Development of L-Lysine Producing Strains from Cellulosic Substrate by the Intergeneric Protoplast Fusion- Conditions for Formation and Regeneration of Protoplast - (속간 원형질체 융합에 의한 섬유질 기질로부터 L-lysine 생산균주 개발 -원형질체의 형성 및 재생 -)

  • In order to produce L-lysine from cellulosic substrates by the intergeneric protoplast fusion between cellulolytic bacteria, Cellulomonas flavigena KFCC31221 and amino acid producing bacteria, Brevibacterium flavum ATCC14067, Corynebacteriurn glutamicum ATCC13032, conditions for protoplast formation and regeneration of these strains were investigated. After the strains were mutated with 500$\mu\textrm{g}$/$m\ell$ N-methyl-N'-nitro N-nitrosoguanidine for 30 min and the mutants were enriched by treating 300$\mu\textrm{g}$/$m\ell$ penicillin-G for 2 hrs, B. flavum Hse- Str$^{r}$ , C. glutamicum Met$^{-}$Thr$^{-}$ Rif$^{r}$ and Cellulomonas flavigena Thr$^{-}$Val$^{-}$Kan$^{r}$ were isolated. The rate of protoplast formation ranged from 95 to 98% when strains were treated at the concentration of 500$\mu\textrm{g}$/$m\ell$ of lysozyme, pH 6.5, 33$^{\circ}C$, for 6 hrs. in Tris- malate buffer supplemented with 0.4M sucrose as osmotic stabilizer. Approximately 30-33% protoplast was regenerated on the regeneration complete medium(RCM) containing 1.5% agar and 0.5M sodium succinate overlaid with the same medium except 0.7% agar.

  • The objective of this study was to develop a simultaneous method of 8 penicillin antibiotics including amoxicillin, ampicillin, cloxacillin, dicloxacillin, nafcillin, oxacillin, penicillin G and penicillin V in meat using LC-MS/MS. The procedure involves solid phase extraction with HLB cartridge and subsequent analysis by LC-MS/MS. To optimize MS analytical condition of 8 compounds, each parameter was established by multiple reaction monitoring in positive ion mode. The chromatographic separation was achieved on a $C_{18}$ column with a mobile phase of 0.05% formic acid and 0.05% formic acid in acetonitrile at a flow rate of 0.2 mL/min for 20 min with a gradient elution. The developed method was validated for specificity, linearity, accuracy and precision in beef, pork and chicken. The recoveries were 71.0~106%, and relative standard deviations (RSD) were 4.0~11.2%. The limit of detection (LOD) and the limit of quantification (LOQ) were 0.003~0.008 mg/kg and 0.01~0.03 mg/kg, respectively, that are below maximum residue limit (MRL) of the penicillins. This study also performed survey of residual penicillin antibiotics for 193 samples of beef, pork and chicken collected from 9 cities in Korea. Penicillins were not found in all the samples except a sample of pork which contained cloxacillin (concentration of 0.08 mg/kg) below the MRL (0.3 mg/kg).

  • A preterm newborn affected by congenital syphilis, born to mother not treated during pregnancy is described. The clinical picture was characterized by respiratory distress, cutaneous manifestations, massive hepatosplenomegaly, severe anemia, thrombocytopenia, disseminated intravascular coagulation syndrome and hypoalbuminemia. The patient was treated with daily injections of 190,500 units of crystalline penicillin G for 14 days. Premature infants with these symptoms and signs should be evaluated for congenital syphilis.

  • For producing single cell protein from the agricultural waste, heat treatment and antibiotics on the growth of Cellulomonas sp. KL-6, isolated in rotting leaf and the adjacent soil mixture, were examined. The organism was able to grow until 5 min. at $65^{\circ}C$, 1 min. at $75^{\circ}C$ and 1/4 min. at $85^{\circ}C$ in gradually rising temperatures. It can be Seen that preheating the suspension at $48^{\circ}C$ results in a marked decrease in heat resistance. On heating at temperature of $55^{\circ}C$ for 30 min., strain KL-6 was more resisted in the 0.1 M phosphate buffer when such substrates as casamino acid (1%), yeast extract (1%) or xylose (5%) were added to it whereas this organism was appeared weaker resistances in 0.1 M phosphate buffer when cysteine (0.03 M), sodium citrate (1%) or casein (1%) were in fused into it. Test strain was susceptible to penicillin-G $(1.563\;{\mu}g/ml)$ and ampicillin $(3.125\;{\mu}g/ml)$, but the organism was resisted to kanamycin $(>200\;{\mu}g/ml)$. The treatment of strain KL-6 with sodium dodecyl sulfate (SDS) resulted in the elimination of R-plasmid from the host strain and the elimination rate with SDS $(10{\sim}30\;{\mu}g/ml)$ was about $9.2{\sim}31.2%$, respectively.

