본 논문에서는 열분해 및 촉매반응의 의한 Hydrazine의 Ammonia 전환율을 연구하였다. 원자력발전소 2차 계통은 물/증기 순환계통으로 기기 및 배관의 부식을 억제하고, 증기발생기(Steam Generator, SG)의 부식생성물 유입을 최소하기 위해 전휘발성처리법(All Volatile Treatment, AVT)을 적용하여 계통수의 pH를 약염기성으로 유지하고 있다. 또한 Hydrazine을 이용하여 계통수의 용존산소제거 및 환원성 분위기를 유지하고 있다. 현재 사용되는 AVT는 대부분 단일 아민(Ammine)으로 계통 전 영역에서 pHt를 약염기성으로 유지하기 어렵다. 따라서 복합 아민을 이용하여 단일 아민의 상호단점을 보완한 수처리법을 적용해야한다. 하지만 복합 아민을 적용할 경우 추가 아민 주입설비, 설치부지, 시설유지보수 및 관리가 요구되므로 기존 주입약품을 이용하여 아민을 공급할 수 있는 연구가 필요하다. 따라서 본 연구에서는 Hydrazine의 열분해 및 촉매반응을 이용한 Ammonia 전환율을 조사하였다.
A bacteria strain producing extracellular $\beta$-mannanase was isolated from soil and was identified as Aeromonas sp. A genomic DNA library constructed from Aeromonas, sp that secrets a $\beta$-mannanase was screened for mannan hydrolytic acticity. Recombinant $\beta$-mannanase activity was detercted on the basis of the clear zones around Escherichia coli colonies grown on a LB medium supplemented locust bean gum, EcoRI restriction analysis of plasmid prepared from recombinant E. coli which showed a $\beta$-mannanase activity revealed 10 kb DNA insert, The optimum pH and temperature for the activity of reconmbinant $\beta$-mannanase were 6.0 and $50^{\circ}C$ respectively and were identical to those of the native enzyme.
An alkalophilic bacterium isolated from compost was selected, identified and tested for the production of glutamic acid from ammonium fumarate. The bacterium was closely related to Bacillus brevis. The conditions for glutamic acid production were pH 8.0, 2% fumaric acid, and 0.8% nutrient broth. The mechanism of glutamic acid formation in this strain was postulated as following scheme. (1) Ammonium fumarate longrightarrow Aspartic acid (2) Aspartic acid + $\alpha$-Ketoglutaric acid longrightarrow Glutamic acid + Oxaloacetic acid.
Nitrogen in the form of nitrate was electrochemically reduced with different cathode materials including Fe, Ni, Cu, and Zn. Zn cathode shows the greatest electrocatalytic activity on the transformation of nitrate ions into ammonia and the $NO_3^-$ removal efficiency has highest value at pH 8.5. Nitrogen in the form of nitrate was initially reduced into nitrite and sequentially, converted into nitrogen inside $NH_3$. Nitrogen in the form of ammonia was completely removed by the reaction with HOCl.

The cycloinulooligosaccharide fructanotransferase (CFTase) gene (cft) from Paenibacillus macerans was subcloned into an E. coli-yeast shuttle vector, pYES2.0, resulting in pYGECFTN. The plasmid pYGECFTN (8.6 kb) was introduced into Saccharomyces cerevisiae SEY2102 cells and then the transformants were selected on the synthetic defined media lacking uracil. The cft gene expression in yeast transformant was demonstrated by the analyses cyclofructan (CF) spots on thin-layer chromatogram. The recombinant CFTase was not secreted into the medium and localized in the periplasmic space. The production of CF was observed after 5 min of the enzymatic reaction with inulin. The optimun pH and temperature for CF production were found to be at pH 8.0 and $45^{\circ}C$, respectively. Enzyme activity was stably maintained up to $55^{\circ}C$. The CF was produced from all inulin sources and was most efficiently produced from dahlia tubers and Jerusalem artichokes.
The authentic hGH converted from met-hGH by immobilized ApM was purified by successive chromatographic processes based on the differences in isoelectric points, hydrophobicities and charges. The final recovery yield was about 14.1% and the specific activity of the purified hGH was 2.75IU per mg when assayed by enzyme immunoassay. The purified hGH was verified to be authentic hGH through the analysis of amino acid composition, amino-terminal amino acid sequence, carboxy-terminal amino acid and tryptic peptide map. The purity of purified hGH was higher than that of commercial hGH when assessed by SDS-PAGE, PAGE, IEF and HSGF. In weight-gain assay and tibia test with hypophysectomized rats, the hGH produced in this study showed the same growth effect as the commercial hGH.

