Efficiency of Enriched Rotifer and Artemia nauplius for the Seedling Production of Flounder, Paralichthys olivaceus (넙치 종묘생산을 위한 Rotifer와 Artemia nauplius의 영양강화효과)

  • Survival and growth rates of flounder larvae fed rotifers and Artemia nauplii enriched by various bio-enrichment techniques were investigated. Enriched materials used in the study were $12\omega-yeast$, 2 commercial emulsified oil and microparticulated products. $\omega-yeast$ were added with one kinds of oils (cuttlefish liver oil, sardine oil, Alaska pollock liver oil, linseed oil) at the level of $5\%,\;15\%\;and\;25\%$. The results are as follows: 1. The growth rate of rotifers cultured with $\omega-yeast$ added $15\%$ cuttlefish liver oil was the best among 12 kinds of $\omega-yeast$ and this performance was almost the same with that of rotifers fed on Chlorella. 2. Survival and growth rates of flounder larvae fed on rotifers and Artemia nauplii enriched with $\omega-yeast$ and the other commercial products were higher than that those of larvae cultured with only Chlorella and non-enriched Artemia. The optimum contents of the cuttlefish liver oil in $\omega-yeast$ for rotifers and Artemia enrichment were $15\%\;and\;25\%$, respectively. 3. Optimum enrichment time and supply amount of $\omega-yeast$ for Artemia nauplii were 6 hr., 1.5 g/g cyst, respectively.

  • Population growth equation of scuticociliate Miamiensis avidus was obtained from the experimental results of in vitro culture condition to estimate the growth rate and carrying capacity from the growth equation. In addition, intraperitoneal infections into olive flounder Paralichthys olivaceus were carried out into 2 different conditions: different concentrations of M. avidus in same water temperature and same concentration of M. avidus in different water temperatures. Olive flounder mortality was threshold dependent with both the temperature and M. avidus density parameters. In this paper, we propose a mathematical model to study M. avidus growth in olive flounder based upon the interactions between parasite and host. The mathematical model was logistic growth differential equation (1.2). The parameters were found with Matlab program through the Levenberge-Marquardt method. In theorem, equilibrium values between the infected fish population and dead population could found. Our equilibrium points were a stable equilibrium and an unstable equilibrium. From the equation (1.6), it was possible to predict the amount of cumulative mortality of olive flounder along with the time after M. avidus infection.

Bactericidal Effects of Hypochlorous Acid Water against Vibrio parahaemolyticus Contaminated on Raw Fish and Shellfish (어패류에 오염된 장염비브리오균(Vibrio parahemolyticus)에 대한 차아염소산수의 살균효과)

  • The bactericidal effects of strongly acidic hypochlorous acid water (StAHA) and slightly acidic hypochlorous acid water (SlAHA) against Vibrio parahaemolyticus contaminated on surface of raw fish and shellfish were examined. V. parahaemolyticus contaminated with about 7.0 log CFU/g on the meat chunk of olive flounder (Paralichthys olivaceus), and yellow tail (Seriola quinqueradiata), and 4.0 log CFU/g on the shucked scallop (Patinopecten yessoensis) were not detected after washing with StAHA and SlAHA at a ratio of 30:1 on a sample weight basis. However, 1.0 log CFU/g of V. parahaemolyticus was survived on shucked oyster (Crassostrea gigas) under same treatment conditions. The bactericidal effects of acidic hypochlorous acid water against V. parahaemolyticus contaminated on surface of shucked oyster were not as effective as those against V. parahaemolyticus contaminated on surface of meat chunk of olive flounder, yellow tail, and shucked scallop. Such differences can be attributed to the complicated surface conformation of oyster.

Effect of Feeding Frequency of Extruded Pellet on Growth and Body Composition of Juvenile Flounder, Paralichthys olivaceus During the Winter Season (겨울철에 부상사료 공급횟수가 넙치 치어의 성장과 체성분에 미치는 효과)

