Dietary supplementation with L-glutamine enhances immunity and reduces heat stress in Hanwoo steers under heat stress conditions

  • This study investigated the effects of L-glutamine (Gln) supplementation on growth performance, physiological traits, heat shock proteins (HSPs), and gene expression related to muscle and adipose tissue development in Hanwoo steers under heat stress (HS) conditions. Eight Hanwoo steers (initial body weight [BW] 570.7 ± 43.6 kg, months of age 22.3 ± 0.88) were randomly separated into two groups, control and treatment, and supplied with the concentration (1.5% of BW kg/day/head) and rice straw (1.5 kg/day/head). The treatment group were fed the Gln supplementation (0.5% of concentration, as-fed basis) once a day at 08:00 h. Blood samples for the assessment of haematological and biochemical parameters and the separation of peripheral blood mononuclear cells (PBMCs) were collected four times, at 0, 3, 6, and 10 weeks of the experiment. Feed intake was measured daily. BW to analyze growth performance and hair follicle collection to analyze the expression of HSPs were executed four times at 0, 3, 6, and 10 weeks. To analyze gene expression, longissimus dorsi muscle samples were collected by biopsy at the end of the study. As a result, growing performance, including final BW, average daily gain, and gain-to-feed ratio, were not different between the two groups. Leukocytes including lymphocytes and granulocytes, tended to increase in the Gln supplementation group (p = 0.058). There were also no differences in biochemical parameters shown between the two groups, except total protein and albumin, both of which were lower in the Gln supplementation group (p < 0.05). Gene expressions related to muscle and adipose tissue development were not different between the two groups. As temperature-humidity index (THI) increased, HSP70 and HSP90 expression in the hair follicle showed a high correlation. HSP90 in the hair follicle was decreased in the treatment group compared with the control group at 10 weeks (p < 0.05). Collectively, dietary Gln supplementation (0.5% of concentration, as-fed basis) may not be influential enough to affect growth performance and gene expression related to muscle and adipose tissue development in steers. However, Gln supplementation increased the number of immune cells and decreased HSP90 in the hair follicle implying HS reduction in the corresponding group.

알레르기 환자에서 TLR9 ligand인 CpG-ODN 자극에 의한 IFN-α 분비와 TLR9 발현 (Toll-like receptor 9 expression and interferon-α secretion upon CpG-ODN stimulation in allergic subjects)

  • 목 적 : 알레르기 환자와 정상인에서 형질세포양 수지상세포의 분포, TLR9 mRNA 양과 $IFN-{\alpha}$의 분비능에 차이가 있는지 알아보고자 하였다. 방 법 : 19명의 알레르기 환자와 17명의 건강한 성인을 대상으로 하였다. 말초혈액단핵세포를 채취하여 Lineage Cocktail(CD3, CD14, CD16, CD19, CD20, CD56)음성, HLA-DR 양성이면서 CD123양성을 유세포 분석기로 분석하였다. 말초혈액단핵세포에 TLR9 작용제(agonists)인 CpG-ODN 2216과 음성 대조를 위해 CpG-ODN 2206으로 자극하고 24시간 후 상청액을 추출하여 $IFN-{\alpha}$의 농도를 측정하였다. 또한 real time RT-PCR을 이용하여 TLR9 mRNA 정량분석을 시행하였다. 결 과 : 말초혈액단핵세포에서 형질세포양 수지상세포의 분포는 알레르기 환자가 평균 $0.1{\pm}0.04%$, 대조군이 평균 $0.25{\pm}0.23%$이었다. TLR9 mRNA 상대적인 양을 나타내는 ${\Delta}{\Delta}Ct$는 알레르기 환자에서 $1.29{\pm}0.41$이었고 대조군은 $1.25{\pm}0.23$이었다. TLR9 리간드인 CpG-ODN 2216 자극에 따른 $IFN-{\alpha}$의 분비능은 알레르기 환자에서 $911{\pm}829pg/mL$ 이었고 대조군에서 $1,095{\pm}888pg/mL$ 이었다. 이 세 결과에서 통계적인 차이는 없었다. 결 론 : TLR9을 통한 신호전달이 알레르기 환자의 면역반응을 대표하지는 않는 것으로 보이며, 향후 더 자세한 TLR9의 역할에 대한 연구가 필요하다.

