Classification and Condensation of Nano-sized Airborne Particles by Electrically Tuning Collection Size (포집크기의 전기적 튜닝 기술을 이용한 나노크기의 공기중 입자 분류 및 수농도 응축)

  • It is not easy to detect nano-sized airborne particles (< 100 nm in diameter) in air. Therefore, the condensation of the nanoparticles alongside of the size-classification is needed for their detection. This paper proposes a hybrid (aerodynamic+electrical) particle classification and condensation device using a micro virtual impactor (${\mu}VI$). The ${\mu}VI$ can classify the nanoparticles according to their size and condense the number concentration of nanoparticles interested. Firstly, the classification efficiency of the ${\mu}VI$ was measured for the particles, polystyrene latex (PSL), ranging from 80 to 250 nm in diameter. Secondly, the nanoparticles, NaCl of 50 nm in diameter, were condensed by 4 times higher. In consequence, the output signal was amplified by 4 times (before condensation: 4 fA, after condensation: 16 fA). It is expected that the proposed device will facilitate the detection of nanoparticles.

  • Clouds play an important role in climate change, in the prediction of local weather, and also in aviation safety when instrument assisted flying is unavailable. Presently, various ground-based instruments used for the measurements of the cloud base height or velocity. Lidar techniques are powerful and have many applications in climate studies, including the clouds' temperature measurement, the aerosol particle properties, etc. Otherwise, it is very circumscribed in cloud velocity measurements because there is no Doppler effect if the clouds move in the perpendicular direction to the laser beam path of Doppler lidar. In this paper, we present a method for the measurement of cloud velocity using lidar's range detection and DIC (Digital Image Correlation) system to overcome the disadvantage of Doppler lidar. The lidar system acquires the distance to the cloud, and the cloud images are tracked using the developed fast correlation algorithm of DIC. We acquired the velocities of clouds using the calculated distance and DIC algorithm. The measurement values had a linear distribution.

Fundamental approach to development of plastic scintillator system for in situ groundwater beta monitoring

  • The performance of a plastic scintillator for use in an in situ measurement system was analyzed using simulation and experimental methods. The experimental results of four major pure beta-emitting radionuclides, namely $^3H$, $^{14}C$, $^{32}P$, and $^{90}Sr/^{90}Y$, were compared with those obtained using a Monte Carlo N-particle (MCNP) code simulation. The MCNP simulation and experimental results demonstrated good agreement for $^{32}P$ and $^{90}Sr/^{90}Y$, with a relative difference of 1.95% and 0.43% between experimental and simulation efficiencies for $^{32}P$ and $^{90}Sr/^{90}Y$, respectively. However, owing to the short range of beta particles in water, the efficiency for $^{14}C$ was extremely low, and $^3H$ could not be detected. To directly measure the low-energy beta radionuclides considering their short range, a system where the source could flow directly to the scintillator was developed. The optimal thickness of the plastic scintillator was determined based on the suggested diameter. Results showed that the detection efficiency decreases with an increase in the depth of the water. The detection efficiency decreased drastically to approximately 10 cm, and the tendency was gradually constant.

  • Foot-and-mouth disease (FMD) and avian influenza (AI) are highly pathogenic viral disease which affects the livestock industry worldwide. Outbreak of these viruses causes great impact in the livestock industry; thus, disease infected animals were immediately disposed. Burial is the commonly used disposal method for deceased animals. However, there is potential for secondary environmental contamination, as well as the risk that infectious agents persisting in the environment due to the limited environmental controls in livestock burial sites during the decomposition of the carcasses. Therefore, this study aimed to investigate the detection of FMD and AI viruses from animal carcass disposal sites using real-time reverse transcription PCR. Soil samples of more than three years post-burial from livestock carcass disposal sites were collected and processed RNA isolation using a commercial extraction kit. The isolated RNA of the samples was used for the detection of FMDV and AIV using qRT-PCR. Based on the qPCR assay result, no viral particle was detected in the soil samples collected from the animal disposal sites. This indicates that 3 years of burial and their carcass disposal method is efficient for the control or at least reduction of spread infections in the surrounding environment.

Detection of PED virus by the immunoelectron microscopy and immunogold conjugate immunoelectron microscopy (면역전자현미경(免疫電子顯微鏡)(IEM) 기법(技法) 및 immunogold conjugate 면역전자현미경(免疫電子顯微鏡)(IGC-IEM) 기법(技法)을 이용(利用)한 돼지 분변내 PED 바이러스의 검출(檢出))

  • Both immunoelectron microscopy(IEM) and immunogold conjugate immunoelectron microscopy (IGC-IEM) techniques were developed for the detection of porcine epidemic diarrhea virus(PEDV) from the feces. Fecal samples were incubated sequentially with anti-PEDV monoclonal antibody(MoAb) and immunogold conjugated goat anti-mouse IgG+IgM. Then negatively stained, mounted on the formvar carbon-coated copper EM grids and observed by the transmission electron microscope. By the direct electron microscopy(DEM), coronavirus particles were observed from 17 cases of total 33 fecal samples of grower pigs and sows. The virons of coronavirus were moderately pleomorphic but mostly spherical, with a diameter ranged from 90 to 190nm. PED virus particles were identified from 15 cases of 17 DEM positive samples by the IEM and IGC-IEM techniques. Aggregates of PED virus coated with specific antibody were seen in fecal samples incubated with homologous anti-PED virus MoAb but not in control samples incubated with anti-TGE virus MoAb. Following incubation with immunogold-conjugated secondary antibody, the gold granules were usually distributed around and among the virus particles and soluble and viral particle-associated antigen. So, IEM and IGC-IEM techniques were proved a rapid and sensitive methods for detection and identification of PED virus from fecal and intestinal contents.

