Molecular Weight Distribution of Pullulan and Degrading Enzyme Activity of Aureobasidium pullulans

  • The effects of DO and pH on the mass production of pullulan with high-molecular weight from A. pullulans ATCC 42023 were evaluated. The maximum pullulan production yield (51%) was obtained at pH non control (initial pH 6.5) and DO control (above 50%) condition. The pullulan degrading enzyme was activated when the pH of broth reached lower than 5.0 and portion of low molecular weight pullulan was increased. The formation of a black pigment was observed at the initial stationary phase, 40hr of fermentation. Therefore, the fermentation should be carried out in pH non control and DO control condition and harvested before reaching stationary phase around 40h for the production of high molecular weight pullulan.

  • Solvent extraction experiments of Nd from chloride solution were studied with PC88A. Solvent extraction reaction of Nd with PC88A and the equilibrium constant were evaluated from the extraction experimental data and extraction conditions. Nd$aq^{3+}$ + 1.5 $H_2$$A_2$,org = $NdA_3$,org + 3H/sun $aq^{+}$ , K=0.25 The predicted distribution coefficients of Nd agreed well with the experimental results. The effect of saponification of PC88A on the extraction of Nd and on the change of equilibrium pH was investigated. Saponified PC88A present as a monomer in the organic reaction and enhanced the distribution coefficient of Nd. The initial extraction conditions had a great effect on the equilibrium pH.

A Study on the Distribution Characteristics of Buxus koreana in Kwanaksan Ecological and Scenery Conservation Area (관악산 생태경관보전지역 내 회양목 분포 특성 연구)

  • The purpose of this study was to find out the vegetation community structure and the distribution characteristics of Buxus koreana through analysis of the topography and soil characteristics in Kwanaksan Ecological and Scenery Conservation Area. To do this twenty two plots as $100m^2$ each were set up and investigated. As a result, the vegetation communities were classified in four groups; Pinus rigida community (I), P. rigida - B. koreana community(II), Quercus mongolica community(III), Quercus acutissima - Quercus serrata community(IV). The distribution characteristics in topography appeared that B. koreana distributed more in valley where soil water and nutrition conditions are good. The drainage condition also influenced the distribution, because the ratio of clay was low in the result of soil texture analysis. B. koreana was distributed in high-pitched slope and the high rock ratio area. According to the soil analysis, the mean soil pH of B. koreana distributed groups(pH 4.79) was higher than that of non-B. koreana distributed groups(pH 4.41). It appeared that B. koreana had tolerance to the barren soil, so the value of exchangeable cations such as $Ca^{2+}$ and $Mg^{2+}$were showed lower than that of the average of Korean forest soil.

  • Let p be an odd prime number. We introduce simple and useful decoding algorithm for orthogonal Latin square codes of order p. Let H be the parity check matrix of orthogonal Latin square code. For any x ${\in}$ GF(p)$^{n}$, we call xH$^{T}$ the syndrome of x. This method is based on the syndrome decoding for linear codes. In L$_{p}$, we need to find the first and the second coordinates of codeword in order to correct the errored received vector.

  • The chlorination of municipal drinking water supplies leads to the formation of so-called disinfection by-products(DBPs), many of which have been reported to cause harmful health effects based on animal studies. This study was conducted: 1) to observe seasonal changes in the major DBPs at four sampling sites on a drinking water distribution system located in Chunchon, Kangwon Do; and 2) to examine the effects of major water quality parameters on the formation of DBPs. During the field sampling, the water temperature, pH, and total and free chlorine residuals were all measured. The water samples were then analyzed for total organic carbon(TOC) and eight disinfection by-products in the laboratory. Chloroform, dichloroacetic acid, and trichloroacetic acid were the major constituents of the measured DBPs. The concentrations of the total DBPs were highest in fall, particularly in October, and lowest in summer. The concentrations of the total DBPs increased with increasing TOC concentrations. Multiple regression analyses showed that the concentrations of chloroform, bromodichloromethane, and chloral hydrate were linearly correlated with the pH. Other water parameters were not included in the regression equations. Accordingly, these results suggest that TOC and pH are both important factors in the formation of DBPs.

  • The stress relaxation of poly(methyl acrylate)-poly(acrylonitrile) copolymer samples was carried out in air, distilled water, pH 3, 7 and 11 solutions at various temperatures using a tensile tester equipped with a solvent chamber. The relaxation spectra of poly(methyl acrylate)-poly(acrylonitrile) copolymers were obtained by applying the experimental stress relaxation curves to the equation of relaxation spectrum derived from the Ree-Eyring and Maxwell model. The determination of relaxation spectra was performed from computer calculation using a Laplace transform method. It was observed that the relaxation spectra of these samples are directly related to the distribution of molecular weights and self-diffusions of flow segments.

