Isolation and identification of Cryptosporidium from various animals in Korea II. Identification of Cruptosporidium muyis from mice (한국산 동물로부터 크립토스포리디움의 분리 및 동정 II. 마우스로부터 Cryptosporidium muris의 분리)

  • Each of SPF mice(Scl: ICR strain, 3-week-old males) was inoculated with 5$\times$104 oocysts of Cryptosporidium by stomach tube. The oocysts were large type one which was previously isolated from Korean mice, and passaged in 3-week-old SPF mice. The patterns of oocyst discharge were monitored daily, and in order to observe the ultrastructure of developmental stages the stomach of the mice was examined by transmission electron microscopy (TEM) at 4 weeks post-inoculation. The prepatent period for 6 mice was 5.6 days post-inoculation on the average, and the patent period was 63.2 days. The number of oocysts discharged per day from the mice reached peak on day 36.6 post-inoculation on the average. A large number of oocysts were found in fecal samples obtained from inoculated mice on days 30~50 post-inoculation. C. tsuris was larger than C. parvum at almost every developmental stages, the sixte difference being 1.4 times in oocysts, 2.4 times in sporozoites, 1.6 times in merozoites, and 1.5 times in microgametes. The ultrastructural features of the attachment site of C. tsuris to the mucus cells were remarkably different from those of C. parvum and its closely related species. The anterior projection of the protozoa (C. muris), the outer aspect of which was surrounded by a thick filamentous process of the host cell, has not been reported at any developmental stages of C. parvum or its closely related species. The size of the oocysts of strain RN 66 was larger than that of Korean mice origin. The above results reveal that the large type Cryptosporidium of Korean mice origin is identified as Cryptosporidium muris and this type was named as C. muris (strain MCR).

A Study on the Salinity Variation of Salt Water in an Estuary (하구(河口)의 해수(海水)의 염도변동(鹽度變動)에 관한 연구(研究) - 군산외항(群山外港)부근을 중심(中心)으로 -)

  • Since the estuary is a very complex place in which the sea water and the fresh water meet, it is very difficult to make a general analytical description of salinity distribution in the estuary. As an attempt to investigate the characteristics of salinity variation in the estuary of the Geum River, the field observations were continuously carried out at three points near the Gunsan New Harbor at the time intervals 1 to 1.5 hours during one tidal cycle and the data were analysed. The following results were obtained; 1. It was reconfirmed that most of the ratios of the salinity to the conductivity were widely distributed between the range of 0.5 to 1.0. 2. The salinity showed the peak at the high water, and then it began to decrease gradually and had the lowest value 0 to 2 hours after the low water. 3. The density current was generally the intense mixing type and when the river discharge was very large it was of the moderate type. 4. The vertical salinity distribution was not significantly affected by the wave height. 5. The maximum vertical salinity differences were generally less than 10 g/l and the time of the occurrence of the minimum value was 0 to 3 hours after the low water when in the spring tide and in the neap tide it occurred 2 to 3 hours after the high water.

Analysis of Hydraulic Characteristics of Flood Plain Using Two-Dimensional Unsteady Model (2차원 부정류 모형을 이용한 둔치의 수리특성 분석)

  • Since the cross-sectional shape of the Nakdong river is compound type, the water stage rises up to the top of the flood plane, as the flow discharge increases during the extreme rain storm in summer. The recent increase of rainfall intensity and flood frequency results in the immersions of parks and hydrophilic facilities located in the flood plain. Therefore it is necessary to analyze the hydraulic characteristics evolved by the extreme rain storm in the flood plain. The study reach ranging from the Gangjeong Goryeong Weir and the Dalseong Weir, where several hydraulic facilities are located along the channel, was selected and numerical simulations were conducted for 42 hours including the peak flood of the typhoon Sanba. The 2-D transient model, FaSTMECH was employed and the accuracy of the model was assessed by comparing the water level between the simulation results and the measured ones at a gauging station. It showed a high correlation with $R^2$ of 0.990, AME of 0.195, and RMSE of 0.252. In addition, the inundation time, the inundation depth, the inundation velocity, and the shear stress variation in the flood plain facilities were analyzed.

