Isolation of Cellulolytic Microorganisms and their Physiological Characteristics (섬유소분해균의 분리 및 그의 생리학적 특성)

  • Celluloytic microorgnasims were isolated form the various sources and four of them were identified as Trichoderma roningi, Aspergillus niger, Penicillium chrysogenum, and Streptomyces sp. The induction of extracellular5 cellulase of these species in the liquid culture media containing carboxymethylcellulose (CMC) or Avicel as inducer showed that CMC was a better effective inducer for the production of CMCase(Cx cellulase component) as well as Avicelase(C$_{1}$ cellulase component) than Avicel. It is believed that certain hydrolysis products of cellulose(CMC) could serve as an inducer for an enzyme synthesis. In T. roningi, Asp. niger, and Strptomyces sp., the optimum temperature of CNCase on CMC-culture medium was 50.deg. but temperature around 40.deg.C was found to be optimum for the activities of CMCase prepared from P.ehrysogenum. The optimum temperature for Avicelase activitiles on Avicel-culture media of T. roningi and P. chrysogenum was $40{\circ}C$ whereas temperature $50{\circ}C$ was found to be optimum for Avicelase from A.niger and Streptomyces sp. The optimal activities of these CNCase and Avicelase prepared from. T. ronigi, Pen.chrysogenum and Streptomyces sp. were found similarly to be at pH's around 5.4 and 6.0 while pH 4.8 was optimum for the activities of CMCase and Avicelase from A.niger, indicating that A.niger in acidic media would yield an enzyme of high activity.

Degradation and Ecotoxicity Reduction of Reactive Dye by Using Advanced Oxidation Process (고도산화공정을 이용한 반응성 염료의 제거 및 생태독성 저감)

  • In this study, the deriving optimum conditions for decolorization of Acid Orange II solution was carried using $TiO_2$ advanced oxidation process. After that, on base of the deriving results, the range of dye concentration was estimated. In addition, acute toxicity test was also carried to assess toxicity unit according to decolorization and TOC removal. In case of the blockage of light, 20 mg/L of dye solution, and 0.5 g $TiO_2$, the effect of decolorization at pH 3 was larger than at pH 6 and 10, so it was shown that decolorization is dependent on pH. The use of 5 g $TiO_2$ showed best performance of decolorization, but that of 3 g $TiO_2$ was chosen to optimum condition in considering of economical aspects. Four light sources, sun, fluorescent lamp, BLB lamp, and UV-B lamp, were used and decolorization was 99.4% and 100% at 50 mg/L, 98.6% and 99.7% at 100 mg/L for sun and UV-B lamp, respectively. In spite of the optimum condition of decolorization at pH 3, the evaluation of acute toxicity test showed highly toxic. In conclusion, although the optimum treatment of dye solution is performed, water ecology can be polluted in discharging it into water system. Therefore, it is needed to study of water ecological system with dye water treatment, and it takes all the circumstances into consideration.

Enzyme Activity and Beating Properties for Preparation of MicroFibrillated Cellulose(MFC) (MicroFibrillated Cellulose(MFC) 제조를 위한 전처리 효소의 활성 및 고해 특성)

  • In this study, we evaluated optimum condition of enzyme with pH and temperature for preparation of microfibillated cellulose(MFC). Well-known endo-glucanase, three enzymes were used and CMC was used for substrate. Enzyme activity was evaluated using DNS method and absorbance with UV/VIS spectrophotometer. The enzyme shown the greatest activity was reacted with pulps at optimum condition for 1 hour and treated pulps beated until 100 mL CSF. Enzyme B and Enzyme L was the higher enzyme activity below 0.1% concentration and Enzyme N was the lowest enzyme activity. At various pH and temperature conditions, enzyme activity of Enzyme B was higher than the others at the same concentration. Especially enzyme activity at $50^{\circ}C$ of Enzyme B was almost not changed over pH 6.0. Optimum condition of three enzyme was pH 6 or pH 7 and $50^{\circ}C$ or $60^{\circ}C$. Also beating efficiency of enzyme treated pulps with Enzyme B is 55.6%.

  • To improve extraction of insoluble proteins and functional properties of abolished proteins by protease. It was found that the optimum pH, optimum temperature, optimum treatment time and optimum unit of enzyme far extraction of protein were pH 9.0, $60^{\circ}C$, 8 hrs, 40 units. The foaming capacity and foaming satbility of sesame meal protein after treatment of enzyme were especially higher than control. The emulsion capacity and emulsion satbility of sesame meal protein were higher than control. Coil absorption as well as water absorption capacities of sesame meal protein were higher than control.

The Condition of Optimum Coagulation for Recycling Water from CMP Slurry

  • Water usage in the semiconductor industries is dramatically increased by not only using bigger wafer from 8 inches to 12 inches but also by adapting new process such as Chemical Mechanical Planarization (CMP) process invented by IBM in late '80. However, The document published by International Semiconductor Association suggests the decreasing ultra pure water (UPW) use from 22 gallon/in$^2$in 1997 to 5 gallon/in$^2$ in 2012. The criteria will possibly used as exporting obstacle in the future. Generally, Solid content of CMP slurry is about 15wt%. The slurry is diluted with UPW before fed to a CMP process. When the slurry is discharged from the process as waste, it contains 0.1~0.6wt% of solid content and 9~10 at pH. The CMP waste slurry is discharged to stream with minimum treatment. In this study, to find optimum condition of coagulation for water recovery from the waste CMP slurry various condition of coagulation were examined. After coagulation far 0.1 wt% solid content of waste CMP slurry, the sludge volume was 10~15% after 30 min of sedimentation time. For the 0.5 wt%, sludge volume was 50~55% after one hour of sedimentation time. For more than 80% of water recycling, the solid content should be in the range of 0.1 to 0.2wr%. Based on the result of the turbidity removal, the Zeta Potential and the analysis of heavy metals, the optimum condition for 0.1 wr% of waste CMP slurry was with 20 mg/L of PACI at 4 to 5 of pH. The result showed that the optimum conditions fer the 0.1 wt% waste CMP slurry were 100mg/L of Alum at 4~5 of pH, 100 mg/L of MgCI$_2$at pH 10 to 11 and 100 mg/L of Ca(OH)$_2$at pH 9 to 11, respectively.

