Effect of Nonlamellar-Prone Lipids on Protein Encapsulation in Liposomes

  • We investigated the effect of two nonlamellar-prone lipids, phosphatidylethanolamine (PE) and dioleoylglycerol (DOG), on the efficiency of protein encapsulation in liposomes. When the phosphatidylcholine (PC) matrix was replaced with PE or DOG during liposome formulation, the amounts of glutathione S-transferase and bovine serum albumin entrapped in the vesicles increased with increasing PE or DOG concentration. The presence of PE and DOG synergistically affected protein entrapment. These results suggest that protein encapsulation can be enhanced by the presence of nonlamellar lipids and/or lipid-induced membrane properties.

  • Chlorella ellipsoidea were cultured in "cold" media starvated with the nitrate and phsophate sources. The effects of the nitrate and phsophate starvation on the biosynthesis of phospholipid and the composition of fatty acids in chloroplasts isolated from these cells were analyzed. The syntheses of phosphatidylcholine and phosphatidylinositol in the nitrate and phosphate starvation were similarly inhibited as compared with the control but phsophatidylethanolamine synthesis in the nitrate starvation was extremely lower than that in the phosphate starvation. The major fatty acids utilized in phospholipid formation within chloroplasts were palmitic acid and linolenic aicd. However, palmitic acid and stearic acid were dominant in the condition of the nitrate starvation. The levels of palmitic acid were enhanced 3-fold than that of the control. These results suggest that the biosynthesis of phospholipid and the composition of fatty acids were affected by the nitrate and phosphate starvation in the culture media.ure media.

DPPC Liposome에 미치는 고들빼기 추출물의 DSC 연구 (The Effects of Godulbaegi Extracts on the Fluidity of Phospholipid Liposomes by DSC)

  • Liposomes have been widely employed as biomembrane-mimetic system and drug-delivery system. In these applications, the low stability of liposomes has been the most serious problem. They have relatively short half-lives and easily lysed through interactions with biological components. This study was performed to investigate the effects of godulbaegi extracts on the fludity of phospholipid liposomes. We used dipalmitoyl phosphatidylcholine(DPPC) liposomes which make most stable liposomes among the other phosphatidylcholines. The thermograms of the DPPC liposomal bilayers incorporated with the hexane extract of godulbaegi(Ixeris sonchifolia H.) were obtained, and the enthalpy changes and the sizes of cooperative unit of the transition were calculated. The incorporation of the Ixeris sonchifolia H. into the liposomal bilayers effectively reduced the transition temperature at which the transition from gel state to liquid-crystalline state occurs, broadened the thermogram peaks, and reduced the ratio of van't Hoff to calorimetric enthalpies. These results indicate indicate that the godulbaegi extracts (Ixeris sonchifolia H.) have significant effects on the fluidity of biological membrance.

Preparation of Chitosan Microcapsules by Using Microreactor

  • Chitosan microcapsules were prepared by microreactor method and conventional batch method using glutaraldehyde as a cross-linking agent and L-a-phosphatidylcholine as an emulsifier. The mean particle sizes of microcapsules prepared by microreactor and batch method were 1.10 and 1.96 mm, respectively. The former microcapsules showed narrower size distribution than those of the latter one.

당뇨쥐의 간 Microsome에서 Phopholipase A_{2} 활성과 지질과산화 (Phospholipase $A_{2}$ Activity and Lipid Peroxidation in Liver Microsome of Streptozotocin Induced Diabetic Rats)

  • The purpose of this study was to investigate phospholipase $A_{2}$ activity and lipid peroxidation I streptozotocin induced diabetic rats. Sprague-Dawley male rats weighting-Dawley male rats weighting 300$\pm$10gm were randomly assigned to normal and STZ-induced diabetic group. Diabetes was induced by intravenous injection of 55mg/kg of STZ in sodium citrate buffer(pH 4.3). Animals were sacrificed at the 6th day of diabetic states. Body weight gains were lower in DM group. Phosphatidylcholine hydrolysis in liver was not significantly different between two groups, whereas phosphatidylethanolamine hydrolysis in liver was increased by 69% in DM group comparing with that of normal group. Liver microsomal phospholipase $A_{2}$ activity and level of TBARS was increased by 91%, 109% in DM group compared with that of normal group, respectively. The present results indicate that phospholipase $A_{2}$ activity is specific to PE hydrolysis, leading to lipid peroxidation process in STZ induced diabetic rats.

