무화과 잎의 용매분획 및 항미생물 활성 (Solvent Fractionation of Fig Leaves and its Antimicrobial Activity)

  • 무화과잎으로부터 천연 항미생물 물질의 개발을 검토할 목적으로 무화과잎을 메탄올로 추출하고 ethyl acetate와 buffer용액을 이용하여 용매분획하였고 각 분획에 대한 항미생물 활성을 측정하였으며 활성이 강한 분획에 대한 최소저해농도(MIC)와 최소치사농도(MLC)를 측정하였다. 무화과 잎중의 항미생물 활성은 산성 분획과 페놀성 분획에서 강하게 나타났으며 미생물의 종류에 관계없이 활성범위가 넓었다. 페놀성 분획이 산성 분획에 비하여 MIC와 MLC값이 상대적으로 낮았고 MIC값의 경우 두 분획이 공통적으로 세균보다는 진균류에 대하여 낮았으며 특히 저온세균인 Pseudomonas aeruginosa에 대한 MIC와 MLC값이 대단히 낮음을 알 수 있었다.

우엉 뿌리 추출물의 항산화 및 Tyrosinase 저해 활성과 Phenolic Compound 분석 (Antioxidative Activity and Tyrosinase Inhibitory Activity of the Extract and Fractions from Arctium lappa Roots and Analysis of Phenolic Compounds)

  • In this study, we investigated on antioxidative activities and tyrosinase inhibitory activities of methanol extract and its fractions from roots of Arctium lappa. The total phenolic compound and flavonoid content of the ethylacetate fraction was found to be 818.29 mg/g and 360.59 mg/g as the highest content. In the measurement of DPPH radical scavenging ability and tyrosinase inhibitory activity, the ethylacetate fraction was higher than the other fractions and the extract. In addition, comparative analysis of phenolic compounds by liquid chromatography-mass spectrometry (MS)/MS system under the multiple-reaction monitoring (MRM) with negative-ion electrospray ionization mode. The main phenolic compounds in the extract and fractions of roots from Arctium lappa were cynarin and chlorogenic acid. The main phenolic compound of the ethylacetate fraction was cynarin. n-Butanol fraction had a significantly higher chlorogenic acid content than other samples. In conclusion, DPPH radical scavenging ability and tyrosinase inhibitory activity of the cynarin-riched ethylacetate fraction showed the highest activity.

Thalictrum uchiyamai Nakai의 항균작용에 관한 연구 (Study on the Antimicrobial Activity of Thalictrum uchiyamai Nakai)

  • The methanol extract of Thalictrum uchiyamai Nakai was fractionatedi nto four parts; (1) ether soluble nonphenolic fraction (2) phenolic tertiary bases fraction, (3) chloroform soluble tertiary bases fraction and (4) aqueous quaternary bases fraction. The fractions of this plant showed a total of twenty-four spots on the silica gel TLC. Ether soluble nonphenolic fraction and ether soluble phenolic fraction showed nine alkaloid spots respectively, chloroform soluble fraction showed three alkaloid spots and quaternary bases fraction showed three alkaloid spots. The antimicrobial activity of the four extracts was shown as follows; nonphenolic tertiary bases inhibited the growth of Micrococcus dutica at the concentration of $50\mu$g/ml, in phenolic tertiary bases $500\mu$g/ml concentration was inhibitory to Micrococcus dutica and to Staphyloccus aureus. In aqueous quaternary bases, $500\mu$g/ml concentration was inhibitory to Klebsiella pneumonae. Chloroform soluble tertiary fraction showed slight activity. All of bases showed reproducible activity against tested microorganisms.

  • The free phenolic fraction from Korean white ginseng (Panax ginseng C.A. Meyer) was studied by GLC/MS as trimethylsilyl and methyl derivative. Five phenolic compounds such as 2,6-ditert butyl p-crestal, phloroglucinol, protocatechuic acid, isoferulic acid, quinic acid were identified newly. And additionally 13 organic acids and hydrocarbons were also identified in the fraction.

