Antioxidative Effect of Persimmon Leaves (감잎의 항산화 효과)

  • The production of malondiadehyde(MDA) was significantly decreased when the methoanol extract of persimmon leaves was added to the system. The methanol extract of persimmon leaves was fractionated by using various solvents such as hexane, chloroform, ethylacetate and butanol. Among the above fractions especially the chloroform fraction, ethylacetate fraction revealed the strong antioxidative activities. The hot water extract of the persimmon leaves was less effective than tannin which was extracted from persimmon leaves in antioxidative activity.

Change of Antioxidative Activity at Different Harvest Time and Improvement of Atopic Dermatitis Effects for Persimmon Leaf Extract (감잎(柿葉) 추출물의 채취시기에 따른 항산화 활성 변화 및 아토피성 피부염 개선효과)

  • Objectives : The purpose of this study was to investigate the anti-oxidative and anti-atopic dermatitis effects of persimmon leaf extract obtained from Cheongdo-gun, where more than 60% of Korean persimmon is produced. Methods : Anti-oxidative effects of the crude persimmon leaf extract harvested monthly between May and November were determined by in vitro assay using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide dismutase (SOD)-like reaction. Anti-atopic dermatitis effects of the crude persimmon leaf extract were determined by using collagenase type I inhibition assay and by quantitative assays including serum histamine, prostaglandin E metabolite and leukotriene $B_4$ levels in animal model of atopic dermatitis using Balb/c mice. Results : Persimmon leaf extract harvested in May had higher levels of total phenolic compounds (182.24 mg/g) and flavonoids (23.05 mg/g) than the ones of different month extract. Also, persimmon leaf extract harvested in May showed the most effective extract scavenging activities of DPPH free radical ($13.39{\pm}0.21\;{\mu}g/ml$) and superoxide anion radical ($40.52{\pm}2.32\;{\mu}g/ml$), leading to use the persimmon leaf extract harvested in May for the experiments hereafter. Persimmon leaf extract showed $326.71{\pm}4.6\;{\mu}g/ml$ of 50% inhibitory concentration ($IC_{50}$) for collagenase type I which is responsible for the degradation of extracellular matrix. In addition, persimmon leaf extract application group could decrease serum levels of histamine, prostaglandin E metabolite and leukotriene $B_4$ compared to the negative control in animal model of atopic dermatitis. Especially, persimmon leaf extract showed a significantly decreased serum leukotriene $B_4$ level relative to the levels of histamine and prostaglandin E metabolite. Conclusions : Persimmon leaf extract showed anti-oxidative and anti-atopic dermatitis effects in vitro and in vivo. These results suggest that persimmon leaf extract may have immunoregulatory function for alleviating atopic dermatitis by decreasing collagenase activity and mast cell activation.

  • Kojongsi persimmon (Diospyros kaki) is the major cultivar of astringent persimmon in southern of Korea. Kojongsi persimmon leaf has been traditionally used for acute and chronic diseases in Oriental countries. The purpose of this study was undertaken to investigate the antioxidative activities of the extract of Kojongsi persimmon leaf during growth. We investigated the antioxidant effects of the persimmon leaf extracts during growth on total polyphenol, total flavonoid, electronic donating ability (DPPH), nitrite (NO) scavenging and superoxide dismutase (SOD)-like activity. The next, we investigated the possible cell protective effects of the persimmon extract treatment against ultraviolet B (UVB)-induced injury in HaCaT keratinocytes. The contents of total polyphenol and flavonoid in leaf extract of Kojongsi persimmon were increased in time-dependent manner. In Jun, DPPH and NO radical scavenging and SOD-like activities in the leaf extract of Kojongsi persimmon was increased to the highest. However, the antioxidant activities in persimmon varieties were not any difference. The cell cytotoxicity by UVB irradation in HaCaT keratinocytes was significantly increased with the compared to the control group. However, the treatment of leaf extract of Kojongsi persimmon in HaCaT keratinocytes was shown to protective effect against UVB-induced cell cytotoxicity. These results suggest that the leaf extract of Kojongsi persimmon has potent antioxidant activity, and protective effect against UVB-induced keratinocyte injury. Thus, these properties may be contributed in the care of acute and chronic diseases.

