Cloning and Expression of A Liquefying $\alpha$-Amylase Gene from Bacillus amyloliquefaciens in Bacillus subtilis (Bacillus amyloliquefaciens 액화형 $\alpha$-amylase 유전자의 클로닝 및 Bacillus subtilis에서의 발현)

  • A 5200 basepair DNA fragment containing the Bacillus amyloliquefaciens amyE gene, encoding liquefying $\alpha$-amylase (1,4-$\alpha$-1)-glucan glucanohydrolase, EC 3.2.1.1), has been inserted into BamHI site of the pUB110 and the hybrid plasmid was designated as pSKS3. The pSKS3 was transformed into the Bacillus subtilis KM2l3 as a host which is a saccharifying $\alpha$-amylase deficient mutant of Bacillus subtilis NA64, and the plasmid in the transformed cell was expressed $\alpha$-amylase production and kanamycin resistance. The $\alpha$-amylase production of the transformed cell was reduced to one fifth of that of the donor strain. The Bacillus subtilis KM2l3 tarring pSKS3 indicated that the amyE gene product is a polypeptide which has the same electrophoretic mobility with that of the Bacillus amyloliquefaciens, but different from the saccharifying $\alpha$-amylase of Bacillus subtilis NA64. It means that the amyE gene of pSKS3 originales from the Bacillus amyloliquefaciens.

Evaluation of pig behavior changes related to temperature, relative humidity, volatile organic compounds, and illuminance

  • The objective of this study was evaluation of pig behavior changes related to temperature, relative humidity, volatile organic compounds (VOCs), and illuminance. A total of 24 growing pigs ([Yorkshire × Landrace] × Duroc) were used in the experiment. A sensor was installed at a height of 0.5 m in the center of the pig house. In experiment 1, temperature was changed every four days to 18℃ (T1), 22℃ (T2), 26℃ (T3), and then 30℃ (T4). In experiment 2, relative humidity was adjusted to 45% (low humidity [LH]), 60% (middle humidity [MH]), and then 75% (high humidity [HH]) for four days. In experiment 3, after cleaning the pig house just before experiment, only minimal ventilation was provided. VOCs and pig behaviors were observed for 7 days without cleaning the pig house. In experiment 4, three light bulbs of 40 W (470 lumens / 45 lx; low illuminance [LI]), 75 W (1,055 lumens / 103 lx; middle illuminance [MI]), and 100 W (1,521 lumens / 146 lx; high illuminance [HI]) were used for four days each. Pig behavior analysis was performed for following criteria : Feed intake, Standing, Lying, Sitting, Drink water, Rooting, Posture transition (lying-standing), Posture transition (standing-lying), Wallowing, and Biting. In experiment 1, feed intake time was lower (p < 0.05) for the T3 than other treatment groups. Standing time was highest (p < 0.05) for the T1 and lowest (p < 0.05) for the T3. Lying time was shorter (p < 0.05) in T1 and T2 compared to T3 and T4. Drinking frequency was higher (p < 0.05) for the T4 than other treatment groups. In experiment 2, the frequency of rooting and wallowing increased (p < 0.05) with increasing humidity. LH showed the lowest (p < 0.05) rooting frequency and HH showed the highest (p < 0.05) rooting frequency. In experiment 3, VOCs concentration did not (p > 0.05) change pig behavior. In experiment 4, lying time was the longest (p < 0.05) at LI and shortest (p < 0.05) at HI. Therefore, pig behavior is heavily influenced by the environment, especially temperature and humidity. However, correlation between pig behavior to VOCs and illuminance seems to be needed more research.

