  • pH- and thermo-sensitive hydrogels containing maleilated chitosan(MC) and N-isopropylacrylamide(NIPAAm) were prepared and characterized for their swelling behavior, biodegradability and drug release profiles. The hydrogels exhibited a typical pH-sensitivity due to the carboxylic acid groups of maleilated chitosan. The change of ratio of NIPAAm to MC in weight did not affect on either lower critical solution temperature(LCST) or EWC significantly. The pH sensitivity of the hydrogel, however, depended on the amounts of carboxylic acid groups of MC. MC was degradable up to 80% weight reduction in 2 hours. The in vitro drug relase profiles were established both in buffer solution pH 1.4 and pH 7.4. Only a negligible amount of indomethacin was released at pH 1.4 in 6 hours, while at pH 7.4 more than 90% of the total drug in the hydrogel was gradually released over ca. 5 hours.

Preparation and Swelling Behavior of Stimuli-responsive PHEMA Hybrid Gels (자극감응성 PHEMA 하이브리드 젤의 제조와 팽윤거동)

  • Poly(2-hydroxyethyl methacrylate), PHEMA, hybrid gels containing Pluronic and acrylic acid (AAc) were prepared as new biocompatible and stimuli-responsive hydrogels by photo-polymerization technique. The prepared hybrid gel showed reversible, temperature-responsive swelling behavior due to the presence of Pluronic component, which underwent sol-gel transition at an elevated temperature to cause gel shrinkage. The hydrogel also exhibited increased swelling degrees and pH-sensitivity due to the AAc component with ionizable carboxylic acid groups. The microporous gel morphology and its changes upon stimuli was observed by scanning electron microscopy.

  • 1.형질전환 조건 설정: 페염균을 casein hydrolysate배지에서 배양하여 550nm에서의 흡광도가 0.1이 되었을 때 pH를 7.0으로 낮추고 형질전환 능력이 나타나도록 할 때에는 pH를 7.6으로 높여 줌으로써 형질전환이 원하는 시기에 발현되도록 함. 2.돌연변이원에 대한 감수성 측정: com- mutant는 야생형보다 돌연변이가 일어난 위치에 따라 UV에 대한 감수성의 차이가 나타났으나 EMS에 대해서는 야생형과 큰 차이가 얼었음. 3.온도민감성: 5시간동안 43$^{\circ}C$에서는 37$^{\circ}C$에서의 생장율의 5%만 증가되었으나 5$0^{\circ}C$에서는 전혀 성장하지 않음.

Drug Release from Thermosensitive Liposomes by High-Intensity Focused Ultrasound (고강도 집속 초음파에 의한 온도민감성 리포솜으로부터 약물 방출)

  • Development of liposomes has been actively studied for effective delivery of drug at tumor site. However, despite their preferential accumulation at tumor site, the therapeutic efficacy of such liposomal drug has been limited because of low drug release. Therefore, we developed a temperature-sensitive liposome (TSL), which can be made to maximize release of drug by external stimulation such as ultrasound. Doxorubicin (DOX) as a model drug was encapsulated into TSL by a pH gradient method. The particle size of the TSL was $142.0{\pm}6.24nm$. Surface charge was $-10.55{\pm}1.12mV$. Release of drug from TSLs was up to 80% within 15 min at over $42^{\circ}C$ measured by fluorescence intensity. Cytotoxicity of released DOX from TSLs with ultrasound was highly increased compared to TSLs without ultrasound. Taken together, we demonstrate that temperature sensitive drug release from TSLs with ultrasound, which may be useful for cancer therapy to increase drug concentration at tumor site by external stimulation.

Synthesis and Swelling Kinetics of a Cross-Linked pH-Sensitive Ternary Copolymer Gel System (pH-민감성 삼성분계 공중합체 젤의 합성 및 팽윤 속도론)

  • A pH sensitive ternary copolymer gel was synthesized in the presence of ethylene glycol dimethacrylate (EGDMA) as a crosslinking agent through radical polymerization of vinyl acetate (VA), acrylic acid (AA) and methyl acrylate(MA) with a weight ratio of 1 : 1.3 : 1. A number of experiments were carried out to determine the swelling behavior of the gel under a variety of pH conditions of the swelling medium. As the pH of the swelling medium was changed from 1.0 to 8.0 at $37^{\circ}C$, the gel showed a shift in the pH-dependent swelling behavior from Fickian (n=0.3447) to non-Fickian (n=0.9125). The resulting swelling parameters were analyzed using graphical and statistical methods. The results showed that the swelling of the gel was controlled by the pH of the medium, i.e. $n=n_o{\exp}(S_{C}pH)$, where n is the diffusion exponent, $n_o(=28.9645{\times}10^{-2})$ is the pre-exponential factor and $S_C$(=0.1417) is pH sensitivity coefficient. The swelling behavior of the gel was also examined in aliphatic alcohols. The results showed that the rate of swelling increased with increasing number of carbon atoms in the alcoholic molecular chain.

