The Ergogenic Effects of Red Ginseng and Paeonia radix Mixtures

  • Objectives: This study was designed to identify the ergogenic effects of Red ginseng and Paeonia radix mixtures and optimal ratios. Materials and Methods: This study was conducted by administering treatments of Red ginseng and Paeonia radix mixtures to rats and by measuring the time to exhaustion by treadmill running. Results: The treatment of Red ginseng and Paeonia radix mixtures to the rats increased the time to exhaustion by treadmill running. The most potent inhibition of Red ginseng and Paeonia radix mixtures on the 5-HT synthesis and the TPH expression in the dorsal raphe was observed at the dose of 200 mg/kg and the optimal ratio of Red ginseng and Paeonia radix for the maximum efficacy was 50:50. Under normal conditions (not exercise), long-term treatment of Red ginseng and Paeonia radix mixtures did not affect the 5-HT synthesis and the TPH expression in the dorsal raphe, suggesting that Red ginseng and Paeonia radix mixtures does not alter serotonin level in the normal rats. The suppressive effect of Red ginseng and Paeonia radix mixtures on the 5-HT synthesis and the TPH expression during exercise is a possible ergogenic mechanism of these mixtures. Conclusions : Red ginseng and Paeonia radix mixtures reduce exercise-induced fatigue, and have the effect of acting as ergogenic aids on the time to exhaustion by treadmill exercise and on 5-HT synthesis and TPH expression.

목단줄기를 접목시킨 작약근과 목단피의 생태적 특성조사 및 성분분석에 의한 품질 비교 (Comparison of Quality and Ecological Properties Radix and Moutan Cortex, which were Paeonia Radix Grafted with Moutan Stem)

  • 약용식물의 생산량을 증가시키고 지상부 꽃은 관상용으로 활용할 수 있는 연구의 일환으로 목단 줄기에 작약 뿌리를 접목시켜 생육생태를 관찰하고 유효지표성분외에 무기성분, 지방산, 유기산등 성분변화 및 향미 관능검사등을 비교한 결과 생육생태를 볼 때 지상부는 목단줄기에 목단꽃이 피었으며 지하부 뿌리는 작약뿌리와 목단뿌리가 혼재하여 생성되었으며, 작약뿌리와 목단뿌리의 혼재비율은 약 3:1정도로 나타났다. paeoniflorin및 paeonol 분석결과 건조시킨 작약근의 함량은 대건물 시료(g)중 9.91 mg/g및 0.80 mg/g,접목 작약근은 11.71 mg/g 및 0.35 mg/g, 목단피는 10.81 mg/g 및 2.84 mg/g, 접목 목단피는11.26 mg/g 및 3.28 mg/g으로 유사한 경향이었다. 무기성분별 10종 함량은 대체로 유사한 경향이었으나 Fe함량은 시료별로 차이가 있었으며 작약근은 86.06 mg/g, 접목 작약근 127.36 mg/g, 목단피 75.05 mg/g, 접목 목단피는 140.63 mg/g으로 함량 차이가 현저하였다. 지방산은 linoleic acid, palmitic acid, linolenic acid 및 oleic acid등이 주성분으로 총 11종을, 유기산은 oxalic acid, citric acid 및 malic acid등이 주성분으로 총 6종을 각각 분석하였으나 함량 조성은 대체로 유사한 경향이었다. 한편, 향미 관능평가 결과 작약근과 접목 작약근은 대체로 유사한 향미 패턴을 나타내었으나, 작약근은 작약근 향미 및 떫은맛이 다소 강하였으며, 목단피와 접목 목단피도 거의 유사한 향미를 나타내었으나, 목단피가 목단피 향미, 쓴 맛, 화한 맛 및 떫은 맛이 다소 강하였다.