Assessment of Radical Scavenging Activity and Phenolic Compounds of Xanthium occidentale

  • Common thistle contains water-soluble substances that are antioxidative to foods. Antioxidant activities measured by DPPH method for the ground samples were the greatest in leaves, although was less than that of commonly used antioxidants, BHT and ascorbic acid. Methanol extracts and fractions from Xanthium occidentale plants dose-dependently increased DPPH free radical scavenging activity, in vitro test. The extracts from leaves showed the strongest antioxidant activity. DPPH scavenging activity of the individual fraction was in order of n-butanol>water>ethyl acetate>n-hexane fraction. By means of HPLC analysis, leaf samples of Xanthium occidentale had the highest amount of phenolic compounds, related with antioxidant activity, and followed by stems and roots. Total content of these antioxidant phenolic com­pounds for leaves extracts were detected in water fraction (36.7 mg 100 $g^{-1}$) as the greatest amount, especially chlorogenic acid (39.4 mg 100 $g^{-1}$) was the greatest component. These results suggest that Xanthium occidentale plants had potent antioxidant activity, and their activities were differently exhibited depending on plant part and fraction.

홍삼 페놀성 성분의 GC/MS분석 (Analysis of Phenolic Components in Korean Red Ginseng by GC/MS)

  • To Identify phenolic components known to exist in Korean ginseng (Panax ginseng C.A. Meyer) by GC/MS, three derivatization methods were employed for their analyses. First, phenolic components in ether soluble acidic fraction prepared from Korean red ginseng powder were taimethylsilylated. Secondly, phenolic acids in the same fraction were esterified with diazomethane followed by trlmethylsilylation. Thirdly, acidic components in ginseng powder were extracted and esterified concurrently by methanolic sulfuric acid, followed by fractionation of phenolic components with Silica Sep-Paka and trimethylsilylatlon. All phenolic components found in ginseng except gen tisic acid were identified by retention times and mass spectrums of standards. Besides, 5 phenolic components including salicyl alcohol and 1-H-indole-2-carboxylic acid were first identified from Korea an red ginseng by GC/MS.

재래식 메주 및 된장 중의 항산화성 물질에 관한 연구 4. 페놀화합물의 함량과 항산화력 (Antioxidative Materials in Domestic Meju and Doenjang 4. Separation of Phenolic Compounds and Their Antioxidative Activity)

  • In order to investigate the antioxidant activity of phenolic compounds contained in domestic Meju and Doenjang , the methanolic extract from defatted Meju and Doenjang was fractionated into the phenolic acid and the isoflavone fractions by alumina column and polyamide-6 column chromatography, respectively. Both phenolic acid and isoflavone fractions exhibited an identical antioxidative effect against the oxidation of linoleic acid. GC analysis revealed that the phenolic acid fractio contained vanillic , chlorogenic ,p-coumalic , ferulic, and caffeine acid. The content of caffic acid was greater than 70% in the phenolic acid fraction and the content of vanillic and chlorogenic acids were disappeared during fermentation, and thus, the facts are thought to be reasons for the low antioxidant activity of the phenolic aicd fraction from Doenjang. Similarly, the isoflavone fraction cotained daidzin, glycitin-65-0 glucosider and genistin and their aglycons such as daidzein, glyciten and genistein. The content of daidzin and genistin in Meju dramatically decreased at the early stage of fermentation ,whereas the content of daidzein an dgenistein rather increased ; however, these change in isoflavone contents did not affect the antioxidant activity of isoflavone fractions.

어성초의 화학성분 및 항미생물 활성 (Chemical Composition and Antimicrobial Activities of Houttuynia cordata Thunb.)