  • A feeding trial of two feeding rate (satiation and $80\%$ satiation) $\times$ four feeding frequency (three meals a day, two meals a day, one meal a day and one meal every two days) factorial design with three replications was carried out to investigate the optimum feeding frequency and rate for growth of flounder during the winter season. Juvenile flounder averaging 45 g were fed a extruded pellet for 60 days. Weight gain was affected by both feeding frequency (P<0.001) and feeding rate (P<0.01). The best weight gain obtained from fish fed two meals a day with satiation was not different from that of fish fed three meals a day with satiation and $80\%$ satiation. Feed efficiency and protein efficiency ratio of fish fed the diet three times daily were the highest among groups, but not different to other groups of fish except for one meal every two days with satiation and $80\%$ satiation feeding. Daily feed intake was affected by both feeding frequency and feeding rate (P<0.001), these values of fish fed two and three meals a day with satiation were the highest among the groups. Moisture, crude protein, crude lipid and ash contents of fish were not affected by both feeding frequency and feeding rate (P>0.05). The results of this study indicate that the optimum feeding frequency for the growth of juvenile flounder grown from 45 g to 53 g is two meals a day with satiation feeding during the winter season.

  • Viral hemorrhagic septicemia (VHS) is one of the most serious viral disease of farmed rainbow trout and some marine fishes in Europe and North America. It has been reported in various marine fish species of Asian countries and induced cause mass mortality in Japanese flounder (Paralichthys olivaceus) culturing in Korea. The aims of this study were to monitor VHSV in wild marine fishes and to give critical information for controling the disease through prophylactic methods. Prevalence of the viral disease, geological distribution and reservoir of the virus were investigated using wild marine fishes captured in southern coast and east china sea for two years. (Reverse Transcriptase Polymerase Chain Reaction) RT-PCR results showed that VHSV were detected in 17 (10.6%) out of 160 fish. G gene sequences of viral strains isolated in this study were closely related to that of a reference strain, KVHS01-1, belonging to VHSV genotype Ⅰ. The results suggest that some of wild marine fishes are VHSV carriers and may spread the pathogen directly to fish farmed in coastal area.

Muscle Quality of Cultured Olive Flounder, Paralichthys Olivaceus (양식 넙치의 육질에 관한 연구)

  • Sensory and physical qualities and palatable compounds of cultured flounder muscle were compared with the wild ones. No differences were seen in the contents of the moisture, lipid, protein between cultured and wild dorsal muscles, however, sensory panels preferred the wild fish to cultured ones because of the texture and taste of wild fish, and they could differentiate the degree of difference in the texture and taste property distinctly. Raw meat of the wild fish was harder and more elastic than the cultured ones, however, the cultured meat was harder and drier than the wild ones once they were cooked. The results of physical properties were similar to the results of sensory evaluation. Alanine was the most abundant compound among the free amino acids which make meat palatable and followed by glutamic acid, proline, methionine, and glycine. There was no difference in total content of free amino acids between two fish muscles. ATP was the most abundant among all nucleotides and their related compounds in both fish muscles followed by IMP, ADP, AMP, however, the total content of those was greater in wild fish muscle (9.4 ${\mu}$mole/g) than in cultured fish muscle (6.7 ${\mu}$mole/g).

Comparison of Nitrogenous Extractives, Amino Acids in Wild and Cultured Bastard (천연 및 양식산 넙치의 함질소엑스분과 아미노산조성)

  • Nitrogenous extractives and amino acids were analyzed with the dorsal muscle from wild and cultured bastard(Paralichthys olivaceus), and flounder (Pleuronichthys cornutus). Cultured bastard was higher in moisture content, and lower in crude lipid and protein content than those of wild one. Contents of free amino acids from wild and cultured bastard, flounder were 305.03mg/100g, 253.42mg/100g, and 340.10mg/100g, respectively. The profile of free amino acids in these samples was very similar, although there were a relatively difference in glycine, threonine, proline contents. Content of IMP in wild bastard (408.31mg/100g) and cultured one (356.26mg/100g) were higher than that of flounder (178.61mg/100g). Wild bastard contained higher trimeth-ylamine oxide and total creatinine than cultured one. Total amino acid contents of dorsal muscle from wild and cultured bastard, and flounder were 21.94g/100g, 20.12g/100g, and 17.63g/100g, respectively. The major amino acids of these samples were glutamic acid, aspartic acid, leucine and isoleucine.

