Inhibition of Adipocyte Differentiation and Adipogenesis by Supercritical Fluid Extracts and Marc from Cinnamomum verum (초임계 추출 계피오일의 3T3-L1 지방전구세포의 분화 전사인자 억제에 의한 지방대사 조절)

  • This study was performed to evaluate the antiobesity effect of supercritical fluid extracts (SFC) and marc methanol extracts (SFM) from Cinnamomum verum in 3T3-L1 preadipocytes. In inducing the differentiation of 3T3-L1 preadipocytes in the presence of an adipogenic cocktail, iso-butylmethylanthine (IBMX), dexamathasone, and insulin, treatment with fraction residue SFC and SFM. SFC significantly reduced the mRNA expression of the transcription factor peroxisome proliferator-activate-dreceptor-${\gamma}$ ($PPAR{\gamma}$), the sterol regulatory-element-binding protein-1c (SREBP1c), and the CCAAT enhancer-binding-protein ${\alpha}$ ($C/EBP{\alpha}$) in a concentration-dependent manner. Moreover, SFC markedly down-regulated acyl-CoA synthetase-1 (ASC1), fatty acid synthesis (FAS), fatty acid transport-1 (FATP1), fatty acid binding protein 4 (FABP4), and perilipin. These findings suggest that SFC may be a potential therapeutic adjunct for obesity by targeting the differentiation of preadipocytes, as well as their functions.

Phosphodiesterase III Inhibitor Cilostazol Protects Amyloid β-Induced Neuronal Cell Injury via Peroxisome Proliferator-Activated Receptor-γ Activation (Amyloid β에 의해 유도된 신경세포 손상에 대한 phosphodiesterase III inhibitor인 cilostazol의 신경보호 효과)

  • The neurotoxicity of aggregated amyloid ${\beta}$ ($A{\beta}$) has been implicated as a critical cause in the pathogenesis of Alzheimer's disease (AD). It can cause neurotoxicity in AD by evoking a cascade of apoptosis to neuron. Here, we investigated the neuroprotective effects of cilostazol, which acts as a phosphodiesterase III inhibitor, on $A{\beta}_{25-35}$-induced cytotoxicity in mouse neuronal cells and cognitive decline in the C57BL/6J AD mouse model via peroxisome proliferator-activated receptor (PPAR)-${\gamma}$ activation. $A{\beta}_{25-35}$ significantly reduced cell viability and increased the number of apoptotic-like cells. Cilostazol treatment recovered cells from $A{\beta}$-induced cell death as well as rosiglitazone, a PPAR-${\gamma}$ activator. These effects were suppressed by GW9662, an antagonist of PPAR-${\gamma}$ activity, indicative of a PPAR-${\gamma}$-mediated signaling. In addition, cilostazol and rosiglitazone also restored PPAR-${\gamma}$ activity levels that had been altered as a result of $A{\beta}_{25-35}$ treatment, which were antagonized by GW9662. Furthermore, cilostazol also markedly decreased the number of apoptotic-like cells and decreased the Bax/Bcl-2 ratio. Intracerebroventricular injection of $A{\beta}_{25-35}$ in C57BL/6J mice resulted in impaired cognitive function. Oral administration of cilostazol (20 mg/kg) for 2 weeks before $A{\beta}_{25-35}$ injection and once a day for 4 weeks post-surgery almost completely prevented the $A{\beta}_{25-35}$-induced cognitive deficits, as did rosiglitazone. Taken together, our findings suggest that cilostazol could attenuate $A{\beta}_{25-35}$-induced neuronal cell injury and apoptosis as well as promote the survival of neuronal cells, subsequently improving cognitive decline in AD, partly because of PPAR-${\gamma}$ activation. The phosphodiesterase III inhibitor cilostazol may be the basis of a novel strategy for the therapy of AD.

