Distribution of Toxin Genes and Antimicrobial Resistance of Vibrio parahaemolyticus Isolated from Seafood in Gwangju (광주지역 유통·판매 수산물에서 분리된 장염비브리오의 독소유전자 분포 및 항생제 내성 조사)

  • The purpose of this study is to investigate the distribution of toxin genes and antimicrobial resistance of Vibrio parahaemolyticus isolated from seafood in Gwangju. A total of 335 seafood, including 163 shellfish, 97 fish, and 36 mollusk, were tested in this study. As a result, V. parahaemolyticus was detected in 123 (36.7%) of 335 seafood. The tdh gene was not detected in all strains, while the trh gene was detected in 3 strains (2.4%). According to antimicrobial susceptibility test, 116 strains (94.3%) represent resistance to ampicillin, and 1 strain (0.8%) represents resistance to trimethoprim/sulfametoxazole. However, all strains were sensitive to 9 antimicrobial agents, including amikacin, chloramphenicol, tetracycline, and more. Therefore, the risk of V. parahaemolyticus isolated from seafood in Gwangju is considered low, but continuous monitoring of V. parahaemolyticus in seafood is required.

  • This study was conducted to investigate the vibrio sap contamination of aquarium (water) in Incheon Area The results obtained were as follows : 1. V. parahaemolyticus was detected 20.4% at samples of aquarium water and not detected at underground water 2. V. unlnificus was not detected at all samples 3. Underground water was similar to aquarium water on pH and the concentration of salt 4. Underground water proved it's safety bacteriologically 5. V. parahaemolyticus was common in the sea water between July to October.

  • Of 643 aquarium water samples, 374(58.2%) were less than 1.0$\times$103 SPC/ml, seasonally, were 56.7% of samples in Spring. 38.7% in Summer,84.5% in Autumn, and 57.7% in Winter, respectively. a1so 3(0.47%) were over 1.0 $\times$ 105. E.coli and V. parahaemolyticus distribution was relatively high when SPC/ml was much. At the aspect of the water quality management, 98.3%(177/181) were cleaned over once a week. 69.1%(125/181) were cleaned over twice a week.

A Profile of Naturally Occurring Plasmids from Selected Strains of Vibrios

  • The naturally occurring plasmids of Vibrio species have been isolated in part to investigate their genetic traits. Among six different Vibrio species tested, Vibrio anguillarum, Vibrio fluvialis, Vibrio vulnficus, Vibrio mimicus and Vibrio furnissi did not show any presence of plasmid. One environmental isolate of Vibrio parahaemolyticus harboring plasmid was observed. The isolated plasmid was 8.7 kb by analysis with restriction endonuclease digestion. No common feature was shown relationships between the presence of plasmid and resistance against commonly used antibiotic compounds from the tested Vibrios.

  • V. parahaemolyticus possessed an extracellular alkaline protease activity during the stationary growth phase. Various factors such as initial pH of medium, incubation temperature and shaking rate were investigated far optimizing the production of alkaline protease from V. parahaemolyticus ATCC 17802. Maximal activity of the protease was obtained when the bacteria were grown in 2% skim milk medium in 0.1M tris/HCl buffer (pH 7.6). Maximal activity of the protease was obtained when the bacteria were growls at initial pH of 7.6, incubation temperature 37$^{\circ}C$ and shaking rate of 250 rpm.

Temperature-Dependency Urease Activity in Vibrio parahaemolyticus is Related to Transcriptional Activator UreR

