L-Proline-producing mutant strains were developed by exposing L-glutamic acid-producing bacteria to N-metyl-N-nitro-nitrosoguanidine and UV irradiation. A L-histidine auxotroph of Corynebacterium acetoacidophilum RYU3161(KCTC 0616BP), which was resistant to sulfaguanidine and proline analogs (DHP, AZC, TAC), was isolated. The activity of the mutant strain's $\gamma$-glutamyl kinase was 45% higher than that of the parent strain. The optimum level of L-histidine for production of L-proline was 0.16 g/l. In a 5-1 jar fermenter, the mutant strain produced L-proline at a high concentration (35 g/l) level within 48 h of cultivation.
Acetaminophen, a widely used analgesic, can be formed by N-acetylation and p-hydroxylation of aniline. Interspecific protoplast fusion technique was used to get acetaminophen directly from aniline and to increase the productivity of acetaminophen. Three auxotrophic mutants were obtained from S. griseus(ATCC 13273) and S. hygroscopicus(KCTC 1089) by N-methyl-N'-nitro-N-nitrosoguanidine(NTG) treatment. Regeneration frequencies of S. griseus$(his^-)$, S. griseus$(lys^-)$, S. hygroscopicus$(arg^-)$ were 42%, 45%, and 31%, respectively. Fusion of protoplasts carrying different auxotrophic markers was achieved by treatment with polyethylene glycol. When protoplasts were treated with 50% polyethylene glycol for 3 minutes, the fusion frequency between S. griseus$(his^-)$ and S. hygroscopicus$(arg^-)$ was $3.8{\times}10^{-5}$. The fusion frequency between S. griseus$(lys^-)$ and S. hygroscopicus$(arg^-)$ was $5.6{\times}10^{-4}$. When we checked the production of acetaminophen, thirty-four out of the fifty-six fusants produced larger amounts of acetaminophen than the parent strains did. Nine fusants produced twice more and twenty-five fusants produced one to two times more of acetaminophen than their parents.
To improve biopolymer productivity and properties of Bacillus strains, protoplast fusion was performed between biopolymer producing Bacillus subtilis K-1 and lactose utilizing Bacillus coagulans. The results were as follows; Protoplasts mixed in fusion fluid containing 33% PEG 6000, 1% PVP and 10 mM $CaCl_2$ were reacted for 5 min at $37^{\circ}C$ and then centrifused protoplasts were directly overlaid on the selective media containing $100\;{\mu}g/ml$ antibiotics and incubated for 3 days. At this conditions, the frequency of protoplast fusion was generally in the range of $4.6{\times}10^{-5}\;to\;1.8{\times}10^{-7}$ in ratio. Segregation ratio was observed between 1 to 6% indicating genetic stability of all the fusants. Fusants growth were also observed on the media contained amino acid and antibiotics as required marked materials. DNA contents of the selected fusants were 1.6 to 2.7 times more than that of parent strains. With observation by TEM microscopy, spherical protoplasts were first released from the swollen parental cells and then contracted to fuse in the process of fusion. And fused cells were observed representative vesicle. Originally, the parental cells were observed as in the morphology of thick-walled and double membrane-surrounded rod shape with TEM microscopy.
Wild perennial types of genus Glycine, as sources of genetic diversity, have been recently employed in the soybean breeding programs. This study was carried out to introduce some useful characters of wild perennial Glycine tomentella into common cultivars, G. max. Parent materials used for interspecific hybridization were three common cultivars, Ulsankong, Baemkong and Danyeobkong, and four G. tomentella strains having different diploid(2n) of 38, 40, 78, and 80. Their cross-fertility, effect of reciprocal cross, development comparison of crossing pod and optimum timing to efficient crossing were investigated. Plant were grown in 1/2,000 Wagner pots in greenhouse and field. Crossing efficiency and pod survival rate were the highest in 2n=38 of the four G. tomentella strains when the common cultivars were fixed as female parents. In the reciprocal cross of three common cultivars and four G. tomentella strains, when the former was used as maternal sources, they showed better crossing rate and longer pod survival than the latter. In the interspecific hybridization between G. max cv. Baemkong andG. tomentella, 2n=38, the pod formed from the female G. tomentella had relatively greater size and weight than that from the female Baemkong in comparison with parents. However, the reverse result was true in a number of ovules per pod. More efficient interspecific hybridization in the field was made on August 20, when temperature and relative humidity were higher and day length was shorter, than one or two months earlier.

