Simulation and Experimental Study on the Impact of Light Railway Train Bridge Due to Concrete Rail Prominence (주행면 단차에 의한 경량전철 교량의 충격 시뮬레이션 및 실험)

  • This study pointed on the dynamic impact of AGT (Automated Guide-way Transit) bridge, due to concrete rail prominence. An experiment was done with 30 m P.S.C. bridge in AGT test line in Kyungsan. An artificial prominence with 10 mm hight, was installed at the mid span of concrete rail. And computer simulation was executed for the artificial prominence. As an experiment result, in the case of with prominence, bridge acceleration responses are increased 50% at the speed range of 20 km/h-60 km/h, and bridge displacement responses increased slightly. With these results, the prominence of concrete rail can be induce excess impact and vibration. And the computer program simulated much the same as experiments. So this program can be used for AGT bridge design and formulate the standard of concrete rail management.

Evaluation of the Feasibility of Oyster-Shell and Eggshell Wastes for Stabilization of Arsenic-Contaminated Soil (농축수산 폐기물(굴껍질 및 달걀껍질)을 이용한 비소 오염토양의 안정화 효율 평가)

  • The objective of this research was to evaluate the feasibility of using oyster-shell and eggshell wastes for the stabilization of arsenic-contaminated soil. Artificial As(V) contaminated soil was mixed with 0~5% oyster-shell and eggshell wastes and each sample was incubated for 30 days in a controlled environment. The efficiency of each treatment was evaluated using various single extractants (1 N HCl, 0.1 N NaOH and 0.5 N $H_2SO_4$). The concentration of As(V) was reduced by 10% upon a 5% oyster-shell or eggshell waste treatments based on the Korea Standard Test method (1 N HCl extraction). Analogous trends were observed in the 0.1 N NaOH or 0.5 N $H_2SO_4$ extractions. In addition, the oyster-shell and eggshell waste treatments increased the pH of each soil from 6.54 (Control) to 7.62~7.94. The exchangeable Ca in each soil also sharply increased from 6.87 cmol(+)/kg (Control) to 12.77~20.18 cmol(+)/kg. Further research is needed to increase the effectiveness of the oyster-shell and eggshell waste for the stabilization of As(V) in the contaminated soil.

Isolation and Characterization of Lactic acid bacteria Leuconostoc mesenteroides DB3 from Camellia japonica Flower (백꽃으로부터 분리한 Leuconostoc mesenteroides DB3의 특성)

  • Lactic acid bacteria (LAB) are widespread in a variety of environments including fermented dairy products, gastroinstetinal and urogenital tracts of human and animals, plant, soil and water. Leuconostoc mesenteroides DB3 was detected by the strongest antibacterial activities among 24 Leuconostoc strains isolated from Camellia japonica flowers. Acid tolerance of L. mesenteroides DB3 existed up to pH 2.5, but the resistance did not show at pH 2.0, which relatively excellent acid resistance existed. Bile acid tolerance was very stable within the test range to 1.2%. L. mesenteroides DB3 exhibited the optimal growth at 30℃, and showed a slight slow growth when compared with L. mesenteroides KCTC3505, which reached a stationary phase at 18 hr. The pH was changed along with the growth curve, but was maintained above pH 3.98. L. mesenteroides DB3 had higher initial antibacterial activities when compared to L. mesenteroides KCTC3505, but it showed similar activities with the standard strain after the latter part of the logarithmic growth phase. Although lactic acid production in L. mesenteroides DB3 was induced by lower amount in the initial part to the standard strain, it was exhibited by similar amounts after the late logarithmic growth phase. Muicin adhesion of L. mesenteroides DB-3 maintained superior to L. mesenteroides KCTC3505. Both strains showed excellent emulsification ability for kerosene. In summary, we evaluate that L. mesenteroides DB-3 has a high potential for application as probiotics owing to its excellent antibacterial activity, acid resistance, bile acid resistance, and muicin adhesion.

