세균성 벼알마름병의 연구동향 (Current Status of Bacterial Grain Rot of Rice in Korea)

  • The grain rot of caused by Bukholderia glumae was fist reported in japan in 1955 and then reported in other countries as well as in Korea in 1986. The pathogen causes both seedling and grain rot of rice but it cannot attack any other parts of adult rice plant. Bacterial colonies grow slowly, and are circular and greyish white. The causal bacterium is Gram-negative and rod shape with 1-3 polar flagella, and produce a diffusible yellow-greenish nonfluorescent pigment on King's medium B. Biochemical characteristics such as negative in arginine dehydrolase, oxidase reaction and nitrate reduction and positive in lecithinase, and the utilization of L-arginine and inositol are useful in differentiation of this from other nonfluorescent bacteria pathogenic to rice. This pathogenic bacterium had belonged to the genus of Pseudomonas but recently was transferred to the new genus Burkholderia on the basis of physiological characteristics and DNA-DNA hybridization data. However, other characteristics such as colony heterogenicity or colonial variation after subcultures, phytotoxin, secreting antibiotics, and relationship between yellow greenish pigment production and pathogenicity need to be clarified more. To develop an effective control strategy for this disease, understanding of detailed life cycle of the disease and critical environmental factors affecting disease development is prerequisite. Although 5,435 ha of rice paddy in Korea was infested during 1998, there is no exact estimation of yield losses and distribution of the pathogen. The review will focus on recent progress on the understanding of the bacteriological and ecological characteristics of the causal bacterium and control means of the disease.

Value of clay as a supplement to swine diets

  • The use of practical management factors to maximize pig health improvement cannot guarantee freedom from diseases. Moreover, because of health safety concerns, the use of antibiotics has been restricted in livestock, including pigs. Therefore, the swine industry has been looking for various alternatives to antibiotics to improve pig's health and performance. Clay is a dietary factor generally accepted for improving pig health. It is a naturally occurring material and is primarily composed of fine-grained minerals. It has a specific structure with polar attraction. Because of this structure, clay has the ability to lose or gain water reversibly. In addition, clay has beneficial physiological activities. First, clay has anti-diarrheic and antibacterial effects by penetrating the cell wall of bacteria or inhibiting their metabolism. Second, it can protect the intestinal tract by absorbing toxins, bacteria, or even viruses. When added to the diet, clay has also been known to bind some mycotoxins, which are toxic secondary metabolites produced by fungi, namely in cereal grains. Those beneficial effects of clay can improve pigs' health and performance by reducing pathogenic bacteria, especially pathogenic Escherichia coli, in the intestinal tract. Therefore, it is suggested that clay has a remarkable potential as an antibiotics alternative.

동의보감 처방중 황기 활용에 대한 연구 (Study on the use of Astragalus in Doneuibogam)

  • Objectives : This report describes 142 prescriptions related to the use of Astragalus mainly blended from Dongeuibogam. Methods : The following conclusions were drawn through investigations on the pre-scriptions that use Astragalus as a key component. Results : Prescriptions that Radix Aconiti Lateralis Preparata was taken as a monarch drug are utilized for 31 therapeutic purposes, for example, cold disease, stool disease, child disease. In particular, 17.3% of prescriptions appear in the chapter of cold. Prescriptions that utilize Radix Aconiti Lateralis Preparata as the main component are used in the treatment of cold, diarrhea and child disease, Bi syndrome due to pathogenic wind, cold and damp and they are also used for treating 55 different types of disease. Radix Aconiti Lateralis Preparata is used in pathogenic factors such as wind, cold and dampness and used in pathology related to the kidney and spleen and stomach system. The dosage of Radix Aconiti Lateralis Preparata is 2pun(about 0.75g) to 5don(about 18.8g), however 1don(about 3.75g)~1don 5pun(about 5.63g) has been taken the most for clinical application. Conclusions : Gangbutang and Ijungtang are the most useful base prescriptions which use the Radix Aconiti Lateralis Preparata as the main ingredient.

