A new technique was proposed for the determination of alkylphenols, chlorophenols and bisphenol A in tap water samples. The sample preparation consists of a solid phase extraction (SPE) of alkylphenols, chlorophenols and bisphenol A from a water sample with XAD-4 and subsequent conversion to isobutyloxycarbonyl (isoBOC) derivatives or tert-butyldimethylsilyl (TBDMS) derivatives for sensitive analysis with the CC/MS SIM mode. The recoveries were 86.6 ∼ 105.2% (isoBOC derivatization) and 97.6∼484.5% (TBDMS derivatization), respectively. The limit of quantitation of alkylphenols, chlorophenols and bisphenol h for SIM were 0.001∼0.050 $\mu\textrm{g}$/1 (isoBOC derivatization) and 0.003∼0.050 $\mu\textrm{g}$/1 (TBDMS derivatization). The SIM responses were linear with the correlation coefficient varying 0.9755∼0.9981 (isoBOC derivatization), and 0.9908∼0.9996 (TBDMS derivatization). When these methods were applied to tap water samples, the range of concentrations were 22.8∼31.3 ng/1 in 2,4-dichlorophenol, 28.6∼70.3 ng/1 in pentachlorophenol, 15.2∼17.4 ng/1 in t-butylphenol, 10.8∼13.2 ng/1 in t-octylphenol and 17.6∼36.3 ng/l in bisphenol A, respectively.

We developed and evaluated a method of short-term acute toxicity testing based on the feeding behavior of Ceriodaphnia dubia. In prior toxicity tests, neonates of C. dubia were hatched and cultivated with the addition of yeast only for the preparation of the transparent daphnid's gut. Scenedesmus subspicatus was supplied as food after 1 to 6 h of exposure to toxicants. The effects of 1-h and 6-h exposure time on test sensitivity did not significantly differ. A comparison of the short-term l-h acute toxicity test developed in this study to the standard 48-h acute toxicity test using heavy metals, cyanide, and pentachlorophenol indicated that the 1-h test provided an acceptable sensitivity level in toxicity testing of C. dubia..

Catalytic degradation of pentachlorophenol in soil by heme and hydrogen peroxide has been hypothesized to occur through nonspecific catalytic reactions similar to those involving ligninase. The present study examines the evidence for a heme catalytic mechanism for the oxidation of organic compounds. In the presence of hydrogen peroxide, heme is converted to the ferryl heme radical (Hm-$Fe^{+4{\cdot}}$), which can oxidize organic compounds, such as 5-aminosalicylic acid (5-ASA). A second 5-ASA may later be oxidized by ferryl heme (Hm-$Fe^{+4}$), which reverts to the ferric heme state (Hm-$Fe^{+3}$) to complete the cycle. We believe that this catalytic cycle is involved in the degradation of hazardous pollutants, such as polycyclic aromatic hydrocarbons (PAHs). Remediation via heme catalytic reactions of PAHs in soil from a pole yard was evaluated, and about 96% of PAHs was found to disappear within 42 days after treatment with heme and hydrogen peroxide. In addition, benzo[a]pyrene and six other PAHs were undetectable among a total of 16 PAH compounds examined. Therefore, we propose heme catalysis as a novel technology for the remediation of hazardous compounds in contaminated soil.

Consortia demonstrated the high capacities of anaerobic degradation of various aromatic compounds, which were successfully enriched from gley paddy soils under different conditions. Phenol and cresol was decomposed anaerobically using nitrate, ferric oxide or sulfate as electron acceptors. Biphenyl was degraded to $CO_2$, especially without addition of external electron acceptor. Alkylphenols with middle length of alkyl chain, were co-metaboliocally degraded with the presence of hydroxylbenzoate as the co-substrate under nitrate reducing conditions. The microorganisms responsible for the anaerobic co-metabolism was Thauera sp. Reductive dechlorination activity was also observed for polychlorophenols, fthalide, polychlorinated biphenyls, polychlorinated dibenzo-p-dioxins with the presence of lactate, formate or $H_2$ as electron donor. The fthalide dechlorinator was classified as Dehalobacter sp. Coupling of two physiologically-distinct anaerobic consortia, aromatic ring degrader and reductive dechlorinator, resulted in the mineralization of pentachlorophenol under anaerobic conditions. These results suggested that gley paddy soils harbored anaerobic microbial community with versatile capacity degrading aromatic compounds under anaerobic conditions.
Twelve chlorophenols (CPs) were extracted by liquid phase microextraction (LPME) from the industrial waste-water and analyzed by GC/MS. To establish the optimal conditions, species of extraction solvent, sample amount, pH of sample, salting out effect, a number of sampling and plunger movement speed were investigated. As a result, the linearities of calibaration curves ranged from 0.9913 to 0.9999, while LODs and LOQs were from 0.05 to 10.0 ng/mL except 2,3,4,5-tetrachlorophenol and pentachlorophenol. Using this method, 4-chloro-3-methylphenol confirmed from waste water at the concentration of 784 ng/mL. The method can be applicable to detect chlorophenols from industrial waste-water.
Here in this work, a 4-chlorophenol (4-CP)-degrading bacterial strain Bacillus subtilis (B. subtilis) MF447840.1 was isolated from the drain outside the Hyundai car service center, Agartala, Tripura, India. 16S rDNA technique used carried out for genomic recognition of the bacterial species. Isolated bacterial strain was phylogenetically related with B. subtilis. This strain was capable of breaking down both phenol and 4-CP at the concentration of 1,000 mg/L. Also, the isolated strain can able to metabolize five diverse aromatic molecules such as 2-chlorophenol, 2,4-dichlorophenol, 2,4,6-trichlorophenol, 4-nitrophenol, and pentachlorophenol for their growth. An extensive investigation was performed to portray the kinetics of cell growth along with 4-CP degradation in the batch study utilizing 4-CP as substrate. Various unstructured models were applied to evaluate the intrinsic kinetic factors. Levenspiel's model demonstrates a comparatively enhanced R2 value (0.997) amongst every analyzed model. The data of specific growth rate (μ), saturation constant (KS), and YX/S were 0.11 h-1, 39.88 mg/L, along with 0.53 g/g, correspondingly. The isolated strain degrades 1,000 mg/L of 4-CP within 40 h. Therefore, B. subtilis MF447840.1 was considered a potential candidate for 4-CP degradation.

