BIOASSAY OF HUMNA TOOTH PROTEIN BLOTTED POLYVINYLIDENE DIFLUORIDE(PVDF)MEMBRANE (사람치아 단백질을 분리 흡착한 PVDF막의 생체반응에 관한 연구)

  • Purpose: Human tooth proteins are highly heterogeneous, comprising diverse proteins derived from a number of genes. The attempts to identify protein for activity of tooth matrix proteins have been defied by several factors. First, the amount of proteins within teeth is very small relative to many extracellular matrix proteins of other tissues. Second, the bioassay system is tedious and needed for long time. Therefore we tried to find easy techniques, which increase the product rate, and an assay of small proteins, with which amino acid sequence is possible without additional procedures. Materials and Methods: Total protein were extracted from 300 g enamel removed teeth and 600 g teeth with 4 mol/L guanidine HCl and purified by gel chromatography. Aliquot of proteins was implanted into muscle pouches in Sprague-Dawley rats for bioassay. By SDS-PAGE and membrane blotting, molecular weight of each protein was estimated and a partial amino acid sequence was obtained. Each fraction blotted on the membrane was cut out and inserted in rat ectopic model. Results: In dissociative method, total tooth proteins were obtained 1mg/ml from enamel removed teeth and 3.5 mg/ml from teeth. In SDS-PAGE, four clear bands at the sites corresponding to 66, 40, 20 and 18 kD. Especially The 66 kD band was clearly exhibited. Amino acid sequencing from tooth could be possible using PVDF membrane blotting technique. In amino acid sequencing, 66 kD protein was identified as albumin. Conclusion: Compared with conventional method for extraction of teeth protein and bioassay of proteins, the methods in this study were easy, time-saving and more productive technique. The matured tooth proteins omitting additional procedure of mechanical removal of enamel were simply analyzed using blotted PVDF membrane. This method seems to make a contribution as a technique for bioassay and amino acid sequencing of protein.

Postmorten Changes in the Myofibrillar Proteins of Alkaline and Acid Rigor Rabbit Muscle during Storage (사후강직형태에 따른 근원섬유 단백질의 이화학적 성질변화)

  • This experiment was carried out to investigate some physico-chemical properties of myofibrillar proteins isolated from alkaline and acid rigor muscles of rabbit. The degree of fragmentation was about 0.35 at 3 days of postmortem in acid rigor muscle fiber, whereas it was only 0.3 at 7 days of postmortem in alkaline rigormuscle fiber. $Mg^{2+}$-activated ATPase activities of actomyosin were increased to the highest level at 1st day in acid rigor and at 3rd day in alkaline rigor muscle fiber. $Ca^{2+}$-activated ATPase activities of actomyosin were slightly increased at postmortem. $Mg^{2+}$-, and $Ca^{2+}$-activated ATPase activities were higher in alkaline rigor muscle than those in acid rigor muscle at postmortem. Solubility of actomyosin increased with postmortem but no differences were observed in rigor types. SDS-PAGE(sodium dodecyl sulfate polyacrylamide gel electrophoresis) band patterns of myofibrils did not show significant differences between rigor types, but newly 30.000 dalton proteins were appeared at 3 day postmortem in both rigor types.

Influence of NaCl and pH on Hydrolysis of Chicken Myofibrillar Proteins by Leukocyte Lysosomal Proteinases (Leucocyte lysosomal proteinase에 의한 닭의 근섬유(筋纖維) 단백질(蛋白質) 분해(分解)에 미치는 NaCl과 pH의 영향(影響))

  • The influence of NaCl and pH on degradation of chicken breast muscle myofibrillar proteins by porcine leukocyte lysosomal proteinases was investigated. The degradation reactions were carried out at $38^{\circ}C$ for 24hours under different conditions. The degradation of myofibrillar proteins by leukocyte lysosomal enzymes at various pH values was limited to partial hydrolysis. Reactions at higher pH values resulted in lower molecular weight degradation products while reactions at lower pH resulted in higher molecular weight degradation products. When NaCl was added into the reaction mixture, enzyme activities of degradation were increased at all pH values studied, as evidenced by NPN-analysis and SDS-PAGE. More severe degradation was observed with higher salt concentration. The concentration of 0.5M NaCl in the reaction mixture gave more degradation of myosin heavy chain by enzyme than that of 0.1M NaCl.

