Changes of Non-Cellulosic Neutral Sugars of Cell Wall in Soybean Sprouts (콩나물 생장중 세포벽 비섬유성 중성당의 변화)

  • This study was carried out to investigate the changes and composition of the non-cellulosic neutral sugars in cell wall of soybean sprouts during growth. The composition of non-cellulosic neutral sugars in cell of soybean sprouts was rhamnose, fucose, xylose, arabinose, mannose, galactose and glucose. The galactose content of cell wall was higher than other non-cellulosic neutral sugars, and was remarkably decreased during growth. The major non-cellulosic sugars of pectic substances were rhamnose, arabinose, and galactose. The arabinose content of pectic substance was increased in cotyledon and hypocotyl during growth. The contents of non-cellulosic neutral sugars were decreased in hypocotyl during growth. The galactose content of pectic substance was higher in cotyledon than those in hypocotyl, and was increased in cotyledon. The content of rhamnose was higher in ionically associated pectic substance than that in covalently bounded pectic substance. The major non-cellulosic neutral sugars of hemicellulose were glucose, rhamnose, arabinose and galactose. The galactose of hemicellulose was decreased remarkably during growth.

Physicochemical Properties and Intestinal Bacterial Growth-Promoting Effect of Cell-Wall Polysaccharides from Cucumber Peel

  • Physicochemical properties, intestinal microbial growth, and inhibitory effects of alcohol-insoluble polysaccharide (AIP) extracted from cucumber peel were investigated. AIP was composed of 14.54% crude protein, 1.04% crude lipid, 13.74 % crude ash, 9.1% soluble dietary fiber, and 41.2% insoluble dietary fiber. AIP showed low bulk density (0.18 g/mL) and water-holding capacity (6.39 g/g), and high oil-holding capacity (3.96 g/g). Pectic substance fractions [water-soluble pectic substance (WSP), ethylenediaminetetraacetic acid-soluble pectic substance (ESP), and alkali-soluble pectic substances (ASP)] and hemicellulose fractions [1 M KOH-soluble hemicellulose (KHP1) and 4 M KOH-soluble hemicellulose (KHP4)] were obtained from sequential chemical fractionation of AIP. WSP showed higher total sugar contents than total uronic acid contents, whereas opposite results were observed in ESP and ASP. Molecular weight distributions of three pectic substance fractions were in order of ASP>ESP>WSP. Ion exchange chromatogram pattern of WSP was different from those of ESP and ASP. Major component of WSP was fraction eluted by 0.05 M ammonium acetate buffer, whereas that of ESP and ASP was fraction eluted by 0.2 M NaOH. WSP and ASP showed growth-promoting activities against Lactobacillus brevis, Bifidobacterium bifidum, and B. longum, whereas B. bifidum and B. longum for ESP. KHP1 and KHP4 fractions had significant growth-promoting activities against B. bifidum.

Softening Related Changes in Cell Wall Pectin of Hot Pepper Fruit (고추과실 세포벽 Pectin질의 연화에 따른 변화)

  • Pectic substance of hot pepper crude cell wall was fractionated during ripening and softening. The pectic substance was divided ionically associated pectin(IAP) and covalently bounded pectin with hemicellulose(CBP). And then, the composition and the modification of the pectic substance using gel filtration chromatography were studied. The polyuronide associated with the CBP increased owing to decreasing of pectin bound hemicellulose linked to galactose by the turning stage showed softening. The molecular weight profiles of IAP shifted from low molecular weight profiles of IAP shifted from low molecular weight to high molecular weight polymers. While, the CBP changed from high molecular weight to low molecular weight polymers during ripening. It is suggested that the changes in the pectic substance associated with fruit softening should involve the alteration of CBP-bound hemicellulose like galactan.

  • Pectin, hesperidin and naringin were extracted from hot air-dried peel ($60^{\circ}C$, 1 hr and air velocity 160 fpm) of citrus produced in Korea in order to see the amount of each component contained in the peel. Pectin was extracted by three different methods and the quality and contents of the pectins were determined respectively. 1. The pectin yield by the total pectic substance method was the highest (26.0% for unshiu (U) and 28.5% for natsudaidai (N) expectedly and the soluble pectic substance method the least (13.5%(U) and 15.6% (N)) The yield by method III (extraction by water at pH 1.5 followed by isopropanol precipitation) was intermediate (18.1% (U) and 20.8%(N)). Anhydrouronic acid (AUA) content was the highest (92.0% (U) and 90.3%(N)) in those by method III. The AUA contents of the other pectins were 80.0% for soluble pectin (for both U and N), 71.6% for the commercial pectin (Sunkist Groups Inc., U.S.A.), 58.0%(N) and 63.4%(U) for total pectic substance. 2. The methoxyl content of total pectic substance was the lowest (4.81%(U) and 4.88%(N)). However, there was no significant difference in methoxyl content among the rest which were found to have low levels(5.27-7.20%). 3. The pectin by method III gave the highest jelly strength. The commercial pectin, soluble pectice substance and total pectic substance were next in order. 4. The hesperidin content of unshiu was 5.07% (dry basis) and the naringin content of natsudaidai 3.03% (dry basis).

