백서 전치 주기적절제시 치수내 LDH ISOENZYME에 관한 연구 (LACTIC ACID DEHYDROGENASE ISOENZYME IN THE PERIODIC AMPUTATED RAT INCISOR PULP)

  • 백서의 전치를 주기적으로 절제후 치수내의 LDH isoenzyme을 전기영동법으로 분리하고 그의 활성도를 측정하여 정상과 비교관찰한바 다음과 같은 결론을 얻었다. 1. 정상치수내에서 5개의 LDH isoenzyme의 존재를 확인하였다. 2. LDH 활성도는 주기적으로 절제한 치아의 치수가 정상치수에 비해 훨씬 증가하였다. 3. 치아를 주기적으로 절제함으로써 치수내 LDH isoenzyme의 Pattern을 변화시킬 수가 있다.

Neuroprotective Effects of Ginsenoside Rg3 against 24-OH-cholesterol-induced Cytotoxicity in Cortical Neurons

  • Ginsenoside $Rg_3$ ($Rg_3$), one of the active ingredients in Panax ginseng, attenuates NMDA receptor-mediated currents in vitro and antagonizes NMDA receptors through a glycine modulatory site in rat cultured hippocampal neurons. In the present study, we examined the neuroprotective effects of $Rg_3$ on 24-hydroxycholesterol (24-OH-chol)-induced cytotoxicity in vitro. The results showed that $Rg_3$ treatment significantly and dose-dependently inhibited 24-OH-chol-induced cell death in rat cultured cortical neurons, with an $IC_{50}$ value of $28.7{\pm}7.5\;{\mu}m$. Furthermore, the $Rg_3$ treatment not only significantly reduced DNA damage, but also dose-dependently attenuated 24-OH-chol-induced caspase-3 activity. To study the mechanisms underlying the in vitro neuroprotective effects of $Rg_3$ against 25-OH-chol-induced cytotoxicity, we also examined the effect of $Rg_3$ on intracellular $Ca^{2+}$ elevations in cultured neurons and found that $Rg_3$ treatment dose-dependently inhibited increases in intracellular $Ca^{2+}$, with an $IC_{50}$ value of $40.37{\pm}12.88\;{\mu}m$. Additionally, $Rg_3$ treatment dose-dependently inhibited apoptosis with an $IC_{50}$ of $47.3{\pm}14.2\;{\mu}m$. Finally, after confirming the protective effect of $Rg_3$ using a terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling assay, we found that $Rg_3$ is an active component in ginseng-mediated neuroprotection. These results collectively indicate that $Rg_3$-induced neuroprotection against 24-OH-chol in rat cortical neurons might be achieved via inhibition of a 24-OH-chol-mediated $Ca^{2+}$ channel. This is the first report to employ cortical neurons to study the neuroprotective effects of $Rg_3$ against 24-OH-chol. In conclusion, $Rg_3$ was effective for protecting cells against 24-OH-chol-induced cytotoxicity in rat cortical neurons. This protective ability makes $Rg_3$ a promising agent in pathologies implicating neurodegeneration such as apoptosis or neuronal cell death.

Influence of Prenatal Noise and Music on the Expressions of c-Fos and Nitric Oxide Synthase in the Hippocampus of Rat Pups

  • The expressions of c-Fos and nitric oxide synthase (NOS) represent neuronal activity and play' a crucial role in the shaping of the development of brain. During the late pregnancy, stresses may influence neuronal activity of prenatal rats. In the present study, the effects of prenatal noise and music on the expressions of c-Fos and NOS in the hippocampus of rat pups were investigated. Exposure to the noise during pregnancy decreased c-Fos and NOS expressions in the hippocampus of rat pups, whereas exposure to music during pregnancy increased c-Fos and NOS expressions in the hippocampus of rat pups. The present results show that prenatal music stimulation may increase neuronal activity of rat offspring, whereas exposure to noise during pregnancy may reduce the neuronal activity of offspring. The present study suggests that prenatal stimuli including noise and music could affect the fetal brain development.

시스플라틴 이독성에서 사물탕의 보호효과 (Protective Effect of Samul against Cisplatin in Primary Rat Organ of Corti Explant)

  • The water extracts of Samultang (Samul) has been used for treatment of ischemic heart and brain damage in Oriental traditional medicine. However, little is known about the mechanism by which the water extract of Samul rescues cells from oxidative damages in cisplatin-induced ototoxicity. Cisplatin is a widely used chemotherapeutic agent that is also highly ototoxic. This study was designed to investigate the protective effects of Samul on ciplatin-induced ototoxicity in HEI-OC1 auditory cells and organ of Corti explant culture. Cisplatin markedly decreased the viability of HEI-OC1 auditory cells. However, treatment of HEI-OC1 cells with Samul significantly reduced cisplatin-induced cell death and apoptotic characteristics through reduction of intracellular peroxide generation. Cisplatin induced cytotoxicity in isolated and cultured hair cell progenitors from postnatal rat cochleae. These progenitor cells are isolated from the lesser epithelial ridge (LER, or outer spiral sulcus cell) area of pre-plated neonatal rat cochlear segments. However, Samul completely protected the morphological changes of organ of Corti and LER. Taken together, these data suggest that the protective effects of the water extracts of Samul against cisplatin may be mediated by the reduction of intracellular peroxide generation.
