The water extracts of 83 oriental herbal medicines (Hanbang) which have been clinically used to treat bacterial infections in Korea were screened for in vitro antibacterial activity by the paper disc assay method. Two Gram positive bacteria, Staphylococcus aureus SG 511, Bacillus subtilis ATCC 6633, and two Gram negative bacteria, Escherichia coli 055, Pseudomonas aeruginosa 9027 were used as test organisms. Among the extracts tested, MeOH extract of Baenongtang showed remarkably potent antibacterial activity. Activity-guided chromatographic fractionations of the $CH_2Cl_2$ extract of Baenongtang afforded seven antibacterial compounds.

Objectives: Pleurotus eryngii is used both for edible and medicinal purposes, and has a physiological activity. The purpose of this study is to investigate the antibacterial effect of Pleurotus eryngii against six oral pathogens (Staphylococcus aureus, Streptococcus criceti, Streptococcus mutans, Streptococcus ratti, Streptococcus sobrinus, and Actinomyces viscosus). Methods: The antibacterial activities of various extracts of Pleurotus eryngii were examined by disc diffusion assay and minimum inhibitory concentration (MIC). The disc diffusion assay was performed by putting a paper disc soaked in extracts on plates inoculated bacterial cultures. The MIC of these extracts was determined by using a broth microdilution assay at a concentration ranging between 0.03 mg/ml to 15.00 mg/ml. The growth inhibition effect of extracts was measured at 600 nm for 24 hrs. Results: The antibacterial activity was confirmed against all six tested bacteria at Pleurotus eryngii ethyl acetate extract by the disc diffusion method. Acetone extract showed the antibacterial activity only against 4 strains containing Streptococcus criceti, Streptococcus mutans, Streptococcus ratti, and Actinomyces viscosus. In ethanol extract, no activity was observed against other strains except Staphylococcus aureus. MIC values of ethyl acetate extract were the same, 7.50 mg/ml in all tested bacteria. Conclusions: Pleurotus eryngii exhibited the antibacterial activity against oral pathogens (Staphylococcus aureus, Streptococcus criceti, Streptococcus mutans, Streptococcus ratti, Streptococcus sobrinus, and Actinomyces viscosus). Thus, Pleurotus eryngii may be considered as a natural antibacterial agent for treatment of dental diseases.
To obtain soil microorganisms producing antimigratory activity which is important in controlling the metastasis of cancer cells, more than three hundreds of soil microbes were isolated from sixteen soil sources including Namsan mountain and designated as DGU1001-10338. At first, their antibiotic activities were examined by paper-disc method. More than 40 soil microbes produced compounds with antibiotic activity. Then, antimigratory activities of selected soil microorganisms were examined in a sphingosylphosphorylcholine-induced migration assay in PANC-1 cells. Six of 42 soil microorganisms having antibacterial activity also had more than 45% inhibitory activity on migration of PANC-1 cells. These results suggested that selected soil microorganisms were a useful starting point to find compounds for controlling metastasis of cancer cells.
In this study, we investigated antimicrobial and cytotoxicity effects to each fraction extracted from Solanum lyratum (SL), which were extracted methanol (SLM) and then the extract was fractionated into five different types : hexane (SLMH), ethyl ether (SLMEE), ethylacetate (SLMEA), butanol (SLMB) and aqueous (SLMA). The antimicrobial activity was analyzed by the paper disc method. Among the various solvent fractions, SLMEA showed the strongest antimicrobial activies. The cytotoxicity of SL fractions on HeLa, MCF-7, HT-29 and HepG2 cells was evaluated by MTT assay. Among various partition layers, SLMEE showed the strongest cytotoxic effects to all cancer cell lines. We also observed that quinone reductase (QR) was induced by all fraction layers of SL to HepG2 cells. Since the QR-induced effects of SLMEE on HepG2 cells at $160{\mu}g/ml$ concentration showed 2.1 when compared with a control value of 1.0, inducer of QR for cancer protection may be contained in this fraction.
Histone deacetylase I (HDAC1) works as one of the components in a nucleosome remodeling (NuRD) complex that consists of several proteins, including metastasis-associated protein 1 (MTA1). Since the protein-protein interaction of HDAC1 and MTA1 would appear to be important for both the integrity and functionality of the HDAC1 complex, the interruption of the HDAC1 and MTA1 interaction may be an efficient way to regulate the biological function of the HDAC1 complex. Based on this idea, a yeast two-hybrid system was constructed with HDAC1 and MTA1 expressing vectors in the DNA binding and activation domains, respectively. To verify the efficiency of the assay system, 3,500 microbial metabolite libraries were tested using the paper disc method, and KB0699 was found to inhibit the HDAC1 and MTA1 interaction without any toxicity to the wild-type yeast. Furthermore, KB0699 blocked the interaction of HDAC1 and MTA1 in an in vitro GST pull down assay and induced morphological changes in B16/BL6 melanoma cells, indicating the interruption of the HDAC1 complex function. Accordingly, these results demonstrated that the yeast assay strain developed in this study could be a valuable tool for the isolation of a HDAC1 complex disruptor.