  • Recently marketing methods of small and medium exporting firms have changed from internet marketing using homepage, or e-catalogs, to search engine marketing. However, there is no specific proof of search engine marketing effectiveness. Therefore the purpose of this research is to explore marketing performance of search engine marketing(SEM) based on search engine optimization. In order to build an optimal SEM strategy, quantitative data are collected from the Google-analytics such as homepage visits, page views, and traffic source for three years. At the same time, this study has carried out a survey to measure the qualitative effectiveness. The result of this quantitative study suggests that the existing carryover effects and lag effects would be maintained through search engine optimization. Meanwhile, the qualitative survey shows that satisfaction and awareness of homepage have been improved after search engine optimization. This can support logically increase of homepage visiting ratio of quantitative analysis. Also exporting companies know very well, that traffic and page views have increased after search engine optimization.

Antigenetic effects of the eluted proteins from house dust mite (Dermatophagoides pteronyssinus) in dogs infested with sarcoptic mite (Sarcoptes scabiei var. canis) (집먼지진드기에서 분리한 용출단백질의 개옴진드기 감염증에 대한 항원효과)

  • Canine sarcoptic mite (Sarcoptes scabiei var. canis) is ectoparasite which burrow usually in the stratum corneum of the skin of dogs. Antigens from the burrowing mites induce humoral and cellmediated immune responses in the hosts. The effect of antigenecity induced by somatic antigens of house dust mite (Dermatophagoides pteronyssinus) isolated by continuous elution has been evaluated in canine sarcoptic mites infestation. Continuous elution was carried out in 7.5% SDS-PAGE to isolate proteins of common antigens from somatic antigens of house dust mite. These eluted proteins from somatic antigens of house dust mite were confirmed by Western blotting in 7.5% SDS-PAGE, and eluted proteins (65, 60 kDa) were isolated. To evaluate the antigenetic effect of eluted proteins, eight dogs were divided as 4 groups such as non-vaccinated and non-challenged control (Group I), challenged control (Group II), vaccinated (Group III), and vaccinatedandchallenged (Group IV) groups. Group II and IV were artificially infested canine sarcoptic mites. Group III and IV were immunized with eluted proteins (65, 60 kDa). At the 6th week of the vaccination, the antibody titers of Group of IV were statistically significant higher than those of Group II (p<0.05). And antibody titers of Group III were also statistically significant higher than those of Group I (p<0.05). From these result, it is possible to replace somatic antigens of canine sarcoptic mites with eluted proteins from somatic antigens of house dust mites in order to diagnose and prevent the canine sarcoptic mite infestations.

Garbage Collection Method for NAND Flash Memory based on Analysis of Page Ratio (페이지 비율 분석 기반의 NAND 플래시 메모리를 위한 가비지 컬렉션 기법)

  • NAND flash memory is widely used in embedded systems because of many attractive features, such as small size, light weight, low power consumption and fast access speed. However, it requires garbage collection, which includes erase operations. Erase operation is very slow. Besides, the number of the erase operations allowed to be carried out for each block is limited. The proposed garbage collection method focuses on minimizing the total number of erase operations, the deviation value of each block and the garbage collection time. NAND flash memory consists of pages of three types, such as valid pages, invalid pages and free pages. In order to achieve above goals, we use a page rate to decide when to do garbage collection and to select the target victim block. Additionally, We implement allocating method and group management method. Simulation results show that the proposed policy performs better than Greedy or CAT with the maximum rate at 82% of reduction in the deviation value of erase operation and 75% reduction in garbage collection time.

  • This study aims to measure Korea's global competitiveness in intelligent information technology, which is the core technology of the 4th industrial revolution. For analysis, we collect patents of each field and prior patents cited by them, which are applied at the U.S. Patent Office (USPTO) between 2010 and 2018 from PATSTAT Online. A global knowledge transfer network was established by grouping citing- and cited-relationships at a national level. The in-degree centrality is used to evaluate technology acceptance, which indicates the process of absorbing existing technological knowledge to create new knowledge in each field. Second, to evaluate the impact of existing technological knowledge on the creation of new one, the out-degree centrality is investigated. Third, we apply the PageRank algorithm to qualitatively and quantitatively investigate the importance of the relationships between countries. As a result, it is confirmed through all the indicators that the AI sector is currently the least competitive.

