Streptozotocin으로 유도된 당뇨 흰쥐에서 하소에 사용되는 한약의 항당뇨 효과 비교 연구 (Anti-diabetic effect of medicinal plants used for lower wasting-thirst in streptozotocin-induced diabetic rats)

  • Objectives : To prove the channel-tropism theory in herbology, we investigated the anti-diabetic effect of six herbal plants used for lower wasting-thirst in streptozotocin-induced diabetic rats. Methods : Diabetes was induced in male Sprague-Dawley rats by consecutive injection of streptozotocin (30 mg/kg i.p.) for 5 days. The rats were divided into normal control, diabetic control, and diabetic treatment with Lycii Radicis Cortex (LRC, 300 mg/kg); Corni Fructus (CF, 300 mg/kg); Bombyx Batryticatus (BB, 50 mg/kg); Lycii Fructus (LF, 300 mg/kg); Phellodendri Cortex (PC, 300 mg/kg); Epimedii Herba (EH, 300 mg/kg); and glibenclimide (10 mg/kg) as a reference drug. Herbal extracts or reference drug were administered orally for 28 days. The changes of body weight, food intake and water intake, and serological markers such as blood glucose, serum total cholesterol, triglyceride (TG), blood urea nitrogen (BUN), and creatinine (Cr) were measured. Results : The decrease of body weight and the increase of food and water intake in STZ-induced diabetic rats was improved by the administration of CF and LF. Also, the enhancement of blood glucose and serum total cholesterol, TG, BUN and Cr in STZ-induced diabetic rats was significantly inhibited by the administration of CF, BB, LF and glibenclimide. On the other hand, EH strongly inhibited the increase of BUN and Cr in the sera of STZ-induced diabetic rats. Conclusions : These results suggest that among six herbal medicines used lower emaciation of emaciation-thirst disease, CF, BB, LF and EH show a characteristics including the channel-tropism theory.

Apply the drug to the affected part is used to be very busy on the angelicae adhuricae radix(白芷) of disperse the edema and drain the pus, and on the arisaematis rhizoma(南星), rhei radix et rhizoma(大黃), olibanum(乳香), phellodendri cortex(黃柏), calomelas(輕粉), glycyrrhizae radix(甘草), angelicae gigantis radix(當歸), myrrha(沒藥) of clearing away heat, activating blood circulation and relieve pain. 7. Apply the drug to the affected part is clearing away heat, activating blood circulation 8. Apply the drug to the affected part is cold and hot. 9. Apply the drug to the affected part is pungent, bitter and sweet. 10. Apply the drug to the affected part is non-toxic. 11. Apply the drug to the affected part is used to be very busy on the chanel of liver, heart, spleen, lung, stomach.

바이칼린 함유생약의 제제화 및 생체이용률 (제 1보): 황금 및 황련 공침물의 확인 및 물리화학적 성질 (Preparation and Bioavailability of Oriental Medicine Containing Baicalin (I) : Identification and Physicochemical Properties of Coprecipitated Product of Scutellariae Radix and Coptidis Rhizoma)

  • Precipitation was formed during the preparation of decoction from a mixture of Scutellariae Radix and Coptidis Rhizoma or Phellodendri Cortex according to the prescription of Hwang-ryean-hae-dog-tang. Baicalin and berberine were identified in coprecipitated product and these components were the active ingredients of two herbal medicine. The coprecipitated product was very slightly soluble in water and sparingly soluble in ethanol. The stoichiometric ratio of baicalin and berberine was found to be 1:1. The lipid-water partition coefficients of coprecipitated product were increased more than baicalin and berberine in chloroform, but were decreased in other organic solvents. The content of baicalin and berberine in coprecipitated product, determined by HPLC, were 23.08% and 26.75%, but the content of active ingredients in supernatant were 0.66% and 0.26%, respectively. The dissolution profile of baicalin of coprecipitated product was increased more than extract of Scutellariae Radix in artificial gastric juice, but was decreased in artificial intestinal juice. The dissolution rate of berberine of coprecipitated product was lower than extract of Coptidis Rhizoma in artificial gastric juice and intestinal juice commonly.