Effects of Dried Leaf Powders, Water and Ethanol Extracts of Persimmon and Green Tea Leaves on Lipid Metabolism and Antioxidative Capacity in 12-Month-Old Rats (감잎, 녹차의 건분 및 물, 에탄올추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향)

  • The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level. Anticancer Effect of Persimmon Leaf Extracts on Korean Gastric Cancer Cell (감잎의 물 및 에탄올 추출물이 한국인 위암 세포주에 미치는 항암효과)

  • This study was performed to investigate the in vitro and in vivo anticancer effects of persimmon leaf extracts on human gastric cancer cells. In vitro anticancer effects of persimmon leaf extracts (water extract at 8$0^{\circ}C$ for 3 hours, water extract at room temperature for 48 hours, 50% ethanol extract at 8$0^{\circ}C$ for 3 hours, 50% ethanol extract at room temperature for 48 hours, 75% ethanol extract at 8$0^{\circ}C$ for 3 hours and 75% ethanol extract at room temperature for 48 hours) on SNU16 (Korean gastric cancer cell) were investigated by MTT assay. Persimmon leaf extracts exhibited strong in vitro anticancer effects. We found that the higher the ethanol content of the solvent, the stronger the in vitro anticancer effects. Extraction yields, contents of flavonoids, vitamin A, vitamin C and vitamin E were measured. We found that the higher the ethanol content of the solvent, the higher the extraction yields and the contents of flavonoids, vitamin A and vitamin E. Among persimmon leaf extracts, 75% ethanol 8$0^{\circ}C$ extract showed the highest extraction yield, the highest contents of flavonoids, vitamin A and vitamin E and exhibitied the strongest in vitro anticancer effect on SNU16. Therefore, 75% ethanol 8$0^{\circ}C$ extract was chosen as the material to investigate in vivo anticancer effects. In vivo anticancer effect of persimmon leaf 75% ethanol 8$0^{\circ}C$ extract was investigated in SNU16 transplanted nude mice. Twenty five female nude mice (BALB/c) were blocked into five groups according to body weight and raised for 4 weeks with diets containing 4% (w/w), 8% (w/w) persimmon leaf 75% ethanol 8$0^{\circ}C$ extract, with IT (intratumoral) injection treatment with 1.65 mg/100 $\mu$1, 3.3 mg/100 $\mu$1 concentration every other day 3 weeks after SNU16 was transplanted. Persimmon 75% ethanol 8$0^{\circ}C$ extract significantly lowered tumor weight and tumor volume in SNU16 transplanted nude mice. Tumor weight and tumor volume in all experimental groups were significantly lower than those in the control group. Helper T cell (CD4) levels of mice injected with 3.3 mg/100 $\mu$1 extract significantly increased. Cytotoxic T cell (CD8) levels in all experimental groups significantly increased and helper/cytotoxic T cell ratios in all experimental groups significantly decreased. Natural killer cell and MHC class II molecule in all experimental groups significantly increased. In conclusion, persimmon leaf 75% ethanol 8$0^{\circ}C$ extract exhibited strong in vitro and in vivo anticancer effects against SNU16 cells and it increased cytotoxic T cell, natural killer cell and MHC classII molecule in experimental groups in SNU16 transplanted nude mice.

