Studies on the resistance monitoring of green peach ahpid, Myzus persicae, its development pattern by artificial selection with imidacloprid and cross-resistance were carried out to develope resistance management strategy. Resistance ratios of M. persicae collected at Hwachon and Dunnae among 5 locations in alpine cultivation area appeared to be high as 37.2 and 16.5, respectively. Resistance of aphid to imidacloprid developed slowly up to 20 time selection, and after that it grew quickly. Imidacloprid-resistant aphid strain showed low cross-resistance ratios(<10) to most of organophosphates, carbamates, and mixed insecticides except pirimicarb(487.8), but high ratios to acetamiprid(143.0) which is one of the neonicotinoids like imidacloprid, and pyrethroids such as deltamethrin(14.9), flucythrinate(12.9) and halothrin(15.9).
The agricultural industry is beset by continuing demands to decrease the use of pest control agents which employ toxic modes of action. Although there are real problems of pesticide resistance, and sometimes overuse or redistribution in the environment, much criticism results from a lack of appreciation of how small is the risk involved. Whatever the background reasons, research and development for pesticide alternatives, particularly within Integrated Pest Management systems, is clearly of high priority. Currently available approaches, including use of natural products and molecular biology, are often regarded with naive optimism and require critical appraisal. For the future, methods of pest control based on chemicals with non-toxic modes of action (e.g. pheromones) continue to offer promise but, for widespread use, will require their integration with biological agents and development by means of plant molecular biology.
Resistance evolution to organophosphate-based pesticides in apple and pear inhabiting arthropods of western North America extends to many classes of pest and some beneficial species. Resistance management programs to minimize resistance in pests while exploiting it in natural enemies have met with mixed success. Among beneficials, resistances have been exploited mostly among predators of pest mites. Evolution of resistant mites, leafminers, leafhopper, aphids, leafrollers and some internal fruit feeders have led to development of new monitoring methods and means to delay or avoid resistance. But it is resistance to azinphosmethyl in codling moth (Cydia pomonella) that is changing the pest control system and moving it from chemical to biologically-based means. Newly merging IPM system will depend more on use of biological, cultural, behavior and genetic controls. But more selective pesticides also will be needed to augment pheromones, resistant host plants and genetically altered organisms. These more biologically-based tactics will be prone to resistance evolution in pests as well, if used too unilaterally and/or too extensively.

Six populations of the diamondback moth, Plutella xylostella, were collected from the different national areas for resistance and reared in laboratory for two sensitive population. These populations of P. xylostella were examined the developed resistance against commercial products of Bacillus thuringiensis. There were 3 products with B. thuringiensis subsp. kurstaki including Tyuneup$^{(R)}$, Thuricide$^{(R)}$ and Geumulmang$^{(R)}$ and 2 products with B. thuringiensis subsp. aizawai including Tobagi$^{(R)}$ and Scorpion$^{(R)}$. The sensitive population of diamondback moths were provided from National Academy of Agricultural Science (NP) and Highland Agriculture Research Center (GR population) and field populations were caught from 6 different national areas. Resistance against Tyuneup$^{(R)}$ was developed 4.8 and 2.5 times in SP and HS compared with GR population of diamondback moth, respectively. In case of Geumulmang$^{(R)}$, it was developed 9.9 and 6.8 times in SP and NM population compared with NP population, respectively. Otherwise, Tobagi$^{(R)}$ was showed higher resistance in HS than any other population compared with GR population, however, Scorpion$^{(R)}$ that is a same strain with Tobagi$^{(R)}$, was showed only double resistance to SP population. It was supposed that the development of resistance to B. thuringiensis might be caused by the continuous application of the specific commercial product at the specific area. So, we need to use the commercial products of B. thuringiensis in rotation with different B. thuringiensis strains. In the other hand, when HS population with highest resistance were reared in laboratory, their resistance ratio was rapidly dropped to 1.1 times at second generation. We have to examined the resistance mechanism of the diamondback moth to B. thuringiensis strains.