Design and Synthesis of p-hydroxybenzohydrazide Derivatives for their Antimycobacterial Activity

  • The main mycobacterial infection in human is tuberculosis caused by Mycobacterium tuberculosis. Tuberculosis is the leading infectious cause of death in the world. Therefore there is continuing and compelling need for new and improved treatment for tuberculosis. The entire logic towards design of new compounds containing 4-hydroxy-N'-(1,3-thiazoldin- 2-yldene)benzohydrazide moiety is basically for superior antimycobacterial activity. The recent advances in QSAR and computer science have provided a systematic approach to design a structure of any compound and further, the biological activity of the compound can be predicted before synthesis. The 3D-QSAR studies for the set of 4-hydroxy-N'-(1,3-thiazoldin- 2-yldene)benzohydrazide and their derivatives were carried out by using V-life MDS (3.50). The various statistical methods such as Multiple Linear Regression (MLR), Partial Least Square Regression (PLSR), Principle Component Regression(PCR) and K nearest neighbour (kNN) were used. The kNN showed good results having cross validated $r^2$ 0.9319, $r^2$ for external test set 0.8561 and standard error of estimate 0.2195. The docking studies were carried out by using Schrodinger GLIDE module which resulted in good docking score in comparison with the standard isoniazid. The designed compounds were further subjected for synthesis and biological evaluation. Antitubercular evaluation of these compounds showed that (4.a), (4.d) and (4.g) found as potent inhibitor of H37RV.

Japanese Medaka, Oryzias latipes as a Test Animal for Marine Ecotoxicological Evaluation (해양생태독성평가를 위한 표준시험생물로서의 송사리(Oryzias latipes)에 관한 연구)

  • Japanese medaka, Oryzias latipes is widely distributed in the North East Asia including Korea, Japan and east China, and commonly used for freshwater toxicity tests and cytotoxicological studies worldwide. In this study, a series of experiments were conducted to identify the potential of the fish as a standard test species for saltwater toxicity evaluation such as marine receiving waters, ocean-dumped materials and sediment pore waters etc. Hatching, growth and mortality rates of the fish were estimated with the wide ranges of salinity from freshwater to seawater (35 psu). Direct exposure of the fertilized eggs in freshwater to the wide ranges of salinity (from 0 to 35 psu) without pre- acclimation to the saltwater revealed no significant differences in hatching rates by salinities (p =0.24). On the other hand, medaka larvae hatched in freshwater and exposed to saltwater directly showed high mortality at > 25 psu treatment groups (p < 0.0001). However, there was no significant difference in mortality of medaka larvae hatched in 13.8 and 14.2 psu at the wide ranges of salinities ($0\~35$ psu). Growth rates of medaka larvae hatched in the above two salinities showed no differences in body length either from 0 to 35 psu treatment groups (p =0.64 for 13.8 psu group and p=0.32 for 14.2 psu group). The number of gill chloride cell in medaka larvae sharply increased when the larvae were exposed to high salinity. Reference tests with zinc chloride revealed 96h $LC_{50}=8.84(7.19\~10.87)mg\;L^{-1}$ using 7~10 day old medaka larvae. These were comparable or better sensitivity in comparison with the other standard test species such as North American sheepshead minnow Cyprinodon variegatus. Based on the results of these experiments, hatching rates and larvalmortality of medaka must be good toxicity parameters for seawater bioassay and the species seems to be a good standard species for both the freshwater and seawater toxicity test.

Stabilization of Heavy Metals-contaminated Soils Around the Abandoned Mine area Using Phosphate (인산염을 이용한 휴.폐광산 주변 중금속 오염토양의 안정화처리에 관한 연구)

  • The objective of this study was to evaluate the efficiency of $(NH_4)_2HPO_4$, $Na_2HPO_4{\cdot}12H_2O$, $CaHPO_4{\cdot}2H_2O$, $Ca(H_2PO_4)_2{\cdot}H_2O$ and $H_3PO_4$ for the stabilization of soils contaminated with multi-metals containing Pb, Cd and As. The application rate of stabilizers to soils was determined based on $PO_4/Pb_{total}$ molar ratio of 0.5, 1, 2, 4. The results of Korea Standard Test and TCLP (EPA Method 1311) showed the reduction of metal leachabilities below the regulatory limits for Pb and Cd when $H_3PO_4$ and $Ca(H_2PO_4)_2{\cdot}H_2O$ were applied. However, stabilization efficiency for Cd was low and in case of As leaching concentration increased rather. It is considered that $PO_4$ reacted effectively $Pb^{2+}$ due to leaching Pb under low pH condition created by adding $H_3PO_4$. Accordingly Pb was stabilized by dissolution and precipitation of hydroxypyromorphite. From the change of metals fraction using sequential extraction procedure when $H_3PO_4$ applied as a stabilizer, we confirmed that residual fraction increased more than 60% and this result was accorded with XRD analysis that detected only hydroxypyromorphite peak in $H_3PO_4$.