Removal of Natural Organic Matter by Mixing Coagulants in Coagulation Process (응집공정에서 혼합응집제 주입에 의한 자연유기물질의 제거)

  • Natural organic matters(NOMs) are found everywhere such as soil, surface and ground waters and consist of both humic and nonhumic components, and their heterogeneith makes each source unique. This study was carried to evaluate the removal characteristics of NOMs by mixing coagulants and the variation of apparent molecular weight distribution(AMWD) in coagulation process. Ratio of optimum coagulants dosage for removal of DOC and turbidity by mixing coagulants was 1.83 mM F $e^{3+}$/mM $Al^{3+}$. DOC removal increased at lower pH. The pH6 control focused on the removal of organic matters could reduce the amount of coagulant consumption by 2 to 3 times based on the pH8.5 of natural water. The dissolved organic matters in the natural water from the mid-stream of Han River were composed of the low molecular weight(LMW,<1 K) of 59.7%, and the medium and high molecular weight(M.HMW, 1~30 K) of 40.3%, respectively. At pH6, the DOC removal efficiencies of LMW(<1 K) and M.HMW(1~30 K) in coagulation process were 27~35%, 62~72%, respectively. The fraction smaller than 1 K was not eliminated to a noticeable degree, while the fraction of 1~30 K was relatively well removed. In conclustion, mixing coagulants were fairly effective in the removal of natural organic matter.r.

Characteristics of Apparent Molecular Weight Distribution and Removal of DOC by Coagulation and Sedimentation Process with Polyaluminum Chloride in Nakdong River Water (응집·침전 공정에서 PAC를 이용한 낙동강 원수의 DOC 제거 및 분자량 분포 특성)

  • The objective of present work is to evaluate the optimum coagulation conditions in order to decrease dissolved organic carbon(DOC) and turbidity at different polyaluminum chloride dosage and pH from Nakdong River water. This studies were carried out to examine distribution on apparent molecular weight(AMW) of DOC in the Nakdong River water and its coagulation-sedimentation water. On the basis of jar tests, at the optimum coagulation pH in order to decrease DOC and turbidity were pH 5.0~6.0 and optimum dosage of polyaluminum chloride were 10~15mg $Al_2O_3/L$. The removal percentage of DOC and UV-254 absorbance were 35~40%, 45~60%, respectively. In pilot plant, at the optimum coagulation pH in order to decrease DOC and turbidity were 5.0-6.5, and the removal percentage of DOC were 30~45%. Distributions of AMW in the Nakdong River, less than 6,800dalton were 60.7% 6,800~11,000dalton were 32.8%, more than 11,000dalton were 6.4%. When the polyaluminum chloride dosage was 12~20mg/L, the removal percentages of each AMW for AMW of Nakdong River water, less than 6,800dalton were 25~28%, 6,800~11,000dalton were 65~68% more than 11,000dalton were 10~60%.

Characteristics of a Titanium-oxide Layer Prepared by Plasma Electrolytic Oxidation for Hydrogen-ion Sensing

  • The characteristics of a titanium oxide layer prepared using a plasma electrolytic oxidation (PEO) process were investigated, using an extended gate ion sensitive field effect transistor (EG-ISFET) to confirm the layer's capability to react with hydrogen ions. The surface morphology and element distribution of the PEO-processed titanium oxide were observed and analyzed using field-emission scanning-electron microscopy (FE-SEM) and energy-distribution spectroscopy (EDS). The titanium oxide prepared by the PEO process was utilized as a hydrogen-ion sensing membrane and an extended gate insulator. A commercially available n-channel enhancement MOS-FET (metal-oxide-semiconductor FET) played a role as a transducer. The responses of the PEO-processed titanium oxide to different pH solutions were analyzed. The output drain current was linearly related to the pH solutions in the range of pH 4 to pH 12. It was confirmed that the titanium-oxide layer prepared by the PEO process could feasibly be used as a hydrogen-ion-sensing membrane for EGFET measurements.

The Effects of pH and Alkalinity Adjustment on Internal Corrosion Control and Water Quality in Drinking Water Pipelines (정수의 pH 및 알칼리도 동시 조절이 상수도관의 내부부식 제어 및 수질에 미치는 영향)

  • This research was carried out to evaluate effect of metallic release and change of water corrosive indices by the pH and alkalinity adjustment using the SDLS (Simulated Distribution Loop System) which consist of six types of pipe loop with DCIP, PVC, PE, STS304, CP, GSP, respectively, and its effects on water quality changes which were microbes quality(SPC), residual chlorine. THMs and other parameters. And it was to propose optimal criteria of water quality control for the field application. According to the results, water control system by pH and alkalinity adjustment showed the changing of corrosive water and reducing of metallic release rate and it was not affects of THMs formation, microbes regrowth and variation of other parameters. Water quality stability and corrosion control were due to calcium carbonate precipitation film formation of pipe inner by water quality control. Therefore, corrosive water control system by pH and alkalinity adjustment can be attributed to effective water quality management in water distribution system according to water quality stability of pH and TIC(Total Inorganic Carbonate concentration) that affect the precipitation and dissolution of solids.