Regulatory Effect of Gigukjihwangtanggami on Cytokine Production in Patients with Cerebral Infarction

  • The Korean genuine medicine, 'Gigukjihwangtanggami (GJT)' has long been used clinically for hypertension and various cerebrovascular diseases. However, experimental study has been carried out very little. Recently cytokines involved in the regulation of inflammatory reactions and immune responses may play an important role in the pathogenesis of cerebral infarction (CI). The aim of this study is to investigate the effect of GJT on the production of pro-inflammatory cytokines and anti-inflammatory cytokines in peripheral blood mononuclear cells (PBMCS) stimulated with lipopolysaccaride (LPS) from Cl patients. The amount of tumor necrosis $factor-{\alpha}\;(TNF-{\alpha}),\;interleukin-1{\beta}\;(IL-1{\beta})$, IL-6, IL-8, IL-4 and IL-10 in PBMC culture supernatant was significantly increased in the LPS treated cells compared to unstimulated cells. GJT inhibited the production of $TNF-{\alpha},\;IL-1{\beta}$, IL-6 and IL-8 induced by LPS. The maximal inhibition rate of the $TNF-{\alpha},\;IL-1{\beta}$, IL-6 and IL-8 production by pretreatment of GJT (1 mg/ml) was $57.32{\pm}2.5%$ (p.0.05), $42.02{\pm}3.5%$ (p.0.05), $40.02{\pm}2.3%$ (p.0.05) and $48.02{\pm}3.1$, (p.0.05), respectively. In the Other hand, GJT increased the production of IL-4 and IL-10. The maximal increase rate of the IL-4 and IL-10 production by pretreatment of GJT (1 mg/ml) was $42.4{\pm}3.3%$ (p.0.05) End $56.4{\pm}2.9%$ (p.0.05), respectively. Taken together, these results indicate that GJT Ray have regulatory effects on the cytokine production and suggest that GJT might use clinically for the treatment of CI.

Therapeutic Effects of Probiotics in Patients with Atopic Dermatitis

  • Recent studies have suggested that oral bacteriotherapy with probiotics might be useful for preventing and managing childhood atopic dermatitis (AD). The purpose of this investigation was to evaluate the efficacy and safety of oral treatment with probiotics for adolescent and adult AD patients as well as for childhood AD patients. Sixty-four patients with mild to moderate AD were recruited for treatment with a mixture of four probiotic strains (Lactobacillus rhamnosus, Lactobacillus plantarum, Lactobacillus casei, and Biftdobacterium lactis) twice daily for 8 weeks. The degree of pruritus was determined by a 10-point visual analog scale every other week, and the patients' global assessments of their clinical responses (i.e., better, unchanged, or worse) was done at the end of intervention. The clinical severity of the eczema was evaluated by eczema area and severity index (EASI) score every other week. As laboratory markers, total immunoglobulin E (IgE), eosinophil cationic protein (ECP) in the serum, and cytokine production [interleukin-4 (IL-4), interleukin-10 (IL-10), and $interferon-{\gamma}\;(IFN-{\gamma})$ by the peripheral blood mononuclear cells (PBMCs) were measured at the beginning and at the end of intervention. Of the 64 enrolled AD patients, only 50 patients finally completed the 8-week study. After 8-week treatment with probiotics, the EASI score was significantly improved (p<0.0001), 50% of the patients experienced improvement of their eczema, and significant improvement of the pruritus was also observed (p=0.0002). The effect was more pronounced for the patients with very high IgE levels (>1,000 ku/l) or for the patients with moderate disease severity. There was no significant difference in the therapeutic effects between the childhood AD and adolescent and adult AD patients. There were no significant changes of cytokines, as well as the total IgE and ECP levels, in the patients' serum. Treatment with the mixture of four probiotic strains was generally well tolerated. Our results suggest that the treatment with the mixture of four probiotic strains is beneficial for the management of the adolescent and adult AD patients, as well as for the childhood AD patients.

Subsets of Inflammatory Cytokine Gene Polymorphisms are Associated with Risk of Carcinogenic Liver Fluke Opisthorchis viverrini-Associated Advanced Periductal Fibrosis and Cholangiocarcinoma

  • Opisthorchis viverrini infection induces chronic inflammation, and a minor proportion of infected individuals develop advanced periductal fibrosis (APF) and cholangiocarcinoma (CCA). Inflammatory cytokines and/or their gene polymorphisms may link to these biliary pathologies. We therefore investigated associations among cytokine gene polymorphisms and cytokine production in 510 Thai cases infected with O. viverrini who presented with APF+ or APF-, as established by abdominal ultrasonography as well as in patients diagnosed with CCA. Levels of pro-inflammatory and anti-inflammatory cytokines were determined in culture supernatants after stimulation of peripheral blood mononuclear cells (PBMCs) with O. viverrini excretory-secretory (ES) products. Pro-inflammatory cytokines, IL-$1{\beta}$, IL-6, IFN-${\gamma}$, LT-${\alpha}$, and TNF-${\alpha}$ were significantly increased in CCA patients compared with non-CCA (APF- and APF+) cases. Polymorphisms in genes encoding IL-$1{\beta}$-511C/T, IL-6-174G/C, IFN-${\gamma}$+874T/A, LT-${\alpha}$+252A/G, and TNF-${\alpha}$-308G/A were then investigated by using PCR-RFLP or allele specific-PCR (AS-PCR) analyses. In the CCA cases, LT-${\alpha}$+252A/G and TNF-${\alpha}$-308G/A heterozygous and homozygous variants showed significantly higher levels of these cytokines than the wild type. By contrast, levels of cytokines in wild type of IFN-${\gamma}$+874T/A were significantly higher than the variants in CCA cases. IFN-${\gamma}$+874T/A polymorphisms were associated with advanced periductal fibrosis, whereas IL-6-174G/C polymorphisms were associated with CCA. To our knowledge, these findings provide the first demonstration that O. viverrini infected individuals carrying several specific cytokine gene polymorphisms are susceptible to develop fibrosis and CCA.