자시에 關한 文獻的 考察(原因과 治方을 爲主로) (A literature studies on the ja-shi(자시))

  • This study has been carried out to investigate pathogenic factor and medicine of ja-shi(자시) by referring to literatures. The results were obtained as follows. 1. In oriental medicine, Chashi(차시), Hamshichang(함시창), Hamshi(함시), Jabal(자발), Shichang(시창), Nojaon(로자온), Hamaon(하마온), Jokshiong(적시옹), Hamshidok(함사독), Tabshijong(탑사종), Shijong(시종) were the other names of ja-shi(자시). 2. The pathogenic factors of ja-shi(자시) are wind-heat, wetness-heat, Kihyeolungche(기혈응체), stomach-heat. wetness, wind wetness, and liking greasy and sweet food. 3. In the internal therapy of ja-shi(자시), Bojesodokum(普濟消毒飮), Sasunchunglyangum(四順淸凉飮), Takljsodoksan(托裏消毒散), Shihogalguntang(柴胡葛根湯), Sungsohwan(醒消丸), Gamisodokum(加味消毒飮) and etc. were used. And Radix glycyrrhizae(甘草), Fructus forsythiae(連翹), Radix angelicae gogantis(當歸), Radix scutellariae(황기), Radix bupleuri(柴胡), Radix p]atycodi(桔梗), Radix Ledebounellae(防風), Frustus arctii(牛蒡子), Rhizoma cimicifugae(升麻) and ete. were used as medicine. 4. In the external therapy of ja-shi(자시), Youigumhwangsan(如意金黃散), Okiogo(玉露膏), Igumsan(二金散), Gumhwangsan(金黃膏), Nungo(嫩膏) and etc. were used. And Radix angelicae dahuricae(白芷), Cortex phellodendri(黃栢), Rhizoma rhei(大黃), Indigo maturalis(靑黛), Radix glycyrrhizae(甘草) and etc. were used as medicine.

Comparative Transcriptome Analysis of Caryophyllene-Treated Helicobacter pylori

  • Helicobacter pylori (H. pylori) establishes long-term infections associated with severe gastric diseases such as peptic ulceration and gastric cancer. Exposure to an antibacterial agent can help regulate the expression levels of its pathogenic genes. In this study, we analyzed the transcriptional changes in H. pylori genes induced by β-caryophyllene. We used next-generation sequencing (NGS) to analyze RNA expression changes, and reverse transcription-polymerase chain reaction (RT-PCR) was performed as required to verify the results. The NGS results showed that 30 out of 1,632 genes were expressed differentially by β-caryophyllene treatment. Eleven genes associated with DNA replication, virulence factors, and T4SS components were significantly downregulated. RT-PCR confirmed that treatment reduced the expression levels of 11 genes. RT-PCR showed the reduced expression of 11 genes (dnaE, dnaN, holB, gyrA, cagA, vacA, secA, flgE, virB2, virB4, and virB8) following β-caryophyllene treatment. These results suggest that β-caryophyllene can modulate various H. pylori pathogenic determinants and be a potential therapeutic agent for H. pylori infection.

폐장생리(肺臟生理)의 운기적(運氣的) 인식과 그 생리계통(生理系統)의 임상적 의의에 관한 연구(硏究) (A study on the Physiology of the Lung by 'Un Qi' and the clinical meaning of its Phyological Systems)

  • This paper documents a study of the physiology of the lung. The aim of this paper is to better comprehend the physiological function of lung. To this end, the relationship between the physiological function of the lung and the characteristics of the 'Keum Un Qi Hua'(金運氣化), and the functional changes of the lung and its surrounding physiological systems have been studied in their relationship to the concept of the organism as a whole. The results of this study are as follows; The function of the lung is comprehended as corresponding to the 'Keum Un Qi Hua'. In visceral phenomenon, the lung has specific relations with the large intestine(大腸), nase(鼻), skin and hair(皮毛), sweat gland(汗孔), nasal mucus(涕), spirit(氣魄), grief and melancholy(憂 悲), the lung channel of hand-taiyin(手太陰肺經), the large intestine channel of hand-yangming(手陽明大腸經). This is called ‘the physiological systems of the lung’, and because these mutual relations reflect the functional changes of the lung, it is applied as the elementary knowledge to diagnose and treat the lung. For instance, a deficiency of the 'Qi‘ of the lung brings on an unconsolidated defending energy whose manifestations are sweating spontaneously as well as being susceptible to the common cold due to a diability in fighting against external pathogenic factors. Invasion of the lung by external pathogenic factors blocks the movement of the 'Qi' of the lung, which results in nasal obstraction. watery nasal discharge and a hyposmia. So the physiological functions and pathological changes of the lung can be determined by observing ’the physiological systems of the lung‘ based on the precepts of oriental medicine. As a result, the function of the lung is to be understood based on the characteristics of the 'Keum Un Qi Hua'. Furthermore we can know that the physiological systems of the lung mirrors the functional changes of the lung.