Effect of Dried Leaf Powders and Ethanol Extracts of Persimmon, Green Tea and Pine Needle on Lipid Metabolism and Antioxidative Capacity in Rats (감잎, 녹차, 솔잎의 건분 및 에탄올추출물이 흰쥐의 지방대사와 항산화능에 미치는 영향)

  • The effects of dried leaf powders and ethanol extract of persimmon, green tea and pine needle on lipid metabolism, lipid peroxidation and antioxidative activity were investigated in rats. Forty-nine male Spargue-Dawley rats weighing 107.8$\pm$1.8g were blocked into seven groups according to body weight. Rats were raised for four weeks with diets containing either 5%(w/w) dried leaf powders of one of three different Korea traditional teas, persimmon(Diospyros kaki Thunb), green tea(Camellia Sinensis O.Ktzc)or pine needle(pinus Koreansis Sieb. Et Zucc), or ethanol from equal amounts of each dried tea powder. Food intake, weight gain, food efficiency ratio, and weights of liver, kidney and epididymal fat were significantly higher in the green-tea-powder group, and significantly lower in the pine-needle-powder and pine-needle-extract groups. Persimmon-leaf powder was found to decrease plasma total lipid, triglyceride and cholesterol concentration by increasing fecal total lipid triglyceride and cholesterol excretions. Liver cholesterol concentration was significantly lower in the green-tea and pine-needle-extract groups. Red-blood-cell superoxide dismutase(SOD) and glutathione peroxidase(GSH-px) activities were significantly increased in rats fed green-tea extract. Liver SOD activity was increased in rats fed pine-needle powder or extract, and liver GSH-px activity was increased in rats fed greentea powder. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentration were both decreased in rats fed dried leaf posers or extracts of persimmon or green tea. It is believed that high vitamin E levels in persimmon leaf, and high flavonoid, beta-carotene and vitamin C levels in green tea effectively inhibited lipid peroxidation. In conclusion, persimmon and green tea leaves were effective in lowering lipid levels and inhibiting lipid peroxidation in animal tissue, while pine needles were effective and lowering body weight gain. From these results, persimmon and green tea leaves can be recommended in the treatment and prevention of chronic discorders such as cardiovascular disease, cancer and aging. As ethanol extracts from these teas were also effecitive in lowering tissue lipid levels and inhibiting lipid peroxidation, we recommend the use of discarded tea grounds for this.

Ingredients of Antioxidant Activity from Calyx of Diospyros kaki Thunberg (감꼭지의 항산화 활성 성분)

  • In this study, in order to search for new functional materials from natural products, was carried out the study of antioxidant active ingredients in persimmon calyx(calyx of Diospyros kaki Thunberg). I have experimented with the effect of antioxidant activity of five different extract(MeOH, n-hexane, EtOAc, n-BuOH and $H_2O$ extract) obtained from persimmon calyx. As a result, the butanol extract, that is the main component fraction of antioxidant activity was found. Three compounds were isolated by silica gel column chromatography from the n-BuOH extract of persimmon calyx. Their structures of compound 1, 2 and 3 isolated from n-BuOH extract of persimmon calyx were identified as quercetin, (+)-catechin and gallic acid by using the TLC, $^1H$-NMR and $^{13}C$-NMR.

The Characteristics of Mixed Dyeing Using Persimmons Juice and Onion Outer Skin Extract (감즙과 양파껍질 추출액을 이용한 혼합염색의 특징)

  • Natural dyes are environmentally and human compatible. But they are not various or not fast in color. The mixed dyeing have been attempted to solve these disadventages of natural dyes. The persimmon juice dyed fabrics have brown-color and good hygienic properities however low color fastness. The onion dyeing show similar brown-color and have good color fastness caused by querectin existed in onion outer skins. Mixed dyeing was carryied out on silk fabrics using persimmion juice and onion outer skin extract in this study. The mixing method were persimmon juice dyeing and then onion dyeing(P-O), onion dyeing and then persimmon juice dyeing(O-P) and dyeing in the mixture of persimmon juice and onion outer skin extract simultaneously(P+O). The mordants were none-mordent, gallic acid after-treatment and alum after-treatment. Several persimmon juice dyed fabrics were irradiated for 2 hours before onion dyeing(PU-O). The color values of dyed silk fabrics were as follows. The persimmon juice dyed silk fabric developed to yellow-red color after 2 hours of uv irradiation. Onion dyed fabrics show similar yellow-red color after dyeing without uv irradiation. The effect of alum after-treatment on color difference were highest in onion dyeing. The dyeabilities of both P-O and O-P were higher than persimmom juice dyeing and onion dyeing. The dyeabilities of P+O was lower than persimmom juice dyeing and onion dyeing. The value of color difference of alum-treated fabric was the highest. The color difference of P-O and O-P caused from 2 to 4 hours of uv-irradiation were lower than those of persimmon juice dyed fabrics. Onion skin extract could prevent the color-change of persimmon juice dyed fabrics in mixed dyeing. The color difference of PU-O was higher than the P-O.