  • Vibrio parahaemolyticus possessing urease-positive property is relatively rare, but such strains consistently exhibit the TDH-related hemolysin (TRH) gene. In this study, we examined the effects of incubation temperature on urease activity expression, using the TH3996 and AQ4673 strains where the enzyme activity is known to be temperature-dependent and -independent, respectively. In the TH3996 strain, $\beta$-galactosidase activity was 4.4-fold lower after $30^{\circ}C$ cultivation than after $37^{\circ}C$ in a ureR-lacZ fusion strain, but temperature dependency was not found in ureD- or nikA-lacZ fusion strains. However, ureR-, ureD-, and nikA-lacZ fusions of the AQ4673 strain was not influenced by incubation temperature. We compared the promoter sequences of ureR between the above two strains. Intriguingly, we detected mismatches of two nucleotides between the two strains located at positions -66 and -108 upstream of the methionine initiation codon for UreR. Additionally, urease activity was not affected by culture temperature at either $30^{\circ}C$ or $37^{\circ}C$ by allelic introduction of the AQ4673 ureR gene into the TH3996 ureR deletion mutant. Taken together, our study demonstrates that the transcriptional factor UreR is involved in the temperature dependency of urease activity, and two nucleotides within the ureR promoter region are of particular importance for the urease activity dependency of V. parahaemolyticus.

Study on the Distribution of Vibrio parahaemolyticus from Various Kinds of Shells in Kunsan Bay (군산만에서 서식하는 패류의 장염 비브리오에 관한 분포연구)

  • The distribution of V. parahaemolyticus was surveyed from various kinds of shells in Kunsan Bay from July to September, 1987. 1be morphological, biochemical and serological characteristics of the isolated strains were studied. The results were as follows: 1. 41 strains were isolated from 1,350 specimens of shells (Crassostrea gigas, Tapes philippinarum, Meretrix Iusoria) 2. The isolation rates of V. parahaemolytieus were 3% in July, 3.8% in August, and 2.2% in September, respectively. 3. V. parahaemolytkus was more frequently isolated from Kunsan (20 strains) than Bideukgi (12 strains) and Gae Hwa-do (9 strains). 4. V. parahaemolyticus was more frequently isolated from C gigas (23 strains) than other shells. 5. Kanagawa hemolysis reactions were all negative. 6. 6 Strains positive to K pooled antiserum included K IV, K V, K VI and K vn type.

Antimicrobial Activity of Pine Needle Extract and Horseradish on the Growth of Vibrio (솔잎 추출물과 고추냉이의 Vibrio에 대한 항균활성)

  • The effects of pine needle extract and horseradish on the growth of Vibrio isolated from crab and flat fish were investigated. The isolated Vibrios were identified as Vibrio parahaemolyticus HY I and V. vulnificus FST I by Api 20E kit. The growth of V. parahaemolyticus HY 1, V. vulnificus FST I and V. perahaernolytich ATCC17802 were inhibited in tryptic soy broth (TSB) containing 1% pine needle ethanol extract. The growth of the Vibrios was more 2 log inhibited in TSB containing 1% pine needle extract and 1% horseradish than in TSB containing 1% horseradish alone. Viable cells of tile Vibrios were decreased more rapidly about 2~3 log in soysauce containing 1% pine needle extract and 1% of horseradish than in soysauce and in soysauce containing 1% horeseradish. Sensory quality of horseradish sauce containing 1% of pine needle extract was similar to that of horseradish sauce (p<0.05).

Heterogeneity Analysis of the 16S rRNA Gene Sequences of the Genus Vibrio (Vibrio 속 16S rRNA 유전자 염기서열의 이질성 분석)

  • Bacterial 16S rRNA gene sequences have been widely used for the studies on molecular phylogeny, evolutional history, and molecular detections. Bacterial genomes have multiple rRNA operons, of which gene sequences sometimes are variable. In the present study, heterogeneity of the Vibrio 16S rRNA gene sequences were investigated. Vibrio 16S rRNA sequences were obtained from GenBank databases, considering the completion of gene annotation of Vibrio genome sequences. These included V. cholerae, V. harveyi, V. parahaemolyticus, V. splendidus, and V. vulnificus. Chromosome 1 of the studied Vibrio had 7~10 copies of the 16S rRNA gene, and their intragenomic variations were less than 0.9% dissimilarity (more than 99.1% DNA similarity). Chromosome 2 had none or single 16S rRNA gene. Intragenomic 16S rRNA genotypes were detected at least 5 types (V. vulnificus #CMCP6) to 8 types (V. parahaemolyticus #RIMD 2210633, V. harveyi #ATCC BAA-1116). These suggest that Vibrio has high heterogeneity of the 16S rRNA gene sequences.
