참깨 종자의 발육단계별 중성지질의 변화 (Changes of Neutral Lipids in Developing Sesame(Sesamum indicium L.)Seeds)

  • These studies were undertaken to investigate changes of neutral lipid content and fatty acid composition were determined. Also accumulation process of monoglyceride, diglyceride and triglyceride content, and fatty acid composition were investigated during the development. The results were summarized as follows ; Changes of lipid during development sesame seeds, glycolipid contents which showed the highest in the early ripening stage and after that rapidly decreased, and phospholipid contents showed a similar pattern as glycolipid occurred. In contrast, the content of neutral lipid was rapidly increased by 29.21% 10 days after flowering(DAF), and showed the highest value by 91.84% at 40th day after flower. The neutral lipid, triglyceride content was rapidly increased as the seeds developed, and consisted of over 60% of the neutral lipid since 30 DAF. In the changes of neutral lipid, phospholipid and glycolipid, stearic acid and palmitic acid decreased during the seed ripening. However, oleic acid and linoleic acid increased during the same periods. Linolenic acid, which showed relatively higher value in the early ripening stage, but rapidly decreased as much as 1% at the later ripening stage.

지방산과 인지질(DOPC)의 혼합 LB막의 광이성질화 현상에 관한 연구 (Photoisomerization of Mixture LB Films of Fatty Acid and Phospholipid(DOPC))

  • We carried out this subject to observe photoisomerization using 1,2-dioleoyl-sn- glycero-3-phosphocholine(DOPC) mixed with fatty acid containing azobenzene group which has reversible to cis-trans by light irradiation. Spreading solutions for the LB films were prepared in chloroform($5.0{\times}10^{-5}$mol/L).We investigated the photoisomerization and property of the organic ultra thin film of fatty acid containing azobenzene was prepared on the hydrophilic ITO(idium tin oxide) glass plate by LB method. As a result, the absorption spectra of 8A5H and DOPC of mixture LB films was induced to photoisomerization by alternating irradiation of ultraviolet and visible light, because the condensation of pure azobenzene monolayers was loosened by the introduction of phospholipid into the monolayers, and the molecular high aggregation in pure azobenzene monolayers is also weakened by the introduction of phospholipid. We found that it was reversibly induced to cis-trans photoisomerization in several solvents and mixture LB films.

지방산과 인지질 혼합 유가초박막의 전기화학적 특성 (Electrochemical Properties of Organic Ultra Thin Films of Fatty Acid and Phospholipid Mixture)

  • We investigated the electrochemical properties for Langmuir-Blodgett (LB) films mixed with fatty acid (8A5H) and phospholipid (DLPE, DMPC, and DPPA). LB films of 8A5H monolayer and 8A5H-phospholipid mixture were deposited using the Langmuir-Blodgett method on the indium tin oxide(ITO) glass. The electrochemical properties measured using cyclic voltammetry with three-electrode system, an Ag/AgCl reference electrode, a platinum wire counter electrode and LB film-coated ITO working electrode at various concentrations(0.1, 0.5, and 1.0 mol/L) of $NaClO_4$ solution. A measuring range was reduced from initial potential to -1350 mV, continuously oxidized to 1650 mV and measured to the initial point. The scan rate was 50, 100, 150 and 200 mV/s, respectively. As a result, LB films of fatty acid and phospholipid (8A5H/DLPE and DPPA) appeared irreversible process were caused by only the reduction current from the cyclic voltammogram and LB film of 8A5H-DMPC mixture was found to be caused by a reversible oxidation-reduction process.

벼의 내냉성과 잎조직인지질의 지방산 조성과의 상관관계 (Relationship between Fatty Acid Composition of Phospholipid from Leaves and Cold Tolerance of Rice Plants)

  • $28^{\circ}C$에서 자란 $3{\sim}4$엽기 벼잎조직의 인지질지방산분석을 통해 벼품종별 상대적내냉성정도와 인지질지방산조성간에 성립되는 밀접한 상관관계를 확인하였다. 즉 내냉성이 강한 품종일수록 불포화도 및 불포화지방산의 구성비가 노은 인지질을 함유하고 있다. 인지질의 불포화상태를 나타내는 모든 지표들이 각품종의 냉해저항성과 잘 일치된다는 사실은, 곧 식물세포가 저온에서 생체막과 관련된 생리적활성을 유지하는데 있어서 필수조건으로 알려진 '생체막의 유동성'은 인지질지방산조성에 의해 절대적으로 지배받는다는 것을 분명히 나타낸다. 한편 벼의 인지질은 최소한 유묘기의 잎에서는 모든 품종에서 공히 phosphatidyl serine과 phosphatidyl choline을 주성분으로 phosphatidyl inositol을 부성분으로, 그밖에 3종의 미량성분을 포함하고 있으며, 인지질의 종류 및 조성은 내냉성과 무관한 것으로 판단된다.