Characteristics of Thirty-Six Bacillus thuringiensis Isolates and a New Serovar of B. thuringiensis subsp. kim (Serotype H52)

  • Thirty-seven strains of Bacillus thuringiensis were isolated from Korean soil and examined for H-antigen serotyping, toxicity, and different spectra of biological activities. The isolate HL-175 bore a specific H-antigen, different from the 51 known serotypes, a spherical $\delta$-endotoxin crystal, and minor different biochemical characteristics. It was resistant to ampicillin, colistin, and penicillin G. Therefore, it was classified as a new serotype, H52, with the name kim. The other 36 isolates also produced endotoxin crystals and endospores. The crystal shape of eight strains was cuboidal while the others were bipyramidal. Biochemical characteristics of the isolates were only slightly different from the known serotypes of B. thuringiensis. The flagellar (H) antigens of the 36 isolates were identified as: one colmeri (H21), three galleriae (H5a,5b); two pakistani (H13); one toumanoffi (H11a, 11b); and twenty-nine kurstaki (H3a,3b). All 36 isolates were resistant to ampicillin, colistin, penicillin, cephalothin, and chloramphenicol.

  • ${\beta}-Lactamase$ was isolated from the culture filtrate of the penicillin producing strain, Penicillium chrysogenum Q176. When the pH of the medium was adjusted to 5.0 at the start of culture, a rapid increase in pH accompanied by the synthesis of penicillin was observed in the first $2{\sim}4$ days. When the pH of medium was brought to 6.0 or 7.0 the opposite was observed: high yield of the enzyme and little of the antibiotics in the medium. The optimum enzyme activi­ty was at a temperature of $40^{\circ}C$ and around pH 7.0. A partially purified enzyme was assayed on several different substrates including penicillins V and G, 6-aminopenicillanic acid, cephalospo­rin C. The V max values calculated were 24.5, 20.4, 7.6, and 6.1 mmoles/hour, and the $K_m$, values were 16.4, 12.6, 7.5, and 6.9 mM in the order given.

Antimicrobial Susceptibility Patterns of Staphylococcus intermedius Isolated from Skin Lesions of Thoroughbred Horse. (더러브렛 말의 피부병변에서 분리한 Staphylococcus intermedius의 약제 감수성 양상)

  • Staphylococcus intermedius (S. intermedius) has been rarely isolated from horse. In this study, we investigated the antimicrobial susceptibility patterns of S. intermedius isolated from skin lesions of Thoroughbred horse in Daegu Equestrian Association. The skin lesions were showed with dehairing and slight purulent inflammation. Bacteria were isolated from skin lesions and identified as S. intermedius by biochemical tests and MicroLog (BIOLOG, California, USA). Antimicrobial susceptibility test of S. intemedius isolates was performed with 33 antimicrobial agents (BBL, Maryland, USA) by using the agar disk diffusion method. It showed a high sensibility in the amikacin, amoxycillin/clavuramic acid, ampicillin, cefoxitin, cephalothin, chloramphenicol, ciprofloxacin, enrofloxacin, florfenicol, kanamycin, neomycin, nitrofurantoin, norfloxacin, ofloxacin, oxacillin, penicillin G, tetracycline and vancomycin. The horse was treated with penicillin, and cured completely after two weeks. The present results showed valuable information for treatment and prevention of skin disease in horse.

Psammaplin A, a Natural Bromotyrosine Derivative from a Sponge, Possesses the Antibacterial Activity against Methicillin-resistant Staphylococcus aureus and the DNA Gyrase-inhibitory Activity

  • Psammaplin A, a natural bromotyrosine derivative from an associated form of two sponges (Poecillastra sp. and jaspis sp.) was found to possess the antimicrobial effect on the Gram-positive bacteria, especially on methicillin-resistant Staphylococcus aureus (MRSA). The minimal inhibitory concentration of psammaplin A against twenty one MRSAs ranged from 0.781 to 6.25 ${\mu}g/ml$, which that of ciprofloxacin was 0.391~3.125${\mu}g/ml$. Psammaplin A could not bind to penicillin binding protein, but inhibited the DNA synthesis and the DNA gyrase activity with the respective 50% (DNA synthesis) and 100% (DNA gyrase) inhibitory concentration 2.83 and 100 ${\mu}g/ml$. These results indicate that psammaplin A has a considerable antibacterial activity, although restricted to a somewhat narrow range of bacteria, probably by inhibiting DNA gyrase.

