phospholipids such as DPPC:DSPC and DPPC:DPPA were investigated with differential scanning calorimeter. The thermotropic properties of DPPC and DSPC mixtures did not deviate much from the ideal curves that was obtained on varying either the temperature or the relative proportions of phospholipids. This means that the two phospholipids are completely miscible in the liquid and solid phase. But the thermotropic properties of DPPC and DPPA mixture deviated much from the ideal phase diagram. It resulted from the repulsion between the head groups of acidic phospholipids. Heat capacity suggested by Harden McConell was calculated. The larger $C_p$ was, the sharper the endothermic peak became.

Alteration of Phospholipids during the Mitophagic Process in Lung Cancer CellsS

  • Matrix assisted laser desorption ionization (MALDI)-time of flight/mass spectrometry (TOF/MS) was applied to investigate alterations in phospholipids in mitophagic cancer cells. Several phospholipids, including phosphatidylcholines (PCs), sphingomyelins (SMs), and phosphatidylinositols (PIs), were successfully analyzed in control and mitophagy-induced H460 cells in the positive and negative ion modes. Principal component analysis was applied to differentiate the two groups. The upregulated and downregulated phospholipid species in the mitophagic cells were also represented in a heatmap. In the volcano plot (fold change > 1.3 and p value < 0.01), individual species of seven PCs, two SMs, and three PIs were selected as differentially regulated phospholipids. In particular, almost all the molecular species of PC, SM, and PI were downregulated in the mitophagic cells. Quantification of these lipids indicated that mitophagy induces altered metabolism of phospholipids. Therefore, phospholipid alterations during the mitophagic process of lung cancer cells were well characterized by MALDI-TOF/MS.

북방 조개의 유지에 관한 연구 (Studies on the Lipids of 'Bugbangjohgae' Spisula sachalinensis)

  • 북방 조개는 우리나라 동해안과 일본 동북 지방에 널리 분포하는 백합과에 속하는 조개로서 일본에서는 식용으로도 이용되나 여기에 대한 지질학적 연구(脂質學的硏究)가 없으므로, 이 조개에 대한 지질학적 연구를 행하여 그 결과를 보고한다. 1) 총 지질중 인지질(燐脂質)이 $43.1\%$로 제일 많았고, 다음 트리그리 세라이드가 $36.2\%$ 스테롤이 $10.3\%$였다. 2) 인지질의 구성을 보면, 포스화터질코린, 포스화터질에나놀아민, 포스화티달에타놀아민, 포스화티질세린이 중요한 것이었다. 3) 중성지질의 지방관은 20 : 5, 16 : 1, 16 : 0이 제일 많았고, 에타놀아민 함유(含有) 인지질에는 20 : 5, 18 : 0, 22 : 6, 코린함유(含有) 인지질에는 16 : 0, 20 : 5, 22 : 6이 중요한 것이었다. 4) 프라스마로겐은 대부분 에타놀아민 함유 인지질에 편재해 있었고, 코란함유 인지질에는 소량 함유되어 있었다. 5) 중요한 스테롤은 22-트란스-24-놀코레스타-5.22-디엔-$3\beta$-을, 22-데하이드로콜레스테롤, 코레스테롤브라시카스테롤, 24-메치렌코레스테롤 및 $\beta$-시토스테롤로 구성되어 있었다.

초임계 $CO_2$와 에탄올 혼합물에 의한 대두인지질의 선택적 추출 (Selective Extraction of Phospholipids from Soybeans with Mixture of Supercritical Carbondioxide and Ethanol)

  • Supercritical carbondioxide is very effective in removing oils from a variety of seed matrices, devoid of any appreciable amount of phospholipid content. However, the limited solubility of phosphalipids in supercritical carbondioxide leaves behind a potentially valuable by-product in spent seed matrix. Any phospholipid extraction process from the spent matrix must maintain the structure and the quality of phospholipids and must be compatible with the end use of the seed protein meal an animal feed or for human consumption. An initial supercritical carbondioxide extraction of soybean flakes was performed at 32 MPa and $80^{\circ}C$ to extract the oils, leaving the phospholipids in the deflatted soybean flakes, A second step was performed on the defatted soybean flake using $X_{eth}$=0.10, Varying the pressure from 175 MPa to 70.6 MPa and temperature from $60^{\circ}C$ to $80^{\circ}C$. For all supercritical carbon dioxide/ethanol mixture extractions, a fraction rich in phospholipids was obtained. The fractions extracted from defatted soybean flakes were dried and them redissolved in chloroform before HPLC-ELSD analysis. Quantitative and qualitative analysis of phospholipids on soybean seeds, defatted soybean flake, and different extracted phospholipid fractions was carried out, to ascertain the effect of extraction pressure and temperature.
