In Vitro Culture and Transformation by Agroinfiltration of Lisianthus (Eustoma russellianus) Pollen (Lisianthus 화분의 기내배양 및 Agroinfiltration에 의한 형질전환)

  • Optimized conditions for Agrobacterium-mediated lisianthus pollen transformation were adjusted using various factors such as temperature, pH and sucrose concentration. Pollen tube growth was successfully achieved in a medium (pollen germination medium; PGM) containing $7-15\%$ sucrose with pH in the range of 5.5-7.0 at temperature of $20-27^{\circ}C$. Lisianthus pollen was vacuum-infiltrated with Agrobacterium cell suspension for 20 min, and transformed pollen was confirmed by GUS histochemistry and Southern hybridization following RT-PCR. Transgenic pollen system may be utilized for establishing an area of plant transient expression systems based on the convenient pollen transformation procedure presented in here.

  • Horizontal starch gel electrophoresis for 29 populations (n=543) of the wrinkled frog, Rana rugosa, from Korea and Japan was peformed to assess the degree of genic variation and genetic diversity, and to understand the biogeographic pattern of distribution and speciation. A sum of 22 presumptive loci was screened from 17 enzymes and general proteins. Four loci, Aco, Est-3, Me-2, and Pgm, demonstrated high levels of polymorphism. The degree of average genetic variation of R. rugosa was P=22.7% (9.1-40.9%), Ho=0.086 (0.048-0.165) and He=0.090 (0.042-0.168). In the south-eastern region of the Korean peninsula (Chongsong, Yongchon, Ulsan, Kyongju, Pohang, yongdok and Ulchin), a few unique alleles in the Mpi locus were detected and their biogeographic implications were considered. The degree of genetic differentiation among the Korean populations was moderate (S=0.900), whereas the degree of genetic diversity between Korean and Japanese populations was notably high (S=0.687, D=0.293). This result corresponds with the data obtained by the mitochondrial cytochrome b gene sequence (Lee et al., 1999) suggesting that the Korean and Japanese R. rugosa might have evolved a specific level of genetic differentiation since their geographic isolation.

Enhanced Production of Bacterial Cellulose in Komagataeibacter xylinus Via Tuning of Biosynthesis Genes with Synthetic RBS

  • Bacterial cellulose (BC) has outstanding physical and chemical properties, including high crystallinity, moisture retention, and tensile strength. Currently, the major producer of BC is Komagataeibacter xylinus. However, due to limited tools of expression, this host is difficult to engineer metabolically to improve BC productivity. In this study, a regulated expression system for K. xylinus with synthetic ribosome binding site (RBS) was developed and used to engineer a BC biosynthesis pathway. A synthetic RBS library was constructed using green fluorescent protein (GFP) as a reporter, and three synthetic RBSs (R4, R15, and R6) with different strengths were successfully isolated by fluorescence-activated cell sorting (FACS). Using synthetic RBS, we optimized the expression of three homologous genes responsible for BC production, pgm, galU, and ndp, and thereby greatly increased it under both static and shaking culture conditions. The final titer of BC under static and shaking conditions was 5.28 and 3.67 g/l, respectively. Our findings demonstrate that reinforced metabolic flux towards BC through quantitative gene expression represents a practical strategy for the improvement of BC productivity.

Genetic Analysis of 5 Mountain Cultivated Ginseng and Wild Ginseng in Korea (국내 5개 지역의 장뇌삼과 산삼의 유전 분석)

  • ISSR PCR technique was applied to investigate genetic relationship among 5 Mountain cultivated ginseng populations (Jinan, Hongcheon, Punggi, Andong and Yeongju) and cDNA libraries of wild ginseng roots were constructed and analyzed functional genes related to morphogenesis via EST. Twenty four ISSR markers tested produced 127 polymorphic loci from 5 regional Mountain cultivated ginseng. Among the regional samples, Yeongju was made 18 polymorphic loci that were the highest level of variations among the cultivated regions. The range of similarity coefficient was 0.46~0.58 and the regional samples of Punggi and Hongcheon, Jinan and Andong were classified to similar groups respectively, whereas Yeongju was shown to be separate group with high level of genetic variation in UPGMA cluster analysis. As a result, there was no relationship according to geographical distance and genetic similarity. Eleven cDNA clones were consisted of 9 known genes and 2 unknown genes analyzed by BLAST program of NCBI. To recognize expression pattern of Homeodomain transcription factor related genes, Northern Blot analysis was performed for wild ginseng's leaf and root. As a result, the gene was only expressed by Mountain wild ginseng root.