Inhibition of Adipocyte Differentiation by Methanol Extracts of Oenanthe javanica Seed in 3T3-L1 Preadipocytes (돌미나리씨 추출물에 의한 3T3-L1 지방전구세포의 분화 억제)

  • Oenanthe javanica has been used as a food source and also in traditional folk medicine for its detoxifying properties and anti-microbial effects since ancient times. In this study, we evaluated the effect and mechanism of O. javanica seed methanol extract (OJSE) on adipocyte differentiation by 3T3-L1 preadipocytes. Under non-toxic conditions, OJSE treatment resulted in a dose-dependent inhibition of lipid droplet generation and triglyceride accumulation by suppressing adipocyte differentiation, which are associated with the decreased expression of key proadipogenic transcription factors including CCAAR/enhancer binding protein ${\alpha}$, ${\beta}$ ($C/EBP{\alpha}$, $C/EBP{\beta}$) and peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$). OJSE also significantly inhibited proliferation and differentiation of 3T3-L1 preadipocytes through G1-phase arrest, indicating that OJSE blocked mitotic clonal expansion during adipocyte differentiation. Investigation of the alteration of G1 phase arrest-related proteins indicated a dose-dependent increase in the expression of p21 and reduction in expression of cyclin E, Cdk2, E2F-1 and phospho-Rb by OSJE. Taken together, these results suggest that OJSE inhibits adipocyte differentiation by blocking the mitotic clonal expansion, which is accompanied by preadipocyte cell cycle arrest.

Effect of Salicornia herbacea L. Supplementation on Tissue Triglyceride Concentrations and PGC-1α & PPAR-γ Expression of Skeletal Muscle of Rats Fed a High-fat Diet (함초의 보충식이가 고지방식이 흰쥐의 혈청 및 조직의 중성지방 농도와 골격근 내 PGC-1α 및 PPAR-γ 단백질 발현에 미치는 영향)

  • This study examined whether the supplementation of Salicornia herbacea L. (SH), a member of the Chenopodiaceae subfamily, affects tissue specific triglyceride (TG) accumulation and the peroxisome proliferator-activated $receptor-{\gamma}$ $coactivator-1{\alpha}$ ($PGC-1{\alpha}$) and peroxisome proliferator-activated $receptor-{\gamma}$ ($PPAR-{\gamma}$) protein expressions of skeletal muscle in rats with a high-fat diet. Sprague-Dawley male rats were randomly divided into three groups: control normal diet group (CD), high-fat diet group (HD), and 5.0% SH supplemented high-fat diet group (SD). The weights of fat tissue of the SD group were reduced by approximately 25%(p<0.01), while the skeletal muscle weight of the SD group increased approximately 5% compared to those in the HD group (p<0.01). The serum and hepatic TG of the SD group decreased approximately 20% compared to those of the HD group (p<0.05). In the protein expression levels in the skeletal muscle, the $PGC-1{\alpha}$ and $PPAR-{\gamma}$ expressions of the SD group were 1.5-folds higher than those of the HD group (p<0.01). From these results, SH supplementation contributes to the improvement of the serum and hepatic TG concentrations, and the $PGC-1{\alpha}$ and $PPAR-{\gamma}$ protein expression levels in the skeletal muscle of fed a high-fat diet. Thus, SH supplementation was effective in reducing fat mass and increasing muscle mass.

Effects of Essential Oils Extracted from Cnidii Rhizoma on Differentiation and Adipogenesis in 3T3-L1 Adiopocytes (천궁(川芎)의 정유 추출물이 3T3-L1 세포의 분화 및 지방 생성에 미치는 영향)

  • Objectives We investigated anti-obesity effects of essential oils extracted from Cnidii Rhizoma (CR) in immature adipocytes to magnify it's clinical therapeutic usage. Methods Essential oil of CR was extracted with ethyl acetate or petroleum ether and through steam distillation, respectively. Oil red-O staining for monitoring its inhibition effect on adipogenesis and differentiation in murine 3T3-L1 adipocytes and 3-(4,5-methylthiazol-2-yl)-2,5-diphenyletetra zolium bromide (MTT) assay for cell safety were done. Also phospho-adenosine monophosphate (AMP)-activted protein kinase (P-AMPK), AMP-activated protein kinase, phospho-acetyl-CoA carboxylase (P-ACC), acetyl-CoA carboxylase, peroxisome proliferator-activated receptor-${\alpha}$ (PPAR-${\alpha}$), peroxisome proliferator-activated receptor-${\gamma}$ (PPAR-${\gamma}$) and CCAAT/enhancer binding protein ${\alpha}$ (C/EBP-${\alpha}$) expressions as obesity-related factors were measured by western blot analysis. Results Protein expressions of P-AMPK, P-ACC and PPAR-${\alpha}$ were increased in essential oils-treated adipocytes compared to those of control group, respectively. Furthermore, protein expressions of PPAR-${\gamma}$ and C/EBP-${\alpha}$ were decreased in essential oils-treated adipocytes compared to those of control group, respectively. Conclusions These results demonstrate that essential oils of CR inhibit adipogenesis and differentiation. Also they promote the oxidation of fatty acids in adipocytes. Thus, results suggest that essential oils of CR could be used as a valuable material for anti-obesity therapeutics via control of lipid metabolism.