Construction of a High-Quality Yeast Two-Hybrid Library and Its Application in Identification of Interacting Proteins with Brn1 in Curvularia lunata

  • Curvularia lunata is an important maize foliar fungal pathogen that distributes widely in maize growing area in China, and several key pathogenic factors have been isolated. An yeast two-hybrid (Y2H) library is a very useful platform to further unravel novel pathogenic factors in C. lunata. To construct a high-quality full length-expression cDNA library from the C. lunata for application to pathogenesis-related protein-protein interaction screening, total RNA was extracted. The SMART (Switching Mechanism At 5' end of the RNA Transcript) technique was used for cDNA synthesis. Double-stranded cDNA was ligated into the pGADT7-Rec vector with Herring Testes Carrier DNA using homologous recombination method. The ligation mixture was transformed into competent yeast AH109 cells to construct the primary cDNA library. Eventually, a high qualitative library was successfully established according to an evaluation on quality. The transformation efficiency was about $6.39{\times}10^5$ transformants/$3{\mu}g$ pGADT7-Rec. The titer of the primary cDNA library was $2.5{\times}10^8cfu/mL$. The numbers for the cDNA library was $2.46{\times}10^5$. Randomly picked clones show that the recombination rate was 88.24%. Gel electrophoresis results indicated that the fragments ranged from 0.4 kb to 3.0 kb. Melanin synthesis protein Brn1 (1,3,8-hydroxynaphthalene reductase) was used as a "bait" to test the sufficiency of the Y2H library. As a result, a cDNA clone encoding VelB protein that was known to be involved in the regulation of diverse cellular processes, including control of secondary metabolism containing melanin and toxin production in many filamentous fungi was identified. Further study on the exact role of the VelB gene is underway.

아토피 피부염의 東西醫學的 文獻 考察 (A literature study about the comparison of Oriental-Occidental medicine on the Atopic dermatitis)

  • The results of a literature study about the comparison of Oriental-Occidental medicine on the Atopic dennatitis were as follows. 1. In Oriental medicine, Atopic dermatitis belongs to the category of the Naesun(내선), Taeryumchang(胎斂瘡), Eczema(濕疹), Chimumchang(浸淫瘡). In Occidental medicine, the other names of Atopic dermatitis are allergic eczema, IgE dermatitis, flexual eczema etc. 2. In Oriental medicine, the definition of atopic dermatitis includes chang(瘡), sun(선), and pung(風). Occidental medicine, is one of the intrinsic eczema classifications. In fact eczema term circumscribes dermatitis in atopic patients. 3. In Oriental medicine, the etiology and pathogenesis of Atopic dermatitis arose from the state of cogenital defect(稟賦不足), internal accumulation of damp and heat(濕熱內蘊) at first, and then invaded pathogenic wind, damp, heat factors again, and combined endo-exoteric pathogenic factors. So it appears in skin. In Occidental medicine, the etiology and pathogenesis of Atopic dermatitis approaches in genetic, allergic and immunologic, pharmacophysioloic aspects. It is only a hypothesis but there is no known facts about radical aetiology. 4. In Oriental medicine, differentiation of syndromes classifies manifestation aspects, etiology and pathogenesis, and invasion period. In Occidental medicine, it divides into an invasion period, and clinical aspect etc. 5. In Oriental medicine, Internal theraphys of Atopic dermatitis are decoction of ingredients(湯劑), pills(丸), and tablet(片). So, it prescribes as treatments on the ground of differentition of syndrome. In Occidental medicine, there is no radical therapy because Atopic dispositions don't change. But steroid, antihistamine as symptomatic tre atments are generally used in Occidental medicine. 6. In Oriental medicine, external therapies are wet dressings(濕敷), lotion(洗劑), powder(散劑), adhesive plasters(膏劑), oil(油劑), smoking(烟薰法), warm over fire therapy(熱烘療法), acupunture and moxibustion therapy(鍼灸療法).

