Analytical Method Validation and Monitoring of Pesticide Residues in Animal Feeds (유통 사료 중 잔류농약 다성분동시분석법 확립 및 모니터링)

  • Safe animal feed is important for the health of animals, the environment and for the safety of foods of animal origin. Therefore, the aims of this study were: 1) to validate simultaneous analytical method and 2) to investigate the amount of pesticide residues in animal feed from Republic of Korea as a part of official control. A total of 126 samples were collected in 2015 and analyzed for 105 pesticides. According to pesticides monitoring result, no residue was found in 84.1% of the samples, whereas 15.9% of samples contained pesticide residues below the maximum residue limits (MRLs). Pirimiphos-methyl and cyproconazole were the two most frequently found pesticides. The results show that all commercial feed monitored in 2015 were safe under the Korean MRL and occurrence of pesticide residues in animal feed could not be considered serious threats to human and animal health. However, continuous monitoring with tighter regulation for pesticide residues in animal feed is recommended.

Mutagenecity evaluation of insecticidal 2-carbomethoxy-4-chlorodiethyl phosphate in short-term bioassays (살충성 물질 2-carbomethoxy-4-chlorodiethyl phosphate의 유전독성 평가)

  • For evaluating the mutagenic potential of 2-carbomethoxy-4-chlorodiethyl phosphate, three different short-term mutagenicity tests were used; Salmonella typhimurium preincubation assay with and without rat liver microsomal activation, chromosome aberration test in cultured chinese hamster lung fibroblast cell and in vivo micronucleus test in male mice bone marrow. In Salmonella typhimurium reverse mutation assay using TA98, TA100, TAl535 and TAl537, 2-carbomethoxy-4-chlorodiethyl phosphate did not show any mutagenic response in the presence and absence of S9 mix. It did not induce any significant structural chromosome aberrations in the absence of metabolic activation. In micronucleus test using ICR mice, the frequency of micronucleated polychromatic erythrocytes (MNPCE) increased in bone marrow cells treated with positive control, mitomycin-C, but 2-carbomethoxy-4-chlorodiethyl phosphate did not increase micronucleated polychromatic erythrocytes. These results indicate that 2-carbomethoxy-4-chlorodiethyl phosphate does not show any positive responses in short-term genotoxicity assays.

Effects of Pesticide (Fenitrothion) Application on Soil Organisms in Pine Stand (살충제(Fenitrothion) 살포가 소나무림의 토양생물에 미치는 영향)

  • As the pine wilt disease spread rapidly over Korea in recent, aerial pesticide spraying to the infected pine stands increased abruptly. The increasing aerial pesticide application for control of the disease would result in disturbance of soil ecosystem in pine stands. This study was conducted to assess the disturbance, using field experiment in pine stands of Hongleong experimental forests in Seoul with BACI experimental design with three sprayed plots and three unsprayed plots of $25m^2$. We sprayed fenitrothion of 0.0335 kg a.i./ha over each of the sprayed plots, being the same as the pesticide residues of litters after aerial spraying. The pesticide had been applied on 4 June, 11 June, and 29 June in 2002, comparable with the dates of the aerial spraying in the previous year in Busan. We monitored population of soil organisms including arthropods, fungi, bacteria, and actinomycetes at two sampling days before pesticide application, at two sampling days during the application season, and at three sampling days after last application for two months since late May in 2002. The pesticide applications did not make any significant effect on the population of arthropods, fungi, bacteria, and actinomycetes in soils and on the structure of soil organism community, showing low probability of significant effects of the aerial pesticide application on soil organisms in pine stands.