Validations of Analysis Methods for Decursin and Decursinol Angelate of Angelicae gigantis Radix by Reversed-Phase Liquid Chromatography

  • A reversed-phase liquid chromatographic method for decursin and decursinol angelate of Angelicae gigantis Radix, an important crude drug in Korean traditional medicine, was developed and validated. Decursin and decursinol angelate, the structure isomer (pyranocoumarin) each other, are the main organic constituents in Angelicae gigantis Radix. This method was developed using a RP-18 column, UV detector at 280 nm and 50% acetonitrile solution containing 0.01 M sodium dodecyl sulfate and 25 mM sodium dihydrogen phosphate (pH 5.0) as the mobile phase. Various validation parameters were included and evaluated satisfactorily. Linearity was established in range 2-75 mg/ml of decursin and decursinol angelate (correlation coefficient = 0.9997 and 0.9995, respectively). This analytical method showed good accuracy (98.1% and 99.5%, respectively). Precision (repeatability) revealed a relative standard deviation value of 1.71% (decursin) and 3.19% (decursinol angelate). For intermediate precision measure the considered variables were equipment and days. A robustness test showing the influence of deferent counter-ion concentration in mobile phase was also performed.

Study on the Adhesive Properties of Polyesters Reinforcing Materials

  • Polyester cord yarns have been treated in an atmospheric-pressure nitrogen plasma reactor in order to enhance their adhesion to rubber. A thin layer or the plasma was generated in the close vicinity of the yam surface using various types or surface discharge. To assess the effect of the plasma treatment on fiber surface properties, the cord thread/rubber matrix adhesion values measured using the untreated and threads cord threads were compared. The static and dynamic adhesion of the cord thread to rubber was characterized by using the standard Henley test. The dynamic adhesion values for the reference and plasma treated fiber were $7,3{\pm}1,2\;N$ and $83,5{\pm}3,5\;N$. The surface properties were investigated by scanning electron microscopy, infrared spectroscopy and electron spin resonance spectroscopy. It is concluded that both polar group interactions and increased surface area of the fibers are responsible for the improved adhesive strength.

  • Recently, the antiwashout underwater concrete with an antiwashout admixtures has been increasingly used for underwater structures. However, the credibility of antiwashout underwater concrete was brought up as problems because it seldom was applied to fields. In this study, experiments were made on the basic properties of antiwashout underwater concrete replaced with fly ash up to 30% to improve its properties. Resultant to the test, we got the results as follow; funnel flow time was decreased, the slump flow was increased and the elevation of head was decreased rapidly whereas the amount of suspended solids became high, pH became low. In view of 70%, the standard ratio of compressive strengths between cast in water and in air, it was obtained the result that the ratio was over 90% at 28days.

Assay of Midazolam in Human Plasma by Gas-Liquid Chromatography with Nitrogen-Phosphorus Detection

  • A sensitive and specific method is described for the determination of midazolam in human plasma. The drug was extracted from 1 mL of carbonate buffered plasma (pH 9.6) with 8 mL of diethyl ether. Famprofazone was used as internal standard. The organic phase was evaporated to dryness. The residue was dissolved in methanol for the direct analysis by gas chromatograph-nitrogen phosphorus detector system. In the concentration range of 1-5000 ng/mL, the calibration curve was linear. The coefficients of variation from the precision test were <6% at the range of the concentration of 0.10-2.00 μg/mL and the detection limit for midazolam in 1 mL of plasma was o.5 ng. This assay is more sensitive, selective, simple and rapid than earlier methods. Plasma midazolam concentrations were determined by this method after administration of midazolam.