We compared the degrees of stress response of 12 domestic purebred chicken strains that have been bred at National Institute of Animal Science, RDA, Korea since 1980. As a physiological marker of stress response, the expression levels of heat shock protein (HSP)-70, HSP-$90{\alpha}$, HSP-$90{\beta}$, hydroxyl-3-methyl-glutaryl coenzyme A reductase (HMGCR) genes and telomere length were measured by quantitative real-time polymerase chain reaction using the lymphocytes of 1,101 chickens. There was significant difference in HSP-70, HSP-$90{\alpha}$, HMGCR expression and telomere length among the strains. There was also significant difference in HSP-$90{\alpha}$, HSP-$90{\beta}$, and HMGCR expression between male and female chickens. Different age groups of chicken exhibited different expression levels of HSP-70, HSP-$90{\alpha}$ and telomere length. The results of the HSPs expression level suggested that, the strains of R, L and Y were highly resistant to stress, whereas the strains of S, O and W were susceptible to stress. Although the statistical differences in some of HSPs gene expression existed between genders, the HSP expression results varied in different strains that some opposed to the others, and there might be interaction between strains and genders, which conclude that there was no difference in stress response between male and female chickens. Moreover, despite of significant difference in some of HSPs expression level, it was considered that there was no difference in stress response between ages due to the inconsistent trends among HSP markers.
This study aimed to assess the performance of laying hens across twelve crossbreed strains (i.e., CFCK, CFYC, CFYD, CKCF, CKYC, CKYD, YCYD, YCCF, YCCK, YDCF, YDCK, and YDYC) of Korean native chicken (KNC) and compare them with Hy-Line Brown layers. A total of 287 18-week-old laying hens were placed in battery cages by strains (2-5 birds per pen). Results indicated that the YCYD and YDYC strains exhibited numerically heavier body weights than Hy-Line Brown at week 18-64. CKYC and YDYC strains demonstrated more than 94% viability by week 64. The CFYC strain had an age of first egg laying of 127 days, and the YDCF strain reached an age of 50% egg production at 140 days, both earlier than their parent stock. The YDCF strain showed over 70% egg production for up to 60 weeks. Regarding egg quality, the CKCF and YCCF strains had numerically higher egg weights among the KNC groups at week 24-64, with the YDYC strain showing a darker (P<0.05) eggshell color compared to CKCF at week 40. Moreover, KNC crossbreeds showed a higher (P<0.05) egg yolk ratio than Hy-Line Brown. In conclusion, the YDCF and YCCF crossbreeds exhibited the most desirable new synthetic Korean native commercial layer based on egg production and quality parameters. Therefore, these strains could be a viable substitute for Hy-Line Brown layers.

This study was conducted to develop a new synthetic breed of Korean native chicken. The combining ability and reciprocal effects for production traits were estimated on 1,157 hens from a $5{\times}5$ diallel cross-mating design using grand parent stock (GPS) lines of Korean native chicken. Body weight, viability, age at first egg laying, egg weight, and hen-day egg production were measured and analyzed. The results showed that the general combining ability (GCA) of the survival rate during laying periods was -9.6 to 11.1, with the highest value obtained in the W strain. Additionally, the GCA of the body weight at 12 weeks was -209.7 to 162.2, with the highest value obtained in the F strain. The GCA for age at fist egg laying was estimated to be -2.8 to 3.7, while the GCA of egg weight was -0.91 to 0.96, and the GCA of hen-day egg production was -4.9 to 6.0. In the estimation of specific combining ability, the YW combination showed the highest survival rate, FW showed the highest body weight at 12 weeks, and GW showed the highest hen-day egg production. The reciprocal effects were significantly different among crosses for almost all productivity traits. In identical breeding combinations, differences in ability were observed when the maternal or paternal breeds were switched. The mean value based on combining ability was higher in WY, WF, and GW combinations for survival rate; GF, HG, and HF combinations for body weight at 12 weeks; and GW, YW, and FW combinations for hen-day egg production. It is concluded that the GF and HF combinations, which have excellent growth performance and moderate survival rate, are the most desirable paternal parent stock (PS) strains, and the GW and FW combinations, which have great laying performance and moderate body weight, are the most desirable maternal PS strains.