Effects of Polygonatum sibiricum rhizome extract on lipid and energy metabolism in high-fat diet-induced obese mice (고지방 식이 유도 비만 마우스 모델에서 황정 추출물의 지방질 및 에너지 대사 관련 유전자에 대한 효능 연구)

  • In this study, factors involved in lipid and energy metabolism following treatment with ethanolic extract of the Polygonatum sibiricum rhizome (ID1216) were evaluated in high-fat diet-induced obese mice. ID1216-treated mice showed a significant reduction in weight gain compared to non-treated mice. ID1216 treatment increased the protein levels of AMP-dependent protein kinase, sirtuin1, peroxisome proliferator-activated receptor ${\gamma}$ coactivator 1-${\alpha}$ ($PGC1{\alpha}$), peroxisome proliferator-activated receptor ${\alpha}$ ($PPAR{\alpha}$) and uncoupling proteins in the adipose tissue, liver and muscle compared to vehicle treatment. Analysis of downstream signals of the sirtuin1 $PGC1{\alpha}$-$PPAR{\alpha}$ pathway showed that ID1216 regulates the expression of ${\beta}$-oxidation related genes such as acyl-CoA oxidase, carnitine palmitoyltransferase1, acyl-CoA dehydrogenase and adipocyte protein 2. In addition, ID1216 increased the expression of adipose triglyceride lipase. These results suggest that ID1216 has anti-obesity effects by regulating the genes involved thermogenesis, ${\beta}$-oxidation and lipolysis in a diet-induced obesity model.

Inhibitory effect of Allium macrostemon extracts on adipogenesis of 3T3-L1 preadipocytes (산달래 추출물의 3T3-L1 지방전구세포 분화 억제 효능)

  • The aim of this study was to compare the biological activities of whole-plant (WAE), bulb (BAE), and leaf (LAE) extracts of Allium macrostemon. The antioxidant activities, total polyphenol contents, and anti-adipogenic activities of WAE and LAE were superior to those of BAE, whereas the biological effects of WAE and LAE were similar. Therefore, the effect of LAE on adipogenesis was further investigated. Treatment of preadipocytes with LAE at 100 g/mL resulted in the inhibition of intracellular lipid accumulation by 49.64%. Consistent with this result, quantitative reverse transcription-PCR showed that LAE treatment decreased the gene expressions of CCAAT/enhancer-binding protein beta (C/EBPβ), peroxisome proliferator-activated receptor gamma (PPARγ), C/EBPα and stearoyl-CoA desaturase 1 (SCD1). Thus, LAE attenuates the adipogenesis of preadipocytes by suppressing the expression of adipogenic and lipogenic genes. These results suggest that LAE can be potentially useful as a functional ingredient to prevent obesity in the food industry.

Inhibitory Effects of Lyophilized Dropwort Vinegar Powder on Adipocyte Differentiation and Inflammation (미나리 발효 식초의 지방세포 분화억제 및 항염증 효과)

  • Obesity, which is characterized by a state of mild chronic inflammation, is known to cause metabolic diseases. This study was carried out to investigate the effect of lyophilized dropwort vinegar powder (DVP) on adipocyte differentiation and inflammation in T3-L1 preadipocyte and RAW 264.7 macrophage cell lines. DVP inhibited the differentiation of 3T3-L1 preadipocytes induced by a mixture of IBMX, dexamethasone, and insulin (MDI). Western blot analysis of cell lysates showed that DVP decreased the levels of two major transcription factors involved in adipogenesis, peroxisome proliferator- activated receptor-${\gamma}$ (PPAR-${\gamma}$) and CCAAT-enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$). DVP also significantly suppressed lipopolysaccharide (LPS)-induced production of nitric oxide (NO), and this was accompanied by a decrease in inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2) protein expression. These results demonstrate that DVP inhibits MDI-induced adipocyte differentiation of 3T3-L1 cells and LPS-induced inflammation in RAW 264.7 macrophage cells. The findings indicate that this natural product may be a good candidate as to prevent metabolic diseases.