  • A source of D-xylose was required for the enhanced production of D-glucose isomerase of Streptomyces sp. strain K-17. D-glucose supported the luxuriant growth of the organism as well as D-xylose, but D-glucose isomerase activity was hardly detected in the D-glucose-grown cells. When the D-glucose-grown cells were incubated aerobically for a few hours in 0.5% xylose solution in 0.05 M phosphate buffer, pH 7.0, it was found that inductive formation of D-glucose isomerase occurred in the cells without multiplication. In the non-growth phase of cells the inductive formation of D-glucose isomerase occurred because a source of nitrogen for the synthesis of enzymes was obtained from turnover of protein accumulated in cells. D-ribose, L-arabinose, D-glucose, D-mannose, citrate, succinate and tartrate could not induce the formation of D-glucose isomerase, but D-xylose could induce. Inductinn of D-glucose isomerase was repressed by D-glucose and its catabolites : glycerol, succinate and citrate. Inductive formation of the enzymes in the non-growth phase was stimulated by $Ba^{2+}$, $Mg^{2+}$ and $Co^{2+}$, and inhibited by C $u^{2+}$, C $d^{2+}$, A $g^{+}$and H $g^{2+}$. The synthesis of enzymes in the induction system composed of 0.5% xylose solution was disrupted by actinomycin D, streptomycin, chloramphenicol, kanamycin, tetracycline, p-chloromercuribenzo ate, arsenate and 2, 4-dinitrophenol, but not disrupted by mitomycin C and penicillin G.icillin G.

Antibiotics Susceptability of Streptococcus pneumoniae Isolated from Pharynx in Healthy Korean Children and Choice of Proper Empirical Oral Antibiotics Using Pharmacokinetics/Pharmacodynamics Model (국내의 소아에서 분리된 폐구균의 항생제 감수성 양상 및 약력동학 모델을 이용한 적절한 항생제의 선택)

  • Purpose : Pneumococcus is one of the most important causes of invasive infection through the childhood period. In January 2008, the Clinical and Laboratory Standards Institute (CLSI) published revised penicillin breakpoints for Streptococcus pneumoniae and penicillin susceptibility rates of S. pneumoniae increased in Korea. This study was performed to determine the probability of oral amoxicillin for the empirical treatment achieving bactericidal exposure against pneumococcus using pharmacodynamics model. Methods : Twenty-three isolates of pneumococci were subjected to determine minimum inhibitory concentration (MIC) for ${\beta}$-lactams and macrolide. For the ${\beta}$-lactams, exposure of fT >MIC (time that free drug concentrations remain above the MIC) for 50% of the administration interval have determined the probability of target attainment (PTA), and regimens that had a PTA >90% were considered optimal. An analysis was performed by applying MIC of 23 isolates to a 5000-patient Monte Carlo simulation model. Results : Among 23 isolates from healthy children, 7 (30.4%) isolates were MIC ${\leq}$1.0 ${\mu}g$/mL and 19 (82.6%) were MIC ${\leq}$2 ${\mu}g$/mL for amoxicillin. Amoxicillin 40 mg/kg/day achieved PTA >90% at MIC ${\leq}$1.0 ${\mu}g$/mL but PTA decreased to 52% at MIC 2 ${\mu}g$/mL, whereas amoxicillin 90 mg/kg/day can predict 97% of PTA at MIC 2 ${\mu}g$/mL. Overall, oral amoxicillin 90 mg/ kg/day for the empirical treatment against pneumococcus can expect more successful response in Korean children. Conclusion : Considering the resistantce pattern of pneumococci in Korean children, we estimate that oral amoxicillin 90 mg/kg/day will provide a pharmacodynamic advantage for the empirical treatment against pneumococcus. And low dose amoxicillin or macrolide are expected to have higher chance of treatment failure than high dose oral amoxicillin.