Epidemiological Aspects of Pathogenic Microbial Foodborne Disease Outbreaks in Korea and Japan, 1999-2004

  • Pathogenic microbial foodborne disease outbreaks (PMFBDOs) have increased in many countries, the boom in food service establishment is not matched by effective food safety and control. In this study, we investigated the current state and the epidemic aspects of FBDOs in Korea and Japan. In Korea, the average prevalence rate of foodborne disease (FBD) was 15.0 per 100,000 population and cases per outbreak of FBD was 57.0. During the same period in Japan, the prevalence rate showed an average of 24.9, and the cases per outbreak were 16. When both prevalence rate and cases per outbreak were compared, the prevalence rate in Japan was much higher than that in Korea (p<0.01). However, average cases per outbreak of FBD in Japan were much lower than those in Korea (p<0.01). In Korea, outbreaks of FBDs were more common in spring (p<0.01), while in Japan, more frequent in summer and winter (p<0.01). Outbreaks of FBD occurred largely through restaurant and school foods (32.0% and 27.5%) in Korea. In Japan, the proportion of the outbreak cases in the restaurant and home were 23.7% and 12.1%, and cases of unknown causes of FBDs were 48.2%, respectively. Bacteria were the major causes of infection in both countries. The prevalence of PMFBDOs by Salmonella spp. Vibrio parahemolyticus and Staphylococcus aureus were much higher in Korea, while those by Camphylobacter spp. and SRSV were more common in Japan. The causes by virus were more frequent in Japan (13.7%) than in Korea (7.7%). The prevalence of FBDs in Korea and Japan showed characteristic differences, especially in the PMFBDOs due to such factors as geography, climate, culture, diet and food management.

국내 골프장 한국잔디의 라이족토니아마름병 발생 (Occurrence of Rhizoctonia Blight of Zoysiagrasses in Golf Courses in Korea)

  • Incidence of Rhizoctonia blight ranged from 22.2% to 100% in the golf courses at six geographical locations in Korea from 1989 to 1993. Rhizoctonia blight occurred more severly in southern area than in northern area. Fifty seven isolates of Rhizoctonia solani obtained from diseased parts of zoysiagrasses were grouped to AG2-2 by anastomosis test. Pathogenicity testes revealed that this pathogen was strongly pathogenic to Korean lawngrasses(Zoysia japonica, Z. matrella, Z. tenuifolia), but not pathogenic to creeping bentgrass(Agrostis palustris), bermuldagrass(Cynodon dactylon), Kentucky bluegrass(Poa pratensis), perennial ryegrass(Lolium prenne), and creeping red fescue(Festuca rubra subsp. rubra L.). The isolation frequency of R. solani AG2-2 fro sheaths of the infected plants was the highest by 91.67%, and that from stolons and roots was 11.13% and 5.63% respectively. The pathogen was not isolated from the leaves. Population density of R. solani in the lawn of large circular patch was highest on surface soils down to 1 cm deep with the value of 4.9$\times$104 (CFU/g soil), but below 1 cm population density decreased sharply down to 0.8~9.8$\times$103 (CFU/g soil). Horizontal distribution of propagules in turfgrass soil was higher in the margin than in center of patch, where the number of propagules was similar to these of healthy looking soils close to the margin of diseased patch. The meteorological factors influencing the outbreak of the disease were temperature, the number of rainy days and precipitation. Optimum temperature for disease development of Rhizoctonia blight in field was 20~22$^{\circ}C$, and that for hyphal growth of R. solani AG2-2 in vitro was 25~3$0^{\circ}C$. In Pusan area, Rhizoctonia blight first occurred in late April and rapidly developed in late June. The disease slightly decreased during July to August and developed again in late September in 1993. The monthly disease progress in Pusan area was similar to that in Kyeonggi province.
