Permeability of pH-sensitive membranes grafted by Fenton-type reaction: An experimental and modeling study

  • Membrane modification by different concentrations of acrylic acid has been described. Grafting of acrylic acid to the surface of a polypropylene membrane was obtained by a Fenton-type reaction. Membrane permeability seemed to have been dependent on the value of pH in the solution. To explain tendency, a simple theoretical model was developed. The model incorporates explicitly statistical conformations of a polyacid chain grafted onto the pore surface. The charged capillary model with a varying diameter for porous membranes was then used to evaluate the permeability of the membrane. It has been shown both theoretically and experimentally that the permeability of a grafted membrane depends on the pH of the solution.

HQNO-sensitive NADH:Quinone Oxidoreductase of Bacillus cereus KCTC 3674

  • The enzymatic properties of NADH:quinone oxidoreductase were examined in Triton X-100 extracts of Bacillus cereus membranes by using the artificial electron acceptors ubiquinone-1 and menadione. Membranes were prepared from B. cereus KCTC 3674 grown aerobically on a complex medium and oxidized with NADH exclusively, whereas deamino-NADH was determined to be poorly oxidized. The NADH oxidase activity was lost completely by solubilization of the membranes with Triton X-100. However, by using the artificial electron acceptors ubiquinone-1 and menadione, NADH oxidation could be observed. The activities of NADH:ubiquinone-1 and NADH:menadione oxidoreductase were enhanced approximately 8-fold and 4-fold, respectively, from the Triton X-100 extracted membranes. The maximum activity of FAD-dependent NADH:ubiquinone-1 oxidoreductase was obtained at about pH 6.0 in the presence of 0.1M NaCl, while the maximum activity of FAD-dependent NADH:menadione oxidoreductase was obtained at about pH 8.0 in the presence of 0.1M NaCl. The activities of the NADH:ubiquinone-1 and NADH:menadione oxidoreductase were very resistant to such respiratory chain inhibitors as rotenone, capsaicin, and $AgNO_3$, whereas these activities were sensitive to 2-heptyl-4-hydroxyquinoline-N-oxide (HQNO). Based on these results, we suggest that the aerobic respiratory chain-linked NADH oxidase system of B. cereus KCTC 3674 possesses an HQNO-sensitive NADH:quinone oxidoreductase that lacks an energy coupling site containing FAD as a cofactor.

  • Poly(vinyl alcohol)(PVA) and hyaluronic acid(HA) hydrogel membranes were prepared with varying HA contents from 10 to 50 wt% of PVA. The water contents of the resulting PVA-HA hydrogel membranes in various pH conditions were measured. And the permeation coefficient of indomethacin was determined using several PVA-HA hydrogel membranes at various pH conditions and also 37$^{\circ}C$.

  • Alumina tubes suitable for the support of gas separation membranes have been prepared by the slipcasting technique. These supports have the average pore size of 0.1 ${\mu}{\textrm}{m}$ within the narrow distribution. The sol-gel dipcoating process of nanoparticulate sols is very sensitive to microstructure of the support, and the coating on the inside surface of the tube is found to be more successful than on the outside surface. Nanoparticulate silica sols (0.82 mol/ι) have been synthesized by an interfacial hydrolysis reaction between TEOS and high alkaline water. When coating an alumina tube with these sols, the minimum limits of the particle size and the aging time required for forming the coated gel layer at the given pH are provided. It is optimum to coat the support with less concentrated sols stabilized through aging for the appropriate time (more than 22 days) at the lower pH (pH 2.0) for producing a reproducible crack free thin film coating in composite membranes.

On-line monitoring of microorganism cultivation processes using optical sensing membranes for simultaneous detection of dissolved oxygen and pH (용존산소와 pH의 동시 검출용 광학 센서 막을 이용한 미생물 발효공정의 온라인 모니터링)

  • An optical sensing membrane has been fabricated to measure the concentration of dissolved oxygen(DO) and pH value simultaneously. It has employed HPTS as a pH sensitive dye and a ruthenium(II) complex as a DO sensitive dye. The sensing membrane has been applied to wells in a 24-well microtiter plate. Using the 24-well microtiter plate the concentrations of dissolved oxygen and pH values have been on-line monitored during the cultivations of E.coli DH5${\alpha}$, B.cereus 318 and P.pastoris X-33. On-line monitoring of DO and pH in microorganism cultivation processes showed good performance of the sensing membrane containing 5 mM HPTS and 2 or 5 mg/mL Rudpp.

