Studies on Antitumor Components of Wild Pholiota squarrosa (Fr.) Quel. (야생 비늘 버섯의 항암 성분에 관한 연구)

  • In order to investigate the antitumor components of Korean wild higher fungi, the carpo-phores of Pholiota squarrosa collected in Kaung Nung area were extracted with water by refluxing for eight hours at 80-90.deg. C. The extracts were dialized for six days against distilled water and lyophilized to produce crude polysaccharide powders. They were found to have the antitumor activity against sarcoma 180 implanted in mice. Especially, the inhibition ratio of the extract of Pholiota squarrosa was 78.7% in the dose of 20mg/kg/day for the period of ten days. The tumor in two of the ten mice was completely regressed. The components of these aqueous extracts were found to be polysaccharide and protein. The total polysaccharide was 42% by Anthrone method and the protein was 55% by Lowry-Folin method in the antitumor component of Pholiota squarrosa.

  • Present experiments were conducted to determine the possibility of cultivation of 9 edible wild species selected among the higher fungi growing in Korea. In the investigation on the mycelial growth according t6o the different media, the mycelial growth of Coprinus comatus was fast on the CSA medium, when malt extract was added to the basal medium PSA, the mycelial of Lapista nuda and Auricularia auricula-judae was fast in growth and density. In the spawning, the mycelial growth of Pholiota squarrosa on the oak tree's sawdust, Pleurotus cornucopiae on the broad-leaves' sawdust, and Coprinus comatus on the compost was respectably fast and also it shown to be possibility of artificial cultivation owing to their carporphore budding when Coprinus comatus and Lepiota alborubescens cultivated on the rice straw, Auricularia auriculajudae and Pleurotus cornucopiae on the sawdust of the popla and Pholiota squarrosaon the sawdust of the oak tree.

  • To investigate antitumor components in Korean higher fungi, the carpophores of Pholiota squarrosa belonging to the family Strophariaceae were collected and extracted with hot water. A protein-bound polysaccharide fraction was obtained by adding ethanol to the extract and by dialyzing through Visking tube. The fraction was examined for antitumor activity against sarcoma 180 implanted in mice. It showed an inhibition ratio of 78.7% at the dose of 20mg/kg/day. The tumor in two of the ten mice was completely regressed. The chemical analysis of the antitumor fraction by Anthrone and Lowry-Folin methods showed that it consisted of 42% polysaccharide and 55% protein. The enzyme fraction of the carpophores showed no proteolytic activity on casein.

  • To elucidate the composition of phytosterols in the fruit body of Pholiota spp. 6 species(P. adiposa, P. aurivella, P. highlandensis, P. nameko, P. squarrosa and Pholiota sp.) were analyzed with Gas chromatography(GC). Pholiota spp. were contained campesterol, stigmasterol, and $\beta$-sitosterol as the major phytosterols.

  • To find anititumor metabolites in Korean basidiomycetes, the shake-cultured mycelia of eight of the higher fungi were extracted with hot water and the extracts, after being partially purified, were subjected to in vivo antitumor test. When administered i. p. at the dose of 30mg/kg/day for ten consecutive days into the female ICR mice, which had been implanted with $1{\times}10^{6}$ / cells of sarcoma 180 twenty four hours before the first injection, the extracts of Agaricus campestris, Lyophyllum decastes, Lyophyllum ulmarium, Armillaria Tabescence and Calvatia exipuliformis respectively showed inhibition ratios of 64.1%, 65.45, 60.-%, 53.0 and 49.3%. These five species were selected for further study, whereas the extracts of Phallus impudicus, Coprinus comatus and Pholiota squarrosa whih showed the inhibition ratios of 31.2%, 33.5% and 19.0% were discontinued.

Analysis of Cultural Characteristics and Phylogenic Relationships of Collected Strains of Pholiota species

  • Cultural characteristics and phylogenic relationships were investigated and classified among collected strains in Pholiota spp. which contain P. adiposa, P. squarrosa, P. nameko etc. They were tested on the four different media(PDA, MCM, YM, MEA) and sawdust(Alder, Oak, Pine, Popular) substrates. There was a little variation according to the media and sawdust substrates, although PDA and popular sawdust substrate seemed to be better. Most strains showed white colonies, but some strains were brown. Mycelial growth length differed according to the strains. To classify species, the internal transcribed spacer regions(ITS) of the ribosomal DNA(rDNA) repeats from Pholiota spp. were amplified using polymerase chain reaction(PCR) and then sequenced. According to the analysis of ITS sequences, they were classified into five clusters. Their spacer regions were $644{\sim}700$ nucleotides in length. The reciprocal homologies of each ITS region among these strains were ranged from $49.6{\sim}99.9%$. The phylogenic analysis might give a criterion to classify species in the collected strains.

  • 1) The study was made about the condition of mycelial growth of some edible wood rot fungi selecting broad leaved tree as host in nature, on conifer sawdust media. 2) The mycelial growth of other fungi except Pholiota squarrosa is inhibited on the media of conifer sawdust. 3) On the sawdust media added the liquid of bran, mycelial growth is ingreased well with little difference in both extractive and non extractive plots.

  • Twenty species of higher fungi growing in the wild were collected and studied extensively for their lectin activities by using erythrocytes of human, rabbit and mouse blood. In total, 14 species demonstrated hemagglutination with some kinds of erythrocytes. Of twenty species, eight (Boletus edulis, B. splendidus, Clavaria zollingeri, Lactaritis subzonarius, L. volemus, Russula cutefracta, Pholiota squarrosa, and P. aspera) were shown lymphoagglutination with murine splenic lymphocytes. Protein contents were estimated from the crude lectin fraction. Above mentioned eight species contained relatively high amounts of proteins than other mushrooms. Since these species had coincidently, hiig emagglutinating activity as well, we could define them as lectin-containeing mushrooms. Some species also contained mitogenic lectins toward murine splenic lymphocytes.
