Vector Construction and Transformation of Ginseng (Panax ginseng C.A. Meyer) Using Disease Resistant Genes (내병성 관련유전자의 운반체 재조합 및 인삼(Panax ginseng C.A. Meyer)의 형질전환)

  • For study about introduce of gene connected with disease and transformation system of gingseng, chitinase gene cloned from soybene and disease resistant gene were carried out for expression and transformation of plant using Agrobacterium. The disease resistance gene(DR-49), 35S-35S-AMV, has been constructed. The disease resistance gene and chitinase gene were introduced into the binary vector pRD 400, which were mobilized into Agrobacterium tumefaciens faciens strain MP 90 and LBA 4404 harboring disarmed Ti-plasmid. As a result of induce transformants using ginseng embryo and petiole, multi shoots were formed on MS medium supplemented 1 mg/ι 2,4-D and 0.5 mg/ι kinetin. Also transformation by cotyledonwas effective on MS medium supplemented 1 mg/ι 2,4-D and 0.5 mg/ι kinetin, transformation percent of disease resistant gene and chitinase gene were showed 18%, 14% respectively. As transformed tissue is under pre-embryoid condition, normal shoot is required through the process of matured embryo.

A Study of Shrinkage Characteristics of Low Shrinkage Normal Strength Concrete With Boundary Restraint Condition (4변 구속조건을 갖는 초저수축 일반강도 콘크리트의 수축특성 연구)

  • In this study, the replacement effects of cementitious materials (fly ash, blast furnace slag, and blended mixtures) were assessed for normal strength concrete with very low shrinkage properties under $350{\mu}{\varepsilon}$ strain using a powder type shrinkage reducing agent. In addition, through mock-up tests of actual size walls restrained with four sides, the shrinkage characteristics using the power type shrinkage reducing agent were measured and the crack reducing ability was assessed. The slump and air contents were measured as the properties of fresh concrete, and the length changes of the prismatic specimens, $100{\times}100{\times}400mm$ in size, were measured for the shrinkage characteristics. To reduce the shrinkage of concrete, the maximum replacing ratio of the fly ash is effective to 20 percent; however, the use of blast furnace slag and ternary mixtures did not reduce the shrinkage.

The Prediction of tong-Term Creep Behavior of Recycled PET Polymer Concrete (단기 크리프 실험을 이용한 PET 재활용 폴리머콘크리트의 장기 크리프거동 예측)

  • In general, polymer concrete has more excellent mechanical properties and durability than Portland cement concrete, but very sensitive to heat and has large deformations. In this study, the long-term creep behaviors was predicted by the short-term creep test, and then the characteristic of creep of recycled-PET polymer concrete was defined by material and experimental variables. The error in the predicted long-term creep values is less than 5 percent for all polymer concrete systems. The filler carry out an important role to restrict the creep strains of recycled PET Polymer concrete. The creep strain and specific on using the $CaCO_3$ were less than using fly-ash. The creep increases with an increase in the applied stress, but not proportional the rate of stress increase ratio. The creep behavior of polymer concrete using recycled polyester resin is not a linear viscoelastic behavior.

Evaluation on efficacy of β-hemolytic Streptococcus iniae vaccine on olive flounder, Paralichthys olivaceus (β-용혈성 Streptococcus iniae 불활화백신의 넙치에 대한 효능 평가)