Expression of Latent P-Type ATPases and Their Presumptive Roles in Cell Membrane of Helicobacter pylori

  • Cation motive ATPases on cell membranes of Helicobacter pylori were investigated using everted membrane vesicles. Latent ATPases could be ascertained from aggregated vesicle using N, N-dimethylformamide (DMF) and Triton X-100. By contrast, ultrasonication or chloroform treatments caused membranes to be disrupted, resulting in an alteration of sensitivities against azide or vanadate. Considerable amounts of vanadate-sensitive enzymes were identified from vesicle micelles, prepared by the dilution method. These were activated in the presence of either $Ni^{2+}\;or\;NH_4^+$. From studies employing H. pylori intact cell systems, we found that ATPase expression of this bacterium was markedly dependent upon air composition. It was interesting that cellular expression of $Ni^{2+}$- or $NH_4^{+}$-motive ATPases was significantly affected by extracellular pH, suggesting that these unique enzymes may physiologically be involved in cellular $Ni^2$ import and $NH_4^+$ export, respectively.

Preparation of graphene oxide incorporated polyamide thin-film composite membranes for PPCPs removal

  • Incorporating nano-materials in thin-film composite (TFC) membranes has been considered to be an approach to achieve higher membrane performance in various water treatment processes. This study investigated the rejection efficiency of three target compounds, i.e., reserpine, norfloxacin and tetracycline hydrochloride, by TFC membranes with different graphene oxide proportions. Graphene oxide (GO) was incorporated into the polyamide active layer of a TFC membrane via an interfacial polymerization (IP) reaction. The TFC membranes were characterized with FTIR, FE-SEM, AFM; in addition, the water contact angle measurements as well as the permeation and separation performance were evaluated. The prepared GO-TFC membranes exhibited a much higher flux ($3.11{\pm}0.04L/m2{\cdot}h{\cdot}bar$) than the pristine TFC membranes ($2.12{\pm}0.05L/m2{\cdot}h{\cdot}bar$) without sacrificing their foulant rejection abilities. At the same time, the GO-modified membrane appeared to be less sensitive to pH changes than the pure TFC membrane. A significant improvement in the anti-fouling property of the membrane was observed, which was ascribed to the favorable change in the membrane's hydrophilicity, surface morphology and surface charge through the addition of an appropriate amount of GO. This study predominantly improved the understanding of the different PA/GO membranes and outlined improved industrial applications of such membranes in the future.

  • 일반적으로 고분자전해질은 고유의 독특한 특성으로 고분자 분리막, 이온교환수지, 의료용고분자등에 널리 응용이 되고 있다. 이러한 고분자전해질은 음이온성과 양이온성이 있으며, 그 이온적성질으로 인하여 반대의 전하를 띄는 고분자전해질들끼리 결합하는 성질을 가지고 있다. 이러한 고분자전해질 착체는 고분자전해질과는 또 다른 고유한 특성을 나타내고 있으며, 특히 고분자분리막과 의료용재료 분야에 응용가능성이 큰 물질이다. 특히 양이온성과 음이온성을 동시에 보유하고 있는 구조적 특성에 의하여 산성과 염기성 모두에서 감응성을 나타낼 수 있다. 일반적으로 고분자 겔의 경우 산성 또는 염기성 영역에서 해리되는 특성에 의하여 pH 감응성을 나타내지만 한쪽 영역에서만 특성을 나타내고 있다. 본 실험에서는 키토산과 폴리아크릴산으로 제조된 고분자전해질착체를 이용하여 pH변화에 따른 함수거동을 관찰함으로써 양쪽의 pH 감응성을 고찰해보고자 하였다.

  • pH-ISFETs, the semiconductor pH sensors, were combined with immobilized enzyme membranes to prepare FET type urea and glucose sensors and its operational characteristics were investigated. Photolithography techniques were applied to immobilize enzymes on the $H^{+}$ sensing membrane of the pH-ISFET with photo-sensitive polymers, PVA-SbQ. Fabricated urea and glucose sensors could determine $0.5{\sim}50{\;}mg/dl$ urea concentrations and $10{\sim}1000{\;}mg/dl$ glucose concentrations, respectively.

  • ISFETS for physiological applications have been developed using the techniques for integrated circuit fabrication. The silicon nitride layer was used as a H+ sensing membrane. However, K+, Na+ and Ca++ sensing ISFETS were fabricated by forming tach specification sensing membranes over the silicon nitride gate insulator. The sensitivities of the fabricated devices were very good. The typical values of measured sentivities were iEmV/pH, 42mv1, pH,5 gmV/pNa and 28mv1p0a. However, the selectivity and stability should be somewhat improved for practical physiological uses with good reliability. The response times were, less than one second, short enough for the practical uses in physiological applications.
