Background: The biological activities of ginseng saponins (ginsenosides) are associated with type, number, and position of sugar moieties linked to aglycone skeletons. Deglycosylated minor ginsenosides are known to be more biologically active than major ginsenosides. Accordingly, the deglycosylation of major ginsenosides can provide the multibioactive effects of ginsenosides. The purpose of this study was to transform ginsenoside Rb2, one of the protopanaxadiol-type major ginsenosides, into minor ginsenosides using ${\beta}$-glycosidase (BG-1) purified from Armillaria mellea mycelium. Methods: Ginsenoside Rb2 was hydrolyzed by using BG-1; the hydrolytic properties of Rb2 by BG-1 were also characterized. In addition, the influence of reaction conditions such as reaction time, pH, and temperature, and transformation pathways of Rb2, Rd, F2, compound O (C-O), and C-Y by treatment with BG-1 were investigated. Results: BG-1 first hydrolyzes 3-O-outer ${\beta}$-$\text\tiny{D}$-glucoside of Rb2, then 3-O-${\beta}$-$\text\tiny{D}$-glucoside of C-O into C-Y. C-Y was gradually converted into C-K with a prolonged reaction time, but the pathway of Rb2 ${\rightarrow}$ Rd ${\rightarrow}$ F2 ${\rightarrow}$ C-K was not observed. The optimum reaction conditions for C-Y and C-K formation from Rb2 by BG-1 were pH 4.0-4.5, temperature $45-60^{\circ}C$, and reaction time 72-96 h. Conclusion: ${\beta}$-Glycosidase purified from A. mellea mycelium can be efficiently used to transform Rb2 into C-Y and C-K. To our best knowledge, this is the first result of transformation from Rb2 into C-Y and C-K by basidiomycete mushroom enzyme.
In this study, we report the room temperature synthesis and luminescence properties of white-light-emitting Rb(V,P)$O_3$. The vanadate phosphor, RbV$O_3$, was synthesized by simple mixing of $RbCO_3$ and $V_2O_5$ at room temperature in air. New direct room temperature solidstate reaction is a cost-effective method to synthesize the above luminescent materials.

In order to determine the stability of ginseng components in this salt concentration when used to ginseng as additive ingredient of sauces or seasonings, we study on the content and charactristic of ginsenosides and changes in pH and color, ginseng tail and ginseng extract were treated with various concentration of NaCl solution. In this experiment, extract of ginseng tail were increased in pH as NaCl concentration were increased, but ginseng extract have not changed evidently. The both solution were decreased in color as the salt concentration were increased. Yield of n-butanol extract was decreased in 5% NaCl concentration, while it was increased in the above concentration, and ginseng extract was changed higher than ginseng tail. Ginsenosides content were increased in 5% NaCl concentration, both $ginsenosied-Rb_1$, $-Rb_2$, -Rc, -Rd of diol line and ginsenoside-Re of triol line and increased in above NaCl concentration. Especially ginsenoside-Re showed to sensitive response to the changes of the salt concentration.
We present the double resonance optical pumping(DROP) spectra according to the laser power, the polarization combination of lasers, and the alignment of lasers in the transition $5P_{3/2}-4D_{5/2}\;and\;5P_{3/2}-4D_{5/2}$ of $^{87}Rb.$ We observed obviously changing DROP spectrum according to the laser power in the transition $5P_{3/2}-4D_{5/2}$ involved the cycling transition. The laser power effects are attributed to the low optical-pumping-effect in the cycling transition. We observed changing DROP spectrum depending on the polarizations of the lasers. The laser polarization effects are attributed to the changing transition probability according to the polarizations of lasers. We compared the co-propagation with the counter-propagation and the spectral linewidths were 12.2 MHz and 6.9 MHz, respectively.