Insecticidal activity of Diamondback Moth, Plutella xylostella against Bacillus thuringiensis and Neem oil (Bacillus thuringiensis와 Neem oil에 대한 배추좀나방의 살충활성)

  • For the environmental friendly management of diamondback moth, Plutella xylostella (L.), Bacillus thuringiensis subsp. kurstaki ($30{\times}10^8\;cfu/mg$) and neem oil (0.5% azadirachtin) were used as green control agencies with mixed and alternative treatments on the chinese cabbage. When Bacillus thuringiensis subsp. kurstaki was applied to 1st and 2nd larva of P. xylostella with recommended concentration, their mortalities were reached to 100% by 2 days after treatment. In case of azadirachtin, its effect of mortality was continued for 7 days, and reached to 100% mortality. $LC_{50}$ values of Bacillus thuringiensis subsp. kurstaki and azadirachtin against 1st, 2nd, 3rd and 4th larva of P. xylostella were $2.8{\times}10^4$, $3.1{\times}10^4$, $3.4{\times}10^4$ and $1.5{\times}10^5\;cfu/ml$, and 2.7, 3.9, 4.7 and 7.1 ppm, respectively. The number of laid eggs of P. xylostella was reduced to 57.5 at 25 ppm of azadirachtin compared with control treatment. The hatch ratio was not significantly different with Bacillus thuringiensis subsp. kurstaki treatment in comparison with control treatment. However, when azadirachtin was applied, their hatch ratio were reduced to 25.8 and 45.4% at 25 and 50 ppm, respectively. On the other hand, emergence rate of eggs was not different with Bacillus thuringiensis subsp. kurstaki treatment, but 45.4% was shown in azadirachtin treatment with 50 ppm in comparison with control. When the mixture with Bacillus thuringiensis subsp. kurstaki and azadirachtin was applied to adults of P. xylostella, their mortality was higher than Bacillus thuringiensis subsp. kurstaki treatment only. These results are supposed that the mixture of Bacillus thuringiensis subsp. kurstaki and azadirachtin might be used as green control agents for reducing the demage of diamondback moth in the Chinse cabbage.

Antifungal Activity of Streptomyces griseofuscus 200401 against Pathogens causing Late Blight and Anthracnose on Pepper (역병균과 고추 탄저병에 대한 Streptomyces griseofuscus 200401의 항균활성)

  • To select microorganisms that exhibit antifungal activity against the fungal pathogens of pepper, Phytophthora capsici and Colletotrichum acutatum, soil samples from a forest and natural fields of Gajang-Dong, Sangju-city were tested in vitro and in vivo. Streptomyces griseofuscus 200401 was finally selected throughout antifungal activity test with dual culture, culture broth, and fruits. For the identification of the strain, nucleotide sequences of 16S rDNA and whole cell fatty acids were analyzed. It is like that the strain 200401 may be a novel biological control agent that can reduce application of chemical fungicides to control late blight and anthracnose on pepper.

Selection and Bioactivity of Tank Mix Combinations of Pesticides for Aerial Application (항공방제용 농약의 혼용가능 조합 선발 및 생물효과)

  • This study was conducted to select excellent tank mix combinations of pesticides for aerial application by manned helicopter. Among 209 pesticide combinations of 3-way tank-mixing for aerial application, a total of 93 recommendable combinations including tricyclazole SC+validamycin-A SL+imidacloprid SL were finally selected for the simultaneous control of key pests on paddy rice such as blast, sheath blight, brown planthopper and moth. The selected combinations were not phytotoxic to rice plants and nearby non-target crops, and excellent in physicochemical properties of ultra low volume (ULV) spray solutions. The efficacies on sheath blight, brown planthoppers and white-backed planthoppers of pesticides sprayed by aerial application were similar to those of pesticides by conventional spraying. Total cost of aerial application in paddy rice was very economical as one fourth level of that of conventional spraying.