Sensitivity Analysis of Runoff-Quality Parameters in the Urban Basin (도시 배수유역의 유출-수질 특성인자의 민감도 분석)

  • The purpose of the study is to analyze the sensitivity of the parameters that affect the runoff and water quality in the studied drainage basins. SWMM model is applied to the four drainage basins located at Namgazwa and Sanbon in Seoul and Gray Haven and Kings Creek in the USA. first of all, the optimum values of the parameters which have least simulation error to the observed data, are detected by iteration procedure. These are used as the standard values which are compared against the procedure. These are used as the standard values which are compared against the varied parameter values. In order to catch the effectiveness of the parameters to the computing result, the parameters are changed step by setp, and the results are compared to the standard results in flowerate and quality of the sewer. The study indicates that the discharge is greatly affected by the types of runoff surface, i.e., impervious area remarkably affects the peak flow and runoff volume while the surface storage affects the runoff volume at mild sloped basins. In addition, the major parameters affecting the pollution concentrations and loadings are the contaminant accumulation coefficient per unit area per time and the continuous dry weather days. Furthermore, the factors that affect the water quality during the initial rainfall period are the rainfall intensity, transport capacity coefficient and its power coefficient. Consequently, in order to simulate the runoff-water quality, it is needed to evaluate previous data in the research performed for the studied basins. To accurately estimated from the tributary areas and the rational computation methods of the pollutants calculation should be introduced.

Regional frequency analysis using spatial data extension method : II .Flood frequency inference for ungaged watersheds (공간확장자료를 이용한 지역빈도분석 : II. 미계측 유역의 홍수빈도 추론)

  • In order to infer regional flood frequency for ungauged watersheds, index flood method was applied for this study. To pursuit this given purpose, annual peak flood data for 22 watersheds located at the upstream of the Chungju Dam watershed were obtained from the spatial extension technique. The regionalization of mean annual flood was performed from extended flood data at 22 points. Based on the theory that flood discharge and watershed size follows the power law the regionalization generated the empirical relationship. These analyses were executed for the full size of the Chungju Dam watershed as one group and three different mid-size watersheds groups. From the results, the relationship between mean annual flood and watershed sizes follow the power law. We demonstrated that it is appropriate to use the relationship between specific flood discharges from the upper and lower watersheds in terms of estimating the floods for the ungaged watersheds. Therefore, not only the procedure of regional frequency analysis but also regionalizaion analaysis using finer discretization of the regions interest with respect to the regional frequency analyisis for the ungauged watersheds is important.

  • Wastewater treatment (WWT) technology has been developing from simple pollutant treatment to energy and resource-saving advanced technology, and various technologies combined with IT and BT are developed to minimize the amount of pollutant and toxic substance discharge to the public water areas and to improve operational efficiency. To examine the development trend of domestic wastewater treatment technology, the registered patent technologies were surveyed, classified and analyzed by year and sector. This paper considers the status of patent registration related to WWT from 2010.1 to 2017.5 in terms of the number of specific technical areas, and the trends are analyzed based on the 10 categorization field such as biological and physicochemical treatment process, equipment and device, material, sludge treatment, membrane, process control and 42 specific technical areas. A total of 3,356 patents have been registered since 2010, and the number of patents has been decreasing since the peak at 2013 and maintains 3~400 per year. The total number of patents has not yet been less than other countries, but the number of patents of more advanced technologies, which can lead the global market, such as process monitoring, new concept processing and equipment technologies is still insufficient compared to developed countries.

Flood Simulation with the Variation of Runoff Coefficient in Tank Model (탱크모형의 流出孔 乘數 변화를 고려한 홍수모의)

  • Rainfall intensity under storms affects peak discharge or its time of occurrence in watershed runoff. Thus, it is reasonable to reflect the effect on the parameters of rainfall-runoff models or the governing equations of the models. This paper relates the change of the runoff coefficient of the first tank in tank model to rainfall intensity under storms. The standard four tanks have made the basic structure of the flood event model. and its modifications are as follows: it has two equal runoff coefficients in the first tank: the runoffs from first and second tanks produce delayed response through a simple delaying parameter. Applying the event simulation model to flood data from Naerinchon. runoff coefficients were estimated and their relation to rainfall intensity was analyzed. The results showed the Weak relation of the two factors. The trend of the two was fitted with the equation a1=kI$. where a1is the runoff coefficient of the first tank: I is rainfall intensity; k and m are fitting coefficients. In the verification. the model used moving averages for the calculation of I(t). If the value I(t) gave more greater value of a1(t) than that of previous time(t-1). the flood simulation was performed again from the beginning with the updated greater value of a1. The reflection of rainfall intensity on the runoff coefficient showed far better results than that of a fixed parameter.

