A convergence study on the association between obesity and periodontitis in Korean adults aged 30-79 (30-79세 한국 성인의 비만과 치주염의 관계에 대한 융합연구)

  • The aim of this study was to confirm the association between obesity and periodontitis in adults and identify the convergence relationship between periodontal disease and health-related behavior at each stage of obesity among 10,058 adults aged 30-79 years from the 7th National Health and Nutrition Survey data. We performed chi-square tests and multivariate logistic regression analyses, adjusting for demographic characteristics and health status. We found that the higher the obesity stage, the higher the risk of periodontitis. Smoking, not using oral care products, and not attending dental check-ups were associated with periodontitis. In addition, we found a difference in influence factors according to the stage of obesity. Increase in obesity stage is a negative influence on the prevalence of periodontitis. Hence, an oral health program should be applied to the obesity management process.

Influence on Oral Health Condition by Visits to Dental Clinics and Dental Education Experience of East Asian Immigrant Women in Korea (동아시아 국내 이주여성의 치과방문여부와 구강건강교육유무가 구강건강상태에 미치는 영향)

  • Objectives : The purpose of this study is to figure out and establish the basic data that can help improve management of oral healthcare for the international marriage immigrant women. Methods : In this sense, we surveyed questions on 237 women out of 1,300 immigrant women, who have participated in the program by multi-cultural household supporting center, in a fashion of face-to-face investigation and on-the-spot direct cavity inspection simultaneously from May 1 to October 31, 2010. Collected data were electro-statistically computerized under SPSS 17.0 program and analyzed with frequency analysis, recurrence analysis and logistic regression analysis respectively. Results : The number of filled teeth got lower when they had lower number of dental visits. Women from the Philippines had the highest number of defect teeth, and the number of fixed prosthetic appliance became less when they had less number of dental visits. When they had no experience of oral health education, they had less fixed prosthetic appliances. There was higher percentage of periodontal disease when they did not have dental visit compared with the dental visit cases. Conclusions : Preparation of a policy for east Asian immigrant women for them to visit dentist comfortably and expansion of systematic oral health education are highly recommended.

The protective effect of coenzyme Q10 on cytotoxicity of regin monomer of odontoblast caused by TEGDMA (코엔자임 Q10 처리에 따른 TEGDMA에 의해 유발된 치아 세포 사멸 억제 효과)

  • Objectives : The purpose of the study is to investigate the protective effect of coenzyme $Q_{10}$ on cytotoxicity effect of dental monomers in odontoblast(MDPC-23). Methods : MDPC-23 was incubated with the(co)monomers triethylene glycol dimethacrylate (TEGDMA) with and without addition of coenzyme $Q_{10}$. The cell proliferation and survival was determined using WST-1 assay. The level of reactive oxygen species(ROS) was measured by immunofluorescent staining for DCF-DA. Results : TEGDMA treatment decreased the cell proliferation by dose dependently(0.1, 1, 2.5, 5, 10 mM) on the growth of MDPC-23 cells. Coenzyme $Q_{10}$ showed cell proliferation from 5 to $500{\mu}M$ by WST-1 assay. Pre-treatment coenzyme $Q_{10}$ showed the antioxidant effect on proliferation and viability of MDPC-23 after 48h(p<0.05). The positive cells were observed in non-coenyme $Q_{10}$ treatment group(group 2) in comparison with coenyme $Q_{10}$ pre-treatment group(group 1) by DCF-DA. The fluorescence positive cells showed 14.715(group 1) and 19.788(group 2) using image J system. Conclusions : TEGDMA induced cytotoxicity. The MDPC-23 cell death was associated with the increasing ROS. Coenyme $Q_{10}$ showed the antioxidant effects by decreasing ROS. This effects may contribute to the treatment of periodontal disease induced by TEGDMA after operation.