Linker Scanning Analysis of the BPV-1 Upstream Regulatory Region

  • The upstream regulatory region (URR) of bovine papillomavirus type 1 (BPV) contains promoters and a conditional transcriptional enhancer that is trans-activated by the viral E2 protein. After deleting the 5' and 3' ends of BPV URR, BamHI linkers were inserted into several positions of BPV URR without causing an addition or a deletion of URR sequences. Most linker scanning mutations did not show any effects on the transcription of P7940 and P89 promoters in BPV URR. However, several mutants showed reduced transcriptional activities. Based on our results we found that the AP-2 and Sp1 binding sites were important for basal level transcription of BPV URR in the absence of the E2 protein and that the CTF/NF-1 site is dispensable for E2 transactivation of BPV URR transcription.

  • Among biological hydrogen production processes, fermentative processes have some advantages. In this research, the hydrogen producing bacterium was isolated from domestic landfill area and identified as Enterobacter sp. The strain was named Enterobacter sp. SNU-1453. Important parameters for the hydrogen process include pH, temperature, concentration of initial glucose, and kind of sugars. The pH of the culture medium significantly decreased as fermentation proceeded due to the accumulation of various organic acids, and this inhibited the $H_2$ production seriously. When pH was controlled at pH 7.0, hydrogen production was 2614.5 m1/1 in 17 hours. The increase of glucose concentration resulted in higher $H_2$ production. The productivity of this strain was 6.87 mmol $H_2/l$ per hi on concentration of 25g glucose/l. Enterobacter sp. SNU-1453 could utilize various sugars. These results indicate that Enterobacter sp. SNU-1453 has a high potential as a fermentative $H_2$ producer.

AN EVALUATION OF FRACTURE TOUGHNESS OF DENTAL CERAMICS BY INDENTATION FRACTURE METHOD (압자압입법에 의한 치과용 도재의 파괴인성 평가)

  • This study was performed to evaluate the fracture toughness of seven commercially available dental ceramics by indentation fracture method. All specimens were fabricated to the final dimensions of approximately 12mm in diameter 2mm in thickness. The characte-ristic indentation dimensions of Vickers or Knoop indentation were measured to calculate the fracture toughness values and Young’s moduli. The results obtained were summarized as follows; 1. Young’s modulus and Vickers hardness of Vita In-Ceram showed the maximum values of $154.4{\pm}49.2$ Gpa and $12.60{\pm}0.71$Gpa, respectively. Results of Scheff test exhibited the significant difference between Vita In-Ceram group and others(p<0.01). 2. Maximum fracture toughness of $2.562{\pm}0.37 MPam^{1/2}$ for Vita In-Ceram and the maxi-mum one of $0.908{\pm}0.132MPam^{1/2}$ for Vitadur-N were calculated. Results of Scheff test showed the significant difference between Vita In-Ceram group and Vita Hi-Ceram(p<0.05) : also between Vita In-Ceram or Vita Hi-Ceram and others(p<0.01). 3. The alumina-based core ceramics showed the aspect of Palmqvist crack for the indentation load of 49.0N, but others showed the median/lateral crack for the indentation load of 9.8N.

  • Purpose: This treatment planning study was undertaken to evaluate the impact of beam angle configuration of intensity-modulated radiotherapy (IMRT) on the dose of the normal liver in hepatocellular carcinoma (HCC). Materials and Methods: The computed tomography datasets of 25 patients treated with IMRT for HCC were selected. Two IMRT plans using five beams were made in each patient; beams with equidistance of $72^{\circ}$ (Plan I), and beams with a $30^{\circ}$ angle of separation entering the body near the tumor (Plan II). Both plans were generated using the same constraints in each patient. Conformity index (CI), homogeneity index (HI), gamma index, mean dose of the normal liver (Dmean_NL), Dmean_NL difference between the two plans, and percentage normal liver volumes receiving at least 10, 20, and 30 Gy (V10, V20, and V30) were evaluated and compared. Results: Dmean_NL, V10, and V20 were significantly better for Plan II. The Dmean_NL was significantly lower for peripheral (p = 0.001) and central tumors (p = 0.034). Dmean_NL differences between the two plans increased in proportion to gross tumor volume to normal liver volume ratios (p = 0.002). CI, HI, and gamma indices were not significantly different for the two plans. Conclusion: The IMRT plan based on beams with narrow separations reduced the irradiated dose of the normal liver, which would allow radiation dose escalation for HCC.

