A Study on the Removal Efficiency of VOCs and Operating Characteristics by Using of Bioscrubber Trickling Filter(BSTE) (Bioscrubber Trickling Filter(BSTF)의 VOCs 제거 및 운전 특성에 관한 연구)

  • Volatile organic compounds (VOCs) and odor materials are major sources of air pollution in Ulsan city, where much chemical plants are located. Therefore, it is necessary to develop a new reactor which can remove VOCs and odor materials effectively and be equipped at the end of pipe easily. A modified reactor (bioscrubber trickling filter, BSTF), which have both characteristics of biofilter and bioscrubber, was developed and tested on its reactivity with several VOCs using two types of media, fiber and activated carbon 4- ceramic(A/C). It was observed that the removal efficiencies of several types of VOCs such as acetaldehyde, ethylalcohol, butanol, diethylamine and triethylamine were up to $95\%$ when they had about 100 ppm of initial concentration and 80 seconds of residence time. Good attachment of microorganisms to both media, where it is thought the reaction efficiency can be determined according to the amount of microorganisms attachment, observed with scanning electron microscopy(SEM). Initial pressure drops of the packed bed with both media were 229 $mmH_2O/m$ at A/C column and 670 $mmH_2O/m$, respectively. However, maximum pressure drop of fiber column during the operation was over 1,647 $mmH_2O/m$. Therefore, it was thought that the fiber material would not suitable to use in the local plant as a packed bed media.

Removal Charateristics of Erythrosine by Activated Carbon Adsorption (활성탄 흡착에 의한 Erythrosine의 제거 특성)

  • Erythrosine is used a food coloring, ink and dye, etc. but erithrosine is rarely used in United States due to its known hazards. The adsorption characteristics of erythrosine by granular activated carbon were investigated in the batch adsorber and the packed column. The adsorptivity of activated carbon for erythrosine were largely improved by pH control. When the pH was 11 in the sample, the erythrosine could be removed 98% of initial concentration. It was estabilished that the adsorption equilibrium of erythrosine on granular activated carbon was successfully fitted by Freundlich isotherm equation in the concentration range from 10mg/L to 1,000mg/L. The characteristics of breakthrough curve of activated carbon packed column depend on the design variables such as initial concentration, bed height, and flow rate.

Optimization of Fractionation Conditions for Natural Organic Matter in Water by DAX-8 Resin and its Application to Environmental Samples (DAX-8 레진의 수중 자연유기물의 분획조건 최적화 및 환경시료에의 적용)

  • Natural organic matter (NOM) is a heterogeneous mixture of organic matter with various polarities and molecular weights in an aquatic environment. This study investigated the effects of separation conditions (resin volume, organic matter, etc.) and the repeated use of the resin for the fractionation of organic components in the DAX resin fractionation method. The distribution characteristics of the organic components ((hydrophilic [Hi], hydrophobic acid [HoA], and hydrophobic neutral [HoN]) under the derived fractionation conditions were also analyzed. Constant fractionation results (i.e. HoA/Hi ratio) were obtained in the column capacity factor (i.e. the packed resin volume) in the range of 50 to 100. The resin-packed column maintained constant separation efficiency for up to two repeated uses. The above conditions were applied to wastewater and stream water samples (before and after rainfall). The results showed that the concentration of organic matter in the wastewater effluent was 2-15 times lower with an increased ratio of hydrophilicity to hydrophobicity (i.e. Ho/Hi) compared to the influent depending on the industrial wastewater classification. Particularly, HoN was found to have a high content distribution, 10.2-50.4% of the total dissolved organic matter (DOM), in the effluents. For the stream water, the content of Hi or HoN increased significantly after rainfall, suggesting a correlation with the distribution characteristics of pollutants from the stream watershed. The results provide useful data to enhance the reliability of the DAX resin fractionation and its application to environmental samples.