  • Olive flounder, Paralichthys olivaceus is one of the most important cultured fish in Korea, its farming has been negatively impacted by viral, bacterial and parasitic diseases. Streptococcal infection was considered as a serious problem because of significant economic losses in olive flounder farm industry. The development and evaluation of vaccine for protection against infection by this agent were required. We evaluated the safety and efficacy of ${\beta}$-hemolytic Streptococcus (S.) iniae vaccine on olive flounder Three hundreds of flounders (weight $119.8{\pm}20.7g$, body length $22.6{\pm}1.4cm$) were reared in 0.5 tons aquaria in land-marine tank system. Seawater was provided from the sea of Inchon in Korea, and water temperature was set to $22^{\circ}C$ and $25^{\circ}C$ in the vaccination and challenge test, respectively. We used the formalin-inactivated ${\beta}$-hemolytic S. iniae (F2K) vaccine (M VAC INIAE; Kyoritsu seiyaku, Japan) originated in Japan. The vaccine was intraperitoneally administered to fish. Both of vaccinated group and control group were challenged with intraperitoneally injection by virulent S. iniae SI-36 isolates with $1.0{\times}10^7CFU/fish$ at 3 weeks after vaccination. Difference on mortality of control and vaccinated group (90.0 and 15.0%, 76.5 and 8.0% respectively) at two trials were found significant (p<0.05), and relative percent survival were 83.4% and 89.5%, respectively. The dead fishes were showed dark pigmentation of skin, abdominal extension, hemorrhagic ascites, and liver necrosis, and isolated the S. iniae strain from ascites, liver and kidney. We confirmed the safety and efficacy of ${\beta}$-hemolytic S. iniae vaccine by determinations of the optimal management condition and artificial challenge test in olive flounder.

Incubation Conditions and Physico-Chemical Factors Affecting Aflatoxin B1 Binding of Lactic Acid Bacteria (Aflatoxin B1에 대한 유산균의 결합력에 영향을 미치는 배양조건과 물리화학적 인자)

  • The purpose of this study was to investigate the aflatoxin $B_1$ binding of lactic acid bacteria (LAB) isolated from Korean traditional soybean paste and to evaluate the effect of incubation conditions and physico-chemical factors on the binding ability of LAB to this mutagen. The amount of aflatoxin $B_1$ bound by Enterococcus faecium DJ22, Lactobacillus fermentum DJ35, Lactobacillus rhamnosus DJ42, and Lactobacillus pentosus DJ47 was strain specific with the percent bound ranging from 19.3% to 52.1%. However, Enterococcus faecalis DJ14, Lactobacillus panis DJ29, and Pediococcus halophilus DJ50 strains did not exhibit any of the binding ability to aflatoxin $B_1$. For most strains, the binding ability was significantly affected by the environmental conditions such as the aflatoxin $B_1$ level, incubation time and temperature, and the initial cell count of LAB. The stability of the aflatoxin $B_1$-bacteria complexes was significantly more unstable after washing. In addition, the binding stability between viable and nonviable cells was not statistically significant. Treatment with heating, acidic pH, ${\alpha}$-amylase, protease, lysozyme, or sodium metaperiodate caused a significant (P<0.05) decrease in aflatoxin $B_1$ binding for the tested strains, suggesting that carbohydrates or proteins in the cell walls may be involved in aflatoxin $B_1$ binding ability. Since the aflatoxin $B_1$ binding of LAB was significantly reduced (P<0.05) by the pretreatment of the urea, the binding force observed in this study may have resulted from hydrophobic interaction.

$pep^{27}$ and lytA in Vancomycin-Tolerant Pneumococci

  • Vancomycin therapy failure due to the emergence of tolerance in pneumococci is increasing. The molecular mechanism of tolerance is not clear, but lytA and $pep^{27}$ are known to be involved. Our aim was to evaluate the expression of both genes in vancomycin-tolerant Streptococcus pneumoniae (VTSP) strains. Eleven VTSP strains from a total of 309 clinical isolates of S. pneumoniae from 1997 to 2006 were classified according to the criteria of Liu and Tomasz. All VTSP strains were evaluated for susceptibility according to CLSI criteria, serotype by the Quellung test, and clonality by PFGE. The expressions of lytA and $pep^{27}$ were analyzed in different growth phases by RT-PCR with and without vancomycin. Eighty-two percent of VTSP strains showed resistance to penicillin, and 100% were sensitive to vancomycin and cefotaxime. The most frequent serotypes of VTSP strains were 23F (4/11) and 6B (3/11). Clonal relationship was observed in only two strains. No significant changes were observed in $pep^{27}$ expression in the three phases of growth in VTSP strains with and without vancomycin. Interestingly, $pep^{27}$ expression in the stationary phase in the non-tolerant reference strain R6 was significantly higher. However, no significant differences in lytA expression were observed between VTSP and R6 strains during the phases of growth analyzed. The absence of changes in $pep^{27}$ expression in VTSP strains in the stationary phase may be related to their ability to tolerate high antibiotic concentrations, and thus, they survive and remain in the host under the antibiotic selective pressure reflected in therapeutic failure.