치료실패 폐결핵 환자의 세포성면역반응에 관한 연구 (Evaluation of the Cell-Mediated Immunity in Treatment Failure Pulmonary Tuberculosis)

  • 연구배경: 결핵은 대식세포와 T 림프구가 주로 관여하는 세포성 면역에 의하여 발생되는 대표적인 질환이다. 특히 Th1 또는 Th2 림프구 가농에 의하여 이루어지는 면역반응의 결과에 따라 결핵균에 대한 감수성 또는 저항성이 결정된다. 본 연구는 치료실패 폐결핵환자의 말초혈액단핵구를 PPD 또는 수용성 TSP 항원으로 결핵의 보호면역과 관계가 있는 Th1 반응과 결핵의 감수성과 관계가 있다고 알려진 Th2 반응을 관찰하였다. 방법: 수용성 TSP 항원과 대조항원인 PPD 항원으로 건강인, 폐결핵으로 진단되어 단기치료지침에 의하여 균음 전화된 치료반응 환자 및 치료실패 환자의 말초혈액 단핵구를 대상으로 단핵구 증식반응과 Th1 반응 및 Th2 반응과 각각 관계가 있는 IFN-$\gamma$ 및 RANTES와 MCP-1 mRNA 발현 빈도를 역전사효소 중합효소연쇄반응으로 조사하였다. 결과: PPD 피부반응 양성 건강인 모두는 PPD 또는 TSP 항원에 의하여 자극지수 4 이상의 유의한 증식반응을 보였으나 PPD 음성 건강인 모두는 PPD 또는 TSP 항원에 의하여 자극지수 4 미만의 증식반응을 보였다. 치유된 환자는 80%의 증식반응을 보였으나 치료실패 환자는 PPD 에 의하여 30.8% 그리고 TSP 항원에 의하여 15.4% 만이 자극지수 4 이상의 유의한 증식반응을 보였다. 치유된 환자의 1FN-$\gamma$ mRNA 발현빈도는 90.0% 이었으나 치료실패 환자는 PPD 또는 TSP 항원에 의하여 23.1% 만이 유도되었다. PPD 양성 건강인의 말초혈액단핵구를 PPD, TSP 또는 PHA로 자극하면 RANTES가 모두 발현되었다. 치료실패 환자, PPD 피부반응 음성 및 치유된 환자를 PPD로 발현을 유도한 경우 각각 76.9%, 80.0%로 대상간에 차이가 없었다. 그러나 TSP 항원으로 유도하면 건강 대조군 및 치유된 환자에 비하여 치료실패 환자는 46.2%로 발현빈도가 유의하게 감소되었다. 또한 PHA로 자극한 경우에서도 치료실패 환자는 69.2%로 감소하여 IFN-$\gamma$ mRNA 발현율 감소 경향과 유사하였다. 치료실패 환자는 MCP-1의 발현빈도가 치유된 환자에 비하여 유의하게 증가되었다. 치료실패 환자에 있어서 PHA 자극의 53.8% 보다는 PPD 또는 TSP로 자극한 경우에 발현빈도가 각각 76.9%로 높았다. 그러나 PPD 양성 건강인 및 치유된 환자는 PPD 또는 TSP로 유도한 결과 40% 이었으며 PHA로 유도한 경우는 각각 80%와 90%로 결핵균 항원에서 낮은 발현 빈도를 보여 치료실패 환자와 상반되는 결과를 보였다. 결론: 치료실패 환자는 PPD 피부반응 양성 건강인 및 치유된 환자에 비하여 말초혈액단핵구의 증식능, 1FN-$\gamma$ 및 RANTES mRNA 발현빈도가 현저히 감소되어 Th1 반응이 억제되어 있었다. 반면에 MCP-1 mRNA의 발현빈도는 현저히 증가되어 Th2 반응의 증가로 결핵균 사균 능력이 치료실패 환자는 감소되어 있다고 생각된다.