개량(改良)메주의 숙성과정(熟成過程) 중 지질(脂質)조성의 변화(變化)에 관(關)한 연구(硏究) (Studies on Change of Lipids Improvement-Meju during the Fermentation)

  • Aspergillus oryzae에 의한 개량메주의 숙성과정에 있어서 지질의 조성변화를 체계적으로 규명하고저 silicic acid column chromatography에 의한 당지질, 중성지질, 인지질의 분획정량 및 gas liquid chromatography에 의해서 중성지질의 성분 변화를 실험한 결과는 다음과 같다. 1. 대두 및 삶은 콩의 총 지질을 분획한 결과 중성지질 93~94%로 대부분이었고 인지질 4~5%, 당지질 2.0~2.1%였다. 개량메주 발효과정 중에 있어서 중성지질의 함량은 차차 감소하였으며 상대적으로 당지질 및 인지질은 증가하였다. 2. 대두 및 삶은 콩의 비극성 지질은 triglyceride가 88-89%로 주요 구성 성분이었고 sterol ester, 유리지방산, diglyceride 및 sterol은 삶은 콩보다 대두에서 함량이 높았다. 또한 메주의 숙성과정에 있어서는 triglyceride의 함량은 현저히 감소하는 경향을 나타내었으며 sterol ester, 유리지방산 및 diglyceride는 차차 증가하였다. 3. 조지질, 중성지질, 당지질 및 안지질의 지방산조성에 있어서 삶은 콩 및 대두에 있어서 linoleic acid 54~70%, oleic acid 20.0~22.6%, palmitic acid 11.0~12.4% 순이었으며 myristic acid는 trace로 검출되었다. palmitic acid는 당지질과 인지질에서 조지질 및 중성지질 보다 그 함량이 조금 높았다. 4. 메주의발효과정에 있어서 조지질, 중성지질, 당지질 및 인지질의 지방산의 변화는 대체로 linoleic acid 및 linolenic acid가 차차 감소하였고 palmitic acid, stearic acid 및 oleic acid는 차차 증가하여 대체로 4 일째에 최대였다. 5. 당지질과 인지질의 지방산의 변화에 있어서 다같이 미확인 지방산 3 종류를 포함하여 9종류가 검출되었으며 2 종류의 미확인 지방산은 발효과정에서 생긴 것으로 앞으로 계속 검토할 흥미있는 과제라 하겠다.

어유 및 식물 종자유의 급이가 흰쥐가 간장, 뇌조직의 지방산 조성에 미치는 영향 (Effects of Fish Oil and Some Seed Oils on Fatty Acid Compositions of Liver and Brain Tissue in Rats)

  • S-D계 흰쥐에 중성지방 혈증 유발을 위해 12% 야자종실유와, 시험유지로서 올리브유, 돈지(豚脂), 어유, 들깨유, 옥수수유, 고추종자유 및 달맞이꽃종자유를 각각 3%씩 급여하여 4주간 사육한 후 간장 및 뇌지질의 지방산 조성을 분석 검토한 바, 간장 인지질의 지방산 조성은 SFA의 함량이 많고 MUFA는 적었으며, 중성지질에서는 MUFA의 함량이 SFA보다 약간 많은 편이었고 콜레스테롤 에스테르는 SFA 함량은 적고 MUFA는 월등히 많았다. 각 획분 모두 PUFA의 함량은 적었으며 PUFA중 $C_{18:2}(n-6)$의 함량이 가장 많았다. n-3계 PUFA는 어유 및 들깨유 급여군에 비교적 많은 반면 n-6계 PUFA는 옥수수유, 고추종자유 및 달맞이꽃종자유 급여군에 많았다. 뇌인지질의 지방산 조성에서 SFA의 함량은 적은 반면 PUFA의 함량이 현저히 많았으며, n-3계 PUFA로서는 $C_{20:5}와\;C_{22:6}$이 n-6계 PUFA로서는 $C_{18:2}$가 주요 지방산을 이루고 있었다.