To evaluate mutagenicity of Phosphinotricin Acetyltransferase(PAT) which is expressed by the glufosinate-resistance gene pat, in vitro reverse mutation test using Salmonella typhimurium, chromosome aberration test using chinese hamster lung(CHL) cells and in vivo micronucleus test of mice were performed. In the reverse mutation, the PAT did not induce mutagenicity in Salmonella typhimurium TA 98, TA 100, TA 1535, TA 1537 with and without metabolic activation at $5000{\mu}g/plate$. In the chromosome aberration test, the results showed no incidence of increased structural and numerical chromosome aberrations at any doses tested(100, 10, $1{\mu}g/mL$). In micronucleus test, the ratio of micronuclei was measured in polychromatic erythrocytes of bone marrow of male ICR mice intraperitoneally administrated with PAT(1250, 625, and 313 mg/kg), the results showed no incidence of increased micronucleated polychromatic erythrocytes (MNPCE). These results indicate that PAT might not have mutagenic potential in vitro and vivo systems.
To obtain the basic information for degradation of remaining pesticide accumulated in cultivated soil of Honam area, the resistant bacterial strains were investigated in Chlorothalonil(TPN). Mancozeb, Bentazone, and Butachlor levels of 100, $500{\mu}g\;ml^{-1}$, and degradation of TPN by TPN-resistant bacteria in sterilized soil was studied under TPN levels 0, 10, 50 and $100{\mu}g\;g^{-1}$. A number of resistance strains were decreased with higher at concentration level of pesticide, and were higher in greenhouse than upland or paddy soil. The resistance of bacteria was strong in other of Bentazone> Butachlor> TPN> Mancozeb. The percentage of bacterial strains of resistance for pesticides isolated from the cultivated soil were the highest in Acinetobacter spp. and Corynebacterium spp., and the lowest in Moraxella spp. A number of TPN-resistant strains were the highest at the TPN level of $10{\mu}g\;g^{-1}$, and 5 days after strains inoculation, and were higher in Pseudomonas spp. TD-25 than TC-23 or strains in non-sterilized soil. The degradation of TPN was fast in order of strain TD-25>strain in non-sterilized soil >TC-23.
Fungicide resistance study in Korea is still in its infancy, and most of those resistance studies are largely limited to newness of the detected resistant strains. In future, detection of fungicide-resistant strains has to be based on sensitivity distribution of pathogen populations to certain fungicides, and standard levels of certain fungicides for resistance should be determined under the basis of this data. Most of the early research on fungicide resistance in Korea has overlooked this point, and resulted in inconsistency and confusion for monitoring sensitivity shift of pathogen population among individual researchers. Fungicide resistance detected in vitro tests has to be documented in field trials by examining control efficacy against resistant and wild-type pathogen populations. Resistance detection in wife has to be correlated with lower activity in practice. Using this process, fungicide resistance will have a practical meaning. Fitness evaluation of resistant strains for survival is, in particular, of importance to determine the future stability of the resistance in the pathogen population. In fields, sensitivity change of pathogen populations should be carefully monitored with and without fungicide selection pressures to establish long-term management strategies against fungicide resistance. It is becoming an urgent task to provide information through research for designing and implementing successful counter-measures against fungicide resistance problems in Korea.
Through the agar dilution method on V-8 juice agar, sensitivity of Phytophthora capsici causing pepper Phytophthora blight to metalaxyl was investigated by using isolates obtained from infected pepper plants during 3 years from 2005 to 2007. By the lapse of time, $EC_{50}$ value to metalaxyl was decreased, showing 1.45, 0.83, and $0.32{\mu}g\;mL^{-1}$ in 2005, 2006, and 2007. None of 2007 isolates was found to be resistant to metalaxyl. Compared the sensitivity of P. capsici isolates to metalaxyl with those to mandipropamid and dimethomorph, there is not a cross resistance response between metalaxyl and mandipropamid/dimethomorph. The resistance to metalaxyl in pepper Phytophthora blight pathogen was not related with the mycelial growth on V-8 agar medium and the pathogenicity on pepper plants.
Karyotype of apterous green peach aphid (Myzus persicae SuIBer) populations occurring in tobacco fields and their resistance to insecticides were investigated. All of the 16 clones collected from tobacco fields had same chromosome number of 2n=12. Among them, 5 clones had the normal karyotype, regardless of color morph, but chromosomal translnations between #1 and #3 autosomal chromosomes were noted in the other 11 clones. All of the aphid clones were highly resistant tolambda cyhalothrin, a pyrethroid pesticide, having over 20 times higher concentrations of LD90 than recommended concentration (10ppm) for the aphid. However, their resistance to acephate, demeton 5-methyl and pirimicarh varied depending on the clones, among which green-colored clones were generally more resistant to the pesticides than red-colored ones. No association was found between the insecticide resistance and the karyotype of the aphid in this experiment.
