Establishment of a live vaccine strain against fowl typhoid and paratyphoid

  • To develop a live vaccine strain against fowl typhoid and paratyphoid caused by Salmonella serovar Gallinarum biovar Gallinarum (Salmonella Gallinarum) and Salmonella serovar Enteritidis (Salmonella Enteritidis), respectively, several nalidixic acid resistant mutants were selected from lipopolysaccharide (LPS) rough strains of Salmonella Gallinarum that escaped from fatal infection of a LPS-binding lytic bacteriophage. A non-virulent and immunogenic vaccine strain of Salmonella Gallinarum, SR2-N6, was established through in vivo pathogenicity and protection efficacy tests. SR2-N6 was highly protective against Salmonella Gallinarum and Salmonella Enteritidis and safer than Salmonella Gallinarum vaccine strain SG 9R in the condition of protein-energy malnutrition. Thus, SR2-N6 may be a safe and efficacious vaccine strain to prevent both fowl typhoid and paratyphoid.

  • 동물(動物)의 Paratyphoid의 원인균(原因菌)으로서 각종동물(各種動物)에서 여러학자(學者)들에 의(依)하여 많은 Salmonella속균(屬菌)이 분리보고(分離報告)되고 있으나 우리나라에서는 아직 동물(動物)에 있어서 Salmonella속균(屬菌)에 대(對)한 관심(關心)이 극(極)히 희박(稀薄)하여 돈(豚)의 Paratyphoid와 추백리(雛白痢)를 제외(除外)한 그 외(外)의 가축(家畜)의 Salmonella증(症)은 거이 간과(看過)되고 있는 상태(狀態)에 있으며 인간(人間)의 Paratyphoid 및 식중독등(食中毒等)에 밀접(密接) 관계(關係)가 있음을 생각할 때 가축(家畜)의 Salmonella증(症)은 단지 수의학(獸醫學) 뿐만 아니라 공중위생상(公衆衛生上) 경시(輕視)못할 문제(問題)이다. 특(特)히 견(犬)은 애완용(愛玩用), 수렵용(狩獵用), 또는 가정(家庭) 경비용등(警備用等)으로 인간생활(人間生活)에 있어 접촉(接觸)하는 기회(機會)가 가장 많은 동물(動物)이므로 이로 인(因)한 인간(人間)의 폐해(幣害)을 생각하지 않을수 없다. 필자(筆者)는 견(犬)에 있어서의 Salmonella속균(屬菌)의 분포상태(分布狀態)를 조사(調査)할 목적(目的)으로 1958년(年) 하계(夏季) 대구(大邱) 시내(市內) 광견병(狂犬病) 예방접종시(豫防接種時) 가정상양견(家庭飼養犬) 100두(頭)의 분변(奮便)을 검사(檢査)하여 6두(頭)에서 Salmonella속균(屬菌) 각(各) 1주(株)씩을 분리(分離)하였으며 다음과 같이 균형(菌型)을 결정(決定)하였다. Sal. typhi-murium 1주(株) Sal. cholerae-suis 1주(株) Sal. paratyphi A 1주(株) Sal. bartord 1주(株) 균형미결정(菌型未決定) 2주(株) (경북대학교(慶北大學校) 논문집(論文集) 제3집(第三輯)에 발표(發表)하였음).

제주도 내 돼지 살모넬라증의 발생 양상 분석 및 병리학적 연구 (Prevalence and pathologic study of porcine salmonellosis in Jeju)

  • Salmonella (S.) Typhimurium is highly contagious, and its infection may rapidly spread within pig populations of herd. According to the survey (1,191 pigs) from 2003 to 2012, 155 pigs (13.0%) were diagnosed as salmonellosis in Jeju. Major porcine salmonellosis cases (88.4%) were concentrated in 4- to 12-week-old weaned pigs, but 6 pigs (3.9%) under 4 weeks old were also diagnosed. Based on the histopathologic examinations, ulcerative enteritis (63.9%) in the large intestine and/or paratyphoid nodules formation (57.4%) in the liver were most prevalent lesions in porcine salmonellosis. Single infection of S. Typhimurium and mixed infection with more than 2 pathogens were detected in 38 (24.5%) and 117 (75.5%) in pigs, respectively. Co-infections of Porcine reproductive and respiratory syndrome virus and Porcine circovirus type 2 were very common in porcine salmonellosis in Jeju and detected in 84 (54.2%) and 59 (38.1%) pigs, respectively. Based on the serotyping tests using 41 bacterial isolates, S. Typhimurium and S. Rissen were confirmed in 39 (95.1%) and 2 (4.9%) cases, respectively.