Aqueous extract of Paeonia radix suppresses lipopolysaccharide-induced expressions of cyclooygenase-2 and inducible nitric oxide synthase in mouse BV2 microglial cells

  • Paeonia radix is the root of Paeonia aliflora Pallas, which is a perennial plant classified in the family Paeoniaceae. Paeonia radix possesses several pharmacological effects such as analgesic, anti-inflammatory and anti-allergic, anti-oxidative, and anti-coagulant activities. In this study, we investigated the effect of the aqueous extract of Paeonia radix on the lipopolysaccharide-induced inflammation in mouse BV2 microglial cells. The aqueous extract of Paeonia radix at respective concentration was treated one hour before lipopolysaccharide treatment. In the present results, the aqueous extract of Paeonia radix suppressed prostaglandin $E_2$ synthesis and nitric oxide production by inhibiting the lipopolysaccharide-stimulated mRNA expressions of cyclooxygenase-2 and inducible nitric oxide synthase in mouse BV2 microglial cells. These results demonstrate that Paeonia radix exerts anti-inflammatory and analgesic effects probably by suppressing mRNA expressions of cyclooxygenase-2 and inducible nitric oxide synthesis. The present study demonstrates that Paeonia radix may offer a valuable mean of therapy for brain inflammatory diseases.

Protective Effect of Red Ginseng and Paeonia radix against Nitric Oxide-Induced Apoptosis in Human Neuroblastoma SK-N-MC cells

  • Objectives : Nitric oxide(NO) is a reactive free radical and a messenger molecule in many physiological functions. However, excessive release NO of induces neurotoxicity. We investigated whether a mixture of red ginseng and paeonia radix prossesses a protective effect against sodium nitroprusside(SNP)-induced apoptosis in the human neuroblastoma cell line SK-N-MC. Methods : We performed 3- (4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay, 4,6-diamidino-2-phenylindole(DAPD) staining, terminal deoxynucleotidyl transferase(TdT)-mediated dUTP nick end labeling(TUNEL)assay, DNA fragmentation assay, reverse transcription-polymerase chain reaction(RT-PCR), Western blot analysis, and caspase-3 enzyme activity assay in SK-N-HFC cells. Result : MTT assay showed that SNP treatment significantly reduced the viabilities of cells and that pre-treatment with the red ginseng and paeonia radix mixture alleviated SNP-induced cytotoxicity. The cells treated with SNP exhibited several apoptotic features, while those pre-treated fir 1 h with the mixture of red ginseng and paeonia radix 1 h prior to SNP expose showed reduced apoptotic features. In addition, the cells pre-treated with the red ginseng and paeonia radix mixture for 1 h prior to SNP expose increased bel-2 expressions, decreased Bax expressions, and decreased caspase-3 enzyme activity. Conclusions : These results show that the red ginseng and paeonia radix mixture exerts a protective effect against SNP-induced apoptosis in SK-N-MC cells.

The Protective Effect of Paeoniae Radix Extract against 1-methyl-4-Phenylpyridium-induced Apoptosis on SK-N-MC Neuroblastoma Cells

  • Background: There are increasing neuro-degenerative disorders with aging. Paeoniae Radix(PR) possesses various pharmacological effects such as sedative, analgesic, anti-inflammatory, anti-stress and neuro-protective actions. Also antiaging and anti-cancer effects of PR were reported. Our purpose was to investigate whether PR is useful on the treatment of Parkinson's disease, one of the neuro-degenerative disorders. Objective: We investigated whether Paeonia Radix possesses a protective effect against 1-methyl-4 phenylpyridine(MPP+)-induced cytotoxicity in neuronal cells. Methods: 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay, flow cytometry, DNA fragmentation assay, reverse transcription-polymerase chain reaction(RT-PCR), and Western blotting were performed on SK-N-MC neuroblastoma cells. Results: Cells treated with MPP+ exhibited several apoptotic features, while cells pre-treated with Paeonia Radix prior to MPP+ exposure showed s decrease in the occurrence of apoptotic features. Conclusions: These results suggest that Paeonia Radix may exert a protective effect against MPP+-induced apoptosis in SK-N-MC neuroblastoma cells.