Sensory and Texture Properties of Gamdanja (감단자의 관능적 및 텍스쳐 특성)

  • The objective of this study was to seek the best recipe for making Gamdanja. Gamdanja is a kind of rice cake made from persimmon extract and winy flour, with a little salt mixed into the waxy flour. The method involved boiling the persimmon extract and beating in the waxy rice powder until all the waxy flour had been added. The procedure was as follows: Gamdanja containing different ratios of ingredients, such as persimmon extract (23, 23.5 and 24 hrs) and waxy rice flour (160 and 180g). The most desirable recipe was determined after sensory examination and mechanical tests using texture, moisture content and colorimeter measurements. The results of the sensory evaluation showed that Gamdanja made using persimmon extracted for 23.5 hrs, with the additional of 160g of winy flour gave the higher scores for QDA profile, chewiness, delicacy and after swallowing preference than the other ratios. The results of the acceptance test also showed that the Gamdanja made using the persimmon extracted for 23.5 hrs with 160g of waxy rice flour was the best mix in every respect, such as for color and overall acceptability. In the textural analysis of Gamdanja, the hardness, adhesiveness, gumminess and chewiness were increased by the additiong of furtherway lour. The moisture content of Gamdnja with persimmon extracted for 23 hrs was higher an those extracted for 23.5 and 24 hrs. The L-, a- and values of Gamdanja were increased with decreasing waxy flour. The sensory and mechanical examinations showed the overall quality of Gamdanja had positive correlations with delicacy and color, but negative correlations with hardness, adhesiveness, gumminess, and chewiness. As a result of these tests, the most desirable recipe for the Gamdanja was persimmon extracted for 23.5 hrs with the additional of 160g of waxy rice flour, containing $1\%$ salt and a moisture content of $45.87\%$.

Effect of Color Developing by Alkali and Heating of Cotton Fabrics Dyed with Persimmon Extract (알칼리와 열처리에 의한 면직물의 감즙염색 발색효과)

  • The purpose of this study is to improve the method of color developing with alkali solution as a promotor of color developing for feasible use. Cotton fabric was dyed with persimmon extract ranged with 0~3% alkali component with 5 types of strong to mild alkali solution. Heat treatment for color developing was applied to fabric dyed with persimmon extract and alkali mixing solution. Tests were carried out to analyze the change of surface color, ${\Delta},Ea^*b^*$, and water repellent of the dyed cotton fabric. The alkali mixing sample showed higher ${\Delta},Ea^*b^*$ value than control one without alkali mixing on the base of dyed fabric due to high color developing by alkali in the initial step of dyeing process. As alkali concentration increased, deeper dark color appeared on the fabric. The fabric color was changed to more dark in the application of sodium hydroxide, sodium carbonate, potassium carbonate in the initial step of dyeing process but color was not changed by increased heating time. However, the fabric showed a slight dark color with sodium acetate and more color change than that of the fabric dyed with persimmon extract without alkali. Therefore, sodium acetate seemed to a suitable promotor for color developing in persimmon extract dyeing. Property of water repellent was showed after color developing by heating with low concentration of alkali treatment.