2012-2021 전수감시 대상 수인성·식품매개감염병의 발생 신고 특징 (Characteristics of Water- and Foodborne Disease's Reports in Korea National Notifiable Infectious Disease Surveillance System, 2012-2021)

  • 질병관리청에서는 「감염병예방법」에 따라 법정 감염병을 지정하여 감염병 감시체계를 운영하고 있으며 그 중 전수감시 대상 수인성·식품매개감염병은 총 6종으로 제2급감염병 중 콜레라, 장티푸스, 파라티푸스, 세균성이질, 장출혈성대장균 감염증과 제3급감염병인 비브리오패혈증이 있다. 전수감시체계는 1954년 국내에 도입되었고 질병관리청에서는 감염병 신고 활성화를 위해 많은 노력을 기울이고 있으며 최근까지 국가 감시체계 신고 현황을 분석한 연구는 부족하였기 때문에 본 연구를 통해 최근 감염병 신고 현황을 이해하고자 한다. 본 연구에서는 지난 10년 동안 전수감시 체계를 통해 수집된 수인성·식품매개감염병의 신고자료 특징을 살펴보았다. 질병관리청에서 운영하는 질병보건통합관리시스템을 통해 수집된 신고정보 중 2012년 1월 1일부터 2021년 12월 31일까지 신고된 것을 분석에 사용하였으며, 이 중 신고 기준에 적합했던 12,296건을 바탕으로 적시 신고 여부와 확인진단 검사 결과 3가지에 따라 자료를 구분하여 분석하였다. 전체 신고 건수는 10년 동안 꾸준히 증가하고 있으며, 그 중 장티푸스와 파라티푸스, 장출혈성 대장균감염증의 신고 건수가 크게 증가하였다. 전체 신고된 건의 96%가 진단일로부터 1일 이내에 적시 신고된 것을 확인하였으며, 지연 신고된 4% 중 비브리오패혈증이 지연 신고 비율이 가장 높았으며 콜레라에서 가장 낮았다. 또한 콜레라는 지연된 건의 평균 지연 일수가 2.7일로 가장 낮은 것으로 나타났다. 확인진단 검사 결과에 따라 살펴보았을 때, 장티푸스와 파라티푸스는 과거에 비해 검사 결과가 '음성'인 건의 비율이 증가한 것을 확인할 수 있었고, 비브리오패혈증은 검사 결과가 '양성'으로 확인된 건의비율이 높았으며, 장출혈성대장균감염증은 확인진단 검사를 실시하지 못한 경우가 특히 많았다. 최초 의심 신고된 이후 최종 검사 결과를 확인할 때까지 소요된 시간을 확인했을 때 검사 결과가 '양성'인 경우에서 소요 일수가 가장 작게나타났고, 검사를 실시하지 못한 경우에서 소요일수의 사분위수 범위가 가장 크게 나타났다. 특히 모든 감염병에서 검사 결과가 양성인 경우의소요 일수가 0∼1일이었으며, 그 중 파라티푸스는음성, 검사 미실시의 경우에서 다른 감염병에비해 높은 소요 일수를 나타냈다. 전반적인 수인성·식품매개감염병의 신고 건수 증가와 더불어 확인진단 검사 결과가 음성인 건의 비율이 증가함에 따라 전수감시체계의 모니터링 기능이 향상되고 있다고 추정할 수 있었다. 단, 일부 감염병의 경우 신고의 적시성과 확인진단 검사 결과 확인의 적시성 측면에서 개선이필요한 것으로 사료된다.

수인성 전염병의 현황 및 효율적 관리 (Present status and effective control measure of water-borne infectious diseases in Korea)

  • Water- borne infectious diseases can be acquired by contact with contaminated water or by ingestion of contaminated water. There are many water- borne infectious agents such as bacteria, virus, and parasite. Among many of water- borne infectious diseases, health authorities of Korean government has particularly intensified to prevent and control typhoid fever(class I ), shigellosis(class I ), cholera(class I ), paratyphoid fever(class I), amebiasis(class II ) and leptospirosis(euivalent to class II ) under the communicable disease control law. Water- borne disease Prevention and control guideline itself has been also well provided by the health authorities. However, in practical public health point of view, there are still many problems remained to be solved out; no prospective investigation project to survey water borne infectious diseases under the national disease prevention and control programmes, incredible statistic data of annual notifiable disease report frequent appearance and varieties of drug resistance water- borne infectious agents, little cooperation and information- exchange system in between the related government authorities( the health authorities, the environment sanitation authorities and the food hygiene authorities) which should be closely collaborated, lack of health consciousness of the people, necessity of evaluation and Hndification on to the outcomes of performed health activities and programmes, neglect activities for water quality investigation, shortage of expertise and human resources in the related field, and poor investment of the government budget to develope and improve public health and sanitation field. In order to prevent and control water- borne infectious diseases effectively, it is emphasized that all the above indicated should be considered and performed to improve under the national health and sanitation development programmes.