Sensing of the Insecticide Carbofuran Residues by Surface Plasmon Resonance and Immunoassay (표면플라즈몬공명과 효소면역분석법을 이용한 살충제 카보후란 잔류물 검출)

  • The pesticide is raising public interest in the world, because it causes damage to an environmental pollution and the human health remaining agricultural products and an ecosystem, in spite of the advantages. Particularly, each country restricts the residual pesticide and induces observance about the safety and usage standard so that they can control the amount of pesticide used and defend the safety of agricultural products. The habitual practice for the analysis of the residual pesticide depends on GC (gas chromatography), HPLC (high performance liquid chromatography) and GC/MS (gas chromatography/mass spectroscopy), which triturate the fixed quantity of samples, abstract and purify as a suitable organic solvent. These methods have the highly efficient in aspects of sensitivity and accuracy. On the other hand, they need the high cost, time consuming, much effort, expensive equipment and the skillful management. Carbofuran is highly toxic by inhalation and ingestion and moderately toxic by dermal absorption. As with other carbamate compounds, it is metabolized in the liver and eventually excreted in the urine. The half-life of carbofuran on crops is about 4 days when applied to roots, and longer than 4 days if applied to the leaves. This research was conducted to develop immunoassay for detecting carbofuran residue quickly on the basis of surface plasmon resonance and to evaluate the measurement sensitivity. Gold chip used was CM5 spreaded dextran on the surface. An applied antibody to Immunoassay was GST (glutathione-s-transferase). The association and the dissociation time were 176 second and 215 second between GST and carbofuran. The total analysis time using surface plasmon resonance was 13 minutes including regeneration time, on the other hand HPLC and GC/MS was 2 hours usually. The minimum detection limit of a permissible amount for carbofuran in the country is 0.1 ppm. The immunoassay method using surface plasmon resonance was 0.002 ppm.

Diversity of Spider Communities in a Pesticide-treated Pine (Pinus densiflora) Forest

  • The diversity of spider communities was investigated in the pine forest of Pinus densiflora in Songri-san, where chemical pesticides had been applied to control pine gall midge (Thecodiplosis japonensis). Spider communities were surveyed in four areas: a pesticide-untreated natural forest (area A), a forest with vinyl-covered ground surface (area B), an aldicarb-treated forest (area C) and a forest treated with a systemic pesticide (phosphamidon) (area D). A total of 74 spider species from 17 families were collected from the four survey areas. There were 54 species from 15 families in area A, 27 species from 12 families in area B, 29 species from 9 families in area C and 34 species from 9 families in area D, respectively. The species diversity of spider communities was highest in pesticide-untreated area A, and much lower in the other three areas. The monthly species diversity of spider communities was highest in May and lowest in January. The similarity of the spider communities was highest in areas B and D. The monthly similarity of the spider communities was highest in November and February. The dominant species was Clubiona jucunda (12.71%, N=304 individuals). According to our results, the application of aldicarb and phosphamidon dramatically decreased spider diversity and the number of individuals in the forests. Thus, the application of these two pesticides to natural forests should be restricted, or alternative pesticides need to be developed. Our results also indicated that application of the pesticides should be avoided in May and June when high spider diversity is expected.

Development of a Biocontrol Agent Using Monacrosporium thaumasium to Control a Root Knot Nematode, Meloidogyne incognita (Monacrosporium thaumasium을 이용한 뿌리혹선충 (Meloidogyne incognita) 방제용 미생물제제의 개발)

  • The created microbial pesticide was used to examine its effects using the inside-pot test method. The selected microbial pesticide KBC3017 particle pesticide manufactured by using Monacrosporium thaumasium was used in the farm-house outdoor test to find the optimum consistency and its effects. The more amount used, the better effect it showed. However, the optimum consistency was 2% and the KBC3017 particle pesticide for which the diatomite and raw jade powder were used as an increaser, when used 2% level of the total amount of soil, showed 71% effect on nematode prevention. The root and the stem of crops were better compared to those without any pesticide used.

  • Agrobacterium vitis causes a crown gall disease in grapevine and that is one of the major hindrances for the wide cultivation and production of grapevine. We studied the possibility of biological control using selected biological control agent. One isolate from the infected soil, named as strain 27, was able to inhibit the biovar 1; A. tumefaciens C58 and Ach5, biovar 2; A. rhizogenes 13264, and biovar 3; A. vitis, in vitro and in vivo test. The putative biological control agent, A. radiobacter strain 27 was carrying the plasmid and the size of isolated plasmid was very similar to that of pAgK84 of A. radiobacter K84.