국내 연근해 및 환자로부터 분리된 vibrio vulnificus의 세균학적 특징

  • Vibrio vuln$cus has been recognized as a pathogen of septicemia and wound infection, when the organism attacks high-risk persons with a history of hepatic disease. alcohol abuse. diabetes or any debilitative disease. Forty six strains of K vulnzjicus. isolated from 1025 marine specimens from May to Novemver for three years. from 1985 to 1987. were studied for their biochemical properties. growth requirements, serotype and drug susceptibilities. The isolates were different in their various biochemical reactions. Ninety-five percent of isolated strains were able to ferment lactose, while most strains didn't utilize sucrose in their biochemical test, for example ornithine, gelatin and mannitol were quite dit'ferent composition than those described in other reports. It was found that the biochemical test wasn't useful for identifying strain. The type of somatic 0 antiserum was determined in isolates from marine sources and in patients with Vibrio septicemia. In patient isolates. 1-2 group were 24% and 1-4 group were 42%. However. 02 group(33%) were more abundant in isolates from marine sources. Minimal inhibitory concentrations(M1Cs) of chloramphenicol, tetracycline. erythromycin and ampicillin were determinef for V vuln~ficus by broth dilution method. MIC90 was I , 0.25, :! and 4,ug/ml in patient isolates. 1, 0.25, 2 and 2 ,ug/ml in marine isolates. The divalent chelating agent, IDTA. inhibited the growth of V. vuln!'ficus at 6.25 mMlml of MIC90.

Pulsed Field Gel Electrophoresis Profile of Erythromycin-Clindamycin Resistant Streptococcus pyogenes Isolated in Korea (국내분리 Erythromycin-Clindamycin 내성 Streptococcus pyogenes에 대한 Pulsed Field Gel Electrophoresis 양상 분석)

  • Ninety two strains of Streptococcus pyogenes were isolated from patients with pharyngitis, scarlet fever, skin infection, and invasive streptococcal infections in Seoul, Korea from January to December, 1998. All isolates were epidemiologically characterized by T protein serotype, and serum opacity factor (OF) detection to phenotypes. To analyze the genetic relationship, fifty two isolates including 32 erythromycin-clindamycin (Em-Cm) resistant strains, 20 antimicrobial susceptible strains were attempted to the pulsed-field gel electrophoresis (PFGE). T protein serotype showed 16 kinds in distribution including T12 and T4. Among the total isolates, 40 strains (43.5%) belonged to the T12 serotype and twenty strains (21.7%) to T4 serotype. On the other hand, when infection aspect of S. pyogenes isolates were analysed by T serotype distribution, T12 type was predominant for pharyngitidis which contributed to 21 strains (53%) and for skin infection isolates which contributed to 11 strains (28%), respectively. In case of T4 type, it was the most predominant pharyngitidis isolates which contributed to 8 strains (40%). In T serotype distribution of Em-Cm resistant strains, 27 strains (84%) of the thirty two showed T12 serotype. In minimum inhibitory concentration (MIC) values of Em-Cm resistance isolates, thirty two isolates showed resistant to erythromycin 27 strains (84%), had high MIC of >$128\;{\mu}g/ml$. And also to clindamycin, twenty two strains (69%) had high MIC of >$128\;{\mu}g/ml$. When OF detection of Em-Cm resistance of S. pyogenes isolates were analyzed by T serotype distribution, T12 serotype isolates revealed that all of the isolates except one strain were OF negative. In PFGE profile analysis to Em-Cm resistance isolates, of the twenty seven, Em-Cm resistance of T12 serotype isolates, 26 strains showed identical PFGE profile and all of these isolates revealed that OF negative. Eighty four percent of Em-Cm resistance S. pyogenes isolates had identical phenotype and PFGE profile. These results strongly suggested that the Em-Cm resistant S. pyogenes isolates from Seoul area showed close genetic correlation and PFGE could be available tool for molecular epidemiology.