Removal of Quinoline Yellow by Granular Activated Carbon (입상 활성탄에 의한 Quinoline Yellow의 제거)

  • The adsorption characteristics of quinoline yellow by granular activated carbon were investigated experimently in the batch adsorber and packed column. The adsorptivity of activated carbon for quinoline yellow were largely improved by acidic pH and higher temperature. When the pH was 3 at $60^{\circ}C$, quinoline yellowcould be removed 97 percent of initial concentration(10 mg/L). It was estabilished that the adsorption equilibrium of quinoline yellow on granular activated carbon was successfully fitted by Freundlich isotherm equation in the temperature range from $25^{\circ}C$ to $60^{\circ}C$. The estimated values of k and ${\beta}$ are 38.71~166.60, 0.380~0.490, respectively. The breakthrough curve of activated carbon-packed column depends on the design variables such as initial concentration, bed height, and flow rate.

Electron Microscopic Study on the Development of the Lateral Motor Column in the Spinal Cord of the Human Fetus (인태아 척수 외측운동주의 발육에 관한 전자현미경적 연구)

  • The prenatal development of lateral motor columns in the lumbar spinal cord was studied by electron microscopy in human embryos and fetuses ranging from 9 mm to 260 mm crown-rump length ($5{\sim}30$ weeks of gestational age). At 9 mm embryo, the lateral motor column were developed from ventro-lateral projection into the marginal layer and composed of primitive neuroblasts. At 20 mm embryo the primitive motor neurons were packed closely together and could readly be distinguished from primitive glioblasts by a presence of large nuclei. The primitive multipolar neurons were observed in lateral motor column at 40 mm fetus. At 80 mm fetus multipolar neurons were characterized by their many dendrites and axons in the vicinity of motor neuron perikarya. At 260 mm fetus, the motor neurons were large and contained all intracytoplasmic structures in the cytoplasm which were also found in mature motor neuron in lateral motor column. The first axo-dendritic synapses found at 40 mm fetus and increased in number throughout fetal development. Axo-somatic synapses with spherical vesicles were first observed at 80 mm fetus. A few axo-somatic synapses were found at next prenatal stages. Axo-dendritic and axo-somatic synapses contained mixed populations of spherical and flattened vesicles by 120 mm fetus. These findings indicate that axo-dendritic synapses develop prior to axo-somatic synapses in the spinal cord during neurogenesis.

  • The optimum resolution condition of 11 organophosphorus pesticides by gas-liquid chromatography was studied using a capillary column. The injection modes and types of vaporization tubes were evaluated in relation to pesticide analysis. The best reproducibility of peak heights of pesticides was obtained by the split mode with the vaporization tube packed with quartz wool. A 25m SE-30 capillary column provided better resolution than SE-54 column for their optimum resolution. The column at $200^{\circ}C$ initially was heated by $5^{\circ}C/min\;to\;230^{\circ}C$ followed by $10^{\circ}C/min\;to\;270^{\circ}C$ and the temperatures were held constant for 5 minutes at $230^{\circ}C\;and\;270^{\circ}C$, respectively. The relative standard deviations of their retention times were less than 0.19%.

Removal Characteristics of Ethyl Acetate and 2-Butanol by a Biofilter Packed with Jeju Scoria

  • The removal characteristics of ethyl acetate and 2-butanol were investigated in a bench-scale down-flow biofilter packed with Jeju scoria medium. Various inlet concentrations and gas flow rates were tested. The adaptation times of microorganisms to the change of the influent concentration of ethyl acetate and 2-butanol gas were found to be about 3 days. At the inlet concentration of 300 ppmv and empty bed contact time (EBCT) of 15 see, the removal efficiencies of the biofilter for ethyl acetate and 2-butanol were above $99.9\%$. The maximum removal capacity of the biofilter for ethyl acetate was $316-318\;g/m^3/h$ and that for 2-butanol was $245-251\;g/m^3/h$. Overall, the removal capacity of the biofilter for ethyl acetate was $50-70\;g/m^3/h$ larger than that for 2-butanol. During the operation of 65 days, the pressure drop through the biofilter column was maintained below $13\;mmH_{2}O/m$. Although the pH in the drain water decreased from 7.2 to 5.0, the pH drop did not affect the removal of the gases. From the above results, the biofilter using Jeju scoria as a packing material seemed to very effectively treat waste gases such as ethyl acetate and 2-butanol.