  • The changesin hardness activities of pectinestrase and polygaacturonase and amounts of pectic substances of cucumber during salting at 1$0^{\circ}C$ were investigated, The hardness of the cucumber was decreased dramatically after 3 weeks whereas activities of pectinestrase and polygalacturonase were increased until 3 weeks and 2 weeks respectively and then decrea-sed. The level of alcohol insoluble solid and protopectin in the cucumber were decreased but those of pectic acid and water soluble pectin were increased during the whole salting periods, Protopectin fractionated from alcohol insoluble solid during salting of cucumber was separated using Sephacryl S-500 It showed that high average molecular weight(AMW) of 100,000 was decreased however lower molecular weight compounds was increased. Pectic acid was observed to be decomposed from AMW 200,000 to AMW 500,000 Water soluble pectin from fresh cucumber contained higher level of pentose with peak I of AMW 2,000,000, however after 6 weeks of saltinf peak II which represented AMW 100,000 was separated. From the changes of sugar composition, the phenomena of softness during the salting was probably caused by solubilization of hemicellulose associated with pectin and decomposition of pectic substances.

  • This paper was carried out to investigate changes in cell wall, cell wall polysaccharides, pectic substances extracted from cell wall of persimmon fruits treated with polygalacturonase and $\beta$-galactosidase in vitro. Degrading degree of cell wall treated with cell wall-degrading enzymes were higher in order polygalacturonase, polygalacturonase+$\beta$-galactosidase and $\beta$-galactosidase. Contents of soluble pectic substances in cell wall treated with cell wall-degrading enzymes showed as the same order as degrading degree of cell wall, while contents of insoluble pectin lower. Contents of versene-soluble pectin and total pectic substance were not affected by cell wall-degrading enzymes. Contents of uronic acid and hexose in soluble material isolated from cell wall treated with polygalacturonase and mixed enzyme were higher than those of untreatment and $\beta$-galactosidase treatment.

  • The changes of methoxy content. compositional sugars. degree of esterification (DE) and pectic substances such as LMP (low methoxy pectin), HMP (high methoxy pectin) IAP (ionically associated pectin) and CBP (covalently bounded pectin) of Fuji apple were investigated during maturity. The contents of LMP+HMP and IAP+CBP in alcohol insoluble solid(AIS) were 70% and 20%. respectively. These pectic substances were increased slightly during maturity. The reductions of hexose and uronic acid were Iron 20-50% and from 2-4%, respectively, but the inclosed of pentose was from 1-9% in LMP, HMP, IAP and CBP during maturity of Fuji apple. The galactose was decreased significantly among hexose. Whereas the relative ratio of uronic acid in HMP and CBP was increased.

  • This study was carried out to investigate changes in the cell wall components of peach during maturation and storage. The hardness of peach was decreased from $24.1\;\pm\;2.2N\;to\;14.6\;\pm\;1.3N$ during maturation and storage. Contents of alcohol-insoluble substance, water-soluble material and cell wall were decreased. Total and insoluble pectic substances were decreased while water-soluble pectin was increased during maturation and storage. Contents of cellulose were increased. Galactose and arabinose contents were decreased during maturation and storage.

  • Pectolytic enzymes were extracted in apple fruits rotted by Botryosphaeria dothidea, and their activities and change of pectic substances were investigated. Exo-polygalacturonase(exo-PG), exo-polymethylgalacturonase(exe-PMG), polygalacturonate-trans-eliminase(PGTE) and pectin-methyl-trans-eliminase(PMTE) were produced by the pathogen. Activities of exo-PG and exe-PMG extracted from rotten apple fruits were high to 21.15 and 24.65 units/mg protein in specific activity at seven days after inoculation, respectively. Activities of PGTE and PMTE showed 5.60 and 7.90 units/mg protein, respectively, but they were lower than those of the exo-type enzymes. Water-soluble and versene-soluble pectins were 11.50 mg/100 mg-AIS and 7.31 mg/100 mg-AIS at 14 days after inoculation, namely, they were increased by 4.23 and 2.16 mg/100 mg-AIS over those of sound apples, respectively. Total soluble pectic substances of rotten apple were 72.4% of total pectic substances and it was higher by 24.8% than sound apple. Insoluble pectic substance was notably decreased from 15.32 to 7.16 mg/100 mg-AIS according to progress of decay while total pectic substances were not changed remarkably.

  • For the purpose of fermentation control and edible period extension of lower salted Kimchi, the contents and compositions of pectic substances in Kimchi, by adding 2 percent salt and 0.4 percent sodium malate buffer(SMB), fermented at $20^{\circ}C$ were investigated. Edible period of SMB added chinese cabbage Kimchi was extended 40 hrs compared to that of control (added 2.5% salt) with good flavor, texture and freshness. The contents of alcohol insoluble solid(AIS) and protopectin(PP) of control were more decrensed during fermentation than those of SMB added Kimchi. But, the contents of pectic adid(PA) and water soluble pectin(WSP) of control were more increased during fermentation than those of SMB added Kimchi. Hexose and pentose from hemicellulose in control, PP and PA respectively, were more decreased during fermentation than those in SMB added Kimchi. Lower polarity and higher molecular weight PP was eluted and higher polarity and lower molecular weight PP was decreased considerably in control compared to those in SMB added Kimchi during fermentation.