Norovirus Targeted Bioreceptor Screening Method based on Lateral Flow Immunoassay (LFIA) (노로바이러스 검출을 위한 측면유동면역분석법 기반의 바이오리셉터 선별기법 개발)

  • Later flow immunoassay (LFIA) is a protein analytical method based on immunoreaction. On the LFIA based protein analytical method, bioreceptor molecule plays a key role, and so a system that evaluates and manages the binding affinity of bioreceptor is needed to secure detection reliability. In this study, Lateral Flow Immunoassay based rapid Bioreceptor Screening Method (rBSM) is presented that provide a simple and quick evaluating method for the binding affinity to the target protein of the antibody as model bioreceptor. To verify this evaluation method, Virus-like particles (VLP) and anti-VLP antibodies are selected as a model norovirus, which is target protein, and the candidate bioreceptors respectively. Among the 5 different candidate antibodies, appropriate antibody could be sorted out within 30 minutes through rBSM. In addition, selected antibodies were applied to two representative LFIA based techniques, sandwich assay and competitive assay. Among these methods, sandwich assay showed more effective VLP detection method. Through applying selected antibodies and techniques to the commercialized mass production lines, an VLP detecting LFIA kit was developed with a detection limit of 1012 copies/g of VLPs in real samples. Since this proposed method in this study could be easily transformable into other combinations with bioreceptors, it is expected that this technique would be applied to LFIA kit development system and bioreceptor quality management.

Dry Magnetic Particle Inspection of Ingot Cast Billets (강편 빌레트의 건식 자분 탐상)

  • Dry magnetic particle inspection(MPI) was performed to detect the surface defects of steel ingot cast billets. Magnetic properties of several materials were characterized by the measurement of the B-H hysteresis curve. The inspection results were evaluated in terms of the magnetizing current, temperature, and the amount of magnetic particles applied to billets. Magnetic flux leakage near the defect site of interest was measured and compared with the results of calculation by the finite element method in the case of direct magnetizing current. Direct and alternating magnetizing currents for materials were deduced by the comparison of the inspections. Results of the magnetic particle inspection by direct magnetizing current were compared with those of finite element method calculations, which were verified by measuring magnetic leakage flux above the surface and the surface defects of the material. For square rods, due to the geometrical effect, the magnetic flux density at the edges along the length of the rods was about 30% of that at the center of rod face for a sufficiently large direct magnetizing current, while it was about 70% for an alternating magnetizing current. Thus, an alternating magnetizing current generates rather uniform magnetic flux density over the rods, except for the region on the face across about 10 mm from the edge. The attraction of the magnetic particle by the magnetic leakage field was nearly independent of the surface temperature of the billets up to $150^{\circ}C$. However, the temperature should have been limited below $60^{\circ}C$ for an effective fixing of gathered magnetic particles to the billet surface using methylene chloride. We also found that the amount of applied magnetic particles tremendously affected the detection capability.

A Study On Radiation Detection Using CMOS Image Sensor (CMOS 이미지 센서를 사용한 방사선 측정에 관한 연구)

  • In this paper, we propose the radiation measuring algorithm and the device composition using CMOS image sensor. The radiation measuring algorithm using CMOS image sensor is based on the radiation particle distinguishing algorithm projected to the CMOS image sensor and accumulated and average number of pixels of the radiation particles projected to dozens of images per second with CMOS image sensor. The radiation particle distinguishing algorithm projected to the CMOS image sensor measures the radiation particle images by dividing them into R, G and B and adjusting the threshold value that distinguishes light intensity and background from the particle of each image. The radiation measuring algorithm measures radiation with accumulated and average number of radiation particles projected to dozens of images per second with CMOS image sensor according to the preset cycle. The hardware devices to verify the suggested algorithm consists of CMOS image sensor and image signal processor part, control part, power circuit part and display part. The test result of radiation measurement using the suggested CMOS image sensor is as follows. First, using the low-cost CMOS image sensor to measure radiation particles generated similar characteristics to that from measurement with expensive GM Tube. Second, using the low-cost CMOS image sensor to measure radiation presented largely similar characteristics to the linear characteristics of expensive GM Tube.

Applicaion of Neural Network for Machine Condition Monitoring and Fault Diagnosis (기계구동계의 손상상태 모니터링을 위한 신경회로망의 적용)

  • The morphologies of the wear particles are directly indicative of wear process occuring in the machine. The analysis of wear particle morphology can therefore provide very early detection of a fault and can also ofen facilitate a dignosis. For this work, the neural network was applied to identify friction coefficient through four shape parameters (50% volumetric diameter, aspect, roundness and reflectivity) of wear debris generated from the machine. The averages of these parameters were used as inputs to the network. It is shown that collect identification of friction coefficient depends on the ranges of these shape parameters learned. The various kinds of the wear debris had a different pattern characteristics and recognized relation between the friction condition and materials very well by neural network. We discuss how the network determines difference in wear debris feature, and this approach can be applied for machine condition monitoring and fault diagnosis.

Sizing of Spray Particles Using Image Processing Technique

  • The image processing technique is simple and, in principle, can handle particles with various shapes since it is based on direct visualization. Moreover, a wide measurement area can be covered with appropriate optical arrangement. In the present paper, various techniques of image processing for sizing and counting particles are reviewed and recent developments are introduced. Two major subjects are discussed in detail: identification of particles (i.e., boundary detection and pattern recognition) and determination of in-focus criteria. Finally, an overall procedure for image processing of spray particles is suggested.