Relationship between National Exam Stress and Subjective Oral Symptom in Allied Health Students (보건계열 대학생의 국가시험 스트레스와 주관적 구강이상증상)

  • The aim of this study was to confirm for relationship between national exam stress and oral health status on allied health students. For this, 829 of allied health students were surveyed in four college and university located in Chungbuk and Kyungbuk. Survey tool were constituted general characteristic(4 items), national exam stress(16 items) and oral health status(34 items). The result of study, national exam stress of female were higher than male and that of department of dental hygiene, nursing, emergency medical service students were higher than other students(p<0.05). Subjective oral symptom were higher on high stress group than other groups(p<0.05). Relationship between stress and subjective oral symptom were related each other(p<0.01). Therefore it is necessary to education for manage oral symptom affected national exam stress.

Resveratrol inhibits cell growth via targeting the Bmi-1 pathway in YD-10B human oral squamous cell carcinoma cells

  • Resveratrol has been reported to exert anticancer activity via modulation of multiple pathways and genes. In this study, we examined the effect of resveratrol on YD-10B human oral squamous cell carcinoma cells and its molecular mechanisms of action. We found that resveratrol inhibited the proliferation of YD-10B cells in a dose- and time-dependent manner. The suppressive effect of resveratrol was accompanied by a reduction in Bmi-1 gene expression. We observed that silencing the Bmi-1 gene by small interfering RNA effectively downregulated the levels of GLUT1 mRNA and protein, which were also repressed by resveratrol. Bmi-1 silencing increased the number of YD-10B cells in S-phase arrest by approximately 2.3-fold compared with the control. In conclusion, the results of the present study demonstrate, for the first time, that resveratrol suppresses Bmi-1-mediated GLUT1 expression in human oral squamous cell carcinoma cells and suggest that the specific molecular targeting of Bmi-1 and/or GLUT1 expression can be combined with a chemotherapeutic strategy to improve the response of oral cancer cells to resveratrol.

Next-generation sequencing analysis of exosomal microRNAs: Fusobacterium nucleatum regulates the expression profiling of exosomal microRNAs in human colorectal cancer cells

  • Colon cancer is one of the most common malignant tumors, but there are still a few validated biomarkers of colon cancer. Exosome-mediated microRNAs (miRNAs) have been recognized as potential biomarkers in cancers, and miRNAs can regulate a variety of genes. Recently, Fusobacterium nucleatum was discovered in the tissues of human colon cancer patients. Its role in colon cancer was highlighted. F. nucleatum may contribute to the progression of colon cancer through the mechanism of exosome-mediated miRNAs transfer. However, the exosomal miRNAs regulation mechanism by F. nucleatum in colon cancer is not well known. Thus, we performed next-generation sequencing to investigate the overall pattern of exosomal miRNAs expression in the colon cancer cell culture supernatant. We have confirmed the alterations of various exosomal miRNAs. In addition, to investigate the function of exosomal miRNAs, a Kyoto Encyclopedia of Genes and Genomes analysis was performed on the target genes of changed miRNAs. Potential target genes were associated with a variety of signaling pathways, and one of these pathways was related to colorectal cancer. These findings suggested that F. nucleatum can alter exosomal miRNAs released from colorectal cancer cells. Furthermore, exosomal miRNAs altered by F. nucleatum could be potential biomarkers for the diagnosis and therapy of colon cancer.

  • Purpose: Autogenous bones are frequently used because of their lack of antigenicity, but good osteoconductive and osteoinductive properties. This study evaluated the biological behavior of perforated and nonperforated cortical block bone grafts. Methods: Ten nonsmoking patients who required treatment due to severe resorption of the alveolar process and subsequent implant installation were included in the study. The inclusion criteria was loss of one or more teeth; the presence of atrophy of the alveolar process with the indication of reconstruction procedures to allow rehabilitation with dental implants; and the absence of systemic disease, local infection, or inflammation. The patients were randomly divided into two groups based on whether they received a perforated (inner surface) or nonperforated graft. After a 6-month healing period, a biopsy was performed and osseointegrated implants were installed in the same procedure. Results: Fibrous connective tissue was evident at the interface in patients who received nonperforated grafts. However, full union between the graft and host bed was visible in those who had received a perforated graft. Conclusions: We found that cortical inner side perforations at donor sites increased the surface area and opened the medullary cavity. Our results indicate an increased rate of graft incorporation in patients who received such perforated grafts.