Continuous removal of phosphorus in water by physicochemical method using zero valent iron packed column (영가철 충진 컬럼을 이용한 연속적인 물리화학적 수중 인 제거)

  • Excessive phosphorus in aquatic systems causes algal bloom resulting in eutrophication, DO depletion, decline in recreational value of water and foul tastes. To treat wastewater containing phosphorus including effluent of wastewater treatment plant, the continuous experiments were performed by using electrochemical way. The spherical ZVI and silica sand which act as physical filter are packed at appropriate volume ratio of 1:2. Electric potential is applied externally which can be changed as per the operational requirement. The results indicate that optimum hydraulic retention time of 36 minutes (10 mL/min at 1 L reactor) was required to meet the effluent standards. Lower concentrations of phosphorus (<10 mg/L as phosphate) were removed by precipitation by contact with iron. Thus, additional electric potential was not required. In order to remove high concentration phosphorus around 150 mg/L as phosphate, external electric potential of 600 V was applied to the reactor.

Comparative Evaluation of Three Purification Methods for the Nucleocapsid Protein of Newcastle Disease Virus from Escherichia coli Homogenates

  • In the present study, the performances of conventional purification methods, packed bed adsorption (PBA), and expanded bed adsorption (EBA) for the purification of the nucleocapsid protein (NP) of Newcastle disease virus (NDV) from Escherichia coli homogenates were evaluated. The conventional methods for the recovery of NP proteins involved multiple steps, such as centrifugation, precipitation, dialysis, and sucrose gradient ultracentrifugation. For the PBA, clarified feedstock was used for column loading, while in EBA, unclarified feedstock was used. Streamline chelating immobilized with $Ni^{2+}$ ion was used as an affinity ligand for both PBA and EBA. The final protein yield obtained in conventional and PBA methods was $1.26\%$ and $5.56\%$, respectively. It was demonstrated that EBA achieved the highest final protein yield of $9.6\%$ with a purification factor of 7. Additionally, the total processing time of the EBA process has been shortened by 8 times compared to that of the conventional method.

Optimized Conditions for In Situ Immobilization of Lipase in Aldehyde-silica Packed Columns

  • Optimal conditions for the in situ immobilization of lipase in aldehyde-silica packed columns, via reductive amination, were investigated. A reactant mixture, containing lipase and sodium borohydride (NaCBH), was recirculated through an aldehyde-silica packed column, such that the covalent bonding of the lipase, via amination between the amine group of the enzyme and the aldehyde terminal of the silica, and the reduction of the resulting imine group by NaCBH, could occur inside the bed, in situ. Mobile phase conditions in the ranges of pH $7.0{\~}7.8$, temperatures between $22{\~}28^{circ}C$ and flow rates from $0.8{\~}1.5\;BV/min$ were found to be optimal for the in situ immobilization, which routinely resulted in an immobilization of more than 70 mg­lipase/g-silica. Also, the optimal ratio and concentration for feed reactants in the in situ immobilization: mass ratio [NaCBH]/[lipase] of 0.3, at NaCBH and lipase concentrations of 0.75 and 2.5 g/L, respectively, were found to display the best immobilization characteristics for concentrations of up to 80 mg-lipase/g-silica, which was more than a 2-fold increase in immobilization compared to that obtained by batch immobilization. For tributyrin hydrolysis, the in situ immobilized lipase displayed lower activity per unit mass of enzyme than the batch-immobilized or free lipase, while allowing more than a $45\%$ increase in lipase activity per unit mass of silica compared to batch immobilization, because the quantity of the immobilization on silica was aug­mented by the in situ immobilization methodology used in this study.