Flood Simulation using Vflo and Radar Rainfall Adjustment Data by Statistical Objective Analysis (통계적 객관 분석법에 의한 레이더강우 보정 및 Vflo를 이용한 홍수모의)

  • Recently, the use of radar rainfall data that can help tracking of the development and movement of rainfall's spatial distribution is drawing much attention in hydrology. The reliability of existing radar rainfall compared to gauge rainfall data on the ground has not yet been confirmed and so we have difficulties to apply the radar rainfall in hydrology. The radar rainfall for the applications in hydrology are adjusted merging method derived from gage. This study uses the Mean-Field Bias (MFB) and Statistical Objective Analysis (SOA) as correction methods to create adjusted grid-based radar rainfall data which can represent the temporal and spatial distribution of rainfall. This study used a storm event occurred in August 2010 for the adjustment of radar rainfall. In addition, the grid-based distributed rainfall-runoff model (Vflo), which enables more detailed examinations of spatial flux changes in the basin rather than the lumped hydrological models, has been applied to Gamcheon river basin which is a tributary of Nakdong River located in south-eastern part of the Korean peninsular and the basin area is $1005km^2$. The simulated runoff was compared with the observed runoff in an attempt to evaluate the usability of radar rainfall data and the reliability of the correction methods. The error range of peak discharge using each correction method was within 20 percent and the efficiency of the model was between 60 and 80 percent. In particular, the SOA method showed better results than MFB method. Therefore, the SOA method could be used for the adjustment of grid-based radar rainfall and the adjusted radar rainfall can be used as an input data of rainfall-runoff models.

Electrochemical Performances of Acid-Treated and Pyrolyzed Cokes According to Acid Treatment Time (산처리 시간별 산화 코크스와 열분해 코크스의 전기화학적 거동)

  • As an activation procedure, in this study, the oxidation treatment of needle cokes with a dilute nitric acid and sodium chlorate $(NaClO_3)$, combined with heat treatment, was attempted. The structures of acid-treated and pyrolyzed coke were examined with XRD, FESEM, elemental analyzer, BET, and Raman spectroscopy. The behavior of double layer capacitance was investigated with the analysis of charge and discharge. The structure of needle coke treated with acid was revealed to a single phase of (001) diffraction peak after 24 h. On the other hand, thecoke oxidized by heat treatment was reduced to a graphite structure of (002) at $300^{\circ}C$. The distorted graphene layer structure, derived from the process of oxidation and reduction of the inter-layer, makes the pores by the electric field activation at the first charge, and generates the double layer capacitance from the second charge. The cell using pyrolyzed coke with 24 h acid treatment and $300^{\circ}C$ heat treatment exhibited the maximum capacitance per weight and volume of 33 F/g and 30 F/mL at the two-electrode system in the potential range of 0~2.5 V.

An Analysis of Spectral Characteristic Information on the Water Level Changes and Bed Materials (수위변화에 따른 하상재료의 분광특성정보 분석)

  • The purpose of this study is to analyze the reflectance of bed materials according to changes in the water level using a drone-based hyperspectral sensor. For this purpose, we took hyperspectral images of bed materials such as soil, gravel, cobble, reed, and vegetation to compare and analyze the spectral data of each material. To adjust the water level, we constructed an experimental channel to control the discharge and installed the bed materials within the channel. In this study, we configured 3 cases according to the water level (0.0 m, 0.3 m, 0.6 m). After the imaging process, we used the mean value of 10 points for each bed material as analytical data. According to the analysis, each material showed a similar reflectance by wavelength and the intrinsic reflectance characteristics of each material were shown in the visible and near-infrared region. Also, the deeper the water level, the lower the peak reflectance in the visible and near-infrared region, and the rate of decrease differed depending on the bed material. We expect the intrinsic properties of these bed materials to be used as basic research data to evaluate river environments in the future.