Comparison of Lipid Components in Wild and Cultured Bastard (천연 및 양식산 넙치의 지방질성분)

  • The lipid components of cultured bastard(Paralichthys olivaceus) were analyzed and compared with those of wild one and flounder(Pleuronichthys cornutus). Total lipid(TL) content of dorsal muscle in wild and cultured bastard, flounder was 2.0%, 1.6%, and 1.9%, respectively. Wild bastard and flounder were higher in neutral lipid(NL) and glycolipid(GL) contents, while lower in phospholipid(PL) content than those of cultured one. The NL was mainly consisted of triglyceride(70.7-72.6%), and PL was mainly occupied by phosphatidylcholine(53.6-58.3%), phosphatidyl-ethanolamine(25.9-29.5%). In fatty acid composition of TL, cultured bastard showed higher content in polyenes such as docosahexaenoic acid. and lower in monoenes such as palmitoleic, oleic acid than those of wild one. Flounder was higher in Percentage of arachidonic, eicosapentaenoic acid compard with bastard. The major fatty acids of these fishes were palmitic, docosahexaenoic, oleic, eicosapentaenoic and palmitoleic acid in order. These fatty acid composition of NL, GL and PL fractions were not significantly different among wild and cultured bastard, flounder.

Investigation on Sedation and Anesthetic of $AQUI-S^{\circledR}$ for Black Rockfish(Sebastes schlegeli) ($AQUI-S^{\circledR}$의 조피볼락(Sebastes schlegeli)에 대한 진정 및 마취 효과)

  • Along with olive flounder(Paralichthys olivaceus), black rockfish(Sebastes schlegeli) is another very popular maricultured species in Korea. As there is many difficulties in handlling live fish for aquaculturist, use of suitable anesthesia for proper handling of fish is very important in the field. In this view, the effect of $AQUI-S^{\circledR}$ has analysed for its use in the field. $AQUI-S^{\circledR}$, contains 50% isoeugenol, is a new anesthics for fish and zero-withdraw time required since it was approved as a safe additives of food. In the present study, black rockfish adult exhibted sedation effect from 5 ppm at $10^{\circ}C\;and\;15^{\circ}C$, and 7.5 ppm at $20^{\circ}C$, on the other hand, anesthesia was at least required 7.5 ppm at $10^{\circ}C\;and\;15^{\circ}C$, and 10 ppm at $20^{\circ}C$. The fish was recovered from sedation and anesthesia after approximately 5 and 10 minutes, respectively. In case of black rockfish fry, sedation was recorded from 2.5 ppm at $20^{\circ}C$, and 5 ppm at $15^{\circ}C\;and\;20^{\circ}C$. The least concentraion of anesthesia was 2.5 ppm at $10^{\circ}C$, 7.5 ppm at $15^{\circ}C$, and 5 ppm at $20^{\circ}C$. The acute toxic test showed that black rockfish adult and nv showed mortality above 12.5 and 15 ppm concentration of $AQUI-S^{\circledR}$, respectivly. In sum, $AQUI-S^{\circledR}$ is considered as useful anesthetic drug for sedation and anesthesia of black rockfish, it is highly availbale to be prevented from a varity of stress.

Myo-inositol Requirement in Diets for Juvenile Olive Flounder (Paralichthys olivaceus) (넙치 치어의 사료 내 myo-inositol 요구량)

  • A long-term (26 weeks) feeding experiment was conducted to examine the essentiality and requirement of inositol in diets for olive flounder because no information is available in the species. Five casein-gelatin based semi-purified diets were formulated to contain four different levels of myo-inositol (0, 0+antibiotic, 400, 800, and 1600 mg/kg, designated as M0, M0+, M400, M800, and M1600, respectively). One (M0+) of the control diets contained tetracycline hydrochloride (0.4%, wt/wt) as an antibiotic to inhibit biosynthesis of inositol by micro-organism in intestine of fish. Olive flounder at the early juvenile stage (initial body weight 1.22 g) were randomly distributed into fifteen 35 L tanks (48 fish/tank) and fed with one of the experimental diets (3 replicates per diet). At the end of the feeding trial, the weight gain, feed intake, specific growth rate, and protein efficiency ratio of fish fed diets containing higher levels of myo-inositol (M800 & M1600) were significantly higher than those of fish fed the other diets (P<0.05). Feed conversion ratio, survival, hematocrits, and hemoglobin of fish fed experimental diets were not significantly different among all the fish groups. Whole body compositions of fish were not different except for lipid content. The lipid content was significantly different between M0 and M400 diet groups. These results indicate that juvenile olive flounder requires dietary supplementation of myo-inositol in diets for normal growth and its optimum level seems to be approximately 800 mg myo-inositol/kg diet.