  • In order to investigate the action of thermophilic fungi on the xylan and its related substances, xylanase and laminaranase were examined using Myriococcum albomyces. In the extract from bran culture of Myriococcum albomyces, xylanase and laminaranase activity were recognized. 1) The optimum pH for xylanase and laminaranase activity were found to be pH 5.0 and pH 6.0. 2) The optimum temperature for xylanase and laminaranase activity were found to be $55^{\cicr}C$. 3) Thermal stability of xylanase for 55 minutes at $65^{\cicr}C$ and laminaranase for 60 minutes at $65^{\cicr}C$ did not influence their stability.

  • Cylindvocarpon destmtalns isolate CY-92-01, pathogen of root-rot of Panax ginseng showed t the maximum mycelial growth on the Czapek solution agar among the thirteen kinds of media. Five isolates (Isolate CY-92-01, CY-92-03, CY-92-07, CY-94-01, CY-94-02) of C. destructan from various growth stages of p. ginseng recovered from several geographical sites also showed maximum growth in the Czapek-Dox broth compared with potato dextrose broth and V-8 juice broth. Rapid growth rate was maintained until 12 days after inoculation on the Czapek-Dox broth and mycelial weight was somewhat constant until 20 days. After 30 days of incubation, the mycelial weight began to decrease. The fungal growth occurred from 5$^{\circ}C$ to $25^{\circ}C$ and optimum temperature for growth was 2$0^{\circ}C$. Mycelial weight orderly decreased at 15, 25, 10, and 5$^{\circ}C$. Quantitative measurement was impossible at 5$^{\circ}C$. No fungal growth was occurred at the temperature higher than 3$0^{\circ}C$. Growth was observed at all tested pH ranges from 2.8 to 8.0. Optimum pH for growth was 4.0~5.0 followed by pH 3.3~3.5 and 5.4~6.0. The least growth occurred at pH 2.8.

Emulsification Activity of Acinetobacter sp. 2-3A Isolated from Petroleum Oil-Contaminated Soil (유류오염 토양에서 분리한 Acinetobacter sp. 2-3A의 유화활성)

  • Fifty hydrocarbon-metabolizing microorganisms were isolated from soil samples polluted by the petroleum oils in Gamman-dong, Busan. Among them, strain 2-3A, showing strong emulsification activity, was selected by oil film-collapsing method. This bacterium was identified as Acinetobacter sp. and designated as Acinetobacter sp. 2-3A. The optimum temperature and pH on the growth of Acinetobacter sp. 2-3A were $25^{\circ}C$ and pH 7.0, respectively. The carbon and nitrogen sources for the most effective emulsification activity were 3.0% olive oil and 0.5% peptone, respectively. The 0.15% potassium phosphate was the most effective emulsification activity as a phosphate source. The optimum emulsification activity condition was $20^{\circ}C$, pH 7.0, and 2.0% NaCl. The optimum time for the best production of biosurfactant was 27 hrs. The emulsification stability was maintained at the temperature range from $4^{\circ}C$ to $100^{\circ}C$, pH range from 6.0 to 10.0, and NaCl range from 0% to 10%. For the oil resolvability of the biosurfactant, the residual oils were investigated by gas chromatography. As a result, it was verified that the biosurfactant decreased and decomposed crude oils from $_nC_{10}$ to $_nC_{32}$.

Enzymatic Characterization of a Recombinant Levansucrase from Rahnella aquatilis ATCC 15552

  • A 1.25 kb DNA fragment including the lscR gene, which encodes a levansucrase of Rahnella aquatilis ATCC 15552, was subcloned into a high-expression vector, pET-29b, and the recombinant enzyme was overexpressed in Escherichia coli. Most of the levansucrase activity was detected in the cytoplasmic fraction after induction with isopropyl ${\beta}-D-thiogalactoside$. The recombinant enzyme with a tag of six histidine residues at the C-terminus was purified 146-fold by affinity and gel-filtration chromatographies. The molecular mass of the purified LscR was approx. 49 kDa as determined by SDS-PAGE. The optimum pH and temperature of this enzyme for levan formation was pH 6.0 and $30^{\circ}C$, respectively. The optimum substrate concentration for levan formation was 300 mM sucrose. Levan formation was increased by the increase of the enzyme concentrations. Maxium yield of levan formation at optimum substrate concentration, pH, and temperature after 24 h of reaction was approximately 80%.

Extraction of proteins from soymilk residue using the enzymes from Aspergillus of oryzae (코지균 효소를 이용한 두유박의 단백질 용출)

  • To extract insoluble proteins of soymilk residue by microorganism, the soymilk residue was treated with crude enzyme solution from Aspergillus oryzae. Optimum conditions of pH, temperature and digestion time were determined, and amino acid composition of the extract was analyzed. The optimum pH for the extraction was 7.5, and the maximum extraction was obtained at $50^{\circ}C$. Under optimum conditions, the extractability with Koji reached to 70% in 12 hrs. The content of essential amino acids of extract was generally high and the composition of essential amino acid was good.