  • This glucose isomerizing enzyme, which Actinomyces sp. (strain, K-17) produced, was inhibited by citrate, oxalate, ethylene diamine tetraacetic acid and cysteine on the enzyme reaction. This enzyme isomerized xylose to ketose as well as glucose. The Michaelis constant of this enzyme was $7.2X\;10^{-1}M.$. on D-glucose. The enzyme activity of intact cells which were harvested in the none xylose containing medium was very weak. If these intact cells of low activity were treated in the buffered xylose solution, its activity was considerably increased. After fifteen hours at $32^{\circ}C$. on xylose treatment, the enzyme activity was increased to equilibrium and the treating condition was proper at neutral pH and in aerobic state. The adequate concentration of xylose on the treatment was about 0.5 percent.

Studies on tussah silkworm, Antheraea pernyi (작잠에 관한 연구)

  • I. Breeding of tussah silkworm(preliminary report). The preliminary examination for bleeding has been carried out since 1963 in tussah silkworms. 1) The strain(l-MG-B)of the heaviest silk quantity was the green silkworm and brown cocoon in univoltine, and the strains(2-G-B, 2-MG-B) of the heaviest silk quantity were also the green silkwom and brown cocoon in bivoltine in both spring and fall in 1965. 2) It looks like the voltinism, the body color and the cocoon color have reached to pure line up to 1965. II. Best place for the winter of tussah pupa. This work was aimed to find out good ways for the winter of tussah pupa. 1) The hatch of bivoltine was better than that of univoltine. 2) The cocoons covered with the leaves were good in the emergence of moth. 3) The cocoons which were kept at natural temperature till the first emergence of moths would show bad in both hatch and emergence. 4) If some of the pupae kept under natural condition were controled at proper temperature for a few days, hatch and laying eggs were best. 5) The best places for the winter were the egg storage and the rearing room. III. Relation between incubation temperature and voltinism. 1) When the tussah pupa are kept at natural temperature during winter, the moths do not come out of the pupa. 2) There is no difference between about 18$^{\circ}C$ and about 25$^{\circ}C$ during incubation in hatching ratio. 3) The tussah silkworms of univoltine in mortality are stronger than that of bivoltine. 4) There is not any relation between voltinism and high or low temperature for pupa and eggs. IV. Induced mutation by gamma-ray and neutron in tussah silkworm. This work was carried out in order to induce the mutation by treating the pupa or the eggs of tussah silkworm with gamma my and neutron. The results obtained are as follows. 1. Though the whole pupa treated with neutron become moths, the moths have no ability to copulate each other. The only moths emerged from pupa treated with neutron, 4000${\gamma}$ are able to lay all un-fertilized eggs, some of which have a hole on the surface and nothing of contents. 2. The non-diapause eggs are treated with neutron in spring, but the hatching ratio is 50∼60 percent, but the whole eggs treated with gamma ray are never hatched. 3. The sensitivity of the pupa to neutron is weaker than that of the eggs. 4. The hatching ratio is in direct proportion to the gamma ray dose. 5. Author find out a new mutant which is excellent in the cocoon quality, so he will do the progeny test next hear.