Antiadipogenic Effects of Red Radish (Raphanus sativus L.) Sprout Extract in 3T3-L1 Preadipocytes (적무 새싹 추출물의 3T3-L1 지방전구세포에서 지방합성 억제 효과)

  • The red radish (Raphanus sativus L.; RR) sprout is a plant of the cruciferous family. In this study, we elucidated the effect of the water extract of RR sprout (RRSE) against ${\alpha}$-amylase, ${\alpha}$-glucosidase, and pancreatic lipase enzyme activity and adipogenesis in 3T3-L1 preadipocytes. ${\alpha}$-amylase, ${\alpha}$-glucosidase, and pancreatic lipase enzyme activity was inhibited in a concentration-dependent manner by RRSE treatment. RSSE also abolished adipocyte differentiation and lipid and triglyceride accumulation without cytotoxicity in 3T3-L1 adipocytes. In addition, RRSE modulated the expression of the proteins related to adipogenic transcription factors: peroxisome proliferator-activated receptor (PPAR)${\gamma}$, sterol regulatory element-binding protein 1 (SREBP-1), and CCAT/enhancer binding protein (C/EBP)${\alpha}$. RRSE also suppressed expression of the proteins responsible for lipid synthesis, transport, and storage: adiponectin, fatty acid synthesis (FAS), perilipin, and fatty acid bind protein-4 (FABP4). This study showed that RRS treatment has the potential to inhibit obesity by controlling the expression of adipogenic transcription factors and adipogenic proteins.

Expression of peroxisome proliferator activated receptor gamma in the neuronal cells and modulation of their differentiation by PPAR gamma agonists

  • 15-Deoxy-${\Delta}^{12, 14}$-prostaglandin $J_2$ (15-deoxy-$PGJ_2$), a naturally occurring ligand activates the peroxisome proliferator-activated $receptor-{\gamma}(PPAR-{\gamma}$). Activation of $PPAR-{\gamma}$ has been found to induce cell differentiation such as adipose cell and macrophage. Here it was investigated whether 15-deoxy-$PGJ_2$ has neuronal cell differentiation and possible underlying molecular mechanisms. Dopaminergic differentiating PC 12 cells treated with 15-deoxy-$PGJ_2$ (0.2 to 1.6 ${\mu}M$) alone showed measurable neurite extension and expression of neurofilament, markers of cell differentiation. However much greater extent of neurite extension and expression of neurofilament was observed in the presence of NGF (50 ng/ml). In parallel with its increasing effect on the neurite extension and expression of neurofilament, 15-deoxy-$PGJ_2$ enhanced NGF-induced p38 MAP kinase expression and its phosphorylation in addition to the activation of transcription factor AP-1 in a dose dependent manner. Moreover, pretreatment of SD 203580, a specific inhibitor of p38 MAP kinase inhibited the promoting effect of 15-deoxy-$PGJ_2$(0.8 ${\mu}M$) on NGF-induced neurite extension. This inhibition correlated well with the ability of SB203580 to inhibit the enhancing effect of 15-deoxy-$PGJ_2$ on the expression of p38 MAP kinase and activation of AP-1, The promoting ability of 15-deoxy-$PGJ_2$ did not occur through $PPAR-{\gamma}$, as synthetic PPAR-${\gamma}$ agonist andantagonist did not change the neurite promoting effect of 15-deoxy-PGJ$_2$. In addition, contrast to other cells (embryonic midbrain and SK-N-MC cells), $PPAR-{\gamma}$ was not expressed in PC-12 cells. Other structure related prostaglandins, PGD$_2$ and $PGE_2$ acting via a cell surface G-protein-coupled receptor (GPCR) did not increase basal or NGF-induced neurite extension. Moreover, GPCR (EP and DP receptor) antagonists did not alter the promoting effect of f 5-deoxy-$PGJ_2$ on neurite extension and activation of p38 MAP kinase, suggesting that the promoting effect of 15-deoxy-$PGJ_2$ may not be mediated GPCR. These data demonstrate that activation of p38 MAP kinase in conjunction with AP-1 single pathway may be important in the promoting activity of 15-deoxy-$PGJ_2$ cells.