The present experiment was carried out to compare performance, carcass characteristics and meat quality among three major meat-type duck strains in Korea. Two commercial duck (CD) strains, produced from parent stocks (PS) (Cherry Valley and Grimaud), and $F_1$ strains, self-produced by farmers, were used in an 8-week feeding experiment. Both feed conversion ratio and production index were significantly higher in Cherry Valley strain compared with the other two at the ends of 6 and 8 weeks (p<0.05). However, no differences were found in carcass characteristics and meat quality, with the exception of abdominal fat weight where Grimaud were higher than the others. Overall, the results of the current study show that Cherry Valley CD strain had higher performance among three strains tested.
In order to identify the best superior cross combination of breeder stocks for broiler production, combining ability test and analyses of phenotypic performances for parent stocks were examined on records of 1,440 broiler chicken which were produced from 4 parental strains and 3 maternal strains at Hanhyup Poultry Breeding Farm from September 28, 1978 to January 5, 1979. The results obtained were as follows; 1. There was not found heterosis effect in viability but it seems to be desirable to select Hubbard strain in paternal line to improve viability. 2. As the paternal and maternal lines, selection of Ross strain showed the best paternal and maternal performance and the best general combining ability in body weight at 8 weeks of age is expected to be able to improve body weight of it s crossbred And the most superior cross combinations based on the specific combining ability and performance of rack crossbred were identified as Hubbard x Ross ana Ross x Hypeco crossbreds. 3. The best paternal and maternal lines on the smallest feed consumption for 8 weeks were Hubbard and Ross strains, and Hypeco strain, respectively. Especially Hubbard x Hypeco cross combination was proved as the smallest feed consumption compared with other cross combinations. 4. In feed requirement per Kg body weight increase, Hubbard strain for paternal line, Hypeco strain for naternal line, and cross combinations of Hubbard x Hypeco, Hubbard x Ross and Ross x Hypeco were certified as the most superiors. 5. Also superior cross combinations of Hubbard x Hypeco and Hubbard x Ross earned the most profit per bird through economic analysis. According to results as shown above, this experiment seems to be able to reach a such conclusion that production of superior cross combinations Hubbard x Ross, Hubbard x Hypeco and Ross x Hypeco through selection of Ross and Hubbard strains to paternal line and Hypeco and Ross strains for maternal line may become to considerable improvement for important economic characters of broiler; viability, body weight, feed consumption and feed requirement.

This study was conducted to reduce the phenomenon of the biased cultivation of certain mushroom varieties and to develop a competitive variety of Grifola frondosa. We developed the first Korean white commercial mushroom strain, 'Bakyeon', by crossing monokaryons derived from brown strains. We have collected and tested the characteristics of mushrooms from domestic and international genetic resources since 2018. We bred the unique domestic variety, 'Bakyeon', which has the following characteristics. The optimal temperature for mycelial growth was 25~28℃ and the optimal temperature for fruit body growth was 16~18℃. The new variety was similar to the control variety (Daebak) in terms of the pileus, which formed a pine cone shape, and the number of days of cultivation. The yield was 94.1 g/bottle, which was 23% lower than the 108.5 g/bottle yield of the control variety. When incubating the parent and control varieties, the replacement line was clear. Moreover, polymerase chain reaction analysis of mycelial DNA resulted in different band patterns between the parent and control varieties, confirming the hybrid species.
