Effects of Dietary Gum Phospholipid on Lipid Metabolism in Broiler Chicks

  • This experiment was to determine the usefulness of gum safflower phospholipid as a feed ingredient. Forty female broiler chicks were divided into four groups and fed experimental diets containing following fats and oils; beef tallow (Tallow), the blend of safflower oil and palm oil (SP-oil), gum rapeseed phospholipid (Rap-PL), or gum safflower phospholipid (Saf-PL) for 21days. There were no differences in growth performances among the treatments. Abdominal fat weight tended to be reduced in the chicks fed. phospholipids. The activity of hepatic acetyl-CoA carboxylase was significantly reduced in the Rap-PL and Saf-PL as compared to that of Tallow. Feeding dietary phospholipids resulted in a slight reduction in total fat and triglyceride contents in the breast and thigh muscles. In addition, total fat and triglyceride contents in the thigh muscle were significantly decreased by dietary Saf-PL as compared to those of Tallow. These results suggested that dietary gum phospholipids, either from rapeseed or safflower, had desirable effects of lowing abdominal and muscle fats, and could be used as a feed ingredient for broiler diets.

Effects of Phospholipid Extract from Squid Viscera on Lipid Oxidation of Fish Oil

  • Phospholipid(PL), phosphatidylcholine(PC) and phosphatidylcholine free PL(PCF) were extracted from squid viscera and the antioxidant effects of each fraction on the oxidation of refined fish oil were evaluated. Polyunsaturated fatty acid contents were the highest in PC(46.7%) followed by PL(44.8%) and PCF(40.9%). The effects of each phospholipid fraction on stabilizing fish oil were compared by incubating at 40$^{\circ}C$ for 10 days. At the initial period(2 days), changes in peroxide value did not show any significant difference ; however, as incubation time was extended, PC fraction showed the strongest antioxidant activity. PL and PCF added to fish oils also resulted in increased stability against oxidation. Antioxidative effect of PC at the 5% level was equivalent to 0.05% BHT, 1% catechin and 1% tocopherol.

Effects of the Phospholipid separated from Duck Egg Oil on the Rat Brain (청둥오리 난황유에서 분리한 인지질이 쥐 뇌에 미치는 영향)

  • This study was performed to investigate the biocogical activity of the phospholipid separated and purified from duck egg yolk as functional brain food. A quantitative restriction of maternal diet was given to Sprague-Dawely rats during lactation. Animals were separated into three groups, such as control, duck egg phospholipid(PL) or pig-brain phospholipid additions groups. And ad libitum during lactation was given to control groups. All of the offsprings at 3 weeks of age fed a diet ad libitum for 4 weeks of rehabilitation. Body weight and brain weight and protein, phosphplipid, and cholesterol were determined on offsprings of each of group sacrificed at 0, 1, 2, 3, 5 and 7 weeks of age, sacrified body weight of duck egg phospholipid and pig brain phospholipid group were significantly higher than the control group. Body weight and brain weight after a recovery period were significantly at the control group. However, protein, phospholipid and cholesterol contents were significantly high than the control group. Response latency in the passive avoidance task was the lowest in the ${\beta}$-amyloid group and highest in the phospholipid group. It is consequently suggested that phospholipid supplement may be effective in memory improvement.

Rapid Screening of Phospholipid Biomarker Candidates from Prostate Cancer Urine Samples by Multiple Reaction Monitoring of UPLC-ESI-MS/MS and Statistical Approaches

  • Ultrahigh performance liquid chromatography-electrospray ionization-tandem mass spectrometry (UPLC-ESI- MS/MS) provides a high-speed method to screen a large number of samples for small molecules with specific properties. In this study, UPLC-ESI-MS/MS with multiple reaction monitoring (MRM) was employed to screen urinary phospholipid (PL) content for biomarkers of prostate cancer. From lists of urinary PLs structurally identified using nanoflow LC-ESI-MS/MS, 52 PL species were selected for quantitative analysis in urine samples between 22 cancer-free urologic patients as controls and 45 prostate cancer patients. Statistical treatment of data by receiver operating characteristic (ROC) analysis yielded 14 PL species that differed significantly in relative concentrations (area under curve (AUC) > 0.8) between the two groups. Among PLs present at higher levels in prostate cancer urine, phosphatidylcholines (PCs) and phosphatidylinositols (PIs) constituted the major head group PLs (3 PCs and 7 PIs). For technical reasons, PL species of low abundance may be underrepresented in data from UPLC-ESI-MS/MS performed in MRM mode. However, the proposed method enables the rapid screening of large numbers of plasma or urine samples in the search for biomarkers of human disease.