Full mouth rehabilitation of the intellectually disabled patient with collapsed bite using partial removable dental prosthesis: a case report (교합이 붕괴된 지적장애 환자의 국소의치 수복증례)

  • Intellectual disability is an imperfect disorder in which mental development is permanently retarded and development of intellectual ability is inadequate. Having intellectual disabilities makes it difficult to handle their own work and adapt to social life. For this reason, it is difficult to hygienically manage the patient's oral condition, multiple caries are easily observed, and the periodontal disease incidence rate is high. The patient in this case is a 33-year-old female with a mental retardation first grade who had a problem with meals. In first visit, Patients had decreased occlusal vertical dimension and posterior bite collapse due to periodontitis and multiple caries. This case reports a satisfactory functional and esthetic results by reconstructing occlusion using partial removable dental prosthesis.

THE EFFECT OF SUPEROXIDE DISMUTASE ON EXPERIMENTAL GINGIVITIS AND ACTIVITY OF 3T3 FIBROBLAST (Superoxide Dismutase가 백서의 실험적 치은염과 3T3 섬유모 세포의 활성에 미치는 영향)

  • Inflammatory cells may produce active species of oxygen in antimicrobial defense. While such species can directly damage surrounding tissue, their major secondary role may be to mediate important components of the inflammatory response. Superoxide dismutase, antioxidant, have significant anti-inflammatory properties in rheumatoid arthritis, ischemic tissue injury and gastrointestinal disease. Increased oxidative product formation diseases. And superoxide dismutase produced by Porphyromonas Gingivalis is resistant to killing by polymorphonuclear leukocyte. The purpose of this study was to investigate on the effects of superoxide dismutase in 3T3 fibroblast and in experimental gingivitis in the rats. The effect of superoxide dismutase(SOD) to cell morphology and cell activity was measured in cultured mouse 3T3 fibroblast. After experimental gingivitis were induced by lipopolysaccharide(LPb) and bovine serum albumin(BSA), injection of SOD were done. WBC count and histologic findings were observed at 1, 2, 3, and 7 days. The results were as follows; 1. There was a little difference between LPS treated groups and SOD treated groups in 3T3 fibroblast morpholoy. 2. There was no difference between only SOD treated groups (except SOD 150U at 3days) and control in 3T3 fibroblast activity. 3. LPS $0.5{\mu}g/ml$ and SOD treated groups (except 150U) had decreased 3T3 fibroblast activity and no significant difference at 3 days. 4. LPS $5.0{\mu}g/ml$ and SOD treated groups were significantly increased cell activity of 3T3 fibroblast than control group at 1 day(P<0.05). 5. In LPS induced gingivitis, the number of leukocytes in SOD treated was significantly decreased than in saline treated at 1 day(P<0.05). 6. In histopathologic findings of LPS or BSA induced gingivitis, inflammatorycell infiltration in SOD treated groups were less than in saline treated group at 1, 2 and 3 days.

Comparison of the Oral Health Behavior and the Oral Health Status According to Residential Areas in Korean Adults: based on 2012 KNHANES data (거주 지역 간 성인의 구강건강행태 및 구강건강상태 비교)

  • Purpose: This study aimed to evaluate and compare the oral health behavior in adults and their oral health status according to their residential area. Methods: The date of 'The fifth Korea National Health and Nutrition Examination Survey 2012' was analyzed for this study. This study were adults over the age of 18 who participated in interviews with the use of a questionnaire and oral examinations. Of those, 4,273 who met all the variables necessary were selected for the final analysis. The residential areas were categorized into 'metropolitan city', 'city', and 'rural area' based on Dong, Eup and Myon as administrative districts. Results: Adjusted odds ratio of oral health behavior according to residential area, self-recognition of oral health status for subjects in rural areas was 0.75(0.59-0.96), using oral hygiene devices for those in rural areas was 0.75(0.63-0.88). Adjusted odds ratio of oral health status according to residential area, periodontal disease in rural areas was 1.97(1.62-2.41), the necessity of prosthetic treatment in subjects in rural areas was 1.27(1.01-1.60). Conclusion: It was discovered that there was a difference between the oral health behavior and oral health status of adults according to residential area. Therefore, to enhance oral health status, programs for which the characteristics of areas with such differences were considered should be developed and consistent research on strategies to reduce the gaps in the oral health status should be made.