  • To obtain a new yeast strain that is able to efficiently produce ethanol from starch, the glucoamylase gene of Saccharomyces diastaticus was transformed into S. cerevisiae without a cloning vector. The competent cells of S. cerevisiae, induced by the treatment of Li$_2$SO$_4$, were transformed with the partial BamHI-digests of chromosomal DNA of S. diastaticus, and the transformants were selected by their abilities to utilize and ferment starch. The transformants, which appeared at a frequency of 8.5$\times$10$^{-7}$, were able to withstand up to 800 ppm of copper sulfate like the recipient and retained the phenotypic expression of the recipient with the exception of the acquisition of STA gene and MAL gene, as regards fermentation of carbohydrates. The enzymatic properties of glucoamylases produced by transformants were very similar to those produced by S. diastaticus as based on optimium pH and temperature.

  • Intracellular pH was determined by distribution of 5.5-dimethyl-2,4-oxazolidlnedione [DMO]in the skeletal muscle of dogs before and after lactic acidosis induced by intravenous infusion of lactic acid solution. After infusion of lactic acid solution arterial pH decreased from 7.40 to around 7.12 [P<0.001]and metabolic acidosis was induced. However, dose-pH change response was not proportional as in the case of hydrochloric acid infusion. During lactic acidosis, intracellular pH changed very little except when venous blood $pCO_2$ increased significantly. The decrease of intracellular pH in lactic acidosis might be due primarily to the increase of intracellular $pCO_2$. And during lactic acidosis, change of extracellular pH was larger than that of intracellular pH, and this was also the case of change In hydrogen Ion concentration in extracellular and intracellular fluid. The fact was estimated that exogenous lactic acid transported into the cell does not contribute to pH change by the participation in the metabolism. Change in plasma potassium Ion concentration was not eminent as metabolic acid-base disturbances by other origin, and changing pattern of Hi/He ratio was not same as Ki/Ke ratio. In spite of no changes in extracellular potassium ion concentration after exogenous lactic acidosis total amount of potassium ion in extracellular fluid increased from 12.62mEg to 18.26mEg [P< 0.05].

  • The effects of pH on the sequential sorption/desorption of chlorinated phenols (2-chlorophenol, 2.4-dichlorophenol and 2,4,5-trichlorophenol) in HDTMA-montmorillonite were investigated by maintaining pH 4.85 or 9.15 in the sequential batch sorption and desorption experiments. The chlorinated phenols are hydrophobic ionizable orginic compounds; they can exist as either neutral (pH << pKa) or anionic (pH >> pKa) forms. Among the tested chlorinated phenols, 2,4,5-trichlorophenol showed the highest sorption affinity at pH 4.85 as expected by the $K_{ow}$ . Neutral speciation at pH 4.85 exhibited higher sorption affinity than anionic speciation at pH 9.15. Our results indicates that desorption of chlorinated phenols is strongly dependent on pH of the aqueous phase. Freundlich model was used to analyze the single-solute sorption/desorption results. The ideal adsorbed solution theory(IAST) was employed to predict the hi-solute sorption/desorption equilibria.

  • Recent studies demonstrated the ability of carbon nanotube (CNT) to promote electron transfer reactions of important compounds and to impart higher stability onto electrochemical sensors. CNT-based sensors measured by hydroxyl radical concentration or pH value suggest great promise for biosensors. This paper describes a new method for fabricating a very simple and inexpensive pH sensor compose of single walled-carbon nanotubes (SW-CNTs) using an ultra-precision spray. CNT-based sensor shows pH sensitivity in buffer solution at different pH range. Our experimental results show the sensor responses to pH buffer solution and the conductance of depends on the pH values. These results support application possibility of SW-CNTs based pH sensor for mass production.