To investigate the effect of the weathering process of biotite on Cs sorption, sorption experiment of Cs with $10^{-3}M$ solution was carried out on the biotite reacted at different reaction times at pH 2 and 4, and 1 M solutions of Na, K, Ca, Mg, Rb, and Cs. Peak changes were observed for some samples by XRD, indicating that new mineral phase formed by biotite weathering. Among several factors, cations in solutions have the most significant influence on the mineralogical changes. The samples reacted with Na showed the most outstanding change with increasing peak width and appearance of $12{\AA}$ peak and $14{\AA}$ peak. This new peaks indicate the formation of hydrobiotite and vermiculite. The new peaks had stronger peak intensity for the sample reacted at pH 4 than that at pH 2, probably due to the fast dissolution of small particles and edges and resultant decrease in the formation of expandable layers. The biotite reacted at Mg solution showed small intensity at $14{\AA}$. Based on XRD results, the degree of biotite weathering was in the order of Na, Mg, and Ca. The samples reacted with K, Rb, Cs solutions did not show noticeable mineralogical changes caused by weathering. The amount of sorbed Cs on weathered biotite showed close relationship with the degree of weathering indicated by XRD. At both pH 2 and 4, the biotite reacted with Na solution showed the highest Cs sorption, and those with Mg and Ca solutions showed the next highest ones. The sorbed amounts of Cs on the bitote reacted with K, Rb, Cs solutions were relatively low. This indicates that at the Cs concentration ($10^{-3}M$) which we used for this experiment and which was much higher than the maximum Cs concentration sorbed on the frayed edge site, expandable layer plays more important role than frayed edge. In the cases of K, Rb, and Cs solutions, Cs sorption was decreased because K is the same cations as the one in the interlayer or the sorption of Rb and Cs on the frayed edge prevents the formation of expandable layers.
Background: Ginsenoside Rb2, a major active component of Panax ginseng, has various physiological activities, including anticancer and anti-inflammatory effects. However, the mechanisms underlying the rejuvenation effect of Rb2 in human skin cells have not been elucidated. Methods: We performed a senescence-associated β-galactosidase staining assay to confirm cellular senescence in human dermal fibroblasts (HDFs). The regulatory effects of Rb2 on autophagy were evaluated by analyzing the expression of autophagy marker proteins, such as microtubule-associated protein 1A/1B-light chain (LC) 3 and p62, using immunoblotting. Autophagosome and autolysosome formation was monitored using transmission electron microscopy. Autophagic flux was analyzed using tandem-labeled GFP-RFP-LC3, and lysosomal function was assessed with Lysotracker. We performed RNA sequencing to identify potential target genes related to HDF rejuvenation mediated by Rb2. To verify the functions of the target genes, we silenced them using shRNAs. Results: Rb2 decreased β-galactosidase activity and altered the expression of cell cycle regulatory proteins in senescent HDFs. Rb2 markedly induced the conversion of LC3-I to LC3-II and LC3 puncta. Moreover, Rb2 increased lysosomal function and red puncta in tandem-labeled GFP-RFP-LC3, which indicate that Rb2 promoted autophagic flux. RNA sequencing data showed that the expression of DNA damage-regulated autophagy modulator 2 (DRAM2) was induced by Rb2. In autophagy signaling, Rb2 activated the AMPK-ULK1 pathway and inactivated mTOR. DRAM2 knockdown inhibited autophagy and Rb2-restored cellular senescence. Conclusion: Rb2 reverses cellular senescence by activating autophagy via the AMPK-mTOR pathway and induction of DRAM2, suggesting that Rb2 might have potential value as an antiaging agent.

Background: RB1 (retinoblastoma 1) was reportedly one of the major determinative factors for sensitivity to taxanes in previous studies. In this study, we investigated the influence of RB1 single nucleotide polymorphisms (SNPs) on the efficacy of platinum-taxane regimens in advanced NSCLC patients. Materials and Methods: 234 cases of patients with advanced NSCLC who were treated with first-line platinum-taxane agents were enrolled in this study. Genomic DNA was extracted from patients' peripheral blood samples using a QIAamp DNA Maxi Kit, and genotyped by iSelect HD Bead-Chip. Results: Regression analyses were conducted through the univariate and multivariate Cox proportional hazards model in the 234 patients. The results showed that of the eight RB1 tagSNPs, only rs4151510 was a positive predictive factor for the advanced NSCLC patients treated with platinum taxanes regimen. The patients with G/G genotype of RB rs4151510 had longer overall survival (OS) than the non-G/G genotype (p=0.018). The histology was also correlated with OS in the whole advanced NSCLC patients. Three tagSNPs of RB1, rs4151510, rs4151465, rs9568036 were significantly associated with OS in the advanced NSCLC patients with squamous cell histology using Kaplan-Meier overall survival analysis stratified by histology. Conclusions: RB1 genomic variants were correlated with the efficacy of platinum-taxanes regimen. RB rs4151510 is an independent factor of the prognosis of NSCLC patients receiving platinum-taxane chemotherapy.