  • For a studyon the production of ${\gamma}$-linolenic acid(GLA) by fungi 3 fungal strains were isolated from soil. Their cell growth lipid content and fatty acid composition were compared in shake flask culture. Among these fungi the fungus designated as FA-007 has high lipid content(21.1%) and GLA content(15.6% of total fatty acids) The fungal strain FA-007 was tentatively identified as Mucor sp. on the basis of morphological characteristics, Fungal oil produced by this fungus was composed of 75.2% neutral lipid 5.3% glycolipid and 19.5% phospholipid. Although the GLA content in phospholipid was higher than it in neutral lipid the GLA content in neurtal lipid was high as 15.5%.

The Measurement of Maxwell Displacement Current of Phospholipid Monolayers on the Water Surface

  • The organization of phospholipid monolayers and their monolayers mixed with fatty acid containing azobenzene on the water surface was investigated by means of the displacement current measurement method. The phase transition from the gaseous phase to the gaseous-fluid phase which accompanies the polar ordering of phospholipid molecules was detected in the range of immeasurably low surface pressure. The molecular area which gives the onset of the transition was determined for phospholipid monolayers. The Maxwell displacement current(MDC) pulses were generated across mixed monolayers due to the photoisomerization of fatty acid containing azobenzene by alternating irradiation of ultraviolet and visible light, because the condensation of pure azobenzene monolayers was loosened by the introduction of phospholipids into the monolayers. The displacement currents generated during light irradiation were also investgated in connection with monolayer compression cycles. It was found that the maximum of MDC appeared at the molecular area just before the initial rise of surface pressure in compression cycles.

우리나라 콩의 지방산 조성 및 단백질의 전기영동 패턴 (Fatty Acid Composition and Electrophoretic Patterns of Proteins of Korean Soybeans)

  • 우리나라 콩중 장려품종 1품종(봉의) 및 재래종 3품종(KW-12, KLS-77005-1 및 102-B)을 대상으로 일반성분, 지방질의 지방산조성 및 단백질의 전기영동 패턴을 비교하였다. 콩의 단백질 함량은 102-B가 33.7%로서 가장 낮았고, KLS-77005-1이 38.5로서 가장 높았으나 지방질함량은 반대로 전자가 24.2%로서 가장 높았고, 후자가 21.3로서 가장 낮았다. 콩의 지방질은 중성지방질, 당지방질 및 인지방질의 순서로 함량이 낮았으며, 102-B는 다른 콩에 비하여 중성지방질의 함량이 높았고 당지질의 함량이 낮았다. 중성지방질은 트리글리세리드가 주성분이었고 다음이 스테롤이었다. 총지방질의 지방산은 레놀레산이 가장 많았고, 다음이 올레산 및 팔미트산으로서 이들 세 지방산이 85%이상을 차지하였다. 중성지방질, 당지방질 및 인지방질의 지방산조성은 모두 리놀레산의 함량이 가장 높았다. 콩단백질은 전기영동상 7개의 분리대를 보였으며 KW-12는 IIS글로부린의 함량이 낮았고 102-B는 이의 함량이 높았다. 콩단백질을 구성하는 subunit의 조성은 시료간에 큰 차이를 보이지 않았다.

Determination of Saturated and Unsaturated Fatty Acids Amount in Leukocyte Membranes from Subjects Fed with Solid and Fluid Oils

  • Modifications in dietary fatty acid intake might lead to a modification in membrane phospholipid fatty acid composition. The purpose of this study was to investigate relationship between different type of oil consumption and leukocyte membrane phospholipid composition. This study was carried out in subjects utilizing butter (n = 15), margarine (n = 15), fluid oil (n = 15) and mixed types of oils (n = 15) in total 60 subjects. Leukocytes were separated from total blood by dextran sedimentation method. Membrane lipids and proteins were isolated following the cell disruption. Fatty acids of membrane phospholipids were isolated by hydrolysation with phospholipase B under ultrasonic dismembranator. Free fatty acids were identified with gas chromatography at chloroform phase. The results obtained were compared with data obtained by chromatograms of the standards. Results more prominent values of arachidic, dihomo-$\gamma$-linolenic and palmitoleic acids were found in butter-or mixed oil-user groups; eicosadienoic, eicosamonoenoic, dihomo-$\gamma$-linolenic and behenic acids in fluid oil; heptanoic, valeric, eicosadienoic and linolenic acids in margarine groups. The fatty acid composition of mixed oil was similar to butter, while other two oils were so different. From this study, it was concluded that the type of oil consumption might have an influence on phospholipid components of plasma membranes.