  • Salmonella enterica serotype Choleraesuis causes swine paratyphoid, with clinical findings of enterocolitis and septicemia. However, the clinicopathological features of S. Choleraesuis infections in pigs have not been reported in Korea. We describe the pathological findings of two weaned pigs with S. Choleraesuis infections, presenting with diarrhea, cough, and sudden death. Pathological examination indicated severe necrotic colitis in pig 1 and septicemic lesions in pig 2. Multidrug-resistant S. Choleraesuis was isolated from the pigs' lungs and intestinal contents. Further research is required for the surveillance of S. Choleraesuis infections in pigs and the virulence estimation in the S. Choleraesuis isolates.

경남지역에서 발생한 가금티푸스의 역학적 특성 및 진단방법에 대한 비교 시험 (Epidemiological characteristics on fowl typhoid outbreak in Kyongnam province and comparison of diagnostic methods for identification of salmonella gallinarum)

  • An epidemiological survey was conducted to investigate fowl typhoid outbreaks in Kyungnam province of Korea. The causative agent, salmonella gallinarum was isolated from 68 chicken samples of tentatively diagnosed fowl typhoid cases occurred during the period from January 1996 to September 1999. Comparative studies were also carried out to evaluate the diagnostic methods for detection of S gallinam The results obtained were as follows; 1. Of the 68 cases of tentatively diagnosed fowl typhoid, 56 (82%) cases were determined as fowl typhoid by biochemical test and pathological findings. The other 12 (18%) cases were determined as paratyphoid. 2. Fowl typhoid outbreaks occur continuously all seasons in the year, however the incidence was remarkably increased from May to September. 3. The frequency of incidence of fowl typhoid in terms of regional distribution was relatively high in egg-laying hens facilities, and the mode of transmission is likely to be either egg-to-egg or lateral transfer by wild birds or rats. 4. All of 18 isolates from 56 cases were identified as S gallinarum by biochemical and serological test. 5. Antimicrobial drug susceptibility test against 18 isolates showed that the isolates were highly susceptible to ASH, CZ, CF and GM (above 90%), whereas those strains were 100% resistant to EM, NA and PC. 6. S gallinarum rfbS gene was targeted to be amplified by PCR for comparative detection of S gallinarum in the experimentally infected chickens. The amplified 720bp DNA fragment, which is specific in D serogroup strains of S enterica subspecies was confirmed by agarose gel electrophoresis. 7. A comparison made between fecal culture and PCR-method revealed that later-method was relatively higher in detection rate than that of former method for S gallinarum. 8. Comparison of currently applied methods, rapid serum agglutination test (RST) and microplate agglutination test (MAT), with experimentally infected chickens were made to evaluate sensitivity of detection by neutralizing antibody titration. Both methods detected neutralizing antibodies from the challenged chickens of 5 day post infection. However, positive reactions were determined after 7 and 9 days post infection by MAT and RST, respectively.

Pulsed-Field Gel Electrophoresis and Mutation Typing of gyrA Gene of Quinolone-Resistant Salmonella enterica Serovar Paratyphi A Isolated from Outbreak and Sporadic Cases,1998-2002, Korea

  • In early 2002, over 200 people in the city of Pusan. Korea suffered from paratyphoid fever resulting from Salmonella Paratyphi A Infection. Antimicrobial susceptibility tests and Xbal pulsed-field gel electrophoresis (PFCE) were conducted to 54 Salmonella Paratyphi A isolated from humans during the period of 1998 to 2002. Most of the isolates ($83\%$) were only nalidixic acid-resistant and $78\%$ were X 1 PFGE patterns. Also, we measured the MIC of ciprofloxacin and screened gyrA mutation(5) using allele- specific PCR and restriction fragment length polymorphism (AS-PCR-RFLP). The representative 5 isolates in 2002 and 1 isolate in 2000 were $1{\mu}g/ml$ of MIC and had mutation at the 83rd codon in gyrA. These data suggest that the outbreak in the early 2002 might have been due to dissemination of the strain present In 2000. Also, decreased susceptibility to ciprofloxacin was partly due to the mutation at the 83rd codon in gyrA.