A novel ginsenoside-hydrolyzing ${\beta}$-glucosidase was purified from Paecilomyces Bainier sp. 229 by a combination of Q-Sepharose FF, phenyl-Sepharose CL-4B, and CHT ceramic hydroxyapatite column chromatography. The purified enzyme was a monomeric protein with a molecular mass estimated to be 115 kDa. The optimal enzyme activity was observed at pH 3.5 and $60^{\circ}C$. It was highly stable within pH 3-9 and at temperatures lower than $55^{\circ}C$. The enzyme was specific to ${\beta}$-glucoside. The order of enzyme activities against different types of ${\beta}$-glucosidic linkages was ${\beta}$-(1-6)>${\beta}$-(1-2)>${\beta}$-(1-4). The enzyme converted ginsenoside Rb1 to CK specifically and efficiently. An 84.3% amount of ginsenoside Rb1, with an initial concentration of 2 mM, was converted into CK in 24 h by the enzyme at $45^{\circ}C$ and pH 3.5. The hydrolysis pathway of ginsenoside Rb1 by the enzyme was $Rb1{\to}Rd{\to}F2{\to}CK$. Five tryptic peptide fragments of the enzyme were identified by a newly developed de novo sequencing method of post-source decay (PSD) matrix-assisted laser desorption/ionization (MALDI) mass spectrometry. By comparing the five identified peptide sequences with the NCBI database, this purified ${\beta}$-glucosidase proves to be a new protein that has not been reported before.
Laboratory-scale experiments were carried out to investigate the adsorption equilibrium, the adsorption kinetics and facilitated transport of two cationic dyes (Methylene Blue (MB) and Rhodamine B (RB)) on Polymer Inclusion Membrane (D2EHPA-PIM). Different adsorption isotherms (Freundlich, Langmuir and Temkin models) as well as kinetics models indicated that the adsorption process is spontaneous and exothermic. Under the optimal conditions, the adsorption removal efficiencies reach about 93% and 97% for MB and RB respectively. Different extraction values by D2EHPA-PIM were obtained for the two cationic dyes: MB is weakly extracted at pH 2.0 (E% = 18.7%) whilst E% = 82.4% was observed for RB at the same pH. This difference was exploited in a mixture containg both the 2 cationic dyes for the selective extraction of RB at pH 2. Desorption of both dyes was achieved from the membrane by using acidic aqueous solutions and desorption ratio up to 90% was obtained. The formulas of the extracted complexes by the PIMs were, determined by the method of slopes. The dyes transport was elucidated using mass transfer analysis where in it found relatively high values of the initial flux ($J_0$) as 41.57 and $18.74{\mu}mol.m^2.s^{-1}$ for MB and RB respectively.

An light-emitting diode (LED)-based light source was used as a monochromatic light source to determine the responses of raw ginseng roots (Panax ginseng Meyer) to specific emission spectra with respect to the production of ginsenosides. The ginsenoside content in the ginseng roots changed in response to the LED light treatments at $25^{\circ}C$ relative to the levels in the control roots that were treated in the dark or at $4^{\circ}C$ for 7 d. Ginseng roots were exposed to LEDs with four different peak emission wavelengths, 380, 450, 470, and 660 nm, in closed compartments. Compared with the control $4^{\circ}C$-treated roots, roots that were treated with 450 and 470 nm light showed a significantly increased production of ginsenosides (p<0.05), with increases of 64.9% and 74.1%, respectively. The contents of the ginsenosides $Rb_2$, Rc, and $Rg_1$ were significantly higher (p<0.05) in the 450 and 470 nm-treated root samples. The ratio of protopanaxadiol ginsenosides ($Rb_1$, $Rb_2$, Rc, and Rd) to protopanaxatriol ginsenosides ($Rb_1$, $Rb_2$, Re, and Rf) was significantly higher (p<0.05) in the 450 and 470 nm-treated root samples than in the control $4^{\circ}C$-treated roots. This is the first report that demonstrates the increase and conversion of ginsenosides in raw ginseng roots in response to exposure to LED light.