The growth inhibitiory effect of chaff vinegar against various food-borne pathogenic bacteria was evaluated. Bacterial growth was evaluated in chaff vinegar at concentrations of 15, 30, 50, 65, 80, and $100\%$ using the paper disc diffusion method and 0.5, 1.0, 1.5, 1.8, 2.0, 2.2 and $2.5\%$ in broth. In the paper disc diffusion assay, chaff vinegar showed a clear zone on both the Gram-positive bacteria; Listeria monocytogenes, Staphylococcus aureus and Gram-negative bacteria; Escherichia coli O157:H7, Salmonella Typhimurium, and Vibrio parahaemolyticus. Chaff vinegar exhibited the greatest growth inhibition for V. parahaemolyticus. The bactericidal effect of chaff vinegar on the E. coli O157:H7 was tested at concentrations ranging from 0.5 to $2.5\%$ (v/v) in the LB broth media. Chaff vinegar retarded the lag phase time of the growth curve in proportion in a concentration-dependent manner. Chaff vinegar at $2.5\%$ completely inhibited the growth of E. coli O157:H7.
In order to develop an analytical method for amikacin and butirosin in presence of their parent antibiotics, kanamycin A and ribostamycin, high-performance liquid chromatographic technique and microbioassay method were evaluated and compared. Using high performance liquid chromatography, two acylated antibiotics, amikacin and butirosin was partially separated from their parent antibiotics, to provide a qualitative analytical method. In microbioassay using Pseudomonas aeruginosa TI-13, a producer of aminoglycoside-3-phosphotransferase I, only acylated antibiotics were selectively analyzed when paper disc-susceptibility assay was used. The standard curve showed a good correlation between the response and odse in semilogarithmic plat with correlation coefficients above 0.96, and analytical deviation from expected dose was within 10%.

Objectives : The purpose of this study was to research the antibiotic, antioxidant and whitening effects of Jwa Kum-Whan(JKW) and Soo Ryeon-Whan(SRW). Both Jwa Kum-Whan and Soo Ryeon-Whan are composed of Coptidis rhizoma and Evodiae fructus, but the ratios of the two species are different. JKW is composed of Coptidis rhizoma and Evodiae fructus by ratio of 6:1 and SRW's ratio is 1:1. Methods : Antibiotic activities of JKW and SRW's water extracts were studied by paper disc diffusion method. Four kinds of bacteria were applied in paper disc, and each extract was dropped, individually. The diameter of inhibition zone was measured. DPPH assay was used for studying free radical scavenging activity. DPPH is quantitatively decolorized from purple color by antioxidant material. Change of color was measured by spectrophotometer. Whitening activity was analyzed by tyrosinase inhibition assay. Tyrosinase is major enzyme for control process of making eumelanin. Optical Density was measured by spectrophotometer. Results : Candida albicans and Staphylococcus aureus's inhibition zone were made large available by Both prescriptions. Inhibition zone's diameter of JKW was preferable to SRW's. Radical scavenging activity was better at SRW, but JKW's activity was available, too. Tyrosinase inhibition activity was found out available only for JKW, not for SRW. Conclusions : JKW had antibiotic effects for Candida albicans, Staphylococcus aureus. And had antioxidant, whitening effects. SRW had antibiotic, antioxidant activities but not whitening effect.
In this study, antibacterial activity and cytotoxicity of the extracts from pericarp and seed of $Vitis$$coignetiea$, which were extracted with 0.1% HCl-60% ethanol, were analyzed. The antibacterial activity of the extracts was determined by paper disc diffusion method against food spoilage and food-borne pathogens. The pericarp extract showed high antibacterial activity against $Bacillus$$cereus$, $Escherichia$$coli$ O157:H7, and $Pseudomonas$$aeruginosa$, and the seed extract represented the high antibacterial activity against $B.$$cereus$, $E.$$coli$ O157:H7, and $Staphylococcus$$aureus$. The cytotoxicity of the $Vitis$$coignetiea$ extract against human cancer cells was determined using the MTT assay and SRB assay. The pericarp extract represented strong growth-inhibition activity against G361 and Hep3B cells and the seed extract greatly inhibited the growth of HeLa and G361 cells in the MTT assay. In addition, the pericarp extract displayed a high inhibition activity against the growth of AGS cells and the seed extract greatly inhibited the growth of HeLa, Hep3B, and MCF7 cells in the SRB assay. Especially, the cytotoxicities of the seed extract against HeLa were significantly higher than those of the extract against other cancer cells at all test concentrations. This study demonstrates that the extract from pericarp and seed of $Vitis$$coignetiea$ possess high antibacterial activity and cytotoxicity.