작약 생육시기에 따른 약근수량 및 Paeoniflorin함량 변화 (Yield of Paeonia Radix and Changes of Paeoniflorin Concentration in Paeonia Radix with Different Growing Stages)

  • 경북 영천군 독농가가 재배한 정식 4년차인 의성종과 밀양종(P. lactiflora Pall.에 속하는 지방종)작약을 공시재료로 하여 생육시기별로 paeoniflorin 및 내용성분을 조사하고 수확기별로 약근수량을 조사한 결과를 요약해 보면 1. 개화기인 5월에 저장근 건물비율은 약 23%로 가장 낮았고, 수축율은 47%로 가장 높아 서로 상반된 양상을 나타내었으며, 6월 이후는 이들 특성의 변화가 없어 안정적이었다. 2. 전 생육기간 중 내용성분의 변화가 가장 큰 시기는 개화기인 5월이었으며, 약근에 함유된 paeoniflorin, 조단백, 조섬유 및 조회분은 공통적으로 개화기인 5월에 함량이 일시적으로 증가하였고, 총당 및 조지방은 변화가 적었다. 3. Paeoniflorin은 5월 시기에 6.37%로 가장 높았으며 6월 이후 변화없이 안정된 경향을 보였고, 의성종과 밀양종의 품종간 차이는 뚜렷하여 밀양종이 약 2% 높았다. 4. 전 생육기간 중 내용성분의 변화가 심하였던 전기(2∼5월)와 안정적인 후기(6∼10월)의 특성간 상관관계 중 전·후기 모두 정의 유의상관을 가진 것은 전분과 건물중, 조섬유와 paeoniflorin이며 부의 유의상관을 가진 특성은 전분과 조섬유, 전분과 paeoniflorin, 전분과 건물중 이었다. 5. 경장, 주당경수, 건전경비율은 이병으로 인한 고사로 8월보다 10월 수확기에서 현저히 낮았으며, 약근수와 길이도 근부현상이 심하여 10월 수확기에서 감소되었다. 6. 4년생 작약의 수확적기는 8∼9월이 적당하며 지상부 고사가 심하면 수확기를 앞당기는 것이 수량 및 품질면에서 더욱 안정적이다.

Paeonia Radix decreases Intracerebral Hemorrhage-induced Neuronal Cell Death via Suppression on Caspase-3 Expressionin Rats

  • Objective : The inappropriate or excessive apoptosis has been known to be associated with neurodegenerative disorders including intracranial hemorrhage(ICH). Paeoniae radix, in traditional Korean medicine, has played its role as blood­nourisher and yin-astringent. In the present study, the effect of Paeoniae radix on the inhibition of neurodegeneration in the brain of rats after artificial ICH and on the resulting apoptosis was investigated. Methods : 30 rats were divided into 6 equal groups ; the sham-operation group, the hemorrhage-induction group, the hemorrhage-induction with 10, 50, 100, and 200 mg/kg Paeoniae radix-treated group, respectively. Stereotactic surgery was performed and collagenase was infused to induce ICH in the region of CA1 of hippocampus of rats. The sham group took only saline infusion. For 7 days after the surgery, 4 testing groups had intraperitoneal injections of Paeoniae radix extract. The step-down inhibitory avoidance task, measurement of neurodegeneration degree in the CA1 region of the hippocampus, and detection of caspase-3 and newly generated cells in the dentate gyrus were done after animal sacrifice. Results : Rats receiving Paeoniae radix extract showed increased latency time in the inhibitory avoidance task. The extension of neuron-deprived areas in the CA1 region was significantly suppressed in the Paeonia treated groups. Also expressions of caspase-3 in the CA1 region and cortex were significantly inhibited in the Paeonia treated groups. The cell proliferation was evaluated by means of BrdU methods and proved to be decreased in the Paeonia treated groups. Conclusion : These results suggest that Paeoniae radix has potential to suppress short-tenn memory loss after devastating neurologic accidents. Also it was proved that Paeoniae radix has a neuroprotective effect and alleviates central nervous complications following intracerebral hemorrhage. Furthermore, it may imply that this medicinal plant can be widely used for vascular dementia and other neurodegenerative disorders.