A method for rapid acute toxicity test based on temperature control of Ceriodaphnia dubia has been developed and evaluated. A new toxicity test based on temperature control (TTBTC) which are based on temperature control, was developed and compared for the adsorption of the better methodology for short-term toxicity screening. Initially, daphnid larval are exposed to toxicants and at the same time the temperature of the water bath containing media is increased to high temperature $(35.5^{\circ}C).$ After 1.25 hrs of contact time, the number of the daphnids, either living (no toxic effect) or dead (toxic effect), is counted by the naked eyes. Effect of exposure time on test sensitivity was not significantly different between 1 to 1.5 hr. Comparison of the rapid 1.25 hr acute toxicity test developed in this study and the standard 48 hr acute toxicity test using heavy metals, cyanide and pentachlorophenol indicated that the 1.25 hour test provides an acceptable level of sensitivity in toxicity test for C. dubia.
- Some properties of an extracellular cytosine deaminase excreted from an isolate from soil samples were examined after 20~80%' ammonium sulfate fractionation. The enzyme catalyzed the conversion of cytosine and 5-fluorocytosine into uracil and 5-fluorouracil by substrate specificity, respectively. The optimum temperature and storage time on the stability of the enzvme preparation were below $50^{\circ}C$ keeping above 90% of the residual activity and near 4 days keeping above 80% of the residual one in Tris-HCI buffer. The maximum activity was also obtained at 8.0 in pH and 37'C in temperature. The pHs and temperatures for enzyme activity ranged from 8.0~8.5 and from 37~$45^{\circ}C$. respectively. The presence of $Ag^-,Hg^{2-}, Zn^{2-}, Cu^{2-}, Sn^{2-}, \; or\; Pb^{2-}$ in the reaction mixture resulted in the marked inhibition in enzyme activity, but 1 mM of $K^+, Fe^[3+}, Mg^{2+}, \; or \; Na^+$. slightly increased the activity. The enzyme preparation was heavily affected by most of inhibitors tested such as 1 mM of EIITA, NaCN and pentachlorophenol, and completely inactivated by p-CMB and TCA of 1 mM, or 10 mM.
Cadmium pentachlorophenolxanthate has been synthesized with pentachlorophenol, carbondisulfide sodium hydroxide and cadmium chloride and its fungicidal activity was tested to Pyricularia oryzae and Cochliobolus miyabeanus. The results are summarized as followes; 1) The sample V synthesized by acidifying the crude PCP-xanthate solution at pH 8, then treated with $BaCl_2\;and\;CaC_2$ showed the highest purity, and the yield was $85.1\%$. 2) The same sample of $50\%$ wettable dust formulation inhibited conidial germination of P. oryzae and C. miyabeanus completely at 10 and 20 ppm, respectively. (Fig. 1) 3) The sample of $1.5\%$ dust formulation inhibited mycelial growth of P. oryzae and C. miyabeanus with 96 and $65\%$, respectively. The results were similar to those obtained by Ceresan calcium (Fig. 2) 4) Both dust and wettable powder formulations had no phytotoxcity on rice seedlings.
Leachate from municipal solid waste (MSW) landfill, effluent from leachate treatment plant, and ground water sample from a monitoring well near landfill site were tested for an acute toxicity. Microtox toxicity test was used for testing the acute toxicity of leachate and other samples. EC$_{50}$ values which a concentration of pollutant for reducing 50% light output from luminescent bacteria, Photobacterium phosphoreum were determined to assess the toxicity of pollutants as well as the relative toxicity. In addition, characteristics of leachate were studied and compared to those of phenol and pentachlorophenol (PCP) which are typical aquatic toxic pollutants. For leachate, EC$_{50}$ for 30 min incubation was 10.8%, while for phenol and PCP, 46 ppm and 1.2 ppm, respectively. the relative toxicity of treated leachate by in situ aeration with activated sludge was reduced to more than 75% of toxicity of the untreated leachate. Microtox toxicity test was failed to figure out EC$_{50}$ values for groundwater from a monitoring well since the relative toxicity of the unconcentrated sample was too low to estimate EC$_{50}$. Addition of activated carbon to leachate was reduced the relative toxicity. The reduction Pattern of the relative toxicity of leachate by mechanical aeration was similar to that of PCP, but different from that of phenol. These findings suggest that the toxicity of leachate may come from PCP-like toxic compounds rather than phenol-like one. In conclusion, the process of aeration with activated sludge might be very important to reduce the environmental toxicity of leachate. And Microtox test could be a reasonable bioassay for screening and monitoring the environmental toxicity of leachate from municipal solid waste landfill as well as for determining the reduction efficiency of the leachate toxicity by various treatment processes in leachate treatment plant.