Ginsenoside Compound K is a triterpene saponin found in the leafs, stems and roots of Panax ginseng. This study aimed to prepare a valuable ginsenoside Compound K using ginseng extracts with the enzyme(Plantase). Plantase showed very efficient activity to produce Compound K from ginseng extracts. Plantase exhibited the highest activity at pH 5 and 50 ℃, as a result of investigating the yield of Compound K by changing the temperature and pH, while fixing the enzyme concentration to 10% or 15% over 48 hours of reaction time. Under optimium conditions, Plantase produced and accumulated Compound K over 35 wt% of whole ginseng extracts. Antimicrobial activitiy of bioconvertied ginseng extracts showed selectivity against Cutibacterium acnes KCTC 3314. Minimal inhibitory concentration (MIC) of bioconverted ginseng extract (35% of Compound K enriched extract) against Cutibacterium acnes KCTC 3314 strain is 31.25ug/mL. These results suggest that the Compound K enriched extract is potential materials for cosmetic products and Plantase is a very useful enzyme for Compound K production.

Bioconversion and fermentation of onion extract by lactic acid bacteria were carried out to enhance the bioavailability of quercetin through the increase of quercetin recovery and aglycone formation. Lactobacillus casei, L. plantarum, and Kluyveromyces lactis were selected as the optimum strains for bioconversion. The environmental conditions to maximize the conversion ratio between glycoconjugate and quercetin aglycone have been evaluated. The concentrations of quercetin after fermentation of onion slurry by K. lactis and L. casei increased to 260% and 318%, respectively; however, the quercetin concentrations decreased after 48 hours of fermentation. Additionally, the quercetin hexose concentration increased to almost 141%. Controlling the initial pH of the onion juice increased the lactic acid production by L. casei and L. plantarum by more than two-fold. Meanwhile, the concentration of quercetin hexose decreased rapidly with the increased production of aglycones. The scale-up experiments showed the same fermentation efficiency; however, thermal sterilization reduced the quercetin glycone concentrations drastically.
AH alkalophilic Bacillus SP. AIk-7, isolated from soil, produced ethylene. The characteristics of this microorganism is the ability to grow well under the alkaline condition, at pH 10.3. This strain is similar to Bacillus alkalophilus in terms of morphological, physiological and biological characteristics. In observation of relationship of cell growth and ethylene production according to incubation times, the ethylene synthesis mostly occur from the late exponential phase to the death phase of growth. The purpose of this paper is to study the effects of various substrates on the biosynthesis of ethylene in the intact cell and the cell-free system by the Bacillus sp. AIk-7. In both intact cell and cell-free extract, optimum conditions for ethylene production was achieved at pH 10.3 and 3$0^{\circ}C$. Ethylene was effectively produced from L-Met and 1-aminocyclopropane-1-carboxylic acid (ACC). In this case, ACC as the substrate on ethylene production were two fold higher than L-met at each concentration of substrates. On the other hand, the cell-free ethylene-forming system was used as a tool for the elucidation of the biochemical reaction involved in the formation of ethylene by Bacillus sp. AIk-7. Ethylene production in the cell-free system required the presence of manganese and cobalt ion to be stimulated a little. The result obtained in this work suggests that L-met and ACC may be a precursor more directly related to bacterial ethylene production than any other substrates tested.
Oxidations of cyclopentene and of 1-pentene with air have been studied on a V/Mo/P/Al/Ti-mixed oxide catalyst in a fixed bed integral reactor. At high levels of conversion maleic anhydride was in each case produced as the major organic product, along with minor amounts of phthalic anhydride and, only starting from 1-pentene, also of citraconic anhydride. At lower levels of conversion a total of 30 organic products have been identified, some of which may be intermediates on the way from the substrates to the three anhydrides mentioned above. Based on the dependence of selectivities of the organic products on conversion, reaction schemes for the formation of maleic anhydride, phthalic anhydride and citraconic anhydride have been proposed. Oxidation at $310^{\circ}C$ led to increasing conversions and selectivities for maleic anhydride with decreasing space velocities. The highest selectivities for maleic anhydride were obtained at conversion of ca. 100%. Oxidation at a constant space velocity of $2{\cdot}10^4h^{-1}$ led to increasing conversions with increasing temperatures in the range of $300^{\circ}C{\sim}420^{\circ}C$, while the selectivity for maleic anhydride passed through a maximum value of ca. 39% at $370^{\circ}C$ in the oxidation of cyclopentene and a maximum value of ca. 30% at $400^{\circ}C$ in the oxidation of 1-pentene.