황련해독탕(黃連解毒湯)의 아토피 피부염 개선 효과(效果) (Ameliorative Effects of Hwangnyeonhaedok-tang on Atopic Dermatitis)

  • Objectives : The water extract of Hwangnyeonhaedok-tang (HHT), composed of the Scutellariae Radix, Coptidis Rhizoma, Phellodendri Cortex and Gardeniae Fructus has been traditionally used to treat fever, inflammation, gastritis and hypertension in east asia. However, little is known about the ameliorative effects of HHT on atopic dermatitis. Therefore, the purpose of this study was to investigate the therapeutic effect of HHT on atopic dermatitis Methods : We investigated the inhibitory effects of HHT on the production of proinflammatory cytokines in rat peritoneal mast cells (RPMCs), on the scratching behavior in ICR mice, and on atopic dermatitis symptoms in 2,4-dinitrofluorobenzene (DNFB)-induced atopic dermatitis-like model hairless mice. Results : Levels of TNF-${\alpha}$ and IL-$1{\beta}$ increased by PMA plus A23187 co-treatment were significantly inhibited by HHT in a dose-dependent manner. HHT also inhibited the histamine release from RPMCs stimulated by compound 48/80, which promotes histamine release. The oral administration of HHT reduced the scratching behavior induced by compound 48/80 and histamine in ICR mice. Furthermore, the intradermal treatment of HHT reduced the ear edema, skin lesions, and atopic molecular marker (IgE and IL-4) in DNFB-induced atopic dermatitis model mice. Conclusions : These results suggest that HHT may be used as a potential treatment for AD as a prescription for treatment of atopic dermatitis.

황연해독탕(黃連解毒湯)이 Hypoxia-reoxygenation에 의해 손상 받은 Mouse Neuroblastoma 2a Cells에 미치는 영향(影響) (The effects of Hwangryunhaedoktang and herbs on Mouse neuroblastoma 2a cells damaged by hypoxia-reoxygenation)

  • Objective : This study was designed to asses the effect of Hwangryunhaedoktang and herbs on Mouse neuroblastoma 2a cells damaged by hypoxia-reoxygenation. Method : Mouse neuroblastoma 2a (N2a) cells were measured by MTT assay and LDH assay after 48h hypoxia and 6h reoxygenation. Mouse neuroblastoma 2a (N2a) cells were treated by Hwangryunhaedoktang and herbs. Result : 1. Hwangryunhaedoktang was effective on LDH assay of hypoxia and reoxygenation. 2. All of herbs were generally effective on LDH assay of hypoxia and reoxygenation. In LDH assay of hypoxia, the effects of herbs depended on concentration. In MTT assay of hypoxia, Coptidis Rhizoma and Gardeniae Fructus were effective. In MTT assay of reoxygenation most of herbs were not effective. But Phellodendri Cortex was effective in high concentration. Conclusion : The results imply that Hwangryunhaedoktang and all herbs of it nay have protective effect on dementia and aging.

  • The methanol extracts of 340 kinds of Korean medicinal herbs were examined in vitro for the inhibitory effect on the acetylcholinesterase (AChE, E.C. 3.1.1.7) from electric eel using acetylthiocholine as a substrate. Among tested, the extracts of Coptidis Rhizoma, Phellodendri Cortex, Evodiae Fructus, Myrrha, Arecae Semen and Piperis nigri Fructus were found to exhibit a significant inhibition upon the acetylcholinesterase in a dose dependent manner, respectively.

Chemical Constituents from the Bark of Phellodendron amurense and Their Cytotoxic Effects on HL-60 Human Leukemia Cells

  • Phellodendri Cortex, phellodendron bark, has been used as a stomachic for intestinal function control and as an antimicro and anti-inflammatory agent. In this phytochemical study, eight compounds, berberine (1), palmatine (2), syringin (3), (+)-syringaresinol di-O-${\beta}$-D-glucopyranoside (4), salvadoraside (5), citrusin B (6), osmanthuside H (7), and kelampayoside A (8), were isolated from the bark of Phellodendron amurense. Their structures were elucidated by comparing spectroscopic data with reported values. Compounds 1 - 8 were evaluated for cytotoxic activity against HL-60 human promyelocytic leukemia cells in vitro. Among them, compounds 1 and 2 reduced the viability of HL-60 cells significantly, with $IC_{50}$ values of 26.0 and $18.5{\mu}M$, respectively.