Effect of Dietary Phospholipid on the Lipid Components of Serum and Organ Tissues in Rats (식이성 인지질이 흰쥐의 혈청 및 장기조직의 지질성분에 미치는 영향)

  • For the study of the effect of dietary phospholipid (PL) on the lipid components of serum and organ tissues in Sprague-Dawley rats, 56 Male-rats were divided into 8 groups, which was composed of 7. One group was fed with basal diet (normal group). And other experimental groups were fed ad libitum with the mixture of carbohydrate. casein salt mixture : vitamin mixture(60:18:4:1) and at the same time fed administratively with 1 gram of phospholipid-free soybean oil, corn oil and sesame oil, and phospholipid-containing soybean oil, corn oil and sesame oil respectively After 60 days the rats were fasted for 12 hours and then decapitated to collect blood and separate organ tissues . The lipid and protein components of serum and organ tissues were analyzed. The results of this study are summarized as follows The supplementation of dietary phospholipid decreases the food efficiency ratio and the growth rate of experimental rats, it increases the level of serum phospholipid and cholesterol ester, but decreases the value of total-cholesterol (T-chol.)/PL ; it decreases the value of albumin/globulin (hyG ratio) of serum protein and it increases the level of phosphatidyl ethanolamine(PE) in serum and organ tissues. And the correlation coefficients among the contents of T-chol., of HDL-chol. and of phospholipid in serum and liver are negative in general. Therefore 1 think that we must eat dietary phospholipid unpurified from vegetable oil to prevent development of atherosclerosis and fat liver.

  • Anti-phospholipid antihodies (aPL) have important roles in various pregnancy complications such as recurrent miscarrige, growth retardation, placental abruption and stillbirth. However, their biological actions on preimplantation development of oocytes are still unclear. In this study, we investigated whether either aPL containing sera or phospholipids could affect in vitro fertilization and development of mouse oocytes. Sera used in this study were collected from three patients and the presence of aPL in the sera was confirmed by enzymatic-linked immunosorbent assay. When mouse oocytes were cultured in a serum-free, Chatot, Ziomek and Bavister (CZB) medium (Experiment 1), addition of aPL-containing sera (10%) to CZB medium did not. significantly (P>0.05) influence sperm penetration of oocytes. However, development to the blastocyst stage was significantly (P<0.05) inhibited by serum addition, and formation of morulae (16-23% vs. 58%) and blastocysts (0-4% vs. 21%) was markedly reduced compared with no addition (Experiment 2). In Experiment 3, pronuclear stage embryos were cultured for 96 h in GZB medium supplemented with 1 $\mu$g /ml phosphatidyl ethanolamine, 1 $\mu$g/ml phosphatidyl inositol or 1 $\mu$g /ml phosphatidyl choline. No increase in embryo development was found after addition of the phospholipids to CZB medium. These results suggest that 1) aPL have an inhibitory role in preimplantation development of mouse embryos, and that 2) the action of aPL may be related to a specific phospholid (s) rather than the tested phospholipids in the present study.

Effects of Choline Deficient Diets on Serum and Liver Lipid Contents of Rats (콜린결핍식이(缺乏食餌)가 흰쥐의 혈청(血淸) 및 간장지질함량(肝臟脂質含量)에 미치는 영향(影響))

  • This experiment is carried out to study effect of choline-deficient diet on serum and liver lipid contents of male rats. The experimental animals use 36 male Sprague-Dawley rats weighing about $100{\pm}3g$. They are classified into 7 groups and fed to experimental diets which are composed of 0.8% choline-supplement of deficient diets in addition to 14% corn oil, 14% corn margarine and 14% lipids mixed with 4% corn oil and 10% corn margarine, respectively. After feeding for 4 weeks, I measure lipid concentration of serum and liver, and the result are as follows. 1. The choline-deficient diet group decreases slightly the rates of weight gain and feed efficiency as compared with those of the choline-supplement diet group, but increases liver weight. 2. The choline-dificient diet group decreases the serum concentrations of total cholesterol (TC), free cholesterol, HDL-choelsterol, VLDL, LDL-cholesterol and phospholipid (PL), but increases the contents of triglyceride and the ratios of cholesterol/HDL-cholesterol and triglyceride/PL, and indicates no remarkable-difference in the ratio of TC/PL. 3. As compared with the choline-supplement diet group, the choline-deficient diet group contains the higher liver contents of total lipids, free cholesterol and triglyceride, and gives little difference in the liver contents of total cholesterol and phospholipid(PL), and presents the higher ratios of VLDL, LDL-cholesterol/HDL-cholesterol and TG/PL in the liver.4. In the choline-deficient diet group, the coutents of serum and liver lipid is not influenced by the kind of dietary fat. On the other hand, the choline-supplemented diet group indicates a significantly lower content of phospholipid in the corn margarine-added diet group than in the corn oil-added diet group. As aforementioned results, I think that the choline-deficient diet induces fatty liver in male rats without relation to kind of fatty acid, and cholines-upplement diet with saturated fatty acid makes the more decrease of liver phospholipid than that with polyunsaturated fatty acid.