  • Recently, interest in programming and artificial intelligence is continuously increasing, and software education is being implemented as a mandatory education from elementary school. For efficient programming education, it is basically necessary to build a lab environment suitable for students and teachers, but there are performance problems due to the inadequacy of old computers and network equipment. Therefore, in this paper, we propose a web page-based online practice environment and algorithm competition scoring system using React and Spring boot to solve the problem of the programming practice environment. Through this, it is thought that programming learning can be carried out using only a web browser even on low-spec computers. In addition, since various programming languages can be learned irrespective of the language to be learned, it is considered that the time cost for establishing a practice environment can be reduced.

Development of an impact Identification Program in Mathematical Education Research Using Machine Learning and Network (기계학습과 네트워크를 이용한 수학교육 연구의 영향력 판별 프로그램 개발)

  • This study presents a machine learning program designed to identify impactful papers in the field of mathematics education. To achieve this objective, we examined the impact of papers from a scientific econometrics perspective, developed a mathematics education research network, and defined the impact of mathematics education research using PageRank, a network centrality index. We developed a machine learning model to determine the impact of mathematics education research and identified the journals with the highest percentage of impactful articles to be the Journal for Research in Mathematics Education (25.66%), Educational Studies in Mathematics (22.12%), Zentralblatt für Didaktik der Mathematik (8.46%), Journal of Mathematics Teacher Education (5.8%), and Journal of Mathematical Behaviour (5.51%). The results of the machine learning program were similar to the findings of previous studies that were read and evaluated qualitatively by experts in mathematics education. Significantly, the AI-assisted impact evaluation of mathematics education research, which typically requires significant human resources and time, was carried out efficiently in this study.

Biological and biochemical modulation of Trichomonas vaginalis KT9 isolate after shifting of culture medium from TPS-1 into TYM (TPS-1 배지 및 TYM 배지에서 배양된 질편모충의 생물학적 특성의 변화)

  • To evaluate the biological and biochemical characteristics of Trichomonas vaginalis KT9 isolate, the growth and size of trichomonads, pathogenicity in mouse, protein profiles and proteinase activity were examined after shifting the medium from TPS-1 into TYM. Generation time of trichomonads in TYM medium was 4.5 hr in comparison to TPS-1 with 7.1 hr. Size of trichomonads cultured in TPS-1 medium ($8.5{\;}{\pm}{\;}0.9{\;}{\times}{\;}6.0{\;}{\pm}{\;}0.9{\;}{\mu\textrm{m}}$) was significantly smaller than those in TYM medium ($10.9{\;}{\pm}{\;}1.4{\;}{\times}{\;}8.2{\;}{\pm}{\;}0.9{\;}{\mu\textrm{m}}$). Trichomonads cultured in TYM medium produced subcutaneous abscess in 9 out of 10 mice, whereas those in TPS-1 medium produced abscesses in 2 out of 10 mice. In SDS-PAGE, trichomonad Iysates from both media showed ten common bands. However, trichomonads in TYM medium showed additional bands of 136 kDa, 116 kDa and 40 kDa in comparison to those in TPS-1 with 100 kDa. By immunoblot with T. vaginalis-immunized rabbit sera, T. vaginalis cultivated in both TYM and TPS-1 media showed 5 common bands. and unique bands of 116 kDa. 105 kDa. and 86 kDa were observed in trichomonads in TYM while a 140 kDa band in those in TPS-1. In gelatin SDS-PAGE, trichomonads in TYM degraded gelatin stronger than those in TPS-1. Also protease activity of trichomonads in TYM was significantly higher than that of trichomonads in TPS-1 using Bz-Pro-Phe-Arg-Nan as a substrate. According to the results, it is assumed that the shift from TPS-1 into TYM medium for cultivation of T. vaginalis might modulate the biological and biochemical properties of T vaginalis in vitro.