  • The water extracts of 82 Korean medicinal herbs were examined for the binding affinity on the recombinant human muscarinic acetylcholine receptor subtype 1 $(mAChR-M_1)$ produced from the CHO (Chinese Hamster Ovary) cell line. Of those tested, the extracts of Coptidis Rhizoma, Phellodendri Cortex, Hedyotis Herba and of Terminariae Fructus were found to exhibit a significant competition with $[^3H]$ N-methyl-scopolamine for the specific binding to $mAChR-M_1$ in a dose dependent manner, respectively.

자시에 關한 文獻的 考察(原因과 治方을 爲主로) (A literature studies on the ja-shi(자시))

  • This study has been carried out to investigate pathogenic factor and medicine of ja-shi(자시) by referring to literatures. The results were obtained as follows. 1. In oriental medicine, Chashi(차시), Hamshichang(함시창), Hamshi(함시), Jabal(자발), Shichang(시창), Nojaon(로자온), Hamaon(하마온), Jokshiong(적시옹), Hamshidok(함사독), Tabshijong(탑사종), Shijong(시종) were the other names of ja-shi(자시). 2. The pathogenic factors of ja-shi(자시) are wind-heat, wetness-heat, Kihyeolungche(기혈응체), stomach-heat. wetness, wind wetness, and liking greasy and sweet food. 3. In the internal therapy of ja-shi(자시), Bojesodokum(普濟消毒飮), Sasunchunglyangum(四順淸凉飮), Takljsodoksan(托裏消毒散), Shihogalguntang(柴胡葛根湯), Sungsohwan(醒消丸), Gamisodokum(加味消毒飮) and etc. were used. And Radix glycyrrhizae(甘草), Fructus forsythiae(連翹), Radix angelicae gogantis(當歸), Radix scutellariae(황기), Radix bupleuri(柴胡), Radix p]atycodi(桔梗), Radix Ledebounellae(防風), Frustus arctii(牛蒡子), Rhizoma cimicifugae(升麻) and ete. were used as medicine. 4. In the external therapy of ja-shi(자시), Youigumhwangsan(如意金黃散), Okiogo(玉露膏), Igumsan(二金散), Gumhwangsan(金黃膏), Nungo(嫩膏) and etc. were used. And Radix angelicae dahuricae(白芷), Cortex phellodendri(黃栢), Rhizoma rhei(大黃), Indigo maturalis(靑黛), Radix glycyrrhizae(甘草) and etc. were used as medicine.

Obacunone 황백성분의 Candida albicans에 대한 항진균효과 (Antifungal Effect of Obacunone on Candida albicans)

  • In the present study, we determined the antifungal effect of obacunone isolated from Phellodendri Cortex against Candida ablicans, a pathogenic fungus. The antifungal effect was analyzed by an in-vitro susceptibility test and in a murine model of disseminated candidiasis. Possible mechanism of the antifungal activity was also examined. Analyses of data resulting from the susceptibility test revealed that the compound inhibited C. albicans growth. At 25 ${\mu}g$ obacunone/ml, there was app. 45% reduction of CFUs (colony forming units) as compared to obacunone-untreated C. albicans yeast cells (P<0.01). In the murine model of disseminated candidiasis due to C. albicans, obacunone enhanced resistance of mice against disseminated candidiasis. During an entire period of 30-day observation, control animals all died within 14 days, whereas 60% of obacunone-treated mice survived (P<0.05). In addition, obacunone inhibited the hyphal production, a major virulence factor of C. albicans, from the blastoconidial form. Thus, obacunone appears to have antifungal activity for C. albicans infection. This may possibly be mediated by the blockage of hyphal production.