High Fat Diet or Exercise Training Alters Hepatic Total and Phospholipid fatty Acid Compositions in Rats (고지방식이 및 운동훈련이 흰쥐 간조직의 총지방산 및 인지질 지방산패턴에 미치는 영향)

  • Effects of high fat diet and/or endurance exercise training on hepatic total and phospholipid(PL) fatty acid compositions were evaluated in rats fed one of the following diets for 31 days. control diet(CD, 5 wt% corn oil) or high fat diet(HFD, 35 wt% corn oil). Half of the rats in each group were exercise-trained regularly on a treadmill for 90 minutes/day during the entire feeding period. Total and PL fatty acid compositions of hepatic lipid extracts were determined by a gas-liquid chromatograph),. Endurance exercise training did not change the daily food intake, but significantly reduced body weight gain and feed efficiency ratio of rats, which were most prominent in animals fed HFD. Exercise training did not significantly change the percentages of ∑saturated fatty acids (SFA) and ∑polyunsaturated fatty acids(PUEA), but decreased the percentage of ∑monounsaturated fatty acids(MUFA) in hepatic total fatty acids, which might be associated with the decrease in (equation omitted) 9-desaturation index of hepatic total fatty acid metabolism. Exercise training significantly lowered the percentages of 16 : 0 and 22 : 5$\omega$3, and increased the percentages of 20 : 1 and 20 : 3$\omega$3 in both total and PL fatty acid compositions in rat liver. Both total fatty acid and PL fatty acid compositions of rat liver responded more sensitively to changes in dietary fat content than to endurance exercise training in this study. Feeding HFD, whoch contains high level of linoleic acid(LA, 18 : 2$\omega$6), significantly decreased the percentages of ∑SFA and $\Sigma$MUFA, and increased the percentages of ∑PUFA and ∑$\omega$6 fatty acids of hepatic total fatty acids. Hepatic total fatty acid composition was affected by dietary fat content and dietary fatty acid composition more sensitively than those found in hepatic PL fatty acid composition. HFD significantly decreased most of desaturation indices, while exercise training significantly decreased elongation index(20 : 5$\omega$3⇒22 : 5$\omega$3) of hepatic total and PL fatty acid metabolism in rats. Antioxidant Activity of Different Lipid Extracts from Mackerel Viscera (고등어 내장으로부터 추출한 지질성분의 항산화효과)

  • To utilize the mackerel viscera effectively, antioxidant substances were extracted and the effects on the oxidation were evaluated. Total lipid(TL) extracted from mackerel viscera were fractionated into hexane extracts, tetracarbon chloride($CCl_4$)-methanol(1 : 1, 1 : 3, 1 : 9, v/v) extracts, phospholipid(PL), phosphatidylcholine(PC), and PC free fracitons(PCF). Inhibition of fish oil oxidation was investigated using these fractions. Hexane fraction showed lower antioxidant activity compared to the TL or $CCl_4-methanol$ fractions(C-M) ; in addition, antioxidant activities increased as polarity of C-M increased. Significant antioxidant activities were observed with PL, PC, and PCF ; among them, PC was the most effective. To determine the antioxidant activity of PC, effects of natural or synthetic antioxidant were compared. Addition of PC to fish oil proved to enhance antioxidant activity the antioxidant activity of phospholipids was derived mainly from PC.

Effect of Growth Temperature on the Composition of Leaf Lipids in Panax ginseng (인삼잎의 지질조성에 주는 재배온도의 영향)

  • Panu ginseng (6 Year old) was grown $17^{\circ}C$/$15^{\circ}C$ and $27^{\circ}C$ day/$23^{\circ}C$ night in the light room of phytotron for 84 days. The composition of neutral lipid(NL), glycolipid(GL), phospholipid(PL) and fatty acids were investigated in leaves. The contents of NL and GL were higher in $25^{\circ}C$ while PL was lower. Similarity (simple correlation) of lipid composition between $16^{\circ}C$ and $25^{\circ}C$ was not significant for PL and GL but significant for NL(p = 0.001), indicating that PL and GL were important factors in the mostability. Similarity of fatty acid composition between growth temperatures was highly significant (p = 0.001) for all three lipids, while similarity between lipids was significant between NL and PL (p=0.01) and NL and GL (p=0.05), but nonsignificant between GL and PL at $16^{\circ}C$. .In NL digalactosyldiacylglycerol (3->$7^{\circ}$) increased but monogalactosyldiacylglycerol (10%) did not change at $25^{\circ}C$. In PL phosphatidic acid (22 -> 4%) and phosphatidylinositol (18 -> 5%) decreased but phosphatidyl ethanolamine (12->l6%) increased at $25^{\circ}C$. Percent unsaturated acid slightly decreased in NL and PL but greatly increased in GL at $25^{\circ}C$. Percent unsaturated bond slightly decreased in NL but did not change in PL and GL.