Lipidomic analysis of plasma lipids composition changes in septic mice

  • A lipidomic study on extensive plasma lipids in bacterial peritonitis (cecal ligation and puncture, CLP)-induced sepsis in mice was done at 24 h post-CLP. The effects of administration of lysophosphatidylcholine (LPC) and lysophosphatidic acid (LPA), compounds known to have beneficial effects in CLP, on the sepsis-induced plasma lipid changes were also examined. Among the 147 plasma lipid species from 13 lipid subgroups (fatty acid [FA], LPA, LPC, lysophosphatidylethanolamine [LPE], phosphatidic acid [PA], phosphatidylcholine [PC], phosphatidylethanolamine [PE], phosphatidylinositol [PI], monoacylglyceride [MG], diacylglyceride [DG], triacylglyceride [TG], sphingomyelin [SM], and ceramide [Cer]) analyzed in this study, 40 and 70 species were increased, and decreased, respectively, in the CLP mice. Treatments with LPC and LPA affected 14 species from 7 subgroups, and 25 species from 9 subgroups, respectively. These results could contribute to finding the much needed reliable biomarkers of sepsis.

Egg Albumin이 알콜생산의 증진에 미치는 영향 및 기능 (Enhancing Effect of Egg Albumin on Ethanol Production and Its Function)

  • Saccharomyces sake를 이용한 알콜발효에서 배지의 첨가물로 egg albumin 및 phoshatidylcholine이 최종 알콜 농도에 미치는 영향과 그의 기작에 대햐여 분석하였다. Phosphatidycholine-egg albumin이 첨가될 때 최종 알콜 농도는 120g / l 정도 얻어졌으며, 첨가물이없는 경우에 비해 대략 30% 정도, 첨가물 linoleic acid-ergosterol에 비하여 20% 정도 증가되었다. 최종 알콜농도가 높을수록 균체의 알콜내성도 비례적으로 증가하였으며, 두 첨가물 사이의 차이는 지방산보다는 albumin과 ergosterol의 영향으로부터 주로 비롯되었다. 열반성 혹은 가수분해된 albumin을 첨가한 결과로 부터 albumin은 균체외부에 보호막을 형성하기보다는 영양분으로 균체내에 흡수되어 균체성질을 변화시켜 알콜내성을 증진시키며, 이로부터 궁극적으로 최종 알콜 농도를 증가시키는 것으로 판단되었다.

Partial Characterization of Soybean cDNA Encoding CTP: Phosphocholine Cytidylyltransferase

  • As the first step to elucidate the relationship between the structure and function of CTP:phosphocholine cytidylyltransferase (EC 2.7.7.15) in plants, the partial nucleotide sequence of soybean cytidylyltransferase cDNA was determined using a polymerase chain reaction (PCR). Degenerate oligonucleotide primers were synthesized from the conserved region revealed from the rat and yeast cytidylyltransferase DNA sequences. The catalytic domain region showed 78 and 76% homology with the rat and yeast amino acid sequences, respectivly. The hydropathy profile indicated that the C-terminal non-catalytic portion of the protein was very hydrophilic, and in the region between the catalytic domain and the C-terminal region, there was a large amphipathic $\alpha$-helical domain that was believed to bind the membrane surface in the active formation. There are 7 potential sites for phosphorylation by protein kinase C and 4 potential sites for phosphorylation by Ca2+/calmodulin kinase within the determined sequence.

  • We studied on the ultra thin DMPC by Langmuir-Blodgett(LB) method. The $\pi$ -A isotherm of the DMPC was measured at the air-water interface varying with the compressing speed and amounts of solutions for spreading. DMPC molecules have one phosphatidylcholine head group and two long alky groups with carbonyl group. Displacement currents generated during the compression of monolayers of DMPC on the surface of water were investigated. The molecular arrangement of deposited films were evaluated by measuring the absorption with the UVspectrometer. And we made structures of metal(Au)/DMPC/Metal(Au) and examined electron through DMPC LB films by means of current-voltage(I-V) measurement .

  • Monolayers of lipids on a water surface have attracted much interest as models of biological membranes, but also as precursors of multilayer systems promising many technical applications. Until now, many methodologies have been developed in order to gain a better underst. DMPC molecules have one phosphatidylcholine head group and two long alkyl groups with carbonyl group. Displacement currents generated during the compression of monolayers of DMPC on the surface of water were investigated. As results, the displacement pick was generated when the area per molecule was about 190$\AA$$^2$in low pressure, and it was generated when the area per molecule about 190$\AA$$^2$ in for pressure, and it was generated when the area per molecule about 150 $\AA$$^2$ in high pressure. Also. for the study of photo device measured the absorption rate. the maxim value shown was 2800-2900nm.