  • 어성초의 화학조성과 항미생물 활성을 검색하였다. 어성초의 화학조성을 분석한 결과 유리아미노산은 Asp, Glu, Gly, Arg, Ala, Pro, Val, Cys, Phe 등이 함유되었고 유리당은 fructose, glucose, sucrose 및 maltose가 함유되어 있었다. 지방산 조성은 주로 linolenic acid, linoleic acid, oleic acid, palmitic acid로 구성되었으며 무기성분중에서는 K 함량이 특히 높은 것으로 분석되었다. 어성초 추출물을 용매분획하여 각 fraction에 대한 항미생물 활성을 검색한 결과 basic fraction을 제외한 acidic, neutral, phenolic fraction에서 항미생물 활성이 인정되었으며 neutral fraction>acidic fraction>phenolic fraction 순으로 높은 활성을 나타냈다. 사용 미생물중 Pseudomonas aeruginosa, Micrococcus luteus, Bacillus subtilis에 특히 높은 활성을 보였으며 Aspergillus parasiticus, Aspergillus flavus에도 비교적 높은 활성을 나타냈다. 활성이 인정된 각 fraction에 대한 MIC값은 neutral fraction의 경우 Pseud. aeruginosa가 0.0075 g/mL 상당량으로 가장 낮았으며 그 밖의 균주에 대하여는 $0.025{\sim}0.75g/mL$상당량의 다양한 MIC 값을 나타냈다. Acidic fraction과 phenolic fraction은 $0.25{\sim}0.75g/mL$ 상당량으로 neutral fraction에 비해 높은 MIC 값을 나타냈다. MLC값은 neutral fraction의 경우 Pseud. aeruginosa가 0.1g/mL 상당량으로 가장 낮았고 그밖의 균주에 대하여는 $0.25{\sim}2.0g/mL$ 상당량의 MLC 값을 보였으며 acidic fraction과 phenolic fraction의 경우 사용 미생물에 대하여 3.0g/mL상당량까지 치사활성을 보이지 않았다.

밤 삽피의 페놀산 조성과 항산화 활성 (Phenolic Acid Composition and Antioxidative Activity of Chestnut Endoderm)

  • 밤 삽피의 천연 항산화제로서의 이용 가능성을 검토하기 위하여 밤 삽피로부터 유리형(EPA), 에스데르형(EPA), 결합형(BPA) 페놀산 획분을 분리하였다. 각 획분의 수율은 유리형 0.794%, 에스테르형 1.236%,결합형이 1.937%였으며 이중 페놀성 화합물의 함량은 에스테르형이 65.19%로 가장 높았다. Gas chromatography로 각 획분의 페놀산의 조성과 함량을 조사한 결과 유리형 획분에서 gallic acid(4.63%), ellagic acid(3.49%), salicylic acid(3.19%), gentisic acid(1.5%)가, 에스테르형 획분에서 gallic acid(13.4%), ellagic acid(11.6), protocatechuic acid(0.9%)가 주요 페놀산으로 확인 되었으며,결합형 획분에서는 gallic acid는 확인되지 않고 다른 획분에 비하여 sinapic acid(1.33%)가 높은 함량을 보였다. 이들 획분의 항산화성을 DPPH 라디칼 소거능, hydroxyl radical 소거능, 지질과산화물 생성 억제 시험으로 조사한 결과 유리형, 에스데르형, 결합형 획분의 순으로 항산화 활성이 우수한 것으로 조사되었다.

Phenolic Composition and Antioxidant Activities of Different Solvent Extracts from Pine Needles in Pinus Species

  • The purpose of this study was to investigate the phenolic acid, proanthocyanidin (PAs), and flavonol glycoside contents, as well as the antioxidant activities of pine needle extracts from six species of young pine trees. The extracts were prepared from Section Pinus (Diploxylon): P. densiflora, P. sylvestris, P. pinaster and P. pinea, and Section Strobus (Haploxylon): P. koraiensis and P. strobus. Phenolics were extracted from pine needles with 80% acetone to obtain the soluble free fraction, and insoluble residues were digested with 4 M NaOH to obtain bound ethyl acetate and bound water fractions. Phenolics were analyzed by HPLC, and the hydrophilic antioxidant activity was measured using oxygen radical absorbance capacity (ORAC). Total phenolic and flavonoid contents of the soluble free fraction were higher than those of the bound ethyl acetate and bound water fractions. The main phenolics were monomers and polymers of PAs in the soluble free fraction, and phenolic acids and flavonol glycosides in bound ethyl acetate fraction. Flavonol glycosides found in different species of pine needles were qualitatively similar within fractions, but composition varied among Pinus sections. High levels of kaempferol arabinoside and an unknown compound were present in all Strobus species. The soluble free fraction had the highest antioxidant activity, followed by bound ethyl acetate and bound water fractions.