An Association of C/T Polymorphism in the TFF1 Gene and the Susceptibility to Gastric Cancer (TFF1 유전자의 C/T 다형성과 위암 민감성과의 연관성)

  • Purpose: This study investigated whether a single nucleotide polymorphism (SNP) located at position -2 in the Kozak sequence of the TFF1 gene is associated with H. pylori infection and the development of gastric cancer in Koreans. Materials and Methods: We enrolled 167 patients with gastric cancer from January 2000 to December 2003 and also 299 healthy controls during the same period. The genotype of the TFF1 SNP was analyzed by polymerase chain reaction-restriction fragment length polymorphism and single strand conformation polymorphism. We also examined the H. pylori infection by Giemsa staining. Results: No significant difference in the allele or the TFF1 SNP genotype frequency was observed between the patients with gastric cancer and the control subjects (P=0.595 and P=0.715, respectively). When stratified by the histological subtype of gastric cancer and the age of the patients, the risk was not statistically significant between the two study groups (P=0.088 and P=0.551, respectively). H. pylori infection was detected in 39 cases and it was not associated with the TFF1 genotype. Conclusion: These findings suggest that this TFF1 gene polymorphism is not associated with H. pylori infection and gastric cancer in Koreans and so it doesn't contribute to the susceptibility to gastric cancer in Koreans.

Characterization of a pH/Temperature-Sensitive Hydrogel Synthesized at Different pH and Temperature Conditions (pH/온도-동시 민감성 Hydrogel의 합성조건에 따른 특성 연구)

  • A hydrogel, poly(N-isopropylacrylamide-co-N, N-dimethylaminopropylmethacrylamide), sensitive to both pH and temperature, was synthesized and characterized at $^13∼23{\circ}C$ and pH of 10.3∼12.3. The gel was more transparent and mechanically stronger at lower preparation temperature and pH. Large pores observed in scanning electron microscope seem to be responsible for the lower biomolecular separation efficiency. The lower critical solution temperature (LCST) decreased at a higher polymerization temperature. At $25^{\circ}C$, which is lower than the LCST, the gel was swollen regardless of the solution pH. At $40^{\circ}C$, however, the gel was swollen at neutral and acidic pHs even though the temperature was higher than the LCST. The gel collapse pH, defined as the point at which the gel made its largest volume decrease per unit pH increment, increased as the gel preparation temperature increased.

  • Curdlan, a natural $\beta-1,3-glucan,$ has been studied as drug carrier due to its unique properties including its thermal gelling characters. In this study, curdlan was chemically acetylated for pH-sensitive drug delivery. Curdlan acetate microspheres(CAMs) were prepared by the solvent evaporation method. The size of the CAMs was below $200{\mu}m.$ The drug loading efficiency of microspheres was approximately $58.44\%$. In the swelling test, the CAMs showed pH-sensitive behavior. The swelling capacity of microspheres at pH 7.4 was much greater than at pH 1.4. Also, release rate of indomethacin(IND) at pH 7.4 from the CAMs was faster than that at pH 1.4. Therefore the CAMs have potential for the controlled release of IND over an extended period of time.

  • mCherry is one of the well-understood red fluorescent proteins which has a similar tertiary structure as GFPs, but pH resistant due to the lack of hydrogen bond network. Whereas mCherry-I202T showed far-red fluorescence and also pH sensitive property because of the additional hydrogen bond formed by substituting Ile of 202 amino acid sequence on mCherry with Thr. In order to verify the pH sensitive characteristic of mCherry-I202T owing to the extension of hydrogen bond, UV-vis spectrum was measured over the range of acidic to basic pH. We also demonstrate further possibilities of applying mCherry-I202T as a pH sensor.

Bactericidal Effects of Food-borne Bacteria using Chlorine Dioxide and Electrolyzed Water (이산화염소수와 전해수를 이용한 식중독균의 살균효과)

  • The present study investigated the bactericidal effects of chlorine dioxide (CD) and electrolyzed water (EW) on pathogenic bacteria, such as Bacillus cereus, Staphylococcus aureus, Salmonella Typhimurium, and Escherichia coli O157:H7, by treatment them with CD and EW, respectively, for 0, 2, 4, 6, 8, and 10 min. Additionally, the sensitivities of Gram-positive (B. cereus and S. aureus) and Gram-negative (S. Typhimurium and E. coli O157:H7) to CD and EW were compared, respectively. In CD, the D-values for B. cereus, S. aureus, S. Typhimurium, and E. coli O157:H7 were 1.85±0.64, 2.06±0.85, 2.26±0.89, and 2.59±0.40 min, respectively. In EW, the D-values for B. cereus, S. aureus, S. Typhimurium, and E. coli O157:H7 were 2.13±0.32, 1.64±0.64, 1.71±0.32, and 1.86±0.36 min, respectively. All strains decreased consistently for 10 min in both CD and EW. However, the D-values of each bacterial species did not differ significantly between CD and EW (P>0.05). When comparing the bactericidal effect of CD and EW, no difference in D-value was observed, even though the pH and available chlorine concentration of CD were significantly lower than those of EW. These data could be used for the application of CD and EW in the food industry, considering characteristics such as the selection of optimal disinfectants, determination of optimal concentrations, and sensitivity to disinfection targets.
